Multifunctional bacterial cellulose-based hydrogel with tannic acid (TA)-modified MXene for antibacterial photothermal therapy and accelerated wound healing

Feng Cheng,Chaofan Yao,Xiaotong Yi,Hongbin Li,Yudong Huang,Jinmei He

This work reports the development of an advanced multifunctional hydrogel system designed to address the challenges of infectious wound healing and tissue regeneration. We constructed a polyvinyl alcohol–borax/GelMA hybrid hydrogel matrix, endowing the photocurable hydrogel with excellent mechanical properties and self-healing capability. Bacterial cellulose (BC), a naturally derived biopolymer synthesized by specific microbial strains, was incorporated into the hydrogel matrix to enhance its mechanical properties, structural stability, and biocompatibility, while also serving as a scaffold for cellular adhesion and proliferation. Additionally, the hydrogel was functionalized with tannic acid (TA)-modified MXene nanosheets, which imparted superior antibacterial efficacy and enabled photothermal therapy under near-infrared (NIR) light irradiation. In vitro evaluations confirmed the hydrogel's excellent biocompatibility and antibacterial performance, while in vivo studies demonstrated its ability to significantly accelerate the healing of infectious wounds. This was achieved by effectively reducing bacterial load, promoting tissue regeneration, and alleviating local inflammation under NIR irradiation. These findings highlight the potential of this BC-based hydrogel system as a robust platform for combating bacterial infections, facilitating wound healing, and supporting tissue regeneration in the context of complex skin injuries.

Counterion-mediated modulation of electroadhesion in polyanionic/polycationic hydrogels: mechanisms and performance

Xianxiao Song,Ziyan Qiu,Hongkai Li,Lei Wang,Shuheng Zhang,Mengke He,Jingtao He,Botian Li,Chengyou Kan

Reversible electroadhesive polyelectrolyte gels have emerged as promising materials for flexible electronic and soft robotic applications. While current research predominantly emphasizes polymer design and structural optimization to enhance both the reversibility and strength of electroadhesion, fundamental limitations persist in elucidating ion-mediated interfacial mechanisms. Herein, the synergistic effects of ion species selection and interfacial engineering were systematically investigated through the development of distinct polyelectrolyte hydrogel assemblies. By varying counterion types (anions and cations) and ion concentrations, the ion-mediated mechanism governing reversible adhesion between polyanionic and polycationic hydrogels was elucidated. Combined electrochemical impedance spectroscopy and molecular dynamics simulations revealed that interfacial electroadhesion primarily resulted from electrostatic interactions within the ionic double layers. Optimizing counterion diffusion rates through ion species selection can reduce the interfacial ionic resistance and increase the capacitance of the ionic double layer, thereby enhancing the electroadhesive performance. The results indicated that Li Cl hydrogels are suitable for single-use high-adhesion patches due to the accelerated Li + /Cl − migration, whereas Na Cl hydrogels are ideal for reusable electroadhesive patches. This counterion-mediated modulation strategy provides a universal framework for designing adaptive hydrogel interfaces with tunable adhesion properties.

Injectable ROS homeostasis protective hydrogel inhibiting microglial ferroptosis through the Nrf2/Slc7a11/Gpx4 to alleviate neuropathic pain and promote spinal cord injury repair

Lu Li,Yu Cao,Xiangsheng Zhang,Jiayi Guo,Ziqiang Lin,Pengyu Zhou,Chuyin Chen,Jiahao Chen,Yike Liu,Danzhi Luo,Jiurong Chen,Yingdong Deng,Peng Sun,Zhiwen Zeng,Jun Zhou

Spinal cord injury (SCI) induced neuropathic pain (NP) remains a major clinical challenge due to persistent neuroinflammation and oxidative stress. We developed an injectable methacrylated and thiolated gelatin hydrogel loaded with quercetin (MSQ) to synergistically scavenge reactive oxygen species (ROS) and inhibit microglial ferroptosis for NP alleviation and neural repair. The MSQ hydrogel exhibited rapid photocrosslinking, sustained quercetin release, and robust ROS scavenging via thiol groups and quercetin, maintaining intracellular redox homeostasis. MSQ attenuated LPS-induced ferroptosis in BV2 microglia by upregulating Nrf2 expression, promoting its nuclear translocation, and activating the Slc7a11/Gpx4 pathway, thereby reducing lipid peroxidation and inflammatory cytokine release. Network pharmacology and molecular dynamics simulations confirmed quercetin's high-affinity binding to Nrf2. In a murine SCI model, MSQ implantation significantly reduced lesion area, suppressed microglial ferroptosis, and decreased pro-inflammatory mediators (TNFα, IL-1β, IL-6), while enhancing neuronal survival (Nissl/NeuN + cells) and axonal regeneration (MAP2/5-HT + expression). Motor functional recovery assays revealed improved BMS scores, gait regularity, and mechanical/thermal pain thresholds in MSQ-treated mice. This study highlights MSQ hydrogel as a multifunctional therapeutic platform that targets ROS homeostasis and microglial ferroptosis via the Nrf2/Slc7a11/Gpx4 axis, offering a promising strategy for post-SCI NP management and neural regeneration.

Multimodal Actuation and Precise Control in Liquid Crystal Elastomer Optical Fiber Artificial Muscles

Haojun Liu,Yongcheng He,Jiajia Luo,Nuo Li,Tanxi Wang,Enhai Song,Qianyi Guo,Zhijun Ma,Zhongqiang Yang,Jiulin Gan,Zhongmin Yang

Artificial muscles mimicking the fibrous structure and functionalities of natural skeletal muscles have garnered substantial interest for applications in actuators, soft robotics, and biomedical devices. However, achieving multidirectional actuation and delicate manipulation in confined environments remains challenging. Inspired by the neuromuscular system, a novel liquid crystal elastomer optical fiber (LCEOF) is introduced as artificial muscle with multimodal actuation and precise control. Fabricated through a two-step process, the LCEOFs possess sufficient orientation order (0.65) and low optical transmission loss (0.37 dB cm −1 ), enabling over 40% contraction strain with minimal ambient interference. Bundling multiple LCEOFs yields artificial arms capable of complicated and controllable deformations, including long-distance contraction (≥5 cm), weightlifting (>4000 times their own weight), wide-range torsion (0–180°), and omnidirectional bending (0–360°). Multimodal actuation is precisely and independently regulated via terminal-coupled laser inputs for each LCEOF in bundled arrays, enabling coordinated, crosstalk-free motions. These optical fiber artificial muscles allow precise and controllable operation of an artificial hand for grasping and manipulating objects, without reliance on free-space lateral illumination. Additionally, robotics systems incorporating bundles of LCEOFs have been designed and demonstrated for tasks such as laser writing and object transfer in confined environments, thereby offering new possibilities for the advancement of smart actuators.

3D Bio-Printing and Dynamic Maturation of Heart Valve Organoids Made of Human Induced Pluripotent Stem Cell-Derived Valve Interstitial Cells

Chuan Gao,Kun Yang,Jiahui She,Ruxiang Wang,Xiangfu Meng,Renyuan Sun,Yunjie Jin,Weihua Qiao,Jiawei Shi,Jianglin Wang,Nianguo Dong,Chao Xu,Donghui Zhang,Zhiqiang Luo

Heart valve organoids hold promise for modeling human valve biology, yet existing platforms often lack human-relevant cell sources, preserved multicellular architecture, and physiologic mechanical cues, limiting their capacity to recapitulate extracellular matrix (ECM) formation and structural maturation. Here, a strategy is established that integrates human induced pluripotent stem cell-derived valve interstitial cells (hiPS-VICs), spheroid organization, bio-printable hydrogels, and controlled mechanical conditioning to engineer advanced valve organoids. 3D bio-printing of hiPS-VIC spheroids is conducted within GelMA/ChSMA constructs, systematically compares spheroid-based bio-printing (SBB) with single-cell bio-printing (SCB), and establishes mechanically-stimulated spheroid-based bio-printing (MSSB) to guide ECM remodeling. Spheroid bioprinting preserved cell–cell organization and improved post-print structural integrity, resulting in substantially enhanced ECM deposition and assembly over time when compared with SCB. Superimposing physiologic loading further promoted cellular spreading and ECM deposition, accompanied by transcriptional signatures associated with matrix production, matrix stabilization, and mechano-responsive pathways. Together, this spheroid-based, mechanically matured, human iPSC valve organoid platform provides a robust and physiologically relevant system that supports ECM development and structural organization beyond dissociated-cell approaches, offering a scalable foundation for human valve biology research, disease modeling, and translational applications.

Silk Fibroin-Regulated Shape Memory Composite Hydrogels with Tunable Recovery Time

Zhengdong Feng,Wei Huang,Fei Lv,Wei Li

Shape memory polymers (SMPs) are pivotal for applications in soft robotics, biomedical devices, and flexible electronics. However, their reliance on external stimuli for activation and the difficulty of precisely programming recovery dynamics remain significant limitations. Autonomous SMPs with delayed recovery offer a promising alternative, yet facile control of recovery onset and duration is still difficult to attain. Here, a semi-interpenetrating network (semi-IPN) hydrogel composed of poly(acrylic acid) and silk fibroin (PAA-SF) is presented, fabricated via a facile one-step UV polymerization. By modulating the SF content within the PAA network, the hydrogel exhibits tunable shape recovery kinetics across multiple programming environments, including organic solvents, air, and water, enabling spatiotemporal control over shape restoration. Moreover, the hydrogels show significantly enhanced mechanical properties and strong adhesion compared to neat PAA hydrogels. Benefiting from these combined properties and excellent cytocompatibility, demonstrating two proof-of-concept devices: a smart four-jaw gripper capable of retrieving objects in confined spaces, and a stiffness-switchable needle that reversibly transitions between soft and rigid states. This work not only introduces a robust and multifunctional shape memory hydrogel but also provides a versatile platform for designing stimulus-free, time-programmable soft actuators for biomedical and robotic applications.

In-situ extrusion 3D printing with tea polyphenol crosslinking for Hyaluronic acid sodium salt -based composite hydrogel scaffolds

Reyihanguli Alimu,Lanlan Dong,Patiguli Aihemaiti,Houfeng Jiang,Wurikaixi Aiyiti,Cijun Shuai

High-performance hydrogel biomaterials hold considerable promise for advanced wound care. However, the suboptimal mechanical properties of conventional hydrogel materials limit their practical application. In this study, Hyaluronic acid sodium salt (HA), xanthan gum (XG), and N-acryloyl-glycinamide (NAGA) hydrogels with porous structures were successfully fabricated using in-situ extrusion 3D printing technology, and a functionalization strategy involving tea polyphenol (TP) immersion was proposed to enhance material properties through additional hydrogen bonding. Systematic optimization of TP concentration and immersion time led to significant improvements in the resulting hydrogel: tensile strain increased from 95 % to 266 %, tensile stress improved 6.8-fold, and compressive stress increased 10-fold, with an elastic modulus of 0.68 MPa and toughness of 1.2 MJ/m³ . Furthermore, TP-treated hydrogels exhibited excellent biocompatibility and strong antioxidant activity and antibacterial activity. This study provides a simple yet effective secondary functionalization approach for enhancing the performance of 3D-printed hydrogels, offering a promising approach for multifunctional hydrogel biomaterials with integrated mechanical strength and biocompatibility. These findings open new avenues for advanced wound care systems and personalized medicine applications.

Self-Assembled Bioactive Glass/Starch/GelMA Microspheres for Hemostasis and Bone Regeneration in Bone Trauma Emergency Treatment

Ying Chen,Wenyi Zhao,Ziyang Liu,Huan Zhou,Zhen Yang,Lei Yang

The high incidence of bone trauma has necessitated the development of multifunctional biomaterial implants with both hemostatic and osteogenic properties to address clinical challenges such as bone nonunion and immune rejection associated with nondegradable bone hemostatic materials. In this study, starch/bioactive glass/GelMA-based self-assembling composite microspheres were developed as a novel bone trauma filler exhibiting dual functions of efficient hemostasis and bone regeneration. These micron-scale composite microspheres can be applied to bone trauma sites during surgical procedures or emergency situations. Upon absorption of blood, self-assembly occurs rapidly through hydroxyl/amino-mediated interfacial interactions, preventing dispersion by blood and facilitating the formation of a stable barrier for emergency hemostasis. Furthermore, the bioactive glass component promotes osseointegration via biomineralization-mediated assembly, while the released bioactive ions enhance the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells. This composite microsphere exhibited biodegradable properties and significantly improved cranial bone defect repair in vivo.

Recombinant human collagen microneedle patch loaded with PRP for diabetic wound treatment

Xinyue Wang,Xinyue Zhang,Yihan Zhao,Xue Zhan,Chen Hu,Haihang Li,Xiaoju Fan,Jie Liang,Yafang Chen,Yujiang Fan

Chronic nonhealing wounds represent significant complications of diabetes, bearing a substantial burden and posing risks of disability or mortality. In diabetic wounds, continuous tissue fluid exudation, inflammatory cell migration, fibrosis, and bacterial biofilm formation create a “barrier”, which decreases the treating efficacy of therapeutics. To address these limitations, a recombinant human collagen type III microneedle patch (rhCol III-PRPM) loaded with platelet-rich plasma (PRP) was developed, in which methacrylated rhCol III (rhCol III-MA) loaded with PRP was utilized to form needle tips, while rhCol III-MA formed the base part of patch. RhCol III-PRPM featured adequate mechanical qualities, swelling capacity, and sustained in vitro release of growth factors from the activation of PRP for over 7 days. Leveraging the synergistic effects of rhCol III and PRP, rhCol III-PRPM patches facilitated cell proliferation, migration, and angiogenesis, and reduced oxidative stress. In animal experiment, this microneedle patch effectively promoted the healing of diabetic wounds during a 20-day treatment, partially owing to upregulating integrins and phosphorylated ERK protein levels. Diverging from other microneedle strategy, the rhCol III exhibited “dual functionality,” serving as both the microneedle patch matrix and therapeutic agent, promoting wound healing upon patch dissolution while delivering PRP. The combination of rhCol III and PRP in the form of microneedle patch offered a straightforward and efficacious way for effective diabetic wounds management, and held promise in bringing new possibility in clinical practice.

Enhancement of spinal cord injury repair in rats using photo-crosslinked GelMA hydrogel combined with mitochondrial transplantation

Wenyong Gao,Can Tang,Diancheng Hang,Hao Chen,Dongsheng Li,Youjie Yan,Wuchang Wang,Hao Peng,Xianmin Wang,Enpeng Zhang,Min Wei,Hengzhu Zhang,Xiaodong Wang

Acute spinal cord injury (SCI) induces mitochondrial oxidative stress, cellular bioenergetic crises, impaired protein degradation, and subsequent degeneration, resulting in increased neuronal vulnerability. Transplantation of exogenous mitochondria to the injury site mitigates cellular energy crises and counteracts neurodegeneration; however, the limited efficacy of mitochondrial transplantation alone constrains its therapeutic potential. In this study, we established a right-sided spinal cord hemisection model at the T10 thoracic segment in rats and transplanted a methacrylate-based gelatin (GelMA) hydrogel containing active mitochondria at the injury site to assess its therapeutic effects and underlying mechanisms. Our findings indicate that GelMA hydrogel combined with mitochondrial transplantation provides superior therapeutic benefits for SCI compared to mitochondrial transplantation alone. GelMA hydrogel enables sustained mitochondrial release at the injury site, supplying energy, upregulating NF200 expression, and promoting axonal regeneration. Additionally, it enhances M2 macrophage accumulation and improves the local inflammatory microenvironment. The structural framework of GelMA hydrogel further supports axonal regeneration. Footprint gait analysis and Basso, Beattie, and Bresnahan (BBB) motor scoring demonstrated that GelMA hydrogel combined with mitochondrial transplantation significantly improves motor function in the right hind limb of rats with SCI. Consequently, GelMA hydrogel combined with mitochondrial transplantation offers a viable and promising approach for treating spinal cord injury.

Facile crystallized-AIE-photosensitizer functionalized hydrogel for antimicrobial photodynamic therapy and wound recovery

Gongmeiyue Su,Yangkun Liu,Wenxin Chou,Yilei Xiao,Zhao Li,Rongji Dai,Yulin Deng,Hongyou Zhao

The development of wound dressings with high antibacterial efficacy is critical for the treatment of bacterial infections. Antimicrobial photodynamic therapy (aPDT) is a promising approach, and aggregation-induced emission photosensitizers (AIE-PSs) have garnered significant attention for their enhanced performance in the aggregated state, where the restriction of intramolecular motion serves as the underlying mechanism. However, conventional aggregation often leads to loosely packed PSs, leaving space for intramolecular motion. Crystallization provides a powerful strategy for forming tightly packed molecular aggregates. In this study, an AIE-PS (MeTS 2 IC) was designed by attaching a propeller-like moiety to rigid planar groups. The former enables intramolecular motion and maintains a twisted conformation, while the latter facilitates crystallization. A bactericidal function alone is insufficient for optimal wound healing; thus, a hydrogel matrix was introduced for biocompatibility. The AIE-PS can easily form microcrystals (MCs) when its concentrated solution is added to the pre-gel solution. Subsequent photo-crosslinking in situ forms the light-activated antibacterial AIE-PS-MCs@hydrogel dressing on the wound. Under white light irradiation, the dressing effectively eliminates bacteria, reduces inflammation, promotes vascular network recovery, and accelerates wound healing. The combination of AIE-PS MCs with hydrogel offers a simple, effective strategy for managing infected wounds, with promising potential for future clinical applications.

Engineered nerve guide conduit enhances peripheral nerve regeneration by modulating the microenvironment through macrophage-triggered cascade reactions

Xindi Yang,Xiao Liang,Binyi Wang,Xing Gao,Wenqiang Yang,Jiajun Li,Huaan Cai,Zan Tong,Yun Chen

Nerve regeneration after peripheral nerve injury (PNI) depends on exogenous and endogenous pro-healing signals in the microenvironment. Maintaining immune system homeostasis and remodeling the regenerative microenvironment are crucial prerequisites for promoting nerve regeneration. Here, a double-layer PSM/Gel-SC(IL-4) nerve guide conduit was fabricated by photocrosslinking poly (ethylene glycol) diacrylate (PEGDA) and methacrylic acid-sodium alginate (SAMA) hydrogel as the outer layer, with GelMA encapsulating IL-4 overexpressing Schwann cells as the inner layer. PSM/Gel-SC(IL-4) conduit enabled the sustained release of IL-4, inducing pro-healing macrophages polarization through JAK1/STAT6 pathway in vitro . The polarized macrophages further triggered the cellular cascade reactions, enhancing the pro-healing characteristics of endothelial cells and Schwann cells. In addition, PSM/Gel-SC(IL-4) conduit significantly promoted functional recovery and nerve regeneration in 12-mm rat sciatic nerve defect model, induced pro-healing macrophage polarization at the early stage and accelerated angiogenesis and myelination at the nerve regeneration stage. This study developed a novel immunomodulatory strategy to remodel the regenerative microenvironment, and explored the underlying cellular and molecular mechanism, demonstrating considerable therapeutic potential for long-distance nerve defect.
