概述
固定的目的在于保存细胞和组织的原有形态结构，固定剂能阻止内源性溶酶体酶对自身组织和细胞的自溶、抑制细菌和霉菌的生长。固定剂通过凝固、生成添加化合物等使蛋白质内部结构发生改变，从而使酶失活。未固定前活细胞被包裹在不能穿透的膜内，固定剂能够破坏这一屏障，使大分子物质穿透膜、逸出。
选择固定液对组织染色和免疫组化染色都极其重要，通常将几种固定剂混合配制成复合固定液，这样可以适应多种组织、细胞成分的保存；然而目前还没有一种标准的固定液适用于所有的组织、细胞成分的保存。固定液主要分为醛类固定液、汞类固定液、醇类固定液、氧化剂类固定液、苦味酸盐类固定液等，较为常用的是醛类中的福尔马林、醇类中的乙醇。
FAB 固定液主要磷酸盐、甲醛等组成，pH 值为 6.6~6.8，多用于血液、骨髓细胞涂片的固定，亦可用于免疫组化的固定。该试剂仅用于科研领域，不适用于临床诊断或其他用途。
操作步骤 (仅供参考)
1、按实验具体要求操作。一般血液、骨髓细胞涂片固定 1~2 min。
注意事项
1、组织取材的厚度不同，固定时间也不同，对组织恰当的选材有利于固定液的渗透。
2、取出新鲜组织后，应及时固定，无法及时固定时，应保存于生理盐水中及时送检。
3、为了您的安全和健康，请穿实验服并戴一次性手套操作。
4、试剂开封后请尽快使用，以防影响后续实验效果。
The purpose of fixation is to preserve the original morphological structure of cells and tissues. Fixatives prevent autolysis of tissues and cells by endogenous lysosomal enzymes and inhibit the growth of bacteria and molds. Fixatives alter the internal structure of proteins via coagulation and adduct formation, thereby inactivating enzymes. Living cells are enclosed within an impermeable membrane before fixation; fixatives can disrupt this barrier and allow macromolecules to penetrate or escape from the membrane.
Selection of fixatives is critical for tissue staining and immunohistochemistry. Multiple fixatives are often mixed to prepare compound fixatives for preserving a wide range of tissue and cellular components. Nevertheless, no single standard fixative is suitable for the preservation of all tissues and cellular components. Fixatives are mainly classified into aldehyde‑based fixatives, mercury‑based fixatives, alcohol‑based fixatives, oxidant‑based fixatives, and picrate‑based fixatives. Among them, formalin (aldehyde‑based) and ethanol (alcohol‑based) are most commonly used.
Formalin-Ammonium Bromide(FAB)Fixative is mainly composed of phosphate salts, formaldehyde and other ingredients, with a pH value of 6.6‑6.8. It is mostly used for fixation of blood and bone marrow smears, and can also be applied for immunohistochemical fixation. This reagent is for research‑only use and is not intended for clinical diagnosis or other purposes.
Operating Procedures (For Reference Only)
1. Operate according to specific experimental requirements. Generally, fix blood or bone marrow smears for 1‑2 min.
Precautions
1. Fixation time varies with the thickness of tissue samples. Proper tissue sampling facilitates penetration of fixative.
2. Fresh tissues should be fixed promptly. If immediate fixation is unavailable, store tissues in normal saline for subsequent processing.
3. For personal safety, wear a lab coat and disposable gloves during operation.
4. Use the reagent promptly after opening to avoid adverse effects on subsequent experimental results.
