HDC Human Diploid Cell Low-Serum Medium is a widely used basal medium that supports the cultivation of diploid cells (such as MRC-5, WI38, 2BS, etc.) with reduced fetal bovine serum (FBS) supplementation. Compared with classic MEM medium, the serum supplementation level can be reduced by more than 80% during culture, while the cell growth rate is significantly increased.
Applications
Supports the growth of multiple diploid cell lines (MRC-5, WI38, 2BS, etc.). This basal medium is suitable for research and large-scale biopharmaceutical production based on cell culture, but it is not for direct use in humans or as a drug.
Preparation Method for Powdered Medium
1.Add purified water (20–30 °C) equivalent to 90% of the final volume to the mixing container.
2.Slowly add 14.90 g/L of medium powder and 0.584 g/L of glutamine, and stir-mix for 30–60 minutes until the solution becomes clear.
3.Slowly add 2.20 g/L of sodium bicarbonate powder, and stir-mix for 3–10 minutes until dissolved.
4.Bring to the final volume with purified water, and adjust the pH to 7.1–7.3 using 1 M hydrochloric acid or sodium hydroxide solution.
5.Immediately sterile-filter through a 0.22 μm filter membrane.
Note: Supplement with 1–3% serum before use.

Cell Thawing
1.Pre-warm HDC complete medium (HDC basal medium supplemented with 1–3% serum) in a 37 °C water bath.
2.Spray the medium bottle with 75% alcohol and place it in a biological safety cabinet.
3.Thaw 1 vial of cells by gently shaking the cryovial in a 37 °C water bath for 1 minute.
4.Gently transfer the cells from the cryovial into a centrifuge tube containing 10 mL of pre-warmed HDC complete medium.
5.Centrifuge at 150g–300g (approximately 800–1200 rpm) for 5 minutes. Discard the supernatant and resuspend the cell pellet in 10 mL of fresh pre-warmed HDC complete medium. Adjust the viable cell density to 2–4×10⁴ cells/cm².
6.Take 0.5 mL of the cell suspension and analyze the viable cell density (×10⁶ cells/mL) and cell viability (%) using a cell counter.
7.If cell viability is > 90%, place the cells under the specified environmental conditions for culture.
Cell Passage
Using a T75 flask as an example:
1.Pre-warm HDC complete medium at 37 °C for 20–30 minutes.
2.Use cells in the mid-logarithmic growth phase with a viability ≥ 90% for subculturing.
3.Remove the culture medium from the vessel using a pipette. Add an appropriate volume of DPBS to rinse the cells, remove the washing solution, and repeat the rinse once.
4.Add 3 mL of recombinant trypsin digestion solution to cover all culture surfaces. Digest at room temperature for 3–5 minutes.
5.After the cells detach from the flask surface, tap the flask. Add 3 mL of pre-warmed HDC complete medium, pipette gently to disperse the cells, and collect the cell suspension. Centrifuge the cell suspension at 300g for 5–10 minutes at room temperature.
6.Discard the supernatant and resuspend the cell pellet in complete medium.
7.Calculate the amount of seed stock required based on a seeding density of 2–4×10⁴ cells/cm² and the passage volume.
8.Aseptically transfer the required volume of seed stock into a T75 flask containing the required volume of pre-warmed HDC complete medium, and add 1–3% serum of the final culture volume.
9.Subculture every 2–4 days.
Cell Adaptation
1.Cells routinely cultured in serum-containing medium do not require gradual acclimatization. They can be directly inoculated from MEM + 10% serum medium into HDC complete medium. Refer to the passage steps for seeding density, and adjust according to specific conditions.
2.Continue passaging the cells until they grow stably.
3.The recommended seeding density is 2–4×10⁴ cells/cm², with subculturing every 2–4 days. Generally, cells become fully adapted after 3 passages.
Cell Cryopreservation
1.Prepare cells in the mid-logarithmic growth phase with viability > 90% and in good condition.
2.Determine the viable cell density and calculate the required volume of cryopreservation medium. The final viable cell density should be > 1×10⁷ cells/mL.
3.Prepare the required cryopreservation medium: 90% HDC complete medium + 10% DMSO, and store at 4 °C.
4.Prepare the cell suspension according to the cell digestion procedure.
5.Centrifuge at 300g for 5 minutes, discard the supernatant, and resuspend the cell pellet in cryopreservation medium.
6.Aliquot the suspension into cryovials of appropriate size according to project needs.
7.Following standard procedures, cool the cryovials automatically or manually (at a rate of 1 °C per minute).
8.Transfer the cells to liquid nitrogen storage for preservation.
