Ultrasensitive colorimetric strategy for Hg2+ detection based on T-Hg2+-T configuration and target recycling amplification

Sang Fuming,Yin Suyao,Pan Jianxin,Zhang Zhizhou

A novelty aptasensor for ultrasensitive detection of Hg 2+ is developed, exploiting the combination of plasmonic properties of gold nanoparticles (AuNPs) and exonuclease III (Exo III)-assisted target recycling for signal amplification. In the presence of Hg 2+ , a DNA duplex can be formed due to the strong coordination of Hg 2+ and T bases of single-stranded DNA (ssDNA) probe. Exo III digests the DNA duplex from the 3′ to 5′ direction, resulting in the releasing of Hg 2+ . Then, the released Hg 2+ binds with another ssDNA probe through T-Hg 2+ -T coordination. After Exo III-assisted Hg 2+ cycles, numerous ssDNA probes are exhausted, which promotes poly(diallyldimethylammonium chloride) (PDDA)-induced AuNP aggregation, leading to an obvious color change and aggregation-induced plasmon red shift of AuNPs (from 520 to 610 nm). Therefore, this biosensor is ultrasensitive, which is applicable to the detection of trace level of Hg 2+ with a linear range from 5 pM to 0.6 nM and an ultralow detection limit of 0.2 pM. Furthermore, it enables visual detection of Hg 2+ as low as 50 pM by the naked eye. More importantly, the assay can be applied to the reliable determination of spiked Hg 2+ in sea water samples with good recovery.

A Fluorescence Biosensor Based on Carbon Quantum Dots Prepared from Pomegranate Peel and T-Hg2+-T Mismatch for Hg2+ Detection

Zhao Weiqin,Huang Chun,Guo Xiyu,Zhu Youyu,Li Yuangang,Duan Yingfeng,Gao Jie

Mercury ions (Hg 2+ ) can cause damage to human health, and thus, the study of the detection of Hg 2+ is extraordinarily important in daily life. This work reported a fluorescence biosensor for the detection of Hg 2+ . The key point of this strategy was that the fluorescence of carbon quantum dots made from pomegranate peel (P-CQDs) was quenched by hemin, and restored after G-quadruplex binding with hemin. The presence of Hg 2+ caused thymine (T)-rich DNA fragments to form T-Hg 2+ -T mismatches, and this change allowed the release of G-quadruplex. G-quadruplex could change the fluorescence of hemin/P-CQDs. P-CQDs exhibited excellent properties through characterization analysis, such as transmission electron microscope, X-ray photoelectron spectroscopy and Fourier transform infrared. This proposed fluorescence detection strategy established the linear ranges of Hg 2+ from 1 nM to 50 nM. In conclusion, this simple biosensor had the advantages of strong sensitivity, high selectivity, and low cost for Hg 2+ detection in environmental water samples.

Label-free fluorescence “turn-on” strategy for mercury (II) detection based on the T-Hg2+-T configuration and the DNA-sensitized luminescence of terbium (III)

Zhihong Li,Hongjing Sun,Xinyue Ma,Ruifang Su,Rui Sun,Chuanyu Yang,Chunyan Sun

A novel label-free fluorescence “turn-on” strategy was developed for the sensitive detection of Hg 2+ based on the thymine-Hg 2+ -thymine (T-Hg 2+ -T) coordination and the fact that single-stranded DNA (ssDNA) greatly enhances the fluorescence of terbium (III) (Tb 3+ ), but double-stranded DNA (dsDNA) does not. In the absence of Hg 2+ , the mercury-specific DNA (MSD) hybridized with the corresponding complementary strand (cDNA) to form a double helix structure in solution based on Watson-Crick base pairings, which cannot enhance the fluorescence of Tb 3+ . In the presence of Hg 2+ , MSD preferentially bound with Hg 2+ to form the T-Hg 2+ -T complex due to the strong affinity of Hg(II) for the T bases of DNA, thus avoiding the hybridization of the cDNA to MSD. The free cDNA can greatly enhance the emission of Tb 3+ , leading to a sharp increase in fluorescence intensity . Under the optimized conditions, the increased fluorescence intensity was proportional to the concentration of Hg 2+ in the range from 10 to 600 nM, and this method can detect concentrations of Hg 2+ as low as 0.24 nM. Moreover, satisfactory results were obtained for the detection of Hg 2+ in river water and fish samples, and the results were consistent with those from the atomic fluorescence spectroscopy (AFS). Thus, the fluorescence characteristics of Tb 3+ were here used in a “turn on” approach to detect Hg 2+ , which represents a new opportunity for Hg 2+ analysis in the field of environmental monitoring and food safety.

Dandelion-like CuO microspheres decorated with Au nanoparticle modified biosensor for Hg2+ detection using a T-Hg2+-T triggered hybridization chain reaction amplification strategy

Yujie Yu,Chao Yu,Rufei Gao,Jun Chen,Hangtian Zhong,Yilin Wen,Xingduo Ji,Jiahao Wu,Junlin He

We fabricate a novel electrochemical biosensor based on the specific thymine-Hg 2+ -thymine (T-Hg 2+ -T) base pair for the highly sensitive detection of mercury ions (Hg 2+ ) and utilize toluidine blue (TB) as a redox indicator that is combined with a hybridization chain reaction (HCR) for signal amplification . The dandelion-like CuO (D-CuO) microspheres that were assembled using Au nanoparticles were first introduced as support materials, which produced more active sites for the thiolated probe (P1) combination. Then, the presence of Hg 2+ induced P1 to hybridize with the other oligonucleotide (P2) through Hg 2+ -mediated T-Hg 2+ -T complexes. In addition, the partial sequence of P2 acted as an initiator sequence, which led the two hairpin DNA (H1 and H2) strands to collectively form the extended double-strand DNA through the HCR process on the electrode surface . TB was employed to interact with the double strands and produce an efficient electrochemical signal. The proposed strategy combined the amplification of the HCR and the inherent redox activity of TB and utilized D-CuO/Au composites, which exhibited high sensitivity for Hg 2+ determination. Under the optimum conditions, the proposed biosensor showed a prominent response for Hg 2+ , including a linear range from 1 pM to 100 nM and a detection limit of 0.2 pM (S/N = 3). Moreover, the new biosensor proved its potential application for trace Hg 2+ determination in environmental water samples.

Magnetic beads based colorimetric detection of mercuric ion

Xijing Huang,Yanlin Hao,Hanyin Wu,Qingquan Guo,Liangqia Guo,Jing Wang,Liangshuang Zhong,Tianran Lin,FengFu Fu,Guonan Chen

A simple and sensitive DNAzyme-based colorimetric sensor for detecting Hg 2+ based on T–Hg 2+ -T base pair and magnetic beads (MBs) is proposed. In the presence of Hg 2+ , capture probes (CPs) immobilized on MBs are hybridized with the signal probes (SPs) consisting of G-quadruplex sequences with the help of T–Hg 2+ -T base pairs. The SPs are separated by MBs and used to bind hemin for the formation of catalytically active hemin/G-quadruplex DNAzymes, which catalyzes the H 2 O 2 -mediated oxidation of ABTS to produce a blue-green-colored product. A linear relationship between the absorbance and Hg 2+ concentration is observed over a range of 10–200   nM with a limit of detection 5   nM. Taking advantage of MBs separation/collection process, the sensing system can be regenerated by using EDTA as the chelator.

A sensitive photoelectrochemical biosensor based on DepAu/TiO2 and T-Hg2+-T structure

Li Mengjie,Gong Li,Ding Yuanbo,He Yiwen

Herein, a sensitive photoelectrochemical (PEC) biosensor was designed for the detection of mercury ion (Hg 2+ ) based on deposited gold nanoparticles (DepAu) enhanced titanium dioxide (TiO 2 ) accompanied by thymine-Hg 2+ -thymine (T-Hg 2+ -T) structure. TiO 2 -modified glassy carbon electrode (GCE) exhibited a baseline anodic photocurrent, which was substantially enhanced upon electrochemical deposition of Au NPs. This PEC signal amplification was ascribed to the dual contributions of the improved photon utilization and the facilitated charge transfer. The electrode was then subjected to stepwise modification with T-rich DNA followed by 1-hexanethiol (HT), each incremental layer engendering a concomitant decrement in the photocurrent response. Ultimately, the introduction of target Hg 2+ enabled specific coordination with the T-rich DNA sequence, facilitating the formation of a stable T-Hg 2+ -T complex architecture on the GCE surface. This coordination event substantially amplified interfacial steric hindrance, thereby impeding electron transfer and effectuating pronounced photocurrent suppression. The quenched PEC response provided a quantitative metric for Hg 2+ determination. The sensitive and selective detection of Hg 2+ was achieved in the range of 100 pmol/L to 100 µmol/L, with the detection limit of 33.3 pmol/L. This PEC biosensing platform holds considerable promise for extending to the detection of diverse heavy metal ions and other environmental contaminants by rational design of recognition elements. Exonuclease III assisted and label-free detection of mercury ion based on toehold strand displacement amplification strategy

Lili Yu,Hui Xu,Hou Chen,Liangjiu Bai,Wenxiang Wang

A label-free, exonuclease III (Exo III) assisted and cost-effective Hg2+ fluorometric assay based on a strand displacement amplification strategy was developed with high sensitivity and selectivity. Thymine–thymine (T–T) mismatches between the toehold domains in hairpin DNA and assistant DNA can serve as specific recognition elements for Hg2+ binding with the help of T–Hg2+–T base pairs to initiate a toehold-mediated strand displacement reaction. Exo III provides a reduced background signal, and the method is label-free using an intercalator, Sybr green I (SG). The number of T–T mismatches, the concentration of SG and Exo III, the incubation temperature and time were all optimized. Under the optimized condition, the limit of detection for Hg2+ can be as low as 9.42 pM with a linear range from 0.01 to 1 nM. Moreover, this proposed strategy shows excellent selectivity, and was successfully applied for the detection of Hg2+ in laboratory tap water and Yanjing lake water samples in our university.

Fe3O4/Au NPs-based electrochemical sensor for the detection of Hg ion in Traditional Chinese Medicine

Fang Li,Pengying Zhang,Jiayi Chen,Jiahui Xi,Shuai Gao,Yunhong Liu

Mercury ion (Hg 2+ ), a highly toxic and bioaccumulative pollutant, can contaminate medicinal herbs and pose significant risks to human health via the food chain. With the growing global demand for traditional Chinese medicine (TCM), there is an urgent need to develop reliable and sensitive Hg 2+ detection methods. In this paper, a novel electrochemical sensor based on magnetic nanoparticles and thymine-Hg 2+ -thymine (T-Hg 2+ -T) coordination chemistry was developed for detecting Hg 2+ in TCM. The magnetic Fe 3 O 4 /Au nanoparticles (Fe 3 O 4 /Au NPs) were synthesized and employed as carriers to conjugate with T-rich thiol-modified DNA probes. In the absence of Hg 2+ , the DNA probes hybridized with ferrocene (Fc)-labeled complementary DNA (cDNA), bringing the Fc signal molecules close to the Fe 3 O 4 /Au NPs surface. In the presence of Hg 2+ , these probes preferentially formed stable T-Hg 2+ -T complexes, effectively inhibiting cDNA hybridization. The Hg 2+ concentration was determined by measuring the corresponding changes in the Fc current. The sensor exhibited a good linear response to Hg 2+ in the range of 0.1–100 ng/mL, with a low detection limit of 0.08 ng/mL. Furthermore, the method was successfully applied to determine Hg 2+ in representative TCM matrices (Lotus leaf, Lonicera japonica , Angelica dahurica , and Houttuynia cordata Thunb.) with satisfactory recoveries (96.73 %−104.84 %). This indicates that the proposed method is particularly suitable for Hg 2+ determination in complex TCM samples, providing a practical solution for quality control and safety monitoring in TCM.

Function of Graphene Oxide as the “Nanoquencher” for Hg2+ Detection Using an Exonuclease I-Assisted Biosensor

Ting Sun,Xian Li,Xiaochuan Jin,Ziyi Wu,Xiachao Chen,Jieqiong Qiu

Graphene oxide is well known for its excellent fluorescence quenching ability. In this study, positively charged graphene oxide (pGO25000) was developed as a fluorescence quencher that is water-soluble and synthesized by grafting polyetherimide onto graphene oxide nanosheets by a carbodiimide reaction. Compared to graphene oxide, the fluorescence quenching ability of pGO25000 is significantly improved by the increase in the affinity between pGO25000 and the DNA strand, which is introduced by the additional electrostatic interaction. The FAM-labeled single-stranded DNA probe can be almost completely quenched at concentrations of pGO25000 as low as 0.1 μg/mL. A simple and novel FAM-labeled single-stranded DNA sensor was designed for Hg2+detection to take advantage of exonuclease I-triggered single-stranded DNA hydrolysis, and pGO25000 acted as a fluorescence quencher. The FAM-labeled single-stranded DNA probe is present as a hairpin structure by the formation of T–Hg2+–T when Hg2+is present, and no fluorescence is observed. It is digested by exonuclease I without Hg2+, and fluorescence is recovered. The fluorescence intensity of the proposed biosensor was positively correlated with the Hg2+concentration in the range of 0–250 nM (R2= 0.9955), with a seasonable limit of detection (3σ) cal. 3.93 nM. It was successfully applied to real samples of pond water for Hg2+detection, obtaining a recovery rate from 99.6% to 101.1%.Keywords:positively charged graphene oxide (pGO);exonuclease I;fluorescence quencher;hairpin structure;T–Hg2+–T

A label-free SERS approach to quantitative and selective detection of mercury (II) based on DNA aptamer-modified SiO2@Au core/shell nanoparticles

Yilin Lu,Jie Zhong,Guohua Yao,Qing Huang

A prototype of label-free SERS sensor, based on the formation of T-Hg 2+ -T pairs and the corresponding orientation variation of DNA on Au shell surface, was developed for detection of Hg 2+ in aqueous solution quantitatively and selectively. The DNA aptamer immobilized on the surface of SiO 2 @Au core/shell nanoparticles (NPs) consisted of two segments, namely, the segment which has the consecutive thymines (T) severing as the Hg 2+ recognition elements, and the segment which contains the guanine (G) and adenine (A) bases working as the signal reporter. With interaction of Hg 2+ ions and the thymines between the adjacent single-stranded DNA (ssDNA), the DNA molecule adopted vertical orientation, resulting in increase of Raman intensity ratio I(660   cm −1 )/I(736   cm −1 ) with increase of Hg 2+ concentration, which thus allowed to measure trace amounts of Hg 2+ in aqueous solution selectively and quantitatively. Our results revealed that this label-free SERS sensor could sensitively respond to Hg 2+ ions within a wide concentration range (from 1   ×   10 −8 to 1   ×   10 −3   M). This work therefore demonstrates that proper design of aptamer-modified SiO 2 @Au core/shell NPs can be utilized for label-free SERS detection of heavy metal ions in the environment.

Gold core-satellite nanostructure linked by oligonucleotides for detection of glutathione with LSPR scattering spectrum

Ying-Bo Liu,Ting-Ting Zhai,Yan-Yan Liang,Yue-Bo Wang,Xing-Hua Xia

We demonstrated a sensitive method for detection of glutathione (GSH) based on LSPR scattering spectrum using gold core-satellite nanostructure linked by T-Hg 2+ -T base pair . The core-satellite assembly caused coupling between plasmonic nanoparticles , which inducing distinct change of LSPR peak wavelength. As the interaction between Hg 2+ and GSH, the core-satellite nanostructure would be disassembled, which accompanied with spectral blue-shift of the scattering spectrum. By using this method, GSH could be quantitatively detected, and the detection limits can reach to 0.1 µM.

Electrochemical DNA sensor for specific detection of picomolar Hg(II) based on exonuclease III-assisted recycling signal amplification

Xiaorong Gan,Huimin Zhao,Shuo Chen,Xie Quan

An ultrasensitive methodology was successfully developed for the quantitative detection of picomolar Hg2+ based on the combination of thymine–Hg2+–thymine (T–Hg2+–T) coordination chemistry and exonuclease III-aided recycling signal amplification. Single-strand probe DNA was immobilized on an Au electrode via an Au–S bond. In the presence of Hg2+, the probe DNA hybridized with the target DNA containing four thymine–thymine (T–T) mismatches via the Hg2+-mediated coordination of T–Hg2+–T base pairs. Then the probe DNA in the DNA duplex was specifically recognized and selectively digested by exonuclease III; in contrast the target DNA was safely dissociated from the DNA duplexes to subsequently hybridize with a new signal probe, leading to target recycling and signal amplification. As a result, the peak current caused by the electrostatic interactions of [Ru(NH3)6]3+ cations with the backbone of the probe DNA decreased by different degrees, corresponding to the Hg2+ concentrations. Under the optimum conditions, the proposed electrochemical DNA biosensor showed a robust detection limit as low as 1 pM (S/N = 3), with a wide linear range from 0.01 to 500 nM and good selectivity. In addition, the proposed method was successfully applied to assay Hg2+ in real environmental samples.
