Asymmetric adhesive SIS-based wound dressings for therapeutically targeting wound repair

Yao Wende,Song Zelong,Ma Xiaodong,Huang Yiqian,Zhang Xueying,Li Yunhuan,Wei Pengfei,Zhang Julei,Xiong Chenlu,Yang Sihan,Xu Yujian,Jing Wei,Zhao Bo,Zhang Xuesong,Han Yan

Severe tissue injuries pose a significant risk to human health. Conventional wound dressings fall short in achieving effective tissue regeneration, resulting in suboptimal postoperative healing outcomes. In this study, an asymmetric adhesive wound dressing (marked as SIS/PAA/LAP) was developed, originating from acrylate acid (AA) solution with laponite (LAP) nanoparticles polymerization and photo-crosslinked on the decellularized extracellular matrix small intestinal submucosa (SIS) patch. Extensive studies demonstrated that the SIS/PAA/LAP exhibited higher tissue adhesion strength (~ 33 kPa) and burst strength (~ 22 kPa) compared to conventional wound dressings like Tegaderm and tissue adhesive products. Importantly, it maintained favorable cell viability and demonstrated robust angiogenic capacity. In animal models of full-thickness skin injuries in rats and skin injuries in Bama miniature pigs, the SIS/PAA/LAP could be precisely applied to wound sites. By accelerating the formation of tissue vascularization, it displayed superior tissue repair outcomes. This asymmetrically adhesive SIS-based patch would hold promising applications in the field of wound dressings.

Modification of bitumen emulsion via heterocoagulation with SIS triblock copolymer latex

Chunmiao Xia,Yingwu Luo

Bitumen emulsions have often been modified by styrene–butadiene rubber latex. The modified bitumen can have excellent low-temperature cracking resistance, but rutting resistance at high temperature still remains poor. In the current work, for the first time, a stable poly(styrene- b -isoprene- b -styrene) (SIS) triblock copolymer latex is synthesized by reversible addition–fragmentation transfer (RAFT) emulsion polymerization. Based on this, a simple heterocoagulation process is developed to prepare the bitumen emulsions modified by SIS. The heterocoagulation results in hybrid particles of SIS shell and bitumen core. With addition of 5 wt % SIS, a continuous polymer-rich phase could be formed in the modified bitumen once the modified emulsion was broken down. The bitumen modified by 5 wt % SIS shows a significant increase in complex modulus at high temperature and a significant decrease in loss tangent, suggesting excellent resistance to rutting at high temperature, which is consistent with the significant increase in softening point from 41 °C for the base bitumen up to 64 °C. Meanwhile, the ductility at 5 °C of the modified bitumen is also dramatically increased from 1.4 cm for the base bitumen to 40 cm, indicating the low-temperature cracking resistance should also be much enhanced. © 2017 Wiley Periodicals, Inc. Application of CH4/N2 separation based on poly(styrene-b-isoprene-b-styrene) (SIS)-poly(dimethylsiloxane-co-methylhydrosiloxane) (PDMS-co-PMHS) crosslinked membrane

Xing Yang,Yayun Zheng,Lan Wang,Qingping Guo,Haiquan Shan,Zongxiang Xu,Jujie Luo

This work involves the preparation and characterization of poly(styrene-b-isoprene-b-styrene)-poly(dimethylsiloxane- co -methylhydrosiloxane) (SIS-c-PDMS-co-PMHS) crosslinked membranes towards CH 4 /N 2 separation. SIS-c-PDMS-co-PMHS was synthesized via a simple hydrosilylation reaction with various PDMS-co-PMHS contents. The structure of crosslinked membranes was investigated by Fourier transform infrared spectroscopy , indicating the degree of hydrosilylation reaction. The morphology of sample membranes was observed in detail by means of Scanning Electron Microscope analysis . Thermodynamic stability and the crosslinked behavior of crosslinked membranes were explored by thermogravimetric analysis and Soxhlet extraction experiments. Furthermore, single gas (CH 4 , N 2 ) permeability was measured using the time-lag method. The CH 4 permeability of crosslinked membranes with 70 wt% PDMS-co-PMHS could reach 503 Barrer at 25 °C and 1 bar, which is significantly greater than that of SIS (25.9 Barrer), while the CH 4 /N 2 ideal selectivity hardly changed. Notably, CH 4 permeability further improved by 77.6% (893.4 Barrer) as the temperature increased to 55 °C. For the CH 4 /N 2 (50/50 vol%) mixed gas, the SIS-c-PDMS-co-PMHS-70 sample membrane exhibited ideal gas separation performance relative to that of the pure SIS membrane (CH 4 : 447.4 vs. 24.2 Barrer,N 2 : 188.1 vs. 9.46 Barrer).

Biomimetic SIS-based biocomposites with improved biodegradability, antibacterial activity and angiogenesis for abdominal wall repair

Guangxiu Cao,Cunyang Wang,Yubo Fan,Xiaoming Li

Small intestinal submucosa (SIS) is a widely concerned acellular material for reconstructing tissue defects, but during the restoration of abdominal wall, it has been restricted due to the fast degradation causing poor long-term mechanical properties, the infection caused by bacteria contamination, and insufficient neovascularization post-operation. In this study, we developed a biomimetic SIS-based biocomposite (CS/ES-SIS) for abdominal wall repair, in which chitosan (CS) and elastin (ES) electrospun nanofibers were used to improve the biodegradability, antibacterial activity, and angiogenesis. The CS/ES-SIS composites were examined through a series of testing experiments, especially in vitro degradation was assessed by a constant deformation loading device and the micromechanical properties during enzymatic degradation under biomechanical environment were measured by nanoindentation. In vitro antibacterial test and cytocompatibility, and in vivo biocompatibility, neovascularisation and tissue regeneration were also investigated. The main research results as follows: (1) After 7 days enzymatic degradation under biomechanical environment, the degradation rate of CS/ES-SIS composites was slower than that of SIS by about 24.5%. Moreover, the CS/ES-SIS composites could better maintain the stability of microstructure and micromechanical properties compared with SIS. (2) The antibacterial rates of CS/ES-SIS composites against E. coli and S. aureus were respectively 98.87% and 98.26% while the SIS demonstrated no obvious antibacterial capacity. (3) The CS/ES-SIS composites supported the viability and proliferation of fibroblast cell L929. In vivo studies showed that the CS/ES-SIS composites could promote tissue regeneration upon implantation without serious inflammatory reaction. Additionally, the vascular number in the CS/ES-SIS composites was as 1.69 times as that in the SIS at 4 weeks. Collectively, all the findings suggested that the newly developed CS/ES-SIS composites might be promising and attractive candidates for applications of abdominal wall repair.

Decorated small intestinal submucosa (SIS) based bio-patch with anti-fouling and vascularized capacity to accelerate chronic wound regeneration via TGF-β and MAPK pathway

Yi Wang,Linhao Hou,Yiqian Huang,Pengfei Wei,Liya Sun,Yan Zhang,Xueqiao Yu,Shiqing Ma,Wei Jing,Bo Zhao,Huachong Ma

Chronic wounds in diabetes take a lifetime risk of developing into diabetic foot ulcers (DFUs). Conventional wound dressings and commercial products fail for ideal tissue regeneration in DFUs due to the incapacity to promote vascularization and prevent bacterial infections during wound repair. In this study, the small intestinal submucosa (SIS) with enriched vascularized bioactive factors, was chemically decorated by 2-methacryloyloxy ethyl phosphorylcholine (MPC) through Michael-type addition to be endowed anti-fouling ability, in order to treat chronic wound regeneration in diabetes. In the diabetic skin injury experiment, we established the animal model by 5-mm diameter defects in the SD rats and implanted the bio-patches to the located sites. The SIS-MPC bio-patch showed superior advantages in bacterial anti-adhesion (100-fold foul resistance than commercial patches), while maintained affinity for fibroblast growth and collagen formation, as demonstrated by immunohistochemical staining from the local tissues, outperforming commercial wound dressings in tissue regeneration, which was attributed to acceleration in neovascularization and inflammation modulation through TGF-β and MAPK pathway. This novel SIS-MPC bio-patch would be a promising candidate for medical translations into diabetes and chronic wound treatments.

Molecular structure design and modular synthesis of SIS building blocks with reversible structures

Zheng Peng,Yang Fan,Zhao Zhongfu,Zhang Chunqing

In this work, furan-terminated polystyrene (FPS) and polyisoprene (FPI) building blocks with molecular weights ranging from 3000 to 5000 were synthesized via living anionic polymerization. A series of styrene-isoprene-styrene (SIS) block copolymers with thermoreversible structures (DASIS) were then prepared through the Diels-Alder (DA) reaction between the furan groups of FPS/FPI and N,N'-(1,4-phenylene)bismaleimide (BMI). The microstructure, dynamic bond formation and thermoreversibility of the as-synthesized polymers were characterized by 1H-NMR, FT-IR, DSC and other analytical techniques. The results demonstrated that DASIS block copolymers with molecular weights of 80000–130000 were successfully fabricated. Benefiting from the unique properties of dynamic DA bonds, the precise connection of building blocks to form macromolecules was achieved at a mild temperature of 60 °C, while the efficient dissociation and recovery of building blocks were realized at an elevated temperature of 150 °C, thus enabling the flexible regulation of the monomer ratio in DASIS. With the increase in the mass fraction of the PS phase in DASIS from the initial 20% to 50%, the elongation at break decreased significantly, whereas the Young’s modulus and tensile strength increased by 182.5% and 157.7%, respectively, which conforms to the basic law of the effect of the ratio of rigid to soft phases on the macroscopic properties in multiphase polymer systems. Furthermore, the Young’s modulus and tensile strength of the recycled DASIS remained above 90% of those of the pristine DASIS. This strategy provides a new insight into the precise synthesis of recyclable polymeric materials and exhibits great potential in the fields of polymer modification and elastomer applications.

Killing two birds with one stone: A therapeutic copper-loaded bio-patch promoted abdominal wall repair via VEGF pathway

Nan Zhang,Yiqian Huang,Pengfei Wei,Liya Sun,Wei Jing,Yunxia Xue,Yan Zhang,Bo Zhao,Ziang Yang

Hernia and life-threatening intestinal obstruction often result from abdominal wall injuries, and the regeneration of abdominal wall defects is limited due to the lack of biocompatible, antibacterial and angiogenic scaffolding materials for treating injured tissues. Taking inspiration from the facile preparation of dopamine polymerization and its surface modification technology, in this study, multi-therapeutic copper element was introduced into porcine small intestinal submucosa (SIS) bio-patches through polydopamine (PDA) deposition, in order to regenerate abdominal wall injury. In both in vitro antibacterial assays, cytocompatibility assays and in vivo abdominal wall repair experiments, the SIS/PDA/Cu bio-patches exhibited robust antibacterial efficiency (>99%), excellent biocompatibility to cells (>90%), and enhanced neovascularization and improved collagen maturity compared to other commercially available patches (3.0-fold higher than the PP mesh), due to their activation of VEGF pathway. These findings indicated the bio-patch was a promising application for preventing visceral adhesion, bacterial infection, and promoting soft tissue regeneration.

An adhesive gelatin-coated small intestinal submucosa composite hydrogel dressing aids wound healing

Lu Wang,Fengling Liu,Xinrang Zhai,Wei Dong,Wei Wei,Zhenhua Hu

It is a challenging clinical task to determine how to repair large-area skin defects better. Traditional wound dressings (e.g., cotton and gauze) can only be used as a dressing; consequently, there is an increasing demand for wound dressings with additional properties (i.e., antibacterial and pro-repair) in clinical practice. In this study, a composite hydrogel with o-nitrobenzene-modified gelatin-coated decellularized small intestinal submucosa ( [email protected] ) was designed for the repair of skin injuries. SIS is a natural extracellular matrix with a 3D microporous structure and also contains high levels of growth factors and collagen. GelNB provides this material photo-triggering tissue adhesive property. The structure, tissue adhesion, cytotoxicity, and bioactivity to cells were investigated. Based on in vivo study and histological analysis, we found the combination of GelNB and SIS improved the healing process by promoting vascular renewal, dermal remodeling, and epidermal regeneration. Based on our findings, [email protected] is a promising candidate for tissue repair applications.

Industrial-sized Warpage-Free Inert Ceramic Supported FT-SIS-SOFCs: Electrolyte Densification Strategy Based on Controlled YSZ Shrinkage

Yan-An Li,Jia-Hong Li,Xiang-Xiang Pan,Xiang-Qi Liu,Wan-Ming Li,Chang-Qing Liu,Cheng-Xin Li

Inert ceramic-supported solid oxide fuel cells (SOFCs) are critical for advancing long-life, high-stability SOFC energy systems. Using an MgO-MgAl 2 O 4 (MMA) substrate, we developed the high-performance industrial-sized MMA-supported flat-tubular segmented-in-series SOFCs (FT-SIS-SOFCs) through a co-sintering process. Dense electrolytes were obtained by implementing bimodal particle engineering to regulate sintering shrinkage behavior. Through precise control of the nanoparticle (∼100 nm) to submicron particle (∼300 nm) mass ratio at 7 : 3, the sintering shrinkage mismatch between the YSZ electrolyte and MMA substrate was reduced from 6.2% to 0.5%, and the warpage deformation decreased from a compressive strain of -0.113 to less than 0.05. The optimized cell demonstrated a maximum power density of 342 mW/cm 2 with an open-circuit voltage of 1.006 V at 850 °C, coupled with exceptional long-term stability. The industrial-sized warpage-free 16-cell FT-SIS-SOFCs exhibited a high open-circuit voltage of 17.50 V and maximum power output of 6.55 W at 850 °C.

Antibacterial and pro-angiogenic SIS-based bio-patches with poly(tannic acid) and magnesium facilitating abdominal wall repair

Lu Peng,Yiqian Huang,Junyan Yang,Yunhuan Li,Xueying Zhang,Pengfei Wei,Wei Jing,Xueqiao Yu,Bo Zhao,Rui Xu

Abdominal wall injury is a common and challenging clinical condition, typically caused by trauma, surgical intervention, or congenital abnormalities. Traditional treatments face significant limitations in preventing infections and promoting tissue integration, highlighting the urgent need for innovative repair materials. In this study, we reported a composite bio-patch based on the decellularized extracellular matrix of small intestinal submucosa (SIS), surface-coated with poly(tannic acid) (PTA) and magnesium ions (Mg 2+ ), which imparted the bio-patch with strong antibacterial properties, enhanced angiogenesis, and tissue regeneration (named as SIS/PTA/Mg). Through in vitro cell assays, antibacterial assays, and rat abdominal wall defect animal model, the SIS/PTA/Mg bio-patch exhibited excellent bacterial resistance (∼98% antibacterial efficiency), favorable biocompatibility, and remarkable repair efficacy (3.83-fold to the commercial mesh). Meanwhile, the SIS/PTA/Mg bio-patch effectively prevented post-operative infections, significantly facilitated angiogenesis, and accelerated tissue regeneration, indicating this SIS/PTA/Mg composite bio-patch as an innovative and clinically promising solution for the treatment of abdominal wall injuries.

New Insights into Baicalein’s Effect on Chlorpyrifos-Induced Liver Injury in Carp: Involving Macrophage Polarization and Pyropto sis

Jing Liu,Wenyue Zhang,Xiaojing Li,Shiwen Xu

Chlorpyrifos (CPF) is widely used in agriculture, plants, and buildings to kill pests and worms. Excessive environmental residues of CPF will result in soil and ecological contamination and toxicity to animals and humans. Baicalein (Bai), derived from the root of natural Scutellaria baicalensis, is a potent anti-inflammatory, antioxidant, and antitumor agent. The objective of this paper is to investigate the molecular mechanism by which Bai prevents CPF-induced hepatotoxic injury. Carp were kept in water containing CPF (23.2 μg/L) and/or fed diets containing Bai (0.15 g/kg). We found that Bai attenuated liver tissue damage and vacuolization caused by CPF. We confirmed that CPF causes M1/M2 polarization imbalance in macrophages and hepatocyte pyroptosis, which ultimately leads to liver injury. Further exploration of the internal mechanism shows that CPF participates in liver toxicity damage by destroying the AMPK/SIRT1/pGC-1α pathway and causing mitochondrial biogenesis and mitochondrial dynamics imbalance. Notably, Bai significantly attenuated CPF-induced inhibition of the AMPK/SIRT1/pGC-1α pathway. In summary, our results suggest that Bai alleviates CPF exposure-induced inhibition of the AMPK/SIRT1/pGC-1α pathway, thereby attenuating macrophage M1 hyperpolarization and pyroptosis by inhibiting the NF-κB pathway. These results may provide new insights into the detoxification mechanism of Bai on the same type of organophosphorus pesticides.

A Novel UBM/SIS Composite Biological Scaffold for 2-Year Abdominal Defect Repairing and Strength Recovery in Canine Model

Weidong Zhong,Jinshui Chen,Qifeng Xie,Wenyue Cheng,Meibiao Zhao,Yang Sun,Jing Dai,Jian Zhang

Biological scaffolds are widely utilized in hernia treatment due to their exceptional pro-regenerative properties, which mitigate scar formation. However, serious complications occurred, caused by inflammatory response, premature degradation, and mechanical failure. Consequently, improvements of the biological scaffold are necessary to mitigate these risks. In this study, a novel biological scaffold integrating basement membrane-containing urinary bladder matrix (UBM) and small intestinal submucosa (SIS) is developed, and its safety and effectiveness are assessed in comparison to a commercial SIS (c-SIS) scaffold. The introduction of UBM as top surface layers significantly promotes cell adhesion, facilitating rapid formation of isolated regeneration zone. Proteomic analysis has demonstrated a more efficient decellularization of the UBM/SIS scaffold, which subsequently mitigates inflammation in murine models, and promotes the polarization of macrophages toward the pro-healing M2 phenotype in a rat model of abdominal wall muscle defect. Furthermore, a two-year repair trial is conducted on a full-thickness abdominal wall muscle defect in canine model and confirmed that the UBM/SIS scaffold exhibits reduced seroma occurrences and enhanced tissue repair performances. Overall, the efficacy of this novel biological scaffold suggests its potential to minimize hernia recurrence in clinical practice and mitigate patient suffering from severe inflammatory responses.
