A novel α-glucosidase inhibitor polysaccharide from Sargassum fusiforme

Mengqing Zhang,Ruijin Yang,Shuhuai Yu,Wei Zhao

Summary α-Glucosidase inhibitors, which can inhibit α-glucosidase to reduce the release of glucose after a mixed carbohydrate diet, has become a key tool in the management of type 2 diabetes mellitus (T2DM). SFP-1, an α-glucosidase inhibitor derived from Sargassum fusiforme ( S . fusiforme ), was studied as a food-derived hypoglycaemic agent. By binding α-glucosidase and then changing the conformation of the enzyme, SFP-1 with a mixed inhibition type exerted an excellent inhibitory effect (IC 50 value: 0.681 mg mL −1 ). Structural analyses suggested that the average molecular weight of SFP-1 was 1.61 × 10 5 Da, mainly composed of glucuronic acid, fucose, galactose, glucose, mannose and xylose, and the relative molar ratio was 23.07: 15.17: 7.72: 4.92: 4.04: 1.99. FTIR and NMR analysis suggested that SFP-1 was an acidic polysaccharide with both α-type and β-type glycosidic bonds. S . fusiforme polysaccharide (SFP-1) has the potential to develop as functional foods and nutraceuticals in T2DM management.

Comparative Study of Sargassum fusiforme Polysaccharides in Regulating Cecal and Fecal Microbiota of High-Fat Diet-Fed Mice

Bin Wei,Qiao-Li Xu,Bo Zhang,Tao-Shun Zhou,Song-Ze Ke,Si-Jia Wang,Bin Wu,Xue-Wei Xu,Hong Wang

Seaweed polysaccharides represent a kind of novel gut microbiota regulator. The advantages and disadvantages of using cecal and fecal microbiota to represent gut microbiota have been discussed, but the regulatory effects of seaweed polysaccharides on cecal and fecal microbiota, which would benefit the study of seaweed polysaccharide-based gut microbiota regulator, have not been compared. Here, the effects of twoSargassum fusiformepolysaccharides prepared by water extraction (SfW) and acid extraction (SfA) on the cecal and fecal microbiota of high-fat diet (HFD) fed mice were investigated by 16S rRNA gene sequencing. The results indicated that 16 weeks of HFD dramatically impaired the homeostasis of both the cecal and fecal microbiota, including the dominant phyla Bacteroidetes and Actinobacteria, and generaCoriobacteriaceae,S24-7, andRuminococcus, but did not affect the relative abundance of Firmicutes,Clostridiales,Oscillospira, andRuminococcaceaein cecal microbiota and the Simpson’s index of fecal microbiota. Co-treatments with SfW and SfA exacerbated body weight gain and partially reversed HFD-induced alterations ofClostridialesandRuminococcaceae. Moreover, the administration of SfW and SfA also altered the abundance of genes encoding monosaccharide-transporting ATPase, α-galactosidase, β-fructofuranosidase, and β-glucosidase with the latter showing more significant potency. Our findings revealed the difference of cecal and fecal microbiota in HFD-fed mice and demonstrated that SfW and SfA could more significantly regulate the cecal microbiota and lay important foundations for the study of seaweed polysaccharide-based gut microbiota regulators.

Identification and theoretical explanation of chemical composition against α-amylase in the n-hexane extract from Sargassum fusiforme

Jiao Yang,Chang Liu,Hengjiang Cai,Dongyu Gu,Zhenni Ji,Xinfeng Guo,Jing Tian,Jing Meng,Yi Yang

Sargassum fusiforme is an edible alga that possesses a number of bioactivities , such as antidiabetic effects. In the present study, the n -hexane extract of S. fusiforme showed inhibitory activity against α-amylase. To interpret the α-amylase inhibitory activity of this extract, a method based on GC–MS and molecular docking was established. A total of 25 compounds were identified using GC–MS and then individually docked with α-amylase to simulate interactions between the latter and the ligands. Among the compounds determined, phenol, 2,2′-methylenebis[6-(1,1-dimethylethyl)-4-methyl- (PMDM) was completely enfolded in the active site of α-amylase via hydrophilic and electrostatic interactions, van der Waals forces, hydrogen bonds and π-π stacking interactions and showed the lowest binding energy (−6.25 kcal/mol). The activity of PMDM against α-amylase was further tested and verified in vitro, and its IC 50 was found to be 79.96 ± 0.34 μM. Thus, this compound is responsible for the α-amylase inhibitory activity of the n-hexane extract of S. fusiforme .

Multifunctional Sargassum fusiforme Polysaccharide-Based Composite Sponges for Massive Hemorrhage

Muhammad Amir Ali,Xialian Fan,Yongchao Jiang,Keyong Tang

Massive hemorrhage from trauma or surgery is a leading cause of accidental death, and bacterial infection poses a serious additional risk after bleeding. Enhancing hemostatic efficacy while minimizing blood loss in the body has consistently been a primary objective for researchers. In this study, the hemostatic efficacy and tissue adhesion strength of a hemostatic material were improved by incorporating alginate and chitosan with Sargassum fusiforme polysaccharide (SFP) through electrostatic interactions. Tissue and blood cell adhesion assays, along with in vivo hemostasis experiments, demonstrated that the composite sponges significantly promoted blood clot formation and enhanced tissue adhesion strength, effectively reducing hemostasis time and rapidly preventing blood loss. These composite hemostatic sponges were shown to have a good antibacterial activity, with the ACSF 2.5 hemostatic sponges having an antibacterial ability of 92.6% against E. coli and 92.0% against S. aureus, effectively preventing wound infection. In addition, these composite hemostatic sponges exhibited excellent in vitro biocompatibility, with no cytotoxicity, hemolysis, or other adverse biological reactions. Finally, cell proliferation assays and cell staining analyses confirmed the exceptional biocompatibility of the composite hemostatic sponges. Overall, these properties make the composite sponge a promising candidate for rapid hemostasis in penetrating or deep wounds with substantial bleeding.

Polysaccharide–Protein Complex from Sargassum fusiforme: Fractionation, Characterization, and Hypoglycemic Activity

Cuifang Wang,Chaoyang Chen,Sinan Chen,Mengyan Huang,Le Dong,Fang Wang,Chao Zhao

The present study aimed to isolate a polysaccharide–protein complex (SFPr-PS3A) from the edible brown seaweed Sargassum fusiforme and to evaluate its hypoglycemic activity. SFPr-PS3A exhibited strong inhibitory activity against α-glucosidase (87.81 ± 2.41%). Protease degradation of its protein fraction markedly reduced the bioactivity (8.04 ± 2.01%), highlighting the essential role of the protein in the complex. Methylation and nuclear magnetic resonance (NMR) analyses revealed that the polysaccharide backbone of SFPr-PS3A was mainly composed of the following repeating units: →[2)-β-d-Xylp-(1]3 → 3)-α-d-Rhap-(1 → [3)-α-d-Rhap-(1]2 → 4)-α-d-Glcp-(1 → 4)-α-d-Galp-(1→, with branches of β-d-Xylp-(1→ and α-d-Fucp-(1→ linked at specific positions. In the activity assessment, SFPr-PS3A exhibited strong α-glucosidase inhibitory activity with an IC50 value of 5.23 ± 0.85 μg/mL. After simulated gastrointestinal digestion, its inhibitory activity remained stable with no significant decrease. In an insulin-resistant (IR) HepG2 cell model, treatment with SFPr-PS3A effectively promoted glucose uptake and glycogen synthesis, concomitant with the upregulation of the protein expression of Akt and GLUT2.

Antiviral activity of a polysaccharide from Sargassum fusiforme against respiratory syncytial virus

Jin Zhang,Shangzhi Wang,Mingrui Yang,Jinming Ding,Yizhen Huang,Yangdong Zhu,Ming Zhou,Bin Yan

This experiment examined the antiviral activity of polysaccharides from Sargassum fusiforme against respiratory syncytial virus (RSV) in vitro, including their mechanism of action and preliminary structural analysis. Four polysaccharides (SFP1, SFP2, SFP3, and SFP4) were purified from Sargassum fusiforme using a DEAE-52 cellulose column and an NW Super 150 gel column. CCK-8 and western blot were utilized to study the antiviral activities and mechanisms of the polysaccharides. Preliminary structural analysis was conducted using HPLC and NMR techniques. The findings suggest that SFP4 (120 kD) is an acidic chemical compound composed of 88.8 % total sugars, 0.13 % proteins, 10.8 % glucuronidic acids, and 21.1 % sulfates. It contains at least ten monosaccharides, primarily mannuronic acid and fucose. Among the four polysaccharides, SFP4 had the highest anti-RSV activity, with a therapeutic index (TI) exceeding 139. SFP4 exhibited noteworthy antiviral efficacy in both upper and lower respiratory cells that were infected, especially when administered as a prophylactic treatment 2 h in advance. Furthermore, SFP4 showed a dose-dependent antiviral effect, with the highest therapeutic index (TI > 320) observed at a concentration of 7.81 μg·mL −1 during the prophylactic phase. It was speculated that SFP4's antiviral effect is due to its ability to inhibit the attachment of G-proteins to cells.

Enzymatic degradation, antioxidant and immunoregulatory activities of polysaccharides from brown algae Sargassum fusiforme

Qian Wen-Wen,Yang Si-Qi,Hu Shu-Min,Wang Xiao-Ling,Zhu Yuzhu,Zhou Tao

In order to make full use of brown algae Sargassum fusiforme , in the presence of pectinase and glucoamylase, the polysaccharide (SFP) from S. fusiforme was hydrolyzed to improve its biological activity. Single factor tests and response surface methodology were used to optimize enzymatic hydrolysis conditions based upon the degradation rate of SFP. Conditions for the hydrolysis of SFP were optimized as: hydrolysis temperature 54.2 °C, total enzyme concentration 68.5 U/mL, activity ratio of pectinase to glucoamylase 3.3:1, pH 6.0, degradation time 3 h. Under the optimized conditions, the degradation rate of SFP reached 18.6%. The antioxidant activities of the enzymatically degraded S. fusiforme polysaccharide (ESFP) prepared under the optimized conditions were greatly improved when compared to those of SFP. The IC 50 values of ESFP on scavenging HO˙, \({\text{O}}_2^{\cdot -}\) and DPPH˙ were 4.78, 0.412 and 0.53 mg/mL, respectively; while those for SFP were 6.82, 0.723 and 0.768 mg/mL, respectively. The ferric reducing power of ESFP was also significantly higher than that of SFP. After incubation with RAW264.7 macrophages for 24 and 48 h, ESFP was found to possess significantly stronger activities in promoting cell proliferation, phagocytosis and NO release than those of SFP, indicating the superior immunoregulatory activity of ESFP.

Nanoliposome encapsulation of Sargassum fusiforme polysaccharides for enhanced bioavailability and functional skin protection

Yifan Chen,Yanan Wu,Cong Ma,Ping Shao,Lijun You,Kseniya Hileuskaya

Marine-derived polysaccharides are promising bioactive macromolecules, but their high molecular weight limits transdermal delivery and functional efficacy. In this study, a sulfated polysaccharide from Sargassum fusiforme (DSFP, ∼41 kDa) was encapsulated into nanoliposomes (DSFP@Nano) to enhance skin-targeted bioactivity. Results showed that DSFP@Nano exhibited nanoscale size, uniform distribution, negative surface charge, and preserved structural integrity. Stability assessment over 21 days demonstrated minimal changes in particle size, polydispersity, and surface potential, while contact angle measurements indicated favorable interactions with biological interfaces. In vitro and in vivo permeation studies further demonstrated that DSFP@Nano exhibited enhanced skin penetration and significantly improved the transdermal delivery efficiency of DSFP. DSFP@Nano improved anti-photoaging activity and reduced oxidative stress in skin cells compared with free DSFP. In vivo , topical application of DSFP@Nano preserved dermal collagen and mitigated UV-induced structural damage. These findings demonstrate that nanoliposome encapsulation effectively enhances the delivery and functional performance of high-molecular-weight polysaccharides, providing a versatile platform for the development of skin-targeted bioactive materials.

Characterization and Antioxidant Activity of Polysaccharides from Sargassum Fusiforme via Different Degradation Methods

Ali Muhammad Amir,Jiang Yongchao,Tang Keyong

To enhance the biological activity of Sargassum fusiforme , a brown algae, polysaccharides were extracted and degraded using two methods: ultrasound-assisted hydrochloric acid (yielding U-SFP) and α-amylase-assisted hydrolysis (yielding M-SFP). A Box-Behnken response surface design (BBRS) was used to optimize the degradation conditions. Structural changes before and after treatment were analyzed, including molecular weight variations during in vitro digestion. The relationship between the structural characteristics and antioxidant activity of the degradation products was established, revealing significant impacts on surface morphology, solubility, crystallinity, and triple-helix structure. The DPPH radical-scavenging activities of U-SFP and M-SFP were 0.106 mg/mL and 0.045 mg/mL, respectively, compared to 0.029 mg/mL for native SFP. Similarly, the ferric reducing power of both U-SFP and M-SFP was significantly higher than that of native SFP. These results demonstrate that controlled degradation enhances the antioxidant capacity of Sargassum fusiforme polysaccharides, highlighting their potential for industrial and biomedical applications.

Antioxidant and antimicrobial evaluation of carboxymethylated and hydroxamated degraded polysaccharides from Sargassum fusiforme

Yin-Ting Li,Bing-Jie Chen,Wei-Du Wu,Kun Ge,Xiao-Yi Wei,Li-Min Kong,Yuan-Yuan Xie,Jin-Ping Gu,Jin-Chao Zhang,Tao Zhou

In order to improve the bioactivity of the polysaccharide from Sargassum fusiforme (PSF), the degraded polysaccharide (DPSF) was modified by carboxymethylation, yielding carboxymethylated degraded polysaccharides (CDPSF), which were further modified to generate hydroxamated derivatives (HCDPSF). Both CDPSF and HCDPSF were characterized by Fourier transform infrared spectroscopy . The molecular weight of CDPSF and HCDPSF was found to be 354 kDa and 375 kDa, respectively. The in vitro antioxidant activity of CDPSF and HCDPSF was evaluated by determining the radical scavenging ability and total antioxidant activity. The results indicated that the antioxidant activity of CDPSF and HCDPSF was significantly improved when compared to those of DPSF. Antimicrobial assays indicated that both CDPSF and HCDPSF possessed a marked antimicrobial ability, while DPSF did not exhibit such effects under the same conditions. Such polysaccharide derivatives have potentials in the pharmaceutical and food industries.

Improved antioxidant and anti-tyrosinase activity of polysaccharide from Sargassum fusiforme by degradation

Bing-Jie Chen,Mei-Jia Shi,Shuai Cui,Shu-Xian Hao,Robert C Hider,Tao Zhou

An efficient method for the degradation of polysaccharides isolated from Sargassum fusiforme (PSF) was developed by using ascorbic acid in combination with H 2 O 2 . The degradation conditions were optimized using a Box-Behnken response surface design (BBRS). The optimum conditions were established as: concentration of ascorbic acid (V C ) and H 2 O 2 17.26   mM, degradation temperature 51   °C and degradation time 1.6   h. The DPPH radical scavenging rate of the degraded polysaccharides from S. fusiforme (DPSF) obtained under the optimal conditions was determined to be 75.22   ±   0.02%, which was well matched with the value (75.21%) predicted by the BBRS model. In vitro antioxidant activity of the polysaccharides was evaluated by determining their radical (hydroxyl radical, superoxide anion radical and DPPH radical) scavenging abilities, and ferric iron reducing power. The inhibitory activity on tyrosinase of DPSF was also evaluated. The results indicate that the degraded polysaccharide has superior antioxidant activity and anti-tyrosinase effect to those of the original polysaccharide.

Equilibrium and Kinetic Studies of Cd2+ Biosorption by the Brown Algae Sargassum fusiforme

Hui-Xi Zou,Nan Li,Li-Hua Wang,Ping Yu,Xiu-Feng Yan

A fundamental investigation of the biosorption of Cd 2+ from aqueous solution by the edible seaweed Sargassum fusiforme was performed under batch conditions. The influences of experimental parameters, such as the initial pH, sorption time, temperature, and initial Cd 2+ concentration, on Cd 2+ uptake by S. fusiforme were evaluated. The results indicated that the biosorption of Cd 2+ depended on the initial Cd 2+ concentration, as well as the pH. The uptake of Cd 2+ could be described by the Langmuir isotherm model, and both the Langmuir biosorption equilibrium constant and the maximum biosorption capacity of the monolayer decreased with increasing temperature, thereby confirming the exothermic character of the sorption process. The biosorption kinetics follows the pseudo-second-order kinetic model, and intraparticle diffusion is the sole rate-limiting step for the entire biosorption period. These fundamental equilibrium and kinetic results can support further studies to the removal of cadmium from S. fusiforme harvested from cadmium-polluted waters.
