Longitudinal multi-omics analysis of the gut-liver axis: Unraveling the molecular mechanisms of metabolic homeostasis regulation by Pd@Pt nanozymes

Yanan Wang,Nan Cheng,Qi Zhang,Fei Chang,Teng Wang,Minrui Kan,Yutong Han,Baiqiang Zhai,Kunlun Huang,Xiaoyun He

Recently, the nanozyme Pd@Pt has garnered attention due to its notable specific surface area and superior enzyme-like catalytic activity, leading to extensive examination and application in previous studies. However, the comprehensive impact of Pd@Pt nanozyme on treating metabolic disorders, such as diabetes and its associated conditions, remains largely unexplored. This research aimed to clarify how Pd@Pt influences metabolic balance at both the transcriptome and microbiome levels and to explore the interactions between microbiota and genes. We conducted an examination of mice subjected to a high-fat diet (HFD) following treatment with Pd@Pt. Transcriptome analysis was performed to identify differentially expressed genes (DEGs), and microbiome analysis was conducted to identify significant bacterial correlations associated with Pd@Pt exposure. The results indicated enhancements in glucose metabolism dysfunctions in the treated mice. Transcriptome analysis revealed that DEGs after Pd@Pt administration were enriched in the PI3K-Akt, NF-κB, and MAPK signaling pathways in the liver. Microbiome analysis identified four significant bacteria that exhibited a strong negative correlation with Pd@Pt exposure, while ten bacteria showed a positive correlation. Furthermore, a correlation network established among the gut microbiota, metabolites, and DEGs demonstrated a robust association. This research enhances our understanding of the mechanisms by which Pd@Pt affects the regulation of metabolic diseases in HFD-exposed environments and proposes a novel strategy for utilizing nanozymes in human health management.

Coordination-Driven One-Step Rapid Self-Assembly Synthesis of Dual-Functional Ag@Pt Nanozyme

Zaihui Du,Longjiao Zhu,Pengfei Wang,Xinyue Lan,Shenghao Lin,Wentao Xu

Realizing high-precise and adjustable regulation of engineering nanozyme is important in nanotechnology. Here, Ag@Pt nanozymes with excellent peroxidase-like and antibacterial effects are designed and synthesized by nucleic acid and metal ions coordination-driven one-step rapid self-assembly. The adjustable NA-Ag@Pt nanozyme is synthesized within 4 min using single-stranded nucleic acid as templates, and peroxidase-like enhancing FNA-Ag@Pt nanozyme is received by regulating functional nucleic acids (FNA) based on NA-Ag@Pt nanozyme. Both Ag@Pt nanozymes that are developed not only has simple and general synthesis approaches, but also can produce artificial precise adjustment and possess dual-functional. Moreover, when lead ion-specific aptamers as FNA are introduced to NA-Ag@Pt nanozyme, the Pb 2+ aptasensor is successfully constructed by increasing electron conversion efficiency and improving the specificity of nanozyme. In addition, both nanozyme has good antibacterial properties, with ~100% and ~85% antibacterial efficiency against Escherichia coli and Staphylococcus aureus , respectively. This work provides a synthesis method of novelty dual-functional Ag@Pt nanozymes and successful application in metal ions detection and antibacterial agents.

Boosting the peroxidase-like activity of Pt nanozymes by a synergistic effect of Ti3C2 nanosheets for dual mechanism detection

Hongqi Geng,Zhijuan Li,Quan Liu,Qi Yang,Huimin Jia,Qiang Chen,Aiguo Zhou,Weiwei He

Nanozymes show great promise in bioanalysis and therapeutics, which largely rely on high catalytic efficiency and selectivity. Inspired by the interfacial electronic interaction between noble metals and transition metal carbides, Pt nanozymes are modified with Ti3C2 MXene nanosheets to construct Ti3C2/Pt hybrids with synergistically enhanced catalytic activity. Although Ti3C2 does not have oxidase and peroxidase-like activity, it can greatly selectively enhance the peroxidase-like activity of Pt nanozymes. Near-infrared irradiation can further increase specifically the peroxidase-like activity of Ti3C2/Pt. The optimal peroxidase-like activity of Ti3C2/Pt is 6 times higher than Pt in the dark and 7.9 times higher than Pt under illumination. This catalytic enhancement is attributed to the interplay of the strong interfacial electron effect and unique photothermal effect of Ti3C2. Using the superior peroxidase-like activity of Ti3C2/Pt, dual mechanism colorimetric methods based on cascade reaction and inhibitory effect are developed for specific detection of glucose and glutathione with a limit of detection of 1.0 μM and 0.0089 μM, respectively. Our work provides an effective means to improve the catalytic activity and selectivity of nanozymes by introduction of an ideal supporter, which will be of value for the design of efficient nanozymes.

Ti3C2/CuWO4/Pt nanozyme: photothermal-enhanced chemodynamic antibacterial effects induced by NIR

Simin Yuan,Lianyuan Ge,Yi Li,Xiaohong Wang,Zhongyuan Liu,Yang Cao,Linglin Yang

With the growing issue of antibiotic resistance, it has become increasingly crucial to develop highly efficient antimicrobial materials. While the single-component nanozyme systems exhibited some catalytic activity, their efficiency remains suboptimal. This study presents a Ti3C2/CuWO4/Pt hybrid nanozyme composed of photothermal agents and nanozymes, which leverages the photothermal effect to enhance nanozyme activity and achieve efficient antimicrobial effects. The composite material exhibited peroxidase (POD)-like catalytic activity, effectively converting hydrogen peroxide (H2O2) into hydroxyl radicals (·OH). Meanwhile, the Ti3C2/CuWO4/Pt material demonstrated high photothermal conversion ability, which not only promoted the generation of ·OH under near-infrared (NIR) light irradiation, but also facilitated copper (Cu2+) ions release from the CuWO4 nanozyme, thereby further augmenting its catalytic activity. After 4 to 5 min of light irradiation, the Ti3C2/CuWO4/Pt nanozyme exhibited significant antimicrobial performance against both Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). In summary, this work presents a Ti3C2/CuWO4/Pt nanoplatform that utilizes the photothermal effect to enhance the chemodynamic antimicrobial activity, showcasing its potential applications in antibiotic-free antimicrobial fields.

A novel biomimetic immunoassay method based on Pt nanozyme and molecularly imprinted polymer for the detection of histamine in foods

Xiaofeng Wang,Xiaoqing Song,Lin Si,Longhua Xu,Zhixiang Xu

This study used surface imprinting polymerization to synthesize molecularly imprinted polymer on the surface of PAF-45 material (PAF-45@MIP). SEM, FT-IR, static and kinetic adsorption experiments were adopted to investigate the morphology and properties of the MIP. The results showed that the PAF-45@MIP could rapidly absorb histamine with a high adsorption capacity. Using PAF-45@MIP as a biomimetic antibody and Pt nanozyme as a marker, a novel direct competitive biomimetic immunoassay (BIA) method for rapidly detecting histamine in foods was developed. Under optimal conditions, the limit of detection and sensitivity of the BIA method were 0.128 and 8.306 mg/L, respectively. Vinegar spiked with histamine was determined using the BIA method with recoveries ranging from 81.50% to 90.38%. The BIA method was also applied to determine histamine in dried shrimp and soy sauce samples, the results correlating well with those obtained using high-performance liquid chromatography.

Sensitive SERS assay for glyphosate based on the prevention of L-cysteine inhibition of a Au–Pt nanozyme

Jun Ma,Gege Feng,Ying Ying,Yong Shao,Yongxin She,Lufei Zheng,A. M. Abd EI-Aty,Jing Wang

Herein, an indirect SERS sensing assay was developed for the determination of glyphosate (Gly) in tap water. The mechanism of detection was based on relieving the inhibitory effect of L-cysteine (L-cys) on a Au–Pt nanozyme by combining Gly with L-cys through divalent copper ions (Cu2+). In this method, a novel nano-chain-like Au–Ag composite with good repeatability was successfully fabricated to detect SERS signals of oxTMB without disturbing TMB Raman signals. Under optimal conditions, the SERS signal intensity of oxTMB (at 1605 cm−1) was proportional to the concentration of Gly. The results showed a good linear response over the concentration ranges of 10 μg L−1 to 1000 mg L−1. The limit of detection and limit of quantitation of Gly were found to be 5 μg L−1 and 10 μg L−1, respectively. In addition, good anti-interference ability against interfering cations and structural analogues deserves to be mentioned. This SERS assay can be used for detection of Gly in tap water that can meet the needs of practical detection.

Development of magnetic fluorescence aptasensor for sensitive detection of saxitoxin based on Fe3O4@Au-Pt nanozymes

Lin Zhu,Wei Zeng,Yueqing Li,Yu Han,Jing Wei,Long Wu

In this work, on the basis of Fe 3 O 4 @Au-Pt nanozymes (MAP NZs) and aptamer recognition, a magnetic fluorescent aptasensor (MFA) was developed for sensitive and accurate detection of saxitoxin (STX). With the bridge of STX aptamer (Apt STX ) and complementary DNA (cDNA), Apt STX decorated MAP NZs (MAP/Apt) and cDNA modified green quantum dots (cDNA@g-QDs) were connected to form MAP/Apt-cDNA@g-QDs complex. As STX behaves a strong binding ability towards Apt STX , it will compete with cDNA and hybridize with Apt to release cDNA@g-QDs. With the addition of TMB, MAP will catalyze TMB to the oxidized TMB (ox-TMB), thereby quenching the fluorescence of g-QDs due to the inner filter effect. Based on this finding, the quantitative relationship between the change in fluorescence of gQDs and STX concentration was explored with a limit of detection (LOD, S/N = 3) of 0.6 nM. An internal standard signal of oxTMB was adopted and reduced the fluctuation of fluorescence signal output. Besides, the fluorescence probe can selectively recognize and detect STX among five marine toxins. Eventually, the MFA method behaved good performance in detecting seafood samples with recoveries of 82.0 % ∼ 102.6 % as well as coefficient of variations (CV) of 7.2 % ∼ 10.3 %. Therefore, the method with internal signal is hopeful to be a potential candidate for sensitive and accurate detection of STX in seafood.

Catalytic Performance of Oligonucleotide-Templated Pt Nanozyme Evaluated by Laccase Substrates

Wang Yu,He Chuan,Li Wei,Zhang Jinli,Fu Yan

Abstract Laccases are effective native biocatalysts for the oxidation of o -/ p -diphenols, aminophenols, polyphenols and polyamines. To overcome the intrinsic limitations of native laccase involving pH, temperature and storage, highly dispersive Pt nanoparticles were synthesized by employing different oligonucleotides as stabilizing agents including A 10 , T 10 , C 10 and G 10 , and their catalytic activities were assessed in the oxidation of laccase substrates under ambient air. This kind of Pt nanozyme displays high stability in the range of 20–90 °C and pH 3–9, beyond the range for native laccase. The laccase-like activities of Pt nanoparticles are greatly associated with the size distribution and the surface charge, which can be easily regulated by the sequence composition and the molar ratio of [precursor]/[template]. The most efficient C 10 -templated Pt nanozyme with 4.6 nm in size, exhibit three times higher affinity toward 2,4-dichlorophenol than that of native laccase. DNA-stabilized Pt nanoparticles display excellent catalytic performance in the oxidation of a wide range of substrates including dopamine, catechol, hydroquinone and p -phenylenediamine. This study paves a way to explore artificial nanoparticles to replace native laccase in the fields of biosensing and green synthesis. Graphical Abstract

Acidity-responsive cascade nanoreactor based on metal-nanozyme and glucose oxidase combination for starving and photothermal-enhanced chemodynamic antibacterial therapy

Tongyao Wang,Dong Dong,Tong Chen,Junlun Zhu,Suxiao Wang,Wei Wen,Xiuhua Zhang,Huadong Tang,Jichao Liang,Shengfu Wang,Huayu Xiong

Persistent inflammation and bacterial infection are common during wound healing, and high blood sugar provides enough nutrients for bacterial growth to make wounds more prone to ulcers. Herein, glucose oxidase (GOx) is mineralized with calcium phosphate (CaP) circled Cu 2 O/Pt nanozyme to form a core–shell structural CaP-GOx-Cu 2 O/Pt (GOx-CaPCuPt) nanoreactor. The prepared biodegradable and acidity-responsive GOx-CaPCuPt nanoreactor can be used for near infrared imaging, photothermal enhanced cascade reaction and antibacterial therapy. Oxidation of glucose (Gs) by the GOx can significantly decrease Gs level for starvation therapy, and the generated H 2 O 2 in this reaction is subsequently converted into highly active hydroxyl radicals (•OH) for chemodynamic therapy (CDT) and O 2 for overcoming hypoxia via triple enzyme like activities (peroxidase, oxidase, and catalase) of Cu 2 O/Pt nanozyme. At the same time, the acidic microenvironment due to the generation of gluconic acid will in turn promote the degradation of the GOx-CuCaP nanoreactor and release of copper ions for enhanced angiogenesis. In addition, Cu 2 O/Pt nanoparticles have strong characteristic absorption in near-infrared region, which can induce a strong photothermal effect (PTT) under the irradiation of 808 nm laser. Under the guidance of near-infrared imaging, the multifunctional nanoreactor can achieve the combination of starvation therapy and PTT-enhanced CDT against Gram -negative and Gram -positive bacteria infectious diseases, which is demonstrated in the staphylococcus aureus infected diabetic rat models.

Multi-omics analysis of Au@Pt nanozyme for the modulation of glucose and lipid metabolism

Wang Yanan,Zhang Qi,Kan Minrui,Chang Fei,He Xiaoyun,Cheng Nan,Huang Kunlun

Au@Pt nanozyme, a bimetallic core–shell structure Au and Pt nanoparticle, has attracted significant attention due to its excellent catalytic activity and stability. Here, we propose that Au@Pt improves glucose tolerance and reduces TG after four weeks administration. The transcriptomic analysis of mouse liver tissues treated with Au@Pt nanozyme showed changes in genes related to glucose and lipid metabolism signaling pathways, including glycolysis/gluconeogenesis, pyruvate metabolism, PPAR signaling, and insulin signaling. Moreover, analysis of fecal samples from mice treated with Au@Pt nanozyme showed significant changes in the abundance of beneficial gut microbiota such as Dubosiella , Parvibacter , Enterorhabdus , Monoglobus , Lachnospiraceae_UCG-008 , Lachnospiraceae_UCG-006 , Lachnospiraceae_UCG-001 , and Christensenellaceae_R-7_group . Combined multi-omics correlation analyses revealed that the modulation of glucose and lipid metabolism by Au@Pt was strongly correlated with changes in hepatic gene expression profiles as well as changes in gut microbial profiles. Overall, our integrated multi-omics analysis demonstrated that Au@Pt nanozyme could modulate glucose and lipid metabolism by regulating the expression of key genes in the liver and altering the composition of gut microbiota, providing new insights into the potential applications of Au@Pt nanozyme in the treatment of metabolic disorder. Trimetallic Au@Pd@Pt nanozyme-enhanced lateral flow immunoassay for the detection of SARS-CoV-2 nucleocapsid protein

Yue Sun,Zihao Xie,Fubin Pei,Wei Hu,Shasha Feng,Qingli Hao,Bing Liu,Xihui Mu,Wu Lei,Zhaoyang Tong

The rapid spread of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) seriously threatened global public health. Establishing a rapid and sensitive diagnostic test for early detection of the SARS-CoV-2 nucleocapsid protein is urgently required to defend against the pandemic. Herein, an enhanced lateral flow immunoassay (LFIA) was fabricated by trimetallic Au@Pd@Pt core–shell nanozymes for detection of the SARS-CoV-2 nucleocapsid protein. The Au@Pd@Pt nanozymes (Au@Pd@Pt NZs) synthesized via a one-pot method, with a dendrite morphology and uniform particle size, showed excellent peroxidase-like activity. Due to the perfect enzyme-like catalytic activity toward 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H2O2), the catalytic signal could be generated even by a tiny amount of Au@Pd@Pt NZs accumulated on the test strip. Therefore, rapid detection with higher sensitivity was achieved. The Au@Pd@Pt NZs-based LFIA provided a quantitative range of 0.05–100 ng mL−1 with a limit of detection of 0.037 ng mL−1, which was 17-fold lower than the LFIA without enhancement. The average recoveries from spiked samples were in the range of 92.5–107.9% with relative standard deviations all less than 4%, indicating the reliability and repeatability of the proposed LFIA. Additionally, the proposed LFIA could report results within 30 min using a microplate reader. In conclusion, the Au@Pd@Pt NZs-LFIA is a rapid, simple, and sensitive method for detecting the SARS-CoV-2 nucleocapsid protein.
