Insight into the mechanism of synergy of ultrasound and high pressure homogenization on the emulsion stability and interfacial behaviors of α-lactalbumin with mogroside V

Jinzhe Li,QiuWan Jiang,Ruyi Zhang,Shiyi Guo,Zhanmei Jiang,Juncai Hou

Due to uneven distribution amino acid residues of α-lactalbumin (αLA) at oil-water interface, some measurement should be adopted to improve its interfacial behavior and emulsion stability. Therefore, this study focused on the mechanism of synergy of ultrasound and high pressure homogenization on the emulsion stability and interfacial behaviors of α-lactalbumin with mogroside V (MGV). Four treatments were adopted: ultrasound (US), high pressure homogenization (HPH) and combined treatment (US-HPH and HPH-US). Primarily, emulsion stability of αLA/MGV was analyzed. US-HPH or HPH-US exhibited better emulsion stability than US or HPH. Especially, HPH-US-αLA/MGV emulsion was not stratified at the given time (45 d), which was the most stable emulsion. Moreover, the micromorphology of emulsion showed that MGV and UH/HPH treatment suppressed the aggregation of αLA emulsion droplet. In addition, interfacial adsorption mass and viscoelasticity of physical treated αLA was improved with MGV in the order of HPH-US > US-HPH > US > HPH, specifically, frequency shift (ΔF) and dissipation shift (ΔD) of HPH-US-αLA/MGV was 42.2% and 32.5% higher than that without MGV. This finding may provide a potential approach for obtaining natural and high-efficient protein-based emulsifier in food industry.

Enzymatic hydrolyzation of mogrosides in Luo Han Guo extract by NKA-adsorbed snailase improves its sensory profile

Yongxin Su,Zhenlin Li,Yinghui Zhao,Yijun Chen,Chen Luo,Xuri Wu

Extracts of Siraitia grosvenorii (Swingle), in Chinese known as Luo Han Guo (LHG), is authorized for use as a natural sweetener . LHG is rich in mogroside V that contains five glucoses, but also contains mogroside IIIE and analogues with fewer than three glucose units that cause an unpleasant aftertaste, limiting the use of the extract. Snailase was applied here to convert mogroside V in LHG extract in favor of siamenoside I formation, the sweetest mogroside with a taste similar to sucrose . For application, snailase was immobilized by adsorption to NKA (a macroporous resin), resulting in 10.9 U per g of adsorbed protein. Reuse of the NKA-adsorbed snailase was demonstrated for four cycles, and a continuous production of improved LHG extract at a 0.5 L scale had a productivity of 68.4 g/(L⋅day). The resulting product containing over 50% siamenoside I displayed an improved taste profile with satisfying safety toward HEK293T cells.

Yisa Liu,Juan Wang,Xiao Guan,Dan Yu,Mengjie Huangfu,Tong Dou,Luwei Zhou,Lin Wang,Guoxiang Liu,Xiaojuan Li,Zhaokun Zhai,Mengjie Han,Haiping Liu,Xu Chen

Background Mogroside V, the main ingredient of Siraitia grosvenorii , has been proved to have therapeutic effects on pulmonary diseases . The specific mechanism still remains to be clarified, which hinders the potence of its medicinal value. Purpose Serum and lung metabolomics based on LC-MS analysis were applied to explore the mechanism of mogroside V against lung inflammation. Method In this study, balb/c mice were divided into control, model, mogeoside V and SH groups. We evaluated the protective effects of mogroside V on lung inflammation in asthmatic mice. Suhuang Zhike Jiaonang was used as positive drug . Metabolic profiles of serum and lung samples of mice in control, model and mogroside V groups were analyzed by LC-MS. Results Administration of mogroside V effectively relieved the expression of biochemical cytokines and lung inflammatory infiltration of asthmatic mice caused by ovalbumin (OVA). And visceral index of mice treated with mogroside V was close to control group. These results indicated that mogroside V ameliorated OVA-induced lung inflammation. LC-MS based metabolomics analysis demonstrated 6 main pathways in asthmatic mice including Vitamin B6 metabolism, Taurine and hypotaurine metabolism, Ascorbate and aldarate metabolism, Histidine metabolism, Pentose and glucuronate interconversions, Citrate cycle (TCA cycle) were regulated after using mogroside V. Conclusion The study firstly elucidates the metabolic pathways regulated by mogroside V on lung inflammation through metabolomics, providing a theoretical basis for more sufficient utilization and compatibility of mogroside V.

The metabolism of a natural product mogroside V, in healthy and type 2 diabetic rats

Guisheng Zhou,Yulong Zhang,Yang Li,Mengyue Wang,Xiaobo Li

Mogroside V, a natural compound isolated from the fruits of Siraitia grosvenorii (Swingle), is a promising candidate for anti-diabetic activity. The present study aims to develop a simple and practical strategy for comparing the in vivo metabolite profiling of mogroside V in healthy and type 2 diabetic (T2D) model rats. In this paper, a highly sensitive and rapid ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS) with MetaboLynx™ software combined with mass defect filtering (MDF) method was established and successfully applied to detect and identify the metabolites in plasma, urine, bile and feces samples of healthy and model rats administrated with mogroside V. The distribution of metabolites in plasma, bile, urine and feces of healthy and model rats had obvious differences. A total of 23 metabolites were observed in healthy rats while 26 metabolites were detected in model rats. The results indicated that dehydrogenation , deoxidation , oxidation and isomerization were the major metabolic transformations of mogroside V. Additionally, it was noticed that the peak areas of metabolites in T2D rat plasma samples were much larger than those of metabolites in healthy rat plasma sample, whereas in T2D rat urine samples they were remarkably smaller compared with healthy rat urine sample. These high blood concentrations of metabolites might be beneficial for the treatment of T2D. The results of this study are valuable and important in understanding the metabolic process and therapeutic mechanism of mogroside V.

Mogroside V reduced the excessive endoplasmic reticulum stress and mitigated the Ulcerative colitis induced by dextran sulfate sodium in mice

Tan Yue-Rong,Shen Si-Yang,Li Xin-Yi,Yi Peng-Fei,Fu Ben-Dong,Peng Lu-Yuan

Ulcerative colitis (UC) is an idiopathic, chronic inflammatory condition of the colon, characterized by repeated attacks, a lack of effective treatment options, and significant physical and mental health complications for patients. The endoplasmic reticulum (ER) is a vital intracellular organelle in maintaining cellular homeostasis. Endoplasmic reticulum stress (ERS) is induced when the body is exposed to adverse external stimuli. Numerous studies have shown that ERS-induced apoptosis plays a vital role in the pathogenesis of UC. Mogroside V (MV), an active ingredient of Monk fruit, has demonstrated excellent anti-inflammatory and antioxidant effects. In this study, we investigated the therapeutic effects of MV on dextran sulfate sodium (DSS)-induced UC and its potential mechanisms based on ERS. The results showed that MV exerted a protective effect against DSS-induced UC in mice as reflected by reduced DAI scores, increased colon length, reduced histological scores of the colon, and levels of pro-inflammatory cytokines, as well as decreased intestinal permeability. In addition, the expression of ERS pathway including BIP, PERK, eIF2α, ATF4, CHOP, as well as the apoptosis-related protein including Caspase-12, Bcl-2 and Bax, was found to be elevated in UC. However, MV treatment significantly inhibited the UC and reversed the expression of inflammation signaling pathway including ERS and ERS-induced apoptosis. Additionally, the addition of tunicamycin (Tm), an ERS activator, significantly weakened the therapeutic effect of MV on UC in mice. These findings suggest that MV may be a therapeutic agent for the treatment of DSS-induced UC by inhibiting the activation of the ERS-apoptosis pathway, and may provide a novel avenue for the treatment of UC.

Modulation of Gut Microbiota Composition and Short-Chain Fatty Acid Synthesis by Mogroside V in an In Vitro Incubation System

Ruiming Xiao,Wanci Liao,Guangjuan Luo,Zhenning Qin,Shuangyan Han,Ying Lin

Mogroside V (MV), a sweetener, is one of the major components inSiraitia grosvenorii. In our research, after in vitro incubation with MV for 24 h, the human gut microbiota diversity changed, with an enrichment of the genera Bacteroides, Lactobacillus, Prevotella, Megasphaera, and Olsenella and the inhibition of Clostridium XlVa, Dorea, and Desulfovibrio. Moreover, the synthesis of short-chain fatty acids, such as acetate, propionate, and butyrate, was increased by gut microbiota. According to ultraperformance liquid chromatography–mass spectrometry (UPLC-MS) analysis, MV was decomposed into secondary mogrosides, such as mogroside II/I and mogrol, by gut microbiota. Enhanced antioxidant abilities of the metabolites were found in the broth. The results suggested that MV, as a potential prebiotic, could benefit human health through its interaction with gut microbiota.

Insight into binding mechanism between three whey proteins and mogroside V by multi-spectroscopic and silico methods: Impacts on structure and foaming properties

Jinzhe Li,Yuxuan Huang,Wenjuan Zhang,Akhunzada Bilawal,Narantuya Sukhbaatar,Batsukh Tsembeltsogt,Zhanmei Jiang,Zhishen Mu

In this work, the interaction mechanism, structure and foaming properties between three whey proteins (α-lactalbumin (ALa), β-lactoglobulin (BLg) and bovine serum albumin (BSA)) and Mogroside V (Mog) at pH 7.0 were systematically formulated via multi-spectroscopic analysis and silico methods. Mog quenched the fluorescence of three whey proteins in a static mode. Mog had greater binding affinity for ALa than that for BLg or BSA. Hydrogen bonds, van der Waals force and hydrophobic interaction acted as the primary driven forces in ALa/BLg/BSA-Mog system. Moreover, content of α-helix in ALa and β-sheet in BLg decreased after Mog addition, whereas there was no significant change in the secondary structure of BSA. In addition, hydrophobic crevice in ALa, “bottom of β-barrel” in BLg and interfacial region between two domains in BSA were the binding areas for Mog using molecular docking. Molecular dynamics (MD) simulation further demonstrated that the complex of ALa and Mog possessed the lowest binding free energy and most stable state. Furthermore, compared with single proteins, the foaming ability of protein/Mog complexes were improved. Especially, Ala-Mog attained the highest increase rate. Bubble morphology presented that three whey proteins containing Mog possessed compacter, smaller and even bubbles. This research could establish theoretical basis for the binding mechanism between different food proteins and low weight surfactants. Besides, whey protein-saponin complexes could be served as the novel food-grade foaming agents applied in food industry.
