Identification experiments of pathogenic bacteria

Principle

1. pathogenic staphylococci are salt-resistant and can grow on high-salt media; 2. pathogenic staphylococci are able to ferment mannitol; 3. when a particulate antigen (e.g., bacterium) binds to a specific antibody, the antigen-antibody can agglutinate to form clumps that can be seen by the naked eye in the presence of electrolytes.

Move

1. Inoculation ring Take 1-2 rings of pus specimen enrichment solution, inoculate on 6.5% high salt agar medium by zonal scribing, place in a 37℃ warm box and incubate for 24 hours.

2. Observe the bacterial characteristics.

3. take suspicious colonies direct smear microscopy (Gram stain)

4. Take suspicious colonies and inoculate them into mannitol fermentation tubes, place them in a 37℃ warm box, incubate them for 24 hours and observe the results.

5. slide agglutination test (1) clean slides, mark the partition;

(2) Aseptic operation each with saline 2 to 3 rings;

(3) Aseptically add a small amount of bacterial culture to zone 1;

(4) Add 1 drop of bacterial antiserum to each;

(5) Gently shake the slide and observe the results for 5-10 minutes at room temperature.
