1、 选择合适的标记缓冲液(如 0.02M PB7.4、0.05MMES 5.5 等 ),向标记缓冲液中加入量子点,量子点终浓度为 3mg/ml;

Quantum dot fluorescent microspheres are mainly used in quantitative lateral flow detection reagents. They feature large Stokes shift, excellent photostability and high quantum yield, which can effectively reduce background interference and improve detection sensitivity. The microspheres are carboxyl-modified on the surface and can be efficiently conjugated with antibodies or antigens directly via a one-step method. Compared with time-resolved microspheres, quantum dot microspheres have higher density, which simplifies centrifugation steps and reduces production time and equipment costs.
| Catalog No. | Fluorescent Color | Microsphere Size | Functional Group | Excitation Wavelength | Emission Wavelength |
| C1521548-01 | Red | 300 nm | Carboxyl | 300~500 nm | 620±10 nm |
| C1521548-02 | Green | 300 nm | Carboxyl | 300~500 nm | 520±10 nm |
| C1521548-03 | Yellow | 300 nm | Carboxyl | 300~500 nm | 570±10 nm |
Under excitation wavelengths ranging from 300 to 500 nm (with 365 nm as the commonly used wavelength), quantum dot microspheres feature a large Stokes shift and outstanding photostability. Quantum dots with different core materials emit distinct fluorescent colors without optical interference, rendering them superior for lateral flow assays involving multiplex simultaneous detection.
One-step labeling protocol is recommended
1. Select an appropriate labeling buffer (e.g., 0.02 M PB, pH 7.4; 0.05 M MES, pH 5.5), and add quantum dots to the buffer to a final concentration of 3 mg/mL.
2. Add an appropriate amount of Tween 20, with a recommended final concentration of 0.01% (v/v).
3. After sonicating the above mixture to homogeneity, add the labeling antibody at a recommended dosage of 240 μg/mL. Mix well and react with shaking at room temperature for 10 min.
4. Add activation solutions of NHS and EDC (both at 10 mg/mL) to achieve final concentrations of 0.4 mg/mL for NHS and 0.2 mg/mL for EDC, respectively. Mix well and react with shaking at room temperature for 2 h.
5. Add an equal volume of 5% BSA solution to the quantum dot mixture and block for 1 h.
6. Centrifuge at 3500×g for 10 min and discard the supernatant.
7. Redissolve the quantum dots in a resuspension buffer to a final concentration of 0.4 mg/mL.
Precautions
1. Inappropriate buffer selection may lead to labeling aggregation. It is recommended to introduce sonication during the labeling process to disperse the quantum dots.
2. Centrifugation conditions should be adjusted accordingly with the centrifugation volume.
3. Quantum dots must be dispersed by sonication prior to use.
4. Strongly oxidizing conditions will compromise the service life of quantum dots.
5. The protocol described in this instruction is for reference only and should be adjusted according to the specific labeling conjugate.
