4.将冻存管中的细胞轻轻移到含有10mL已预热BHK无血清培养基的离心管中;
5. 170g-190g(约800rpm ~ 1000rpm)离心5 分钟,丢弃上清液并将细胞重悬于10 ~ 30mL 新鲜预热的培养基中,调节活细胞密度至1×10⁶ cells/mL mL;
6.取0.5mL 细胞悬液,用细胞计数仪分析活细胞密度(×10⁶ cells /mL)和细胞活率(%)。
7.如果细胞活率 > 90%,将细胞置于规定的环境条件下进行细胞培养。
细胞传代
1.将BHK无血清培养基放入37℃ 条件下预热20-30 min;
2.取活细胞密度 ≥ 7×10⁶ cells/mL 、细胞活率 ≥ 90% 、处于对数生长期中期的细胞进行传代;
3.根据1×10⁶ cells/mL 的接种密度和传代体积,计算所需种子液量;

BHK Serum-Free Medium is a serum-free medium formulated without serum, proteins, or any animal-derived components. It is prepared with high-quality raw materials to ensure high product quality and stability. The culture process does not require serum supplementation, making it suitable for high‑density suspension culture of BHK cells. It supports rapid BHK cell expansion, high‑density culture, and viral vaccine production under a completely serum‑free and protein‑free system.
Applications
BHK Serum‑Free Medium is suitable for suspension adaptation and large‑scale serum‑free suspension culture of BHK cells. It can be used in the research, development, and production of biologics such as rabies vaccines, supporting processes from R&D to industrial scale. However, it is not intended for direct use in humans or as a pharmaceutical product.
Cell Recovery
1.Pre‑warm BHK Serum‑Free Medium in a 37°C water bath.
2.Spray the medium bottle with 75% alcohol and place it in a biosafety cabinet.
3.Thaw one vial of cells by gently swirling it in a 37°C water bath for 1 minute.
4.Carefully transfer the cells from the cryovial to a centrifuge tube containing 10 mL of pre‑warmed BHK Serum‑Free Medium.
5.Centrifuge at 170–190 g (approximately 800–1000 rpm) for 5 minutes. Discard the supernatant and resuspend the cell pellet in 10–30 mL of fresh pre‑warmed medium. Adjust the viable cell density to 1×10⁶ cells/mL.
6.Take 0.5 mL of the cell suspension and analyze the viable cell density (×10⁶ cells/mL) and cell viability (%) using a cell counter.
7.If the cell viability is >90%, place the cells under the specified environmental conditions for culture.
Cell Passage
1.Pre‑warm BHK Serum‑Free Medium at 37°C for 20–30 minutes.
2.Use cells in the mid‑logarithmic growth phase with a viable cell density ≥7×10⁶ cells/mL and cell viability ≥90% for subculturing.
3.Calculate the required seed culture volume based on an inoculation density of 1×10⁶ cells/mL and the passage volume.
4.Aseptically transfer the required volume of seed culture into a shake flask containing the appropriate volume of pre‑warmed medium.
5.Place the shake flask under the specified environmental conditions for cell culture.
6.Subculture every 48 ± 3 hours with fresh medium following the above procedure.
7.Before passaging, if the viable cell density is below 7.0×10⁶ cells/mL or the cell viability is below 90%, centrifuge the cells at 170–190 g (approximately 800–1000 rpm) for 5 minutes. Gently discard the supernatant, resuspend the cells in pre‑warmed medium, take a sample for counting, and then proceed with passaging.
Cell Adaptation
Direct Adaptation
1.For cells that can be directly adapted, they can be inoculated directly from their current serum‑free medium into BHK Serum‑Free Medium. The seeding density should follow the passaging procedure but may be adjusted based on specific conditions.
2.Continue subculturing the cells until stable growth is achieved.
3.After several passages, when the viable cell density reaches ≥7×10⁶ cells/mL within 48 ± 3 hours of inoculation, cell viability is >90%, and the doubling time is stable, the cells can be considered successfully adapted.
Sequential Adaptation
1.For cells conventionally grown in 5–10% serum‑containing or serum‑free media, use a gradual adaptation method with an initial seeding density of 1×10⁶ cells/mL.
Monitor cell growth until the viable cell density reaches ≥7×10⁶ cells/mL.
2.Dilute the cells with a mixture of BHK Serum‑Free Medium and the original medium at a ratio of 25:75 (BHK:original). Once cells grow well in this ratio, further dilute stepwise by increasing the proportion of BHK Serum‑Free Medium: 25:75 → 50:50 → 75:25 → 90:10 → 100:0.
3.After complete replacement with BHK Serum‑Free Medium, within 48 ± 3 hours of inoculation, the viable cell count should reach at least 7×10⁶ cells/mL with cell viability ≥90%. At this point, the cells are fully adapted to BHK Serum‑Free Medium.
Cell Cryopreservation
1.Prepare cells in the mid‑logarithmic growth phase with viability >90% and in good condition.
2.Determine the viable cell density and calculate the required volume of cryopreservation medium so that the final viable cell density is >1×10⁷ cells/mL.
3.Prepare the cryopreservation medium: 93% BHK Serum‑Free Medium + 7% DMSO, and store at 4°C.
4.Centrifuge at 170–190 g (approximately 800–1000 rpm) for 5 minutes. Discard the supernatant and resuspend the cells in the cryopreservation medium.
5.Aliquot the cell suspension into appropriate cryovials according to project requirements.
6.Follow standard procedures to cool the cryovials automatically or manually (at a rate of approximately 1°C per minute).
7.Transfer the cells to liquid nitrogen for long‑term storage.
