Calcium is a metallic element that appears as a silvery-white crystal at room temperature. Animal bones, shells, and eggshells all contain calcium carbonate. The detection of calcium content in living organisms is primarily achieved by measuring calcium ion concentration. Spectrophotometric detection of calcium is relatively common; this method requires suitable metal indicators or dye compounds that undergo color changes upon selective binding to calcium ions. Commonly used dyes include o-Cresolphthalein Complexone (OCPC), Arsenazo III, Methyl Thymol Blue (MTB), among others.
The Calcium Content Assay Kit (o-Cresolphthalein Complexone, Colorimetric Method) utilizes the principle that calcium ions bind with OCPC under alkaline conditions to form a purple-red complex. Magnesium ion chelating agents are added to eliminate background interference from magnesium ions. Absorbance is measured at 575 nm using a spectrophotometer, and the calcium content is calculated according to the formula.
Advantages of this method: 1. It is the internationally recommended routine method for serum calcium measurement; 2. Single reagent system, simple operation; 3. High sensitivity; 4. Stable color development; 5. Small sample volume required.
This kit is intended for research purposes only and is not suitable for clinical diagnosis or other applications.

Materials to Be Prepared by the User
1. Centrifuge tubes or test tubes, cuvettes, spectrophotometer
Operating Procedure (For Reference Only)
1. Sample Preparation
① Plasma and Serum Samples: Plasma and serum are prepared by conventional methods and can be used directly for determination with this kit. Store at -20°C for Ca detection.
② Cell or Tissue Samples: Take appropriate cells or tissues and homogenize. Centrifuge at low speed and collect the supernatant. Store at -20°C for Ca detection.
③ High-Concentration Samples: If the sample contains high concentrations of Ca, dilute with ddH2O. Ordinary distilled water should not be used for dilution.
④ (Optional) After sample preparation, protein concentration may be determined using a BCA Protein Assay Kit to facilitate subsequent calculation of Ca content per unit protein weight in tissues or cells.
2. Preparation of Ca Working Color Development Solution
Immediately before use, mix equal volumes of Ca Assay Buffer and OCPC Chromogenic Solution to prepare the Ca Working Color Development Solution. Prepare fresh and use immediately. Store at 4°C protected from light; do not store for extended periods.
3. Ca Loading
Use dry test tubes treated with dilute hydrochloric acid and cleaned with deionized water, or disposable sterile polyethylene centrifuge tubes. Set up blank tube, standard tube, and sample tube according to the table below. Solutions should be added sequentially in order, taking care to avoid bubble formation. If the calcium ion content in the sample is too high, reduce the sample volume or dilute appropriately before measurement. It is recommended to set up duplicate tubes for sample testing.

4. Ca Measurement
Mix well and let stand at room temperature for 10 minutes. Zero the spectrophotometer with distilled water. Use a cuvette with a 1 cm light path. Measure absorbance at 575 nm. Record the absorbances of the standard tube and sample tube (designated as Astandard and Asample).
Where:
Asample = absorbance of the sample tube
Astandard = absorbance of the standard tube
Ablank = absorbance of the blank tube
V = total volume of tissue extraction solution (L)
