Developmental effects of fenpropathrin on zebrafish (Danio rerio) embryo-larvae: Toxic endpoints and potential mechanism

Junguo Ma,Chunyu Diao,Yidan Zhang,Huan Kang,Yiyi Feng,Yuanyuan Li,Xiaoyu Li

Fenpropathrin (FEN), a highly efficient and broad-spectrum synthetic pyrethroid insecticide. Although sufficient concern has been given to the negative impacts of FEN on non-target organisms, limited knowledge exists regarding the developmental effects of FEN. In this study, effects of FEN (0.45, 1.35, 4.05, and 12.15 μg/L) on various early life-stage endpoints of zebrafish were investigated from 3 to 144 h post-fertilization (hpf) in order to disclose the developmental effects and underlying mechanisms caused by FEN. The results indicate that exposure to FEN induced developmental toxicity in zebrafish, including decreased heart rate, reduced blood flow, shorter body length, smaller eye size, non-inflated swim bladder, and disrupted craniofacial chondrogenesis, which were possibly due to a significant decrease in the levels of thyroxine (T4), triiodothyronine (T3), insulin-like growth factor-1 (IGF-1), and growth hormone (GH), increase in lipid accumulation, and alteration in the contents of total cholesterol (T-CHO) and triglyceride (TG) in larvae exposed to FEN. Besides, FEN exposure also resulted in the inhibition of spontaneous movement of embryo at 24 hpf, a decline in touch evoke response (TER) at 72 hpf, and a reduction in free-swimming activity at 144 hpf, as well as the larval activity at 144 hpf during the dark-light transition stimulus. Mechanistic examinations have shown that FEN treatment inhibits the activities of AChE and elevates the ACh levels. In addition, FEN exposure increased ROS levels and altered the levels of malondialdehyde (MDA), and induced apoptosis as determined by acridine orange staining and elevated caspase-3 levels, suggesting that the involvement of oxidative stress and apoptosis in FEN-induced developmental toxicity of embryos. Transcriptome sequencing of larvae showed that FEN altered the expressions of multiple metabolic and nervous system pathways, including PPAR signaling pathway, lipid metabolism pathway, carbohydrate metabolism pathway, retinol metabolism pathway, and neuroactive ligand-receptor interaction pathway, demonstrating that FEN alters the normal development of zebrafish embryos, and multiple pathways mediating the FEN-induced developmental toxicity. Overall, these findings enhance our understanding of the developmental toxic effects of FEN and provide fundamental data for assessing the risk of FEN on non-target organisms.

Electrospun nanofibres containing Zn-MOF for electrochemiluminescent determination of fenpropathrin residues in fruit juices

Zhuo Sun,Juan Lu,Xin Zhang,Xiangyu Shan,Qian Wu,Chao Li,Huiling Li,Shuning Yang,Li Tian

In this work, Zn-MOF electrospun nanofibres (Zn-MOF NFs), CdTe and Au@Pt-rGo were applied to Ru(bpy) 3 2+ /tri- n -propylamine electrochemiluminescence (ECL) system. And a novel ECL sensor for the determination of fenpropathrin was constructed for the first time. Due to the large surface area and pores, Zn-MOF NFs can both carry CdTe and Au@Pt-rGo and promote luminescence by facilitating electron transfer and reducing electron transport paths. CdTe participated in the luminescence reaction and improved the system’s sensitivity together with Ru(bpy) 3 2+ . Au@Pt-rGo can promote electron transport in the system. Under the synergistic effect of the three, the ECL sensor had achieved satisfactory results in weak alkaline solution. The constructed ECL sensor can detect fenpropathrin in the range of 4 × 10 −11 –4 × 10 −6 M, and the detection limit was 1.33 × 10 −11 M (S/N = 3). It can be used to detect fenpropathrin in juice samples, which provided a new idea for determining fenpropathrin.

Fenpropathrin exposure induces neurotoxicity in zebrafish embryos

Yu Tingting,Xu Xiaowen,Mao Huiling,Han Xue,Liu Yulong,Zhang Hongying,Lai Jingli,Gu Jianfeng,Xia Mengling,Hu Chengyu,Li Dongming

Fenpropathrin has been a commonly used insecticide to control agricultural and household insects over a few decades. Up to now, fenpropathrin residue in soil and water has been often determined due to its widespread use, which poses serious threat to environment and aquatic organisms. The potential of fenpropathrin to affect aquatic lives is still poorly understood. In this study, we used zebrafish ( Danio rerio ) embryo as an experimental model system to evaluate the toxicity of fenpropathrin to the development of zebrafish nervous system. Zebrafish embryos were separately exposed to fenpropathrin at the dose of 0.016 mg/L, 0.032 mg/L, 0.064 mg/L, starting at 6 h post-fertilizationhpf (hpf) up to 96 hpf. The results showed that fenpropathrin exposure gives rise to physiological, behavioral, and neurodevelopmental impairments in zebrafish embryos, including enhanced acetylcholinesterase (AChE) activity, abnormal swimming behavior, karyopyknosis in brain cells, increased intercellular space, and uneven migration of neuron in brain area. In addition, the expressions of genes concerning neurodevelopment and neurotransmitter system were inhibited following fenpropathrin exposure. We also found that fenpropathrin exposure distinctly induced oxidative stress by increasing reactive oxygen species (ROS) generation and inhibiting the production of antioxidant enzymes catalase (CAT) and superoxide dismutase (SOD). Expectedly, some apoptosis-associated genes were induced and the apoptosis appeared in the brain and heart cells of zebrafish embryos. Moreover, fenpropathrin exposure also inhibited the expressions of genes in Nrf2 signaling pathway, such as heme oxygenase-1 ( HO-1 ) and SOD . In summary, the results of this study indicate that oxidative stress-triggered apoptosis may be an underlying fundamental of fenpropathrin-induced neurotoxicity in zebrafish embryos.

A strip-based immunoassay for rapid determination of fenpropathrin

Xiujin Chen,Liqiang Liu,Hua Kuang,Shanshan Song,Chuanlai Xu

We report the generation of monoclonal antibodies against fenpropathrin, originating from a BALB/c mouse immunized with a conjugate of hapten 1 [(RS)-cyano-3-phenoxybenzyl-2,2,3,3-tetramethylcyclopropane-carboxylic acid)] and keyhole limpet hemocyanin and the establishment of a monoclonal antibody-based immunochromatographic test strip. A coating antigen (test band) and a goat anti-mouse antibody (control band) were separately immobilized on nitrocellulose membrane strips as capture reagents. The anti-fenpropathrin monoclonal antibody was labeled with gold nanoparticles as a detection probe. This test strip had a wide linear range (15.6–250 μg L−1) with a low detection limit of 62 ± 6 μg L−1 for fenpropathrin, evaluated using a strip reader, and could be read within 10 min. This novel test strip was used to test fenpropathrin in fortified samples and found to have a high recovery rate (>80%). In summary, our monoclonal antibody-based immunochromatographic strip offers a rapid screening tool for fenpropathrin detection in agricultural commodities.

Carboxymethyl chitosan–modified UiO-66 for the rapid detection of fenpropathrin in grains

Xiaoyao Wang,Haizhi Huang,Siyao Zhong,Xuping Shentu,Zihong Ye,Xiaoping Yu

Fenpropathrin residues in grain are potentially harmful to humans. Therefore, a fluorimetric lateral flow immunoassay using a zirconium-based organic skeleton (UiO-66) as a signal marker was developed for detecting fenpropathrin. Herein, carboxymethyl chitosan (CMCS) was used to modify UiO-66 and improve its water solubility to facilitate stable binding with sodium fluorescein (NaFL). This resulted in formation of a new fluorescent probe that is more suitable for lateral flow immunoassay (LFIA). The materials were characterized via electron microscopy , Fourier-transform infrared spectroscopy, and powder X-ray diffraction. CMCS and NaFL were successfully bound to UiO-66. Under optimized conditions, the constructed NaFL/UiO-66@CMCS-LFIA exhibited a good linear relationship within the range of 0.98–62.5 μg/L, with a detection limit of 3.91 μg/L. This probe was fourfold more sensitive than traditional colloidal gold nanoparticle-based LFIA. Finally, NaFL/UiO-66@CMCS-LFIA was successfully applied to detect fenpropathrin in wheat and maize samples. The detection limit was 1.56 μg/kg and recoveries ranged from 96.58 % to 118.56 %. This study provides a sensitive, stable, and convenient method for the rapid detection of pesticide residues .

A molecule-imprinted electrochemiluminescence sensor based on CdS@MWCNTs for ultrasensitive detection of fenpropathrin

Wu Qian,Tian Li,Shan Xiangyu,Li Huiling,Yang Shuning,Li Chao,Song Yujia,Li Ruidan,Guo Yanjia,Lu Juan

A molecularly-imprinted electrochemiluminescence sensor was constructed for the determination of fenpropathrin (FPT) by molecular imprinting technology. In this sensing platform, the introduction of CdS@MWCNTs significantly enhanced the initial ECL signal of the luminol-O 2 system. Specifically, MWCNTs was used as a carrier to adsorb more CdS, in which CdS acted as a co-reaction promoter for luminescence. Molecularly imprinted polymer (MIP) containing specific recognition sites of FPT was used as the material for selective recognition. With increasing amount of FPT the ECL signal decreased. Under the optimum conditions, the ECL response was linearly related to the logarithm of FPT concentration. The developed ECL sensor allowed for sensitive determination of FPT and exhibited a wide linear range from 1.0 × 10 − 10 mol L − 1 to 1.0 × 10 − 6 mol L − 1 . The limit of detection was 3.3 × 10 − 11 mol L − 1 (S/ N  = 3). It can be used for the detection of FPT in vegetable samples.

Fumarate mitigates disruption induced by fenpropathrin in the silkworm Bombyx mori (Lepidoptera): A metabolomics study

Xue-Yang Wang,Zi-Qin Zhao,Cheng-Xian Song,Zhi-Hao Su,Mu-Wang Li,Yang-Chun Wu,Byung Rae Jin,Ming-Jie Deng

The silkworm Bombyx mori L. is a model organism of the order Lepidoptera. Understanding the mechanism of pesticide resistance in silkworms is valuable for Lepidopteran pest control. In this study, comparative metabolomics was used to analyze the metabolites of 2 silkworm strains with different pesticide resistance levels at 6, 12, and 24 h after feeding with fenpropathrin. Twenty-six of 27 metabolites showed significant differences after fenpropathrin treatment and were classified into 6 metabolic pathways: glycerophospholipid metabolism, sulfur metabolism, glycolysis, amino acid metabolism, the urea cycle, and the tricarboxylic acid (TCA) cycle. After analyzing the percentage changes in the metabolic pathways at the 3 time points, sulfur metabolism, glycolysis, and the TCA cycle showed significant responses to fenpropathrin. Confirmatory experiments were performed by feeding silkworms with key metabolites of the 3 pathways. The combination of iron(II) fumarate + folic acid (IF-FA) enhanced fenpropathrin resistance in silkworms 6.38 fold, indicating that the TCA cycle is the core pathway associated with resistance. Furthermore, the disruption of several energy-related metabolic pathways caused by fenpropathrin was shown to be recovered by IF-FA in vitro . Therefore, IF-FA may have a role in boosting silkworm pesticide resistance by modulating the equilibrium between the TCA cycle and its related metabolic pathways.

A chromosome-level genome assembly and multi-omics analysis reveal fenpropathrin resistance mechanisms in the turnip aphid, Lipaphis erysimi

Xiudao Yu,Benjamin J Hunt,Shuchao Wang,Bartlomiej J Troczka,Chaozhi Shangguan,Wenhong Zhu,Yinhui Kuang,Chris Bass

BACKGROUND The turnip aphid Lipaphis erysimi is a cruciferous crop pest with the potential to reduce yields by up to 90%. Treatment of L. erysimi infestations using insecticides has resulted in numerous cases of resistance in field populations yet genomic resources are lacking for this species, limiting the ability of researchers to investigate the molecular basis of resistance and develop management strategies. RESULTS Here we describe the creation of a chromosome scale genome assembly for L. erysimi and the characterization of fenpropathrin resistance in this species using multi-omics analysis. A 409 Mb genome was assembled for L. erysimi , with an N50 of 95.2 Mb and 90% of assembled content contained within four chromosome-scale scaffolds. We identified a key M903L mutation in a voltage-gated sodium channel gene that is targeted by fenpropathrin and the accompanying up-regulation of cytochrome P450 and UDP-glycosyltransferase detoxification genes. Using transgenic flies, we further confirmed the ability of L. erysimi CYP4CJ1 to confer fenpropathrin resistance in vivo . CONCLUSION Mechanisms for fenpropathrin resistance in L. erysimi were identified through multi-omics analysis and functional characterization. Collectively, this study provides valuable genomic resources for molecular investigations of L. erysimi and establishes a foundation for developing novel strategies to enhance pyrethroid efficacy in this species. © 2026 Society of Chemical Industry.

The Effect of the Pyrethroid Pesticide Fenpropathrin on the Cardiac Performance of Zebrafish and the Potential Mechanism of Toxicity

Ferry Saputra,Yu-Heng Lai,Marri Jmelou M. Roldan,Honeymae C. Alos,Charlaine A. Aventurado,Ross D. Vasquez,Chung-Der Hsiao

Simple SummaryIn recent years, the human population on Earth has increased, leading to a higher demand for food sources. Pesticides have emerged as a breakthrough to enhance crop yields. However, the excessive use of pesticides has raised concerns regarding their potential impact on aquatic animals. Thus, this study aims to assess the potential cardiovascular toxicity of various pesticides, with a specific focus on fenpropathrin. When compared to other pesticides, fenpropathrin causes cardiomegaly, an increased heart rate, a significant rise in blood flow velocity, and an elevated metabolism rate. Furthermore, it has been found to bind to several ion channels, inducing alterations in cardiovascular-related markers. These findings indicate that fenpropathrin induces cardiotoxicity in the zebrafish larvae model.AbstractFenpropathrin, a pyrethroid insecticide, has been widely used for many years in agricultural fields. It works by disturbing the voltage-gated sodium channel, leading to paralysis and the death of the target animal. While past studies have focused on neurodegeneration following fenpropathrin poisoning in humans, relatively few pieces of research have examined its effect on other peripheral organs. This study successfully investigated the potential toxicity of fenpropathrin on the cardiovascular system using zebrafish as an animal model. Zebrafish larvae exposed to varying doses of fenpropathrin underwent an evaluation of cardiac physiology by measuring the heart rate, stroke volume, cardiac output, and shortening fraction. The blood flow velocity and the dorsal aorta diameter were also measured to assess the impact of fenpropathrin exposure on the vascular system. Furthermore, molecular docking was performed to evaluate the pesticide binding affinity to various proteins associated with the cardiovascular system, revealing the potential mechanism of the fenpropathrin cardiotoxic effect. The findings demonstrated a significant dose-dependent increase in the heart rate stroke volume, cardiac output, shortening fraction, and ejection fraction of zebrafish larvae after 24 h of acute treatment with fenpropathrin. Additionally, zebrafish treated at a concentration of 1 ppm exhibited significantly larger blood vessels in diameter and an increased blood flow velocity compared to the control group. According to molecular docking, fenpropathrin showed a high affinity for various voltage-gated sodium channels like scn1lab, cacna1sb, and clcn3. Finally, from the results, we found that fenpropathrin caused cardiomegaly, which may have been induced by the voltage-gated sodium channel disruption. This study highlights the significant disruption of fenpropathrin in the cardiovascular system and emphasizes the need for further research on the health implications of this pesticide. Keywords:fenpropathrin;zebrafish;cardiovascular toxicity;pesticide

Solid-state Ru(bpy)32+ electrochemiluminescence sensor for trace detection of fenpropathrin using loofah sponge-like carbon nanofibers and CdS

Lu Juan,Li Chao,Guo Yanjia,Feng Yongjia,Song Yujia,Li Ruidan,Tian Li,Wang Jing

Loofah sponge-like carbon nanofibers (LF-Co,N/CNFs) were utilized as a carrier for Ru(bpy) 3 2+ , and then combined with CdS to create a novel solid-state electrochemiluminescence sensor capable of detecting trace amounts of fenpropathrin. LF-Co,N/CNFs, obtained through the high-temperature pyrolysis of ZIF-67 coaxial electrospinning fibers, were characterized by a loofah-like morphology and exhibited a significant specific surface area and porosity. Apart from serving as a carrier, LF-Co,N/CNFs also functioned as a luminescence accelerator, enhancing the system's luminescence efficiency by facilitating electron transmission and reducing the transmission distance. The inclusion of CdS in the luminescence reaction, in conjunction with Ru(bpy) 3 2+ , further boosted the sensor's luminescence signal. The resulting sensor demonstrated a satisfactory signal, with fenpropathrin causing significant quenching of the ECL signal. Under optimized conditions, a linear relationship between the signal quench value and fenpropathrin concentration in the range 1 × 10 –12 to 1 × 10 –6  M was observed, with a detection limit of 3.3 × 10 –13  M (S/N = 3). This developed sensor is characterized by its simplicity, sensitivity, and successful application in detecting fenpropathrin in real samples. The study not only presents a straightforward detection platform for fenpropathrin but also introduces new avenues for the rapid determination of various food contaminants, thereby expanding the utility of carbon fibers in electrochemiluminescence sensors. Chiral pyrethroid insecticide fenpropathrin and its metabolite: enantiomeric separation and pharmacokinetic degradation in soils by reverse-phase high-performance liquid chromatography

Ping Zhang,Qian Yu,Yuhan He,Wentao Zhu,Zhiqiang Zhou,Lin He

The enantiomeric separation of fenpropathrin enantiomers on Lux Cellulose-1, Lux Cellulose-3 and Chiralpak IC chiral columns as well as enantioselective degradation of fenpropathrin in soil were investigated by reverse-phase high-performance liquid chromatography (RP-HPLC). The effects of the chiral stationary phase, mobile phase component, mobile phase ratio, temperature and thermodynamic parameters on resolution were carefully evaluated. Fenpropathrin enantiomers achieve a baseline separation (Rs = 2.30) on the Lux Cellulose-3 column with a mobile phase of methanol/water at a ratio of 85/15. Partial separations were achieved on Lux Cellulose-1 and Chiralpak IC columns with maximum Rs = 1.01 and Rs = 0.64, respectively. Enantioselective degradation of fenpropathrin enantiomers in soil was investigated based on the proposed separation method. The half-life (t1/2) of S-fenpropathrin and R-fenpropathrin was 17.8 days and 12.6 days, respectively, indicating that the degradation of fenpropathrin in soil was enantioselective, with R-fenpropathrin preferentially degraded. Furthermore, the formation and degradation of 3-PBA, the main metabolite of fenpropathrin, were also investigated in three soils. Soil microorganisms were confirmed as the main factor that is responsible for enantioselective degradation of fenpropathrin enantiomers in soil. Such results provide a new approach for fenpropathrin enantiomer separation and contribute to a comprehensive risk assessment of fenpropathrin at the enantiomer level.

Biaobiao Ge,Jie Huang,Qianmin Dong,Fen Yang,Mengmeng Wang,Pei Liang

This work reveals a novel SERS enhancement mechanism of “satellite” structure. • Stable and uniform L-Au@ZIF-67@S-Au NPs were synthesized by self-assembly method. • L-Au@ZIF-67@S-Au NPs substrates was prepared with an enhancement factor of 3.4 × 10 6 . • Determination of fenpropathrin in citrus peel, and its LOD reached 0.995 μg/L. A new hybrid substrate of core-satellite plasma MOF, namely L-au @ ZIF-67 @ S au NPs, was developed in this study. The substrate aims to improve SERS performance by combining the molecular trapping layer of ZIF-67 with double electromagnetic hot spots distributed on the inner surface and the outer surface. The interface self-assembly strategy was adopted to combine MOF with plasma nanostructures, thus combining excellent molecular adsorption ability with strong local field enhancement effect. The results showed that the substrate had a low detection limit of 1.43 × 10 −10  M for rhodamine 6G, achieved rapid adsorption equilibrium within 10 min, and showed high signal reproducibility (relative standard deviation, RSD = 1.66%, n  = 20). The substrate was applied to the detection of fenpropathrin residues in citrus peels, showing good sensitivity, and the detection limit was 0.995 μg·L −1 , with an enhancement factor of 3.4 × 10 6 and excellent reproducibility. This work builds a multifunctional and environment-friendly SERS platform, which effectively combines advanced nano-material design with practical applications of food safety monitoring Download: Download high-res image (146KB) Download: Download full-size image
