X-Ray Activatable Au/Ag Nanorods for Tumor Radioimmunotherapy Sensitization and Monitoring of the Therapeutic Response Using NIR-II Photoacoustic Imaging

Si Zheng,Duyang Gao,Yayun Wu,Dehong Hu,Ziyue Li,Yuenan Wang,Hairong Zheng,Yingjia Li,Zonghai Sheng

Radioimmunotherapy (RIT) is an advanced physical therapy used to kill primary cancer cells and inhibit the growth of distant metastatic cancer cells. However, challenges remain because RIT generally has low efficacy and serious side effects, and its effects are difficult to monitor in vivo. This work reports that Au/Ag nanorods (NRs) enhance the effectiveness of RIT against cancer while allowing the therapeutic response to be monitored using activatable photoacoustic (PA) imaging in the second near-infrared region (NIR-II, 1000–1700 nm). The Au/Ag NRs can be etched using high-energy X-ray to release silver ions (Ag + ), which promotes dendritic cell (DC) maturation, enhances T-cell activation and infiltration, and effectively inhibits primary and distant metastatic tumor growth. The survival time of metastatic tumor-bearing mice treated with Au/Ag NR-enhanced RIT is 39 days compared with 23 days in the PBS control group. Furthermore, the surface plasmon absorption intensity at 1040 nm increases fourfold after Ag + are released from the Au/Ag NRs, allowing X-ray activatable NIR-II PA imaging to monitor the RIT response with a high signal-to-background ratio of 24.4. Au/Ag NR-based RIT has minimal side effects and shows great promise for precise cancer RIT.

“Double-punch” strategy against triple-negative breast cancer via a synergistic therapy of magneto-mechanical force enhancing NIR-II hypothermal ablation

Hui Du,Fang Yang,Chenyang Yao,Wenhao Lv,Hao Peng,Stefan G. Stanciu,Harald A. Stenmark,Young Min Song,Bo Jiang,Aiguo Wu

Triple-negative breast cancer (TNBC) is a form of breast cancer that is more aggressive and harder to treat than others, with a higher probability of relapse. Its nefarious capabilities for migrating and invading other parts of the body together with the current lack of clinically established effective therapies account for a low survival rate. In this work, we demonstrate the in-tandem use of two complementary therapeutic routes to effectively combat TNBC. A versatile magnetic-photothermal converter (MPC) consisting of zinc-doped ferrite nanoparticles and polyethene glycol, is shown to display excellent therapeutic efficiency, being capable to fight TNBC via two distinct routes: magneto-mechanical force (MMF) and near-infrared-II (NIR-II) hypothermal ablation. The combined use of these two complementary and synergistic therapies, which are less aggressive to the human body compared to conventional chemotherapeutic approaches, results in the splendid suppression of TNBC migration and invasion. Remotely controlling the MPCs by an external magnetic field , results in cellular MMF effects that cause direct mechanical destruction to the cancer cell membrane , leading to its necrosis. Furthermore, the MMF disrupts intracellular lysosomes , thereby triggering the release of large amounts of protein hydrolases , which induce intracellular oxidative stress , and accelerate the induction of apoptosis. Complementing the therapeutic approach based on MMF, the excellent photothermal performance of the MPC in the NIR-II region (1064 nm) is exploited to enable effective hypothermal ablation of the tumours, which can be achieved in deep tissue layers. The proposed multifunctional nanocomposites , together with the demonstrated “double-punch” therapeutic approach, hold significant potential to pave the way for future cutting-edge weapons against the dreadful TNBC.

A tumor microenvironment-activatable CuS@TF nanosystem with intrinsic multi-nanozyme properties for NIR-II photoacoustic imaging-guided synergistic PTT/CDT

Liang Zhixing,Wang Wenyi,Liu Yanfei,Yuan Tao,Sun Jinwei,Li Xuliang,Liu Yan-Cheng,Guo Xiaohuan,Mo Yuxuan,Wen Changchun

The efficacy of single-modal chemodynamic therapy (CDT) is often restricted by the antioxidant scavenging of overexpressed glutathione (GSH) within the tumor microenvironment (TME) and insufficient catalytic kinetics. Meanwhile, traditional photothermal therapy (PTT) also faces bottlenecks of a single therapeutic mechanism and low photothermal conversion efficiency, leading to unsatisfactory overall anti-tumor outcomes. To address these challenges, we constructed a smart-responsive, core-shell multifunctional nanotheranostic platform for near-infrared-II (NIR-II) photoacoustic (PA) imaging-guided synergistic PTT/CDT. This platform utilizes copper sulfide nanoparticles (CuS) as the photothermal core, conformally coated in situ with an Fe-tannic acid (TF) metal-phenolic network (MPN) shell (CuS@TF). In the mildly acidic TME, the TF shell dissociates to expose Fe/Cu bimetallic catalytic centers possessing excellent intrinsic peroxidase-like and catalase-like activities. Driven by the synergistic reduction of endogenous GSH and tannic acid, CuS@TF not only effectively depletes antioxidant GSH but also efficiently modulates H 2 O 2 metabolism, triggering a cascade burst of highly toxic hydroxyl radicals (·OH). Furthermore, under 1064 nm laser irradiation, the highly efficient localized hyperthermia generated by the CuS core not only directly ablates the tumor but also thermodynamically boosts the catalytic kinetics of the nanozymes, achieving profound PTT-sensitized CDT. Systematic in vitro and in vivo evaluations demonstrate that, under the precise navigation of PA imaging, CuS@TF exhibits remarkable tumor eradication efficacy (with a tumor inhibition rate of ~ 96.2%) and excellent in vivo biosafety. This work provides a novel design paradigm for the development of precise synergistic tumor theranostics based on multifunctional nanozymes. Graphical

Mono-crystalline Ge1-xSnxSe micro-sheets with hexagonal morphologies for Visible-NIR photodetectors: Increased carrier concentration, narrowed band gap and improved performances

Rui Zhang,Xue Luo,Feiyu Zhao,Qiang Xu,Yang Xu,Yang Xu,Shuo Chen,Xianping Fan,Xvsheng Qiao

GeSe is a narrow band gap semiconductor with feasible application as infrared photoelectric devices . However, challenges are still maintained to improve their carrier concentration and conductivity for broader scope of photodetector applications. Aiming at such scope, the Ge 1- x Sn x Se (0 <  x  < 0.2) were synthesized via a facile “one-pot” wet chemical method and a lattice site substitution strategy. Compared to the pure GeSe micro-sheets the synthesized Ge 1- x Sn x Se micro-sheets possessed a smaller band gap (0.914 eV) and a better electrical conductivity (2.80 × 10 −5  Ω −1 cm −1 ), where the former exhibit greatly improved photo-current. The Ge 1- x Sn x Se micro-sheet was then assembled into a photodetector device to demonstrate its remarkable photo responsivities in visible to NIR spectral region (3614 A/W at 532 nm and 86 A/W at 1319 nm). The photo-detection range broadening and the responsivity enhancement can be attributed the carrier concentration increment as well as the band gap narrowing. Therefore, the Ge 1- x Sn x Se micro-sheets can be considered as an excellent candidate that can be applied as high performance photodetectors in the visible to near infrared region.

Qing Zhong,Gen Li,Hui Long,Jingxian Zhang,Suiping Deng,Langhuan Huang,Shaozao Tan

Temperature-sensitive hydrogels are always hotspot and have shown great potential in the field of biomedicine. Herein, we report N,N-diethylacrylamide (DEA) temperature-sensitive nanocomposite hydrogels with improved mechanical properties composed of carbon nanotubes (CNT) and glycidyl methacrylate functionalized quaternized chitosan (QCGM), followed by deciphering the synergy between CNT and QCGM for the influence on the properties of the hydrogels. The prepared hydrogels not only present a temperature-stimulus response but also feature in NIR-stimulus response performance due to the addition of CNT. In particular, the DQ0.5C1 hydrogel exhibits the optimal performance, including high mechanical strength and thermal behavior, and was subjected to examine the photothermal conversion ability, as well as loading the model drug ciprofloxacin hydrochloride (CIP) to investigate the drug release and antibacterial ability under different conditions. Moreover, the biocompatibility experiments and the in vivo full-thickness skin defect model results signify that the hydrogels have excellent biocompatibility and also can promote wound closure with accelerated wound contraction and improve the wound-healing process. All in all, the prepared temperature-sensitive nanocomposite hydrogels with improved mechanical properties, high biological safety, and accelerated wound healing have considerable practicability in the fields of drug delivery and wound dressing.

A-site cation engineering of Mn5+-activated ABaPO4 (A=Li, Na, K) phosphors for high-performance NIR-II luminescence and optical thermometry

Yutong Wang,Hang Zhao,Jiayin Niu,Anurak Weahayee,Wanwisa Limphirat,Theeranun Siritanon,Jingyi Ren,Peng Jiang,Yanfei Sun,Tingting Lu

The development of high-performance narrowband near-infrared second window (NIR-II) phosphors remains a critical challenge in the fields of advanced bioimaging and optical thermometry. Herein, a series of Mn 5+ -doped ABaPO 4 (A = Li, Na, K) phosphors are synthesized via conventional solid-state method, and the effect of A-site alkali metal cations on their crystal structure and luminescence properties is comparatively investigated. Mn K -edge X-ray absorption near-edge structure (XANES) spectroscopy unambiguously confirms that Mn ions are predominantly stabilized in the +5 oxidation state and exclusively occupy tetrahedral [PO 4 ] sites in all three hosts. All samples exhibit characteristic narrowband NIR-II emission in the 1160-1200 nm range originating from the 1 E → 3 A 2 spin-forbidden transition of Mn 5+ . The lattice distortion induced by different A-site cations significantly modulates the doping limit, crystal field strength, thermal stability and temperature sensing performance of Mn 5+ ions. All three Mn 5+ -doped hosts exhibit good thermal stability. The fluorescence decay lifetime of LiBaPO 4 : Mn 5+ reaches 772.436 μs＀ NaBaPO 4 : Mn 5+ shows unique energy level splitting and achieves a maximum relative temperature sensitivity of 2.43% K −1 based on the fluorescence intensity ratio technique. KBaPO 4 : Mn 5+ possesses a highly symmetric crystal lattice and a higher doping limit, with a thermal quenching activation energy ( ΔE ) of 0.414 eV. This work not only provides three promising NIR-II phosphors for bioimaging and optical thermometry, but also offers new insights into the research of Mn 5+ -activated phosphate systems.

Bacteria-responsive injectable hydrogel based on quaternized chitosan for real-time infection monitoring and synergistic NIR-II photodynamic/photothermal anti-infection therapy

Wen-Zhen Li,Dongyang Li,Huanhuan Wang,Hongli Zhang,Xipeng Li,Xiaoling Yang,Jiamei Wang,Anning Yang,Qin Zeng,Judun Zheng,Yideng Jiang,Yuhui Liao

Green-Synthesized Scaffolds for Smart Hydrogels. Green-synthesized quaternized chitosan (QCS) represents an ideal macromolecular backbone for constructing functional hydrogels, driven by its structural tunability, excellent aqueous solubility, and intrinsic cationic nature. Unlike native chitosan, the targeted introduction of quaternary ammonium groups endows QCS with a permanent positive charge. This modification not only enhances its degradation but also confers superior antibacterial efficacy and mucoid hesiveness. By leveraging QCS as a versatile scaffold to rationally design polysaccharide-based hydrogels with tailored physicochemical and biological profiles, this work provides a sustainable and highly effective platform for advancing next-generation bioactive materials. • Sol-Gel transition and enhanced antibacterial performance. Macrocyclic host TEBox markedly improves EGCG’s (guest) water solubility and antibacterial effect via host-guest interactions. It also effectively drives the Sol-Gel transition of QCS-EC Sol, finally enabling the preparation of bacteria-responsive multifunctional injectable hydrogel (MQCEC-TEB Gel). This gel has excellent skin adhesion and promotes cell proliferation and tissue regeneration. • Real-Time, Visual Infection Monitoring. Key innovation: a chaotropic effect-based bacteria-responsive agent (TEBox-B 12 ). When exposed to bacteria, TEBox-B 12 undergoes in situ reduction to generate stable organic radicals, causing rapid visible color change (pale yellow to deep purple). This provides a revolutionary tool for early, non-invasive, real-time infection diagnosis. • Synergistic Anti-infection Therapy for Deep Tissue. Radicals from the above reaction not only enable color development but also have strong NIR-II absorption, allowing the hydrogel to achieve deep-tissue photothermal (PTT) and photodynamic (PDT) antibacterial therapy. NIR-II light penetrates deeper into tissue, overcoming traditional phototherapy limitations. This study presents a new smart dressing and "diagnosis-and-therapy integration" strategy, offering a new paradigm for managing invasive infection wounds. Smart hydrogel dressings with sustained wound-healing promotion and infection-monitoring capabilities demonstrate significant potential for invasive infection wound repair. Inspired by mussels, a highly adhesive and multifunctional injectable antibacterial hydrogel dressing (MQCEC-TEB Gel) was developed for wound repair and real-time infection monitoring, comprising a viscous hydrogel scaffold (QCS-EC) and a bacteria-responsive agent (TEBox-B 12 ) crosslinked via host-guest interaction. The epigallocatechin gallate (EGCG) was incorporated into quaternized chitosan (QCS) enhancing the viscosity of QCS and forming the QCS-EC scaffold by hydrogen bonding, providing sustained antibacterial activity and promoting wound-healing. Meanwhile, the bacteria-responsive agent TEBox-B 12 self-assembles via a chaotropic effect between box-shaped quaternized pyridinium salt (TEBox) and the superchaotropic anion B 12 H 12 2− . Upon bacterial exposure, TEBox-B 12 underwent in situ reduction, generating stable organic radicals that trigger a rapid visible color change from pale yellow to deep purple for real-time infection monitoring. The resulting radicals also exhibit strong NIR-II absorption, enabling deep-tissue photothermal (PTT) and photodynamic (PDT) antibacterial therapy. The host-guest interactions between TEBox of TEBox-B 12 and EGCG on QCS-EC improves the water solubility and antibacterial efficiency of EGCG, facilitating Sol-Gel transition to form the injectable MQCEC-TEB Gel platform. This system synergistically integrates host-guest interaction, NIR-II PDT, and PTT to efficiently eradicate deep-tissue bacteria and promote wound healing, offering a novel multimodal strategy for early diagnosis, non-invasive monitoring and treatment of invasive infection wounds.

Multi-omics decodes a defect-interface dual-engineered PtPb@SbO3-x nanozyme for NIR-II photothermal-amplified eradication of drug-resistant pneumonia

Zhangwei Qiu,Danyan Wang,Zijun Jin,Jiaping Han,Xueli Wang,Xiaojun He,Liqin Wu

Managing pneumonia caused by multidrug-resistant (MDR) bacteria presents significant clinical challenges. The near-infrared II (NIR-II) laser irradiance exhibits strong photothermal conversion capabilities, making it a promising candidate for photothermal and chemodynamic therapies as non-antibiotic strategies. However, traditional methods are often hindered by issues such as the uncontrollable production of reactive oxygen species and the low efficiency associated with NIR-II photothermal therapy. This underscores the necessity for precisely regulated and highly effective synergistic therapies. To address these limitations, we have innovatively developed a defect/interface dual-engineered nanozyme (PtPb@SbO 3-x ) and comprehensively characterized its atomic-scale structure and catalytic mechanisms using density functional theory calculations and synchrotron radiation techniques. In vitro experiments demonstrated that PtPb@SbO 3-x could efficiently eliminate drug-resistant bacteria and disrupt biofilm structures under low-concentration hydrogen peroxide and NIR-II irradiation while exhibiting excellent biocompatibility. In pneumonia models, the nanozyme enabled rapid infection clearance and significantly reduced inflammatory responses via synergistic photothermal and chemodynamic therapy effects. Furthermore, integrated multi-omics analyses-including metabolomics, transcriptomics, and proteomics-systematically uncovered the molecular mechanisms driving its therapeutic efficacy. This study successfully establishes a non-antibiotic nanozyme-based therapeutic strategy that is efficient, low in toxicity, and non-invasive for treating MDR bacterial infections. It also provides a solid theoretical basis and technical framework for the rational design of defect/interface dual-engineered nanoplatforms.

A NIR-II Laser-Triggered Nb2C-Arg/Gel Composite Multifunctional Hydrogel with Photothermal, ROS Scavenging, and NO Release for Infected Wound Healing

Mi Liu,Weidan Zhao,Denghao Li,Dingyu Cui,Jiali Liu,Qingao Chen,Yan Zhou,Li Jiang,Huaibin Zhou

Infected wound healing remains a significant clinical challenge due to bacterial resistance and excessive inflammation. Herein, a multifunctional hydrogel dressing (Nb2C-Arg/Gel) is developed that integrates two-dimensional Nb2C nanosheets (photothermal agent) and arginine [nitric oxide (NO) donor] into a calcium ion cross-linked sodium alginate matrix. The hydrogel has a relatively high swelling capacity and good ability to scavenge reactive oxygen species (ROS), which are beneficial for absorbing wound exudate and reducing the inflammatory response of wounds. Under irradiation with a low-power NIR-II laser (1064 nm, 1 W cm–2), Nb2C-Arg/Gel achieves rapid photothermal heating (48.6 °C in 1 min), while thermally enhances arginine decomposition-triggered on-demand NO release. This synergistic mechanism provides superior antimicrobial efficacy against Staphylococcus aureus (97.65%) and methicillin-resistant S. aureus (90.05%). In vivo studies on MRSA-infected murine wounds demonstrate that the residual area on day 10 was 2.9%, which accelerated healing compared to that of the control group (17.8%), accompanied by reduced inflammation and enhanced angiogenesis. The excellent biocompatibility of hydrogel is validated by low hemolysis (<5%) and promoted fibroblast proliferation (125% cell viability). Combining with photothermal, NO-mediated antibacterial action, and ROS scavenging to alleviate oxidative stress, this antibiotic-free hydrogel dressing has great potential for clinical application for the treatment of refractory-infected wounds.

Using Gd-Enhanced β-NaYF4:Yb,Er Fluorescent Nanorods Coupled to Reduced TiO2 for the NIR-Triggered Photocatalytic Inactivation of Escherichia coli

Huang Zhou,Fengjiao He

β-NaYF4:Yb,Er,Gd fluorescent nanorods were successfully coupled to a reduced TiO2(UCNPs@R-TiO2) nanocomposite and applied to visible-light catalytic sterilization under 980 nm near-infrared (NIR) light illumination. The UCNPs (β-NaYF4:Yb,Er,Gd) absorb the NIR light and emit red and green light. The visible light can be absorbed by the R-TiO2(Eg = 2.8 eV) for the photocatalytic reaction. About 98.1% ofEscherichia coliwere effectively killed upon 12 min of NIR light irradiation at a minimum inhibitory concentration (MIC) of 40 μg/mL UCNPs@R-TiO2nanocomposite. The bactericidal properties were further evaluated by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis. We found that the high bactericidal activity was due to the synergistic effect between the UCNPs and R-TiO2. Moreover, the UCNPs show excellent upconversion luminance properties, and the introduction of visible-light-absorbed R-TiO2nanoparticles (2.8 eV) was conducive to the efficient separation and utilization of photogenerated electron-hole pairs.

S-nitrosoglutathione functionalized polydopamine nanoparticles incorporated into chitosan/gelatin hydrogel films with NIR-controlled photothermal/NO-releasing therapy for enhanced wound healing

Wenyu Wang,Huan Sheng,Daihong Cao,Fenglian Zhang,Weifen Zhang,Fang Yan,Dejun Ding,Ni Cheng

Nitric oxide (NO) has aroused wide interest in the treating infected wounds due to its characteristic functionalities. However, its utilization is limited due to its volatile properties, high reactivity, direct potential toxicity, and byproducts of NO donors limited its application. Herein, endogenously NO donor S-nitrosoglutathione (GSNO) was connected covalently to polydopamine nanoparticles (PDA-GSNO NPs) to minimize the loss of NO in aqueous medium. Meanwhile, near-infrared (NIR)-controlled NO release and photothermal therapy (PTT) was obtained through the photothermal conversion by PDA. Then chitosan (CS)/gelatin (GE) biocomposite hydrogel films with preferable biocompatibility, surface hydrophilicity, hydroabsorptivity, and mechanical adhesive properties were constructed. By embedding PDA-GSNO NPs into the films, a multifunctional wound dressing was fabricated. Under NIR light irradiation, the combination of PTT, NO-releasing, and CS antibacterial agents can strengthen the in vitro antimicrobial efficacy and in vivo wound healing activities. Meanwhile, the obtained wound dressing presented good biocompatibility. This work outlines an approach for combating bacterial infections and demonstrating the possibility for synergistic NO-releasing wound healing.

Au/MXene hydrogel with NIR photothermal regulation promotes diabetic wound healing through synergistic induction of reactive oxygen species storms and EGCG release

Yumei Li,Xingjun Zhao,Linna Chang,Yanan Hu,Shibo Xu,Xiuli Ren,Xifan Mei,Zhenhua Chen

Antimicrobial hydrogels with NIR and enhanced mechanics benefit diabetic wound healing. • Au/MXene modifies gel network intermolecular interactions, regulating hydrogel mechanics. • NIR-triggered hyperthermia promotes wound healing via ROS/EGCG release. Biological hydrogels are ideal wound dressings due to their moisturizing, biocompatible, and drug delivery properties. Polyvinyl alcohol (PVA) and sodium alginate (Alg)-based injectable hydrogels show great potential in wound repair; however, simple hydrogels lack antimicrobial properties and cannot effectively prevent and control infections. Herein, Au@Ti 3 C 2 -E composites were obtained by anchoring gold nanoclusters on the surface of ultrathin Ti 3 C 2 nanosheets (1.5 nm) and modifying them by grafting epi gallocatechin gallate (EGCG). Under 808 nm laser irradiation, Au@Ti 3 C 2 -E@H exhibited good photothermal effects. Interestingly, elevated temperature can act as a “switch” that significantly enhances the Fenton-like catalytic activity of gold nanoclusters (3–5 nm) and promotes the release of EGCG grafted on the surface. The inhibition results showed that Au@Ti 3 C 2 -E@H achieved 94.5% and 93.0% kill rates of Methicillin-Resistant Staphylococcus Aureus (MRSA) and Ampr Escherichia coli ( E.coli ) through higher Reactive Oxygen Species (ROS) generation efficiency and disruption of bacterial biofilms. Molecular biology results showed that Au@Ti 3 C 2 -E@H significantly upregulated Vascular Endothelial Growth Factor (VEGF), Basic Fibroblast Growth Factor (bFGF), Epidermal Growth Factor (EGF) and Platelet Endothelial Cell Adhesion Molecule-1 (CD31) nearly threefold and downregulated Tumor Necrosis Factor-α (TNF-α), Interleukin-6 (IL-6), and Matrix Metalloproteinase-9 (MMP-9) nearly fourfold, which could promote angiogenesis and epithelial repair. After 21 days of the treatment cycle, the diabetic rats achieved a wound healing rate of 99.2%. Au@Ti 3 C 2 -E enhances the mechanical properties and antibacterial effects of hydrogels, synergizing photothermal and nanozyme activities to provide a promising solution for the repair of diabetic wounds infected with drug-resistant bacteria.
