概述

HBC599是一种非荧光HBC模拟物，在溶液中不发光，但与Pepper RNA适体形成复合物后会激活并发出明亮荧光。可用激发波长为515nm，发射波长为599nm进行检测复合物。

使用HBC599进行活细胞RNA成像的染色

本实验方案根据研究内容整理，仅供参考，请根据实际需求进行调整。

（1）细胞准备与转染：将适当的细胞（例如 HeLa、U2OS）接种到玻底培养皿或盖玻片上，培养至汇合度达到 50-70%；用带有Pepper RNA适配体序列标签的质粒或RNA转染细胞，孵育12-48小时，以确保RNA充分表达。

（2）HBC599溶液配制：使用无水DMSO配制1-5mM的HBC599储备溶液。分装后于-20°C或-80°C避光保存。将HBC599储备溶液用预热的、无血清的细胞培养基（DMEM或Opti-MEM）稀释，通常最终工作浓度在0.5至5µM之间。充分混匀。

（2）细胞染色：用温热的无血清培养基或PBS轻轻洗涤转染后的细胞1-2次。完全吸尽洗涤液，将配制好的HBC599染色培养基加入细胞中。在37°C、5%CO₂条件下，将细胞在染色培养基中孵育15到30分钟。小心吸弃染色液。用温热的无血清培养基或PBS洗涤细胞2-3次。

（3）活细胞成像：细胞中加入少量预热的新鲜培养基。立即在共聚焦或落射荧光显微镜下对细胞进行成像。HBC599荧光通常使用515nm激光激发，并使用599nm发射滤光片进行检测。

注意事项：

（1）HBC599及其荧光信号对光敏感。在染色过程及后续成像中，尽量避光操作，以防止光漂白和荧光猝灭。

HBC599 is a non-fluorescent HBC mimetic that does not emit light in solution but activates and emits bright fluorescence upon forming a complex with the Pepper RNA aptamer. The complex can be detected using an excitation wavelength of 515nm and an emission wavelength of 599nm.

Staining for Live-Cell RNA Imaging with HBC599

This protocol is compiled for research reference only. Please adjust according to your actual experimental requirements.

(1) Cell Preparation and Transfection: Seed appropriate cells (e.g., HeLa, U2OS) in glass-bottom dishes or coverslips and culture until 50–70% confluency. Transfect cells with plasmids or RNA tagged with Pepper RNA aptamer sequences, then incubate for 12–48 h to ensure sufficient RNA expression.

(2) Preparation of HBC599 Solution: Prepare 1–5 mM HBC599 stock solution in anhydrous DMSO. Aliquot and store at -20°C or -80°C protected from light. Dilute the HBC599 stock solution with pre-warmed serum-free cell culture medium (DMEM or Opti-MEM). The typical final working concentration ranges from 0.5 to 5 µM. Mix thoroughly.

(3) Cell Staining: Gently wash transfected cells 1–2 times with warm serum-free medium or PBS. Aspirate the wash solution completely, then add the prepared HBC599 staining medium to the cells. Incubate cells in staining medium at 37°C, 5% CO₂ for 15–30 minutes. Carefully aspirate the staining solution. Wash the cells 2–3 times with warm serum-free medium or PBS.

(4) Live-Cell Imaging: Add a small volume of pre-warmed fresh medium to the cells. Immediately image the cells under a confocal or epifluorescence microscope. HBC599 fluorescence is typically excited by a 515 nm laser and detected with a 599 nm emission filter.

Precautions

(1) HBC599 and its fluorescent signal are light-sensitive. Minimize light exposure during staining and subsequent imaging to prevent photobleaching and fluorescence quenching.

(2) For personal safety, wear a lab coat and disposable gloves during operation.
