Split-aptamer mediated regenerable temperature-sensitive electrochemical biosensor for the detection of tumour exosomes

Dongfang Liu,Jinlu Tang,Hui Xu,Kun Yuan,Aaron Albert Aryee,Cuijie Zhang,Hongmin Meng,Lingbo Qu,Zhaohui Li

In this paper, a s plit-aptamer m ediated r egenerable t emperature-sensitive (SMRT) electrochemical biosensor was constructed for the detection of exosomes. The split-aptamer used in this SMRT biosensor was composed of two fragments, one of which was immobilized on the surface of an electrode via sulfhydryl groups and named split-a and the other was labelled with methylene blue and named split-b. The two fragments could form sandwich structures at the electrode surface via target-induced self-assembly in the presence of target exosomes at 4 °C in PBS, and then realizing the detection of exosomes via voltammetry . In addition, due to the temperature sensitivity of the split-aptamer, the electrode could be regenerated through temperature-induced disassembly of the sandwich structures. Consequently, the SMRT biosensor realized sensitive and specific analysis of target exosomes with a limit of detection of 1.5 × 10 6 particles/mL and could be quickly and easily regenerated by washing with PBS at 37 °C for 30 s without any additives. This is the first study on the construction of a reproducible electrochemical biosensor using a split-aptamer for the specific detection of tumour exosomes, and may provide an innovative strategy for the economical and efficient design of regenerable electrochemical biosensors.

A split aptamer-based biosensor for visual detection of leuco-malachite green

Tianci Xu,Yan Jiang,Fangran Jia,Runze Zhang,Boyi Shao,Qianwen Li,Chengze Li,Han Gao,Hongrui Han,Chenchen Zhang,Yongjie Yang

Leuco-malachite green (LMG), a metabolite of malachite green (MG), exhibits stronger bioaccumulation and higher toxicity to humans. Therefore, development of a rapid and sensitive detection method for LMG is of great significance. In this study, LMG-specific split aptamers (SPALMG) with a dissociation constant (Kd) of 1.538 ± 0.1261 μM and enhanced affinity compared to the parent aptamer are generated using A5b-12C, a high-affinity variant, as the parent aptamer for LMG detection. Subsequently, a colorimetric aptasensor is developed, employing thiol-modified SPAlmg as probes and gold nanoparticles (AuNPs) as signal indicators. In the presence of LMG, the SPAlmg preferentially bind to LMG to form a ternary complex, causing the AuNPs to aggregate and induce a color change from red to blue. The sensor exhibits high sensitivity and specificity, with a limit of detection (LOD) of 46 nM, a linear range of 1–7 μM, and good recoveries of 96–113%. Compared with other studies, a more excellent LOD is achieved. This work provides a novel affinity probe that holds great promise for the development of sandwich-type sensors for LMG detection.

Ratiometric fluorescent detection of pesticide based on split aptamer and magnetic separation

Ying Wang,Fuyan Zhu,Li Yin,Guangxu Qu,Dik-Lung Ma,Chung-Hang Leung,Lihua Lu

Pesticide residues have seriously threatened human health and affected the sustainable development of the environment. It is pivotal to establish an efficient detection platform for pesticide assays. A novel ratiometric fluorescent aptasensor for sensitive determination of pesticide has been successfully established. Carbon dots (CDs) are used as the reference emitter, while the T-rich single-stranded DNA templated copper nanoparticles (CuNPs) provide the increasing signal. More precisely, CDs/magnetic beads (MBs) combined with split aptamer are used in this detection platform to present a label-free and enzyme-free isocarbophos (ICP) detection with high sensitivity and specificity. In the presence of ICP, a sandwich-typed complex is formed which attaches a T-rich DNA. After magnetic separation and addition of Cu 2+ /ascorbic acid (AA), the CuNPs are generated and the strong red luminescent appears. Without ICP, just blue emission from CDs exits. The developed ratiometric aptasensor offers an excellent ICP detection in the linear range of 3–200 nM with a detection limit of 0.81 nM. It provides new ways for the assay of pesticides in agricultural products or environmental samples.

Split aptamer-regulated AuNF nanozymes for colorimetric and electrochemical dual-readout kanamycin biosensing

Hailiang Qin,Xinyan Guo,Yao Zhao,Meng Luo,Minqiao Feng,Haoqian Xue,Pengbo Zhang,Meilin Zhou,Na Wang,Shuxia Xu

Kanamycin (Kana) residues released into aquatic environments due to its extensive use have raised increasing concerns because of their potential risks to ecosystems and human health. In this work, we report a label-free split aptamer-modified gold nanoflower (AuNF)-based aptasensing platform with colorimetric and electrochemical readouts for the determination of Kana in water. Two thiolated split aptamer fragments (SPA1 and SPA2) were immobilized on AuNFs through Au–S bonds. The flower-like AuNFs provided a high-surface-area scaffold for aptamer loading, while the DNA modification further enhanced the peroxidase-like activity of AuNFs toward TMB oxidation. In the presence of Kana, target-induced assembly of SPA1 and SPA2 promoted the aggregation of AuNFs, which inhibited their catalytic activity toward TMB oxidation, thereby reducing oxTMB formation and the resulting colorimetric and electrochemical signals. Based on this signal-off mechanism, the platform enabled quantitative detection of Kana with linear ranges of 100–3000 nM for colorimetry and 50–2000 nM for chronoamperometry, with limits of detection of 0.51 and 0.074 nM, respectively. The method also showed satisfactory selectivity, reproducibility, and storage stability. In spiked environmental water samples, recoveries ranged from 87.7% to 106.2% with relative standard deviations below 5.0%, and the electrochemical results were in acceptable agreement with those obtained by HPLC–MS. This study provides a simple and effective dual-readout aptasensing strategy for Kana determination in environmental water samples.

Rapid detection of kanamycin using cooperative recognition split aptamer and graphene oxide nanosheets

Ye Hua,Wan Tianxiang,Li Xinfu,Li Chao,He Kuo,Guo Yuanxin

Split aptamers are fragments of the parent aptamer which still possess binding ability to its target. They have shown great potential in the development of aptasensors. In this study, a fluorescence aptasensor was constructed employing the split aptamer as a cooperative recognition molecular probe and Graphene Oxide nanosheets (GO) as a quencher for rapid detection of kanamycin in chilled pork. Under optimal conditions, the proposed biosensor showed a good linear relationship in the range of 0.5–50 nM with the detection limit of 0.36 nM. The recoveries of the actual samples ranged from 97.6 to 103.5%. Furthermore, the test can be completed in 25 min. This study provides good reference for developing simple, economical, and rapid assay for other small molecular targets detection.

DNA tweezers aptasensor with split aptamer for the dual detection of Enrofloxacin and Kanamycin

Zixuan Shang,Pengfei Ma,Imran Mahmood Khan,Ningru Yang,Zhouping Wang

Currently, the technology for detecting antibiotics is developing rapidly, with good sensitivity and selectivity. However, some detection methods still have issues with cumbersome pretreatment and high experimental costs. Herein, A “turn-off” bifunctional DNA tweezer was constructed through the design of DNA sequence and self-assembly technology for the dual detection of Enrofloxacin (ENR) and Kanamycin (KANA) in a food matrix. The operation is simple, which increases the detection speed and reduces the detection cost. The two ends of the bifunctional DNA tweezers are connected with specific split aptamer sequences, and the split aptamer forms a specific ternary complex with the target. The tweezers “clip” ENR and KANA at their respective ends, causing a structural change. Under the optimal experimental conditions, the linear range of detection ENR is 0.2 ng/mL-1000 ng/mL with an LOD of 0.18 ng/mL, the linear range of detection KANA is 0.5 ng/mL-1000 ng/mL with an LOD of 0.36 ng/mL. The bifunctional DNA tweezers involved provide a new idea for the construction of DNA nanomachines, which is also suitable for the recognition and detection of other small molecules.

Highly sensitive electrochemical PDGF-BB sensor based on protein-templated split aptamer ligation reaction

Susu Cui,Fan Wang,Weiwei Yang

Split aptamers have attracted extensive attention in sensing benefit of optimized concise structure and low-cost, but with lower binding affinity and stability. Here, we successfully engineer three pairs of split aptamers for PDGF-BB protein and develop a highly sensitive electrochemical sensor based on protein-templated split aptamer ligation reaction. The protein template directs the reassembly of split aptamers and accelerates the ligation reaction between azide and alkyne groups attached to the split fragments. The ligation yields of RS Split1 and RS Split2 with reserved binding sites are found to be 72.6 ± 1.8 % and 82.5 ± 1.3 %, respectively, while broken binding sites also obtain a ligation yield of 61.5 ± 2.1 % for BS Split3. We utilize the chemically modified split aptamers in place of the “capture” and “detection” antibodies used in ELISA, directly enabling the capture of electrochemically sensitive ligated aptamer to report target binding. This protein-templated ligation strategy improves the stability and sensitivity of the electrochemical sensor, with a low detection limit of 7 pM for PDGF-BB. The saturated electrochemical signal is approximately 3 times that of the non-ligated system. The proposed electrochemical sensor is also successfully applied to 50 % serum due to its excellent selectivity and bio-orthogonality. This strategy avoids the signal loss caused by traditional binding interruption during washing step via converting the ternary complex of aptamer/protein/aptamer to the binary complex of ligated aptamer/protein. This research offers a general sensing platform for other protein biomarkers and, more importantly, does not rely on expensive antibodies.

Split aptamer based sensing platform for adenosine deaminase detection by fluorescence resonance energy transfer

Mengke Wang,Junyang Chen,Dandan Su,Guannan Wang,Xingguang Su

In this paper, a split aptamer based fluorescence resonance energy transfer (FRET) platform was constructed for the determination of adenosine deaminase (ADA) activity by using gold nanoclusters (AuNCs) and gold nanoparticles (AuNPs). A single adenosine triphosphate (ATP) aptamer was split into two fragments (referred to as P1 and P2). P1 was covalently attached to the AuNCs at the 5′ end (P1-AuNCs), and P2 was labeled with AuNPs at the 3′ end (P2-AuNPs). In the presence of ATP, ATP bound with the two fragments with high affinity to link P1-AuNCs and P2-AuNPs together, thus the fluorescence of P1-AuNCs was quenched via FRET from P1-AuNCs to P2-AuNPs. With the addition of ADA, ATP was transformed into inosine triphosphate (ITP), and then P1 and P2 were released to cause the fluorescence recovery of the system. So a split aptamer based FRET platform for ADA detection can be established via the fluorescence intensity change of the system. This platform showed a good linear relationship between the fluorescence intensity and ADA concentration in the range of 2–120 U L −1 , and the limit of detection (LOD) was 0.72 U L −1 . Moreover, the detection of ATP in human serum sample demonstrated the accuracy and applicability of the method for ADA detection in real sample.

Split Aptamer-Driven CRISPR-Cas12a biosensor with signal amplification for quantitative detection of adenosine deaminase

Ping Qiu,Longjian Huang,Jiayi Zhang,Wenbo Li,Xudong Xin,Luchang Chen,Qianli Tang,Kai Zhang,Xianjiu Liao,Jihua Wei

We report the development of a novel split aptamer-driven electrochemical biosensor integrating entropy-driven Signal amplification (EDSA) and CRISPR-Cas12a technology for the highly sensitive and specific detection of adenosine deaminase (ADA). ADA is an important biomarker involved in immune regulation and purine metabolism, and its accurate quantification is critical for diagnosing various diseases. In our system, ADA catalyzes the deamination of adenosine to inosine, resulting in the release of Aptamer1 from a split aptamer structure. The liberated Aptamer1 initiates a strand displacement reaction within a Blocker/Bandage/Scaffold complex, triggering a cascade of entropy-driven amplification cycles that generate a DNA duplex capable of activating CRISPR-Cas12a. Activated Cas12a exhibits trans -cleavage activity, specifically cleaving a ferrocene-labeled single-stranded DNA (ssDNA) probe immobilized on a gold electrode, leading to a measurable decrease in electrochemical signal. The biosensor operates under isothermal and enzyme-free conditions, providing simplicity, rapid detection, and low cost. It achieves a low detection limit of 0.688 U·L −1 with a wide linear range from 1 to 20,000 U·L −1 . The system demonstrates excellent specificity against ADA analogues and common interfering proteins and performs reliably in diluted human serum samples. This strategy represents a versatile and modular platform for point-of-care diagnostics. By altering the aptamer sequence, the biosensor can be adapted for the detection of other clinically relevant proteins and enzymes, contributing to advances in personalized medicine.

An electrochemical quinine detection approach based on small molecule promoted split aptamer click ligation reaction

Xuan Luo,Susu Cui,Weiwei Yang,Yongsheng Yu

Quinine is a natural antimalarial drug that also serves as a food additive; however, excessive consumption can result in harmful side effects. Here, we propose a simple and cost-effective electrochemical strategy for quinine detection based on the small molecule-templated split aptamer click ligation (SMT-SpA-CLR), where the integrity of the aptamer is restored through the template-promoted proximity effect. We first investigate the effects of two splitting sites on the affinity of quinine aptamer and find that the loop of the stem with more Watson-Crick base pairs is more suitable for splitting, which can maintain the affinity to a great extent. The affinity of the Split 1 design ( K d  = 48 μM) is slightly reduced compared to the affinity of the parent aptamer. The stable quinine recognition and its sensitive detection are achieved by integrating the SMT-SpA-CLR with an electrochemical signal output. The linear response range for quinine detection is from 0.05 μM to 10 μM with a low detection limit of 25 nM (at S/N = 3). This strategy provides a novel ideal for the design of small molecule-templated split aptamer reaction and can potentially be expanded for detecting other small molecules and proteins.

A split aptamer-labeled ratiometric fluorescent biosensor for specific detection of adenosine in human urine

You Junhui,You Zhengyi,Xu Xin,Ji Jiangrong,Lu Tian,Xia Yuhong,Wang Lipin,Zhang Liying,Du Shuhu

A dual-emission ratiometric fluorometric aptasensor is presented for highly specific detection of adenosine. An adenosine binding aptamer (ABA) was split into two halves (termed as ABA1 and ABA2). ABA1 was covalently bound to blue-emitting carbon dots (with excitation/emission maxima at 365/440 nm) as responsive fluorophore (referred to as ABA1-CDs). ABA2 was linked to red-emitting silica-coated CdTe quantum dots (with excitation/emission maxima at 365/613 nm) acting as internal reference and referred to as ABA2-QDs@SiO 2 . Upon addition of graphene oxide, the fluorescence of ABA1-CDs is quenched. After subsequent addition of ABA2-QDs@SiO 2 and different amounts of adenosine, the blue fluorescence is recovered and causes a color change from red to royal blue. The method represents a ratiometric turn-on assay for visual, colorimetric and fluorometric determination of adenosine. The limit of detection is as low as 2.4 nM in case of ratiometric fluorometry. The method was successfully applied to the determination of adenosine in (spiked) human urine. Recoveries range from 98.8% to 102%. Adenosine binding aptamer1-carbon dots (ABA1-CDs) can absorb on graphene oxide (GO) via π stacking. This causes fluorescence to be quenched by fluorescence resonance energy transfer (FRET). After addition of ABA2-silica-coated quantum dots (ABA2-QDs@SiO 2 ) and adenosine, binding of adenosine to two pieces of aptamers forms a complex (ABA1-CD/adenosine/ABA2-QD@SiO 2 ) which dissociates from GO. As a result, fluorescence is recovered.

Ratiometric electrochemical aptasensor based on split aptamer and Au-RGO for detection of Aflatoxin M1

Honglin Li,Congcong Du,Ting Guo,Hongyuan Zhou,Ying Zhou,Xinrui Huang,Yu Hao Zhang,Shuo Wang,Xiaozhu Liu,Liang Ma

ABSTRACT A novel ratiometric electrochemical aptasensor based on split aptamer and Au-rGO nanomaterials was proposed to detect Aflatoxin M1 (AFM1). In this work, Au-rGO nanomaterials were coated on the electrode through the electrodeposition method to increase the aptamer enrichment. We split the aptamer of AFM1 into 2 sequences (S1 and S2), where sequence S1 was immobilized on the electrode due to the Au-S bond, whereas sequence S2 was tagged with methylene blue (MB) and acted as a response signal. A complementary strand to S1 (CS1) labeled with ferrocene (Fc) was introduced as another reporter. In the presence of AFM1, CS1 was released from the electrode surface due to the formation of the S1-AFM1-S2 complex, leading to a decrease in Fc and an increase in MB signal. The developed ratiometric aptasensor exhibited a linear range of 0.03 μg L −1 to 2.00 μg L −1 , with a detection limit of 0.015 μg L −1 for AFM1 detection. The ratiometric aptasensor also showed a linear relationship from 0.2 μg L −1 to 1.00 μg L −1 , with a detection limit of 0.05 μg L −1 in natural milk after sample pretreatment, indicating the successful application of the developed ratiometric aptasensor. Our proposed strategy provides a new way to construct aptasensors with high sensitivity and selectivity.
