潮霉素 B（Hygromycin B）是由吸水链霉菌（Streptomyces hygroscopicus）产生的一种氨基糖苷类抗生素。其作用机制为干扰 70S 核糖体的易位过程，并诱导 mRNA 模板的错读，从而抑制蛋白质合成，最终导致原核细胞（如细菌）及真核细胞（如酵母、真菌和高等哺乳动物细胞）死亡。
在实验室中，潮霉素 B 广泛用于筛选和维持携带潮霉素抗性基因的原核或真核细胞。该抗性基因编码潮霉素激酶，通过磷酸化作用使潮霉素 B 失去活性。自潮霉素抗性基因被发现以来，潮霉素 B 已成为转基因实验中常用的筛选试剂，尤其适用于植物组织培养，且可与其它筛选试剂配合使用，实现双选择体系。
使用方法
本品是无菌的潮霉素 B 溶液，可直接稀释使用。
工作浓度：细胞培养中为 50-1000 µg/mL。哺乳动物细胞培养通常使用的浓度为 200 µg/mL。细菌/植物细胞 20-200 μg/mL；真菌 200-1000 μg/mL。但最适工作浓度需根据具体细胞类型、培养基组成及实验体系，通过预实验确定。
注意事项
1. 本品为有毒化合物，操作时请避免接触皮肤和眼睛。
2. 请在通风良好的条件下操作，并穿戴实验服和一次性手套，确保安全。
Hygromycin B is an aminoglycoside antibiotic produced by the bacterium Streptomyces hygroscopicus. Its mechanism of action involves interference with the translocation process of the 70S ribosome and induction of misreading of the mRNA template, thereby inhibiting protein synthesis and ultimately leading to the death of both prokaryotic cells (e.g., bacteria) and eukaryotic cells (including yeasts, fungi, and higher mammalian cells).
In the laboratory, Hygromycin B is widely used for the selection and maintenance of prokaryotic or eukaryotic cells harboring the hygromycin resistance gene. This resistance gene encodes a hygromycin kinase that inactivates Hygromycin B through phosphorylation. Since the discovery of the hygromycin resistance gene, Hygromycin B has become a standard selection reagent in transgenic experiments. It is particularly suitable for plant tissue culture and can be used in combination with other selective agents to establish dual‑selection systems.
Instructions for Use
This product is a sterile Hygromycin B solution and can be diluted directly for use.
Working concentrations: 50‑1000 µg/mL for cell culture. A typical working concentration for mammalian cell culture is 200 µg/mL; 20‑200 μg/mL for bacteria and plant cells; 200‑1000 μg/mL for fungi. However, the optimal working concentration should be determined empirically through preliminary titration experiments, as it varies depending on the specific cell type, culture medium composition, and experimental system.
