Haloalkane dehalogenase DhaA catalyzes the hydrolytic cleavage of carbon-halogen bonds and produces alcohol, a proton and a halide. However, DhaA suffers from the poor environmental stability, such as sensitivity to high temperature, low pH, hypersaline and organic solvent. In order to improve the environmental stability of DhaA, DhaA was covalently conjugated with inulin, a hydrophilic polysaccharide in the present study. Each DhaA was averagely conjugated with 7∼8 inulin molecules. The conjugated inulin could form a hydration layer around DhaA, which increased the conformational rigidity and decreased the entropy of the enzyme. Conjugation of inulin maintained 75.5 % of the enzymatic activity of DhaA and slightly altered the structure of DhaA. As compared with DhaA, the conjugate (inu-DhaA) showed slightly different kinetic parameters (K m of 2.9 μmol/L and K cat of 1.0 s −1 ). Inulin conjugation could delay the structural unfolding and/or slow the protonation process of DhaA under undesirable environment, including the long-term storage, low pH, hypersaline and organic solvent stability. As a result, the environmental stability of DhaA was markedly increased upon conjugation with inulin. Thus, inulin conjugation was an effective approach to enhance the environmental stability of DhaA.

Individuals diagnosed with inflammatory bowel disease (IBD) frequently encounter dysbiosis of the intestinal microbiota and are predisposed to psychiatric comorbidities, such as depression and anxiety. The development of oral therapeutics that concurrently attenuate intestinal inflammation, modulate gut microbiota, and ameliorate psychiatric symptoms is imperative for effective IBD management. This study introduced an orally administered Inulin gel incorporating magnesium diboride nanosheets (MgB2@Inulin gel). The hydrolysis of MgB2 nanosheets could generate reductive hydrogen gas (H2), which is critical for scavenging excess reactive oxygen/nitrogen species (RONS), and boron ions that neutralize harmful lipopolysaccharides (LPS). Prebiotic Inulin could effectively protect MgB2 nanosheets from gastric acid degradation, ensuring their persistence in the intestinal tract, where it modulated the gut microbiota and preserved intestinal barrier integrity. Notably, MgB2@Inulin gel markedly alleviated anxiety and depressive behaviors in the model of colitis by regulating the gut–brain axis. Thus, this work underscores the pivotal role of the gut–brain axis in the therapeutic approach to IBD and its associated psychiatric manifestations, proposing a straightforward, safe, and efficacious oral hydrogen treatment strategy for IBD and concurrent mental health disorders.

Pancreatic islet surface engineering has been proposed as an “easy-to-adopt” approach to enhance post-transplantation islet engraftment for treatment against diabetes. Inulin is an FDA-approved dietary prebiotic with reported anti-diabetic, anti-inflammatory, anti-hypoxic and pro-angiogenic properties. We therefore assessed whether inulin would be a viable option for islet surface engineering. Inulin was oxidized to generate inulin-CHO, which would bind to the cell membrane via covalent bond formation between –CHO and –NH2 across the islet cell membrane. In vitro assessments demonstrated enhanced islet viability and better glucose-induced insulin secretion from inulin-coated (5 mg mL−1) islets, which was accompanied by enhanced revascularization, shown as significantly enhanced tube formation and branching of islet endothelial MS1 cells following co-culture with inulin-coated islets. Reduction of cytokine-induced cell death was also observed from inulin-coated islets following exposure to pro-inflammatory cytokine LPS. LPS-induced ROS production was significantly dampened by 44% in inulin-coated islets when compared to controls. RNA-seq analysis of inulin-coated and control islets identified expression alterations of genes involved in islet function, vascular formation and immune regulation, supporting the positive impact of inulin on islet preservation. In vivo examination using streptozotocin (STZ)-induced hyperglycemic mice further showed moderately better maintained plasma glucose levels in mice received transplantation of inulin-coated islets, attributable to ameliorated CD45+ immune cell infiltration and improved in vivo graft vascularization. We therefore propose islet surface engineering with inulin as safe and beneficial, and further assessment is required to verify its applicability in clinical islet transplantation.

Electronic skin is a flexible, stretchable smart material widely used in fields such as healthcare, robotics, and smart wearables. However, electronic skin still faces multiple challenges in sensitivity, conductivity, biocompatibility, and data processing. In this study, we report a multifunctional hydrogel, named PIPC, prepared by semi-interpenetrating networks of acrylamide and inulin, combined with ionic interactions. The PIPC hydrogel exhibits adhesion, conductivity (≈5.2 S/m), high fracture strength (≈60 kPa), and stretchability (with a maximum strain of up to 1552 %). Notably, its gauge factor (GF) reaches 5.54, demonstrating exceptional strain sensitivity. The introduction of inulin significantly enhances the biocompatibility of the PIPC hydrogel, making it more suitable for contact with the human body. Using PIPC e-skin, we have successfully realized human-machine interaction systems such as real-time control of mechanical hand, game interaction, and emergency signal sending. This study highlights the tremendous potential of PIPC hydrogel e-skin in biomedical monitoring, smart sensing, and human-machine interaction technologies.

Background Inflammatory bowel disease (IBD) is a chronic inflammatory bowel disease with no clinical cure. Excessive production of reactive oxygen species (ROS) at the inflammatory sites leads to the onset and progression of IBD. And the current non-invasive imaging methods are not ideal for the diagnosis and monitoring of IBD.Methods Herein, we developed inulin (IN)-coated cerium oxide nanoparticles (CeO2@IN NPs) for treatment and monitoring of IBD guided by computed tomography (CT). The physicochemical properties, ROS scavenging ability and CT imaging capabilities of CeO2@IN were investigated in vitro. Moreover, the therapeutic and targeted inflammation imaging effects of CeO2@IN were validated in dextran sulfate sodium (DSS)-induced colitis model. Results CeO2@IN with catalase (CAT) and superoxide dismutase (SOD) capabilities effectively scavenged ROS, thus protecting the cells against oxidative stress. In colitis model mice, orally administered CeO2@IN successfully traversed the gastrointestinal tract to reach the colon under the protection of IN, and effectively reduced intestinal inflammation, thereby maintaining the intestinal epithelial integrity. Notably, CeO2@IN performed better than conventional CT contrast agents for gastrointestinal tract imaging, particularly in detecting the inflamed areas in the colon. In addition, CeO2@IN exhibited excellent biocompatibility in vitro and in vivo. Conclusion The study provided a novel integrated diagnostic and therapeutic tool for the treatment and monitoring of IBD, presenting great potential as a clinical application for IBD.

Cinnamaldehyde as an antioxidant was encapsulated in inulin-modified nanoliposomes in order to improve its physical and antioxidant stability. The microstructure, particle size and volume distribution of cinnamaldehyde liposomes were characterized by atomic force microscopy (AFM) and dynamic light scattering (DLS). The particle size and polydispersion index (PDI) values of the inulin modified liposomes were 72.52 ± 0.71 nm and 0.223 ± 0.031, respectively. The results showed that the liposomes after surface modification with inulin remained spherical. Raman and Fourier transform infrared (FTIR) spectra analysis showed that hydrogen bonds were formed between the inulin and the liposome membrane. Inulin binding also restricted the freedom of movement of lipid molecules and enhanced the order of the hydrophobic core of the membrane and the polar headgroup region in lipid molecules. Therefore, the addition of different concentrations of inulin influenced the permeability of the liposome bilayer membrane. However, when inulin was excessive, the capacity of the bilayer membrane to load the cinnamaldehyde was reduced, and the stability of the system was reduced. Additionally, the encapsulation efficiency (EE) and retention rate (RR) of cinnamaldehyde from inulin-modified liposomes during storage were determined. The EE value of the inulin modified liposomes was 70.71 ± 0.53%. The liposomes with 1.5% inulin concentration had the highest retention rate (RR) and the smallest particle size during storage at 4 °C. The addition of inulin also enhanced the thermal stability of the liposomes. Based on the results, the surface modification improved the oxidation stability of liposomes, especially the DPPH scavenging ability. In conclusion, these results might help to develop inulin as a potential candidate for the effective modification of the surface of liposomes and provide data and conclusions for it.Keywords:liposomes;surface modification;inulin;physical stability

Colorectal cancer is one of the most common types of tumor in the world. Here we developed a lipoic acid esterified polysaccharide (inulin) delivery system for tanshinone IIA to treat colorectal cancer in vitro . The release of tanshinone IIA in the system was highly responsive to glutathione , which is commonly abundant in cancer cells. In addition, this drug delivery system was proliferative to Bifidobacterium longum , the common inhabitant of human intestine. Thus, this strategy might be useful to improve colon cancer therapy efficacy of anticancer drugs and meanwhile promote the growth of beneficial commensal flora in the gut.

A new biopolymer–silica hybrid material consisting of inulin and tetraethoxysilane (TEOS) for use as an adsorbent was successfully synthesized via the sol–gel method in acidic conditions. The hydrolysis and condensation processes were attained in water/ethanol solution. Three molar ratios of inulin:TEOS (1:1, 1:2, and 2:1) were prepared and dried at various temperatures (50, 60, and 70 °C). The optimized molar ratio of 2:1 with a drying temperature of 70 °C was found to obtain the best morphology and characteristics for absorbent properties. Fourier transform infrared spectroscopy (FTIR) analysis showed a strong interaction between inulin and TEOS, which was also observed using energy dispersive X-ray spectroscopy (EDX). Field emission scanning electron microscopy (FESEM) images revealed the presence of nanoparticles on the rough surface of the hybrid sol–gel. X-ray diffractometer (XRD) analysis showed the amorphous state of the silica network where the inulin existed as an anhydrous crystalline phase. Brunauer–Emmet–Teller (BET) analysis confirmed that the composite was mesoporous, with 17.69 m2/g surface area and 34.06 Å pore size. According to thermogravimetric analysis (TGA) results, the hybrid inulin-TEOS adsorbent was thermally stable under a temperature of 200 °C.Keywords:adsorbent;biopolymer–silica hybrid;hybrid sol–gel;sol–gel process

Fluorescent labeling of long-chain inulin (FXL) is essential for tracking its spatiotemporal dynamics in gastrointestinal systems, yet conventional methods risk altering its native properties. This study systematically compared three labeling strategies: FITC-DB (dibutyltin dilaurate-catalyzed hydroxyl conjugation), FITC-RA (reductive amination), and FA (fluoresceinamine coupling). Multimodal characterization revealed that FITC-DB achieved the highest substitution degree (DS = 0.42 at 50 mg FITC), though fluorescence quenching occurred at elevated loadings. FITC-RA induced significant aggregation (peak size at 50 mg), while FA labeling exhibited minimal structural perturbation but low efficiency (DS < 0.20). Spectroscopic analyses confirmed covalent thiourea bond formation (1720 cm⁻¹ FTIR peak) and aggregation-dependent optical shifts. Crucially, FITC-DB at 50 mg balanced optimal labeling efficiency, colloidal stability (highest zeta potential), and preserved macromolecular architecture. These findings establish design principles for high-fidelity FXL probes in biological tracking applications.

The pervasive contamination by Di(2-ethylhexyl) phthalate (DEHP) and nanoplastics (NPs) poses a significant threat to reproductive health, yet the combined effects of these pollutants on endometrial receptivity remain poorly understood. Our study investigates the impact of chronic DEHP and NPs co-exposure on endometrial ciliary function in mice and explores the potential protective role of dietary inulin. Utilizing a multifaceted approach—including transcriptomic analysis, immunohistochemistry, scanning electron microscopy, and immunofluorescence—we reveal that DEHP and NPs synergistically impair endometrial cilia, crucial for embryo implantation. Remarkably, inulin supplementation restores ciliary structure and function, mitigating the detrimental effects of these pollutants. Our findings suggest that dietary interventions could offer a practical strategy to counteract the reproductive toxicity of environmental contaminants, providing new insights into maintaining reproductive health in polluted environments. This research underscores the urgency of addressing environmental pollutants and highlights the potential of inulin as a protective agent against reproductive toxicity.

Fructooligosaccharides (FOS) and inulin are beneficial for human health. However, their benefits differ in individuals who consume prebiotics. Several factors contribute to this variation, including host genetics and differences in the gut microbiota. Bifidobacterium and Bacteroides are strong carbohydrate-utilizing bacteria in the gut, and the level of the Bacteroides/Bifidobacterium (Ba/Bi) ratio in the gut is closely related to the body's ability to utilize prebiotics. However, how to select the type of prebiotics more beneficial for populations with specific Ba/Bi backgrounds and the underlying regulatory mechanisms remain unclear. Here, we explored the dynamics of the gut microbiota and metabolic functions during the in vitro fermentation of FOS and inulin in two different groups: Bacteroides/Bifidobacterium high (H) and Bacteroides/Bifidobacterium low (L). This study revealed that the baseline Ba/Bi ratio had a greater impact on the gut microbiota compared to prebiotic species. Noticeable differences were observed between the two groups after prebiotic intervention, with the H group being more likely to benefit from the prebiotic intervention. Compared to the L group, the H group exhibited significantly higher microbial α-diversity; the co-abundance response group 1 (CARG1) members Ruminococcus gnavus and Blautia involved in the synthesis of propionic and butyric acids increased significantly, the abundance of pathogenic bacteria such as Escherichia Shigella decreased significantly, and the ability to degrade carbohydrates and synthesize fatty acids was greater. Regression modeling showed that the key microbiota could predict the short-chain fatty acid (SCFA) levels, with FOS associated with the ecological roles of CARG2 and CARG7 and inulin associated with CARG4, which provides the basis for the use of prebiotics in nutritional applications and the stratification of populations based on pertinent microbiota profiles to explain the incongruent health effects in human intervention studies.

The gut microbiota plays a significant role in the pathogenesis and remission of inflammatory bowel disease. However, conventional antibiotic therapies may alter microbial ecology and lead to dysbiosis of the gut microbiome, which greatly limits therapeutic efficacy. To address this challenge, novel nanomicelles that couple inulin with levofloxacin via disulfide bonds for the treatment of salmonellosis were developed in this study. Owing to their H 2 S-responsiveness, the nanomicelles can target the inflamed colon and rapidly release levofloxacin to selectively fight against enteric pathogens. Moreover, the embedded inulin can serve as prebiotic fiber to increase the amount of Bifidobacteria and Lactobacilli in mice with salmonellosis, thus maintaining the intestinal mechanical barrier and regulating the balance of the intestinal flora. Therefore, multifunctional nanomicelles had a better curative effect than pure levofloxacin on ameliorating inflammation in vivo . The pathogen-targeted glycovesicle represents a promising drug delivery platform to maximize the efficacy of antibacterial drugs for the treatment of inflammatory bowel disease.
