The aim of this research was to investigate the antimicrobial characteristics and mechanism of hesperetin againstAlicyclobacillus acidoterrestrisvegetative cells. The results presented show that hesperetin had effective antimicrobial activity onAlicyclobacillus acidoterrestrisvegetative cells, minimum inhibition concentration (MIC) of 0.0625 g/L, and minimum bacterial concentration (MBC) greater than 2 g/L. Moreover, treatment of hesperetin caused significant damage to cell integrity, preventing the growth ofAlicyclobacillus acidoterrestrisvegetative cells, enhancing the leakage of nucleic acid and proteins, and destroying the vegetative cell morphology. To further investigate the mechanism, transcriptomic analysis was carried out, and 3056 differentially expressed genes (DEGs) were detected. Gene ontology (GO) enrichment analysis revealed that hesperetin inhibitsAlicyclobacillus acidoterrestrisby affecting the intracellular nitrogen metabolism and amino acid metabolism. The Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis explained that hesperetin was also able to prevent the growth ofAlicyclobacillus acidoterrestrisby affecting the processes of nutrient transport, energy metabolism, and flagella motility. These results provide new insights into the antimicrobial effects and mechanism of hesperetin againstAlicyclobacillus acidoterrestris, which provides a new method for inactiveAlicyclobacillus acidoterrestrisin the juice industry. Keywords:Alicyclobacillus acidoterrestris;hesperetin;antibacterial activity;antibacterial mechanism;transcriptomic analysis

Hesperetin (Hst) is a common citrus fruit flavonoid with antioxidant, anti-inflammatory, and anti-neurodegenerative effects. To explore the antioxidant and anti-aging effects and mechanisms of Hst, we induced chronic oxidative stress in Caenorhabditis elegans (C. elegans) using low-concentration H 2 O 2 and examined its effects on lifespan, healthy life index, reactive oxygen species (ROS), antioxidant enzymes, and transcriptomic metrics. Hst significantly prolonged lifespan, increased body bending and pharyngeal pumping frequency, decreased ROS accumulation, and increased antioxidant enzyme activity in normal and stressed C. elegans . Hst significantly upregulated daf-18 , daf-16 , gst-2 , gst-3 , gst-4 , gst-39 , hsp-16.11 , sip-1 , clpp-1 , and dve-1 and downregulated ist-1 and kgb-1 mRNAs in stressed C. elegans . These genes are involved in the insulin/insulin-like growth factor-1 signaling (IIS), heat shock protein (HSP), mitochondrial unfolded protein response (mtUPR), and c-Jun N-terminal kinase (JNK) pathways. In summary, Hst increases lifespan and antioxidant ability, correlating with these pathways, during chronic oxidative stress in C. elegans .

The molecularly imprinted solid-phase extraction (MISPE) monolithic column coupled with high-performance liquid chromatography (HPLC) was firstly developed for the extraction of hesperetin in the flesh of Citrus reticulata cv. Chachiensis , which is a traditional Chinese medicine (TCM) . The molecularly imprinted polymers (MIPs) have been prepared by a thermal polymerization method using hesperetin as the template, acrylamide (AM) as functional monomer and ethylene glycol dimethacrylamide (EGDMA) as cross-linker in the mixed porogen of methanol, toluene and dodecanol. The prepared MIPs were characterized in detail by SEM and FTIR. The results confirmed the uniform and open structure of network skeleton with large flow-through pores. The influence of synthesis conditions on the specific recognition properties of hesperetin MIPs were also investigated systematically. The results showed that high adsorption capacity and good selectivity of MIPs were achieved when using non-imprinted polymer monolith (NIP) and structure similarly compound rutin as references. Furthermore, several parameters of the MISPE method have been optimized, and then it was successfully applied to the extraction of hesperetin from the flesh of Citrus reticulata cv. Chachiensis . Good gathering and impurity removing ability of prepared MIP were demonstrated. The MISPE method was proven to be a potentially competitive technique for separation and cleanup of hesperetin in complex TCM with satisfied recovery (90.8 ± 3.2%) and good precision (RSD = 6.48%).

Through simple bioconcentration, salt molecules of NaCl and KCl were introduced into lotus root, then a class of porous carbon materials (CPLR) with 3D pore structure based on the as-prepared lotus root was prepared by direct pyrolysis under different temperature. The CPLR was characterised by scanning electron microscopy, transmission electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy, and Raman spectra (Raman). In addition, the electrochemical performance of the CPLR-based sensors was examined by electrochemical impedance spectroscopy and cyclic voltammetry. Under the first-rank experimental conditions, the response current increased linearly with the concentration of hesperetin in the range from 0.05 to 100.0 μmol/L, and the minimum detectability was calculated to be 0.011 μmol/L ( S / N = 3). The sensor in Chinese herbal medicine buddleja officinalis maxim and healthy human urine samples performed well.

In this study we designed and synthesized a series of new hesperetin derivatives on the basis of the structural characteristics of acetylcholinesterase (AChE) dual-site inhibitors. The activity of the novel derivatives was also evaluated. Results showed that the synthesized hesperetin derivatives displayed stronger inhibitory activity against AChE and higher selectivity than butyrylcholine esterase (BuChE) (selectivity index values from 68 to 305). The Lineweaver-Burk plot and molecular docking study showed that these compounds targeted both the peripheral anionic site (PAS) and catalytic active site (CAS) of AChE. The derivatives also showed a potent self-induced β-amyloid (Aβ) aggregation inhibition and a peroxyl radical absorbance activity. Moreover, compound4fsignificantly protected PC12 neurons against H2O2-induced cell death at low concentrations. Cytotoxicity assay showed that the low concentration of the derivatives does not affect the viability of the SH-SY5Y neurons. Thus, these hesperetin derivatives are potential multifunctional agents for further development for the treatment of Alzheimer’s disease. Keywords:hesperetin derivatives;acetylcholinesterase (AChE);butyrylcholine esterase (BuChE);β-amyloid (Aβ);multifunctional

Inhibition of advanced glycation end products (AGEs) formed in protein glycosylation is crucial for minimizing diabetic complications. Herein, the anti-glycation potential of hesperetin-Cu (II) complex was investigated. Hesperetin-Cu (II) complex strongly inhibited three stages glycosylation products in bovine serum albumin (BSA)-fructose model, especially for the inhibition of AGEs (88.45%), which was stronger than hesperetin (51.76%) and aminoguanidine (22.89%). Meanwhile, hesperetin-Cu (II) complex decreased the levels of BSA carbonylation and oxidation products. 182.50 µg/mL of hesperetin-Cu (II) complex inhibited 66.71% β-crosslinking structures of BSA, and scavenged 59.80% superoxide anions and 79.76% hydroxyl radicals. Moreover, after incubating with methylglyoxal for 24 h, hesperetin-Cu (II) complex removed 85.70% methylglyoxal. The mechanisms of protein antiglycation by hesperetin-Cu (II) complex may be through protecting structure, trapping methylglyoxal, scavenging free radicals and interacting with BSA. This study may contribute to the development of hesperetin-Cu (II) complex as a functional food additive against protein glycation.

A highly sensitive voltammetric sensor, based on reduced graphene oxide on SWCNTs modified glassy carbon electrode (GCE), was constructed and used for sensitive detection of hesperetin . The electrochemical behavior of hesperetin at this sensor was investigated systematically and a novel voltammetric method for determination of hesperetin was proposed. The redox characters of hesperetin was discussed in detail and a reasonable reaction mechanism was proposed also. As the analytical method, the response currents were linear relationship with the hesperetin concentrations in the range of 5.0×10 −8 to 3.0×10 −6 mol L− 1 , with a detection limit of 2.0×10 −8 mol L −1 ( S / N =3). The method was also applied successfully to detect hesperetin in biological samples and Chinese herbal medicine Flos buddlejae with satisfactory results.

Hesperetin (HES) is a key biological active ingredient in citrus peels, and is one of the natural flavonoids that attract the attention of researchers due to its numerous therapeutic bioactivities that have been identified in vitro. As a bioenhancer, piperine (PIP) can effectively improve the absorption of insoluble drugs in vivo. In the present study, a cocrystal of HES and PIP was successfully obtained through solution crystallization. The single-crystal structure was illustrated and comprehensive characterization of the cocrystal was conducted. The cocrystal was formed by two drug molecules at a molar ratio of 1:1, which contained O–H–O hydrogen bonds between the carbonyl and ether oxygen of PIP and the phenolic hydroxyl group of HES. In addition, a solubility experiment was performed on powder cocrystal in simulated gastrointestinal fluid, and the result revealed that the cocrystal improves the dissolution behavior of HES compared with that of the pure substance. Furthermore, HES’s bioavailability in the cocrystal was six times higher than that of pristine drugs. These results may provide an efficient oral formulation for HES.Keywords:hesperetin;piperine;cocrystal;solubility;bioavailability

Background: Exploring new strategies to improve type 2 diabetes mellitus (T2DM) is one of the frontier hotspots in the field of healthy food. Flavonoid–metal complexes have become one of the research hotspots in the field of health foods due to their unique structural and functional properties. Methods: In this study, the effect of hesperetin–copper(II) complex [Hsp–Cu(II)] on the gut microbiota of mice with T2DM was investigated by the 16S rRNA high-throughput sequencing. Results: The analyses of α and β diversity indicated that the richness and diversity of gut microbiota in the T2DM mice decreased and the community structure was significantly different from the normal mice. Hsp–Cu(II) increased the abundances of the beneficial bacteria (Lactobacillus,Ligilactobacillus,Romboutsia,Faecalibaculum, andDubosiella), and decreased the amounts of the harmful bacteria (Desulfobacterota,Corynebacterium, andDesulfovibrio) and the ratio ofFirmicutes/Bacteroidetes(from 44.5 to 5.8) in the T2DM mice, which was beneficial for regulating the composition of intestinal microbiota. The linear discriminant analysis effect size analysis showed that the intervention of Hsp–Cu(II) made the short-chain fatty acid (SCFA) producers (o_Lachnospirales,f_Lachnospiraceae,g_Faecalibaculum,g_Romboutsia, andg_Turicibacter) and the lactic acid bacteria producers (f_Lactobacillaceaeando_Lactobacillales) highly enriched, and the production of its metabolite SCFAs (acetic acid, propionic acid, butyric acid, and valeric acid) were increased in a dose-dependent manner, promoting the SCFA metabolism. Conclusions: Hsp–Cu(II) may improve glucose metabolic disorders and alleviate T2DM by modulating gut microbiota composition, promoting probiotics proliferation and SCFAs production, restoring intestinal barrier integrity, and suppressing local inflammation. These research findings may provide a theoretical basis for developing Hsp–Cu(II) as a new hypoglycemic nutritional supplement, and offer new ideas for the dietary food nutritional regulation to alleviate T2DM.

Hesperetin is the aglycone of hesperidin and is widely found in the Rutaceae plants and herbal medicines. It exhibits antioxidant, detoxifying, anti-inflammatory, and antimicrobial properties, similar to hesperidin. It has also shown potential in the regulation of certain diseases. In order to detect hesperetin in complex matrix samples such as citrus and herbal, we developed an anti-hesperetin monoclonal antibody and established an indirect competitive enzyme-linked immunosorbent assay (icELISA). The half maximal inhibitory concentration (IC 50 ) was determined to be 2.03 ng/mL, the detection range was 0.39–12.73 ng/mL. In practical sample testing, the concentration of hesperidin measured by icELISA is consistent with the result of UPLC-MS/MS, and the correlation coefficient (R 2 ) is 0.97. These results showed that the established method has good accuracy, reproducibility and broad application prospects, and can be used for the detection of hesperetin in complex matrix samples (such as citrus and herbal samples).

Drug-drug cocrystals can improve the physical and pharmaceutical properties of drugs and have been widely studied by many researchers. A new pharmaceutical cocrystal [C 16 H 18 FN 3 O 3 ][C 16 H 14 O 6 ] · 2(C 2 H 5 OH) (HSP-NOR) was synthesized by hesperetin (HSP) and norfloxacin (NOR) with the aim of improving the antibacterial activity and physicochemical properties. The single crystal structure shows that the asymmetric unit comprises both hesperetin and norfloxacin, as well as two ethanol molecules, in which norfloxacin exists as a zwitterion. The cocrystal formation improves the solubilities of hesperetin and norfloxacin in pure water. Preliminary antibacterial experiments showed that the antibacterial activities of the cocrystal on Streptococcus pneumoniae , Escherichia coli , Shigella dysenteriae , and Pseudomonas aeruginosa were stronger than those of norfloxacin. The in vivo study of acute toxicity showed that the minimum lethal dose of the cocrystal to mice was >3000 mg kg -1 BW, and there were no signs of toxicity after oral administration at this dosage.

Ulcerative colitis (UC) is a chronic relapsing inflammatory bowel disease with a rising global incidence rate, which poses a significant challenge to clinical management due to the limited efficacy and adverse reactions of current treatments. To overcome these limitations, we engineered Hes-Zn@IN, a novel core-shell nanoparticle system combining a hesperetin‑zinc (Hes Zn) metal-polyphenol coordination core encapsulated in an inulin (IN) polysaccharide shell. This nanotherapeutic platform exhibits synergistic therapeutic benefits: superior antioxidant capacity through metal-polyphenol coordination, demonstrating a 38% reduction in ROS and a 47% decrease in NO production in LPS-activated macrophages; robust immunomodulatory effects, significantly lowering pro-inflammatory cytokines (46% reduction in TNF-α, 42% in IL-6) while increasing anti-inflammatory IL-10 by 78%; a pH-dependent release profile enabling 1.85 times greater drug accumulation in inflamed colon tissue. In murine DSS-induced colitis models, oral administration of Hes-Zn@IN produced remarkable therapeutic outcomes, including 19.2% improvement in colon length, restoration of ZO-1 tight junction protein expression, and substantial mitigation of histopathological inflammation. Our findings establish this inulin-armored metal-polyphenol hybrid as a breakthrough nanomedicine that synergistically targets colonic redox-immune remodeling and barrier repair, presenting a comprehensive therapeutic approach for UC management.
