PurposeTo screen endophytic Bacillus producing volatile organic compounds (VOCs) with antifungal activity, and to explore their biocontrol properties toward the growth and pathogenicity of Curvularia lunata. Methods Two-sealed-base-plate assays were used to estimate the antifungal activities of Bacillus strains against C. lunata. Conjoint analysis of solid-phase microextraction gas chromatography-mass spectrometry and antagonistic experiments were used to identify the VOCs responsible for the antifungal activity. Effects of individual synthetic VOCs were analyzed along with reactive oxygen species (ROS) accumulation in C. lunata conidia. After exposure to individual VOCs, conidia were also sprayed onto maize leaves to evaluate their pathogenicity. Expression levels of virulence-related genes in C. lunata mycelium following exposure to VOCs were analyzed using quantitative real-time PCR.Results Among the ten endophytic Bacillus strains and two plant growth-promoting rhizobacterial (PGPR) strains, only B. subtilis strain DZSY21 strongly inhibited the growth of C. lunata by producing VOCs. 2-Methylbutyric acid, 2-heptanone, and isopentyl acetate produced by strain DZSY21 showed inhibitory effects on the mycelia growth and conidial sporulation of C. lunata. 2-Heptanone and isopentyl acetate also repressed the germination of conidia and the expression levels of virulence-related genes in C. lunata mycelium. Moreover, isopentyl acetate strongly enhanced the accumulation of intracellular ROS in conidia. The disease indexes of maize leaves sprayed with VOC-treated C. lunata conidia were reduced from 60.52 to 26.64%.Conclusion Endophytic B. subtilis strain DZSY21 displayed the potential to control C. lunata by producing VOCs, especially 2-heptanone and isopentyl acetate.

A phenolic-UDP-glycosyltransferase Bs-PUGT from Bacillus subtilis PI18 was cloned and expressed in Escherichia coli BL21 (DE3). The purified Bs-PUGT could catalyze the glycosylation of tyrosol, 4-hydroxybenzyl alcohol, 2-hydroxybenzyl alcohol, caffeic acid, cinnamic alcohol, ferulic acid, and so on. This enzyme showed a high activity and stability over a broad pH range and was sensitive to temperature. Studies on the kinetic parameters indicated that the affinity of Bs-PUGT to UDP-G ( K m) and its catalytic efficiency ( K cat) increased by 1.5-fold and 1.7-fold, respectively, with the addition of 10 mM Ca 2+ . The most effective glycosylation of caffeic acid catalyzed by whole-cell E. coli /Bs-PUGT was achieved with a molar yield of 78.3% in a system with pH 8.0, 30 °C, 25 g/L sucrose, 10 mM Ca 2+ , and 0.5 g/L substrate concentration. The addition of Ca 2+ increased the molar yield of caffeic acid glucosides and shortened the reaction. This work proposes a strategy for the efficient glycosylation of phenolic compounds by microbe-derived glycosyltransferase assisted by metal ions.

Beneficial plant-associated microbes such as Bacillus species have important potential uses as bioinoculants that reduce dependence on agricultural chemicals. These microbes produce diverse metabolites that enhance plant resilience, but the functions of these compounds and the associated plant hormone pathways remain unclear. Using Bacillus velezensis ES-120, a novel strain isolated from Chenopodium album L., we demonstrate that distinct classes of ES-120 metabolites play different roles. Using Arabidopsis thaliana , we show that non-volatile soluble metabolites produced by ES-120, including lipopeptides identified by reversed-phase high-performance liquid chromatography, exhibit biocontrol activity by inhibiting multiple soil-borne pathogens. In contrast, volatile organic compounds (VOCs) emitted by ES-120 promote plant growth, likely by modulating auxin and brassinosteroid (BR) signalling pathways, with effects sustained across diverse conditions. Headspace solid-phase microextraction coupled with gas chromatography–mass spectrometry identified multiple VOCs produced by ES-120. Individual compounds showed limited activity, suggesting that growth promotion may depend on interactions among multiple VOCs or on an undetected low-abundance compound. In greenhouse trials, ES-120 reduced wilt in tomato ( Solanum lycopersicum ) and increased the biomass of Chinese cabbage ( Brassica rapa var. chinensis ). Thus, B. velezensis ES-120 functions as a dual-purpose bioinoculant: its soluble metabolites suppress soil-borne pathogens and its VOCs enhance plant growth via auxin and BR pathways, highlighting its potential for sustainable agriculture.

Summary Recent investigations have demonstrated that bacteria employ the volatile compounds they produce during interactions with other organisms, such as plants, fungi, nematodes and bacteria. However, studies focused on the antibacterial activity of plant growth-promoting rhizobacteria (PGPR) volatiles against bacterial phytopathogens are still rare. In this study, Bacillus strain D13, which is antagonistic to Xanthomonas oryzae pv. oryzae ( Xoo ), was isolated and screened. Volatile compounds emitted from strain D13 reduced the colony diameter and cell motility of Xoo cultured in divided Petri plates. Transmission electron micrograph analysis showed concentration in cytoplasm and altered surface morphology in the majority of Xanthomonas cells after co-cultivation with strain D13. Transcriptional expression of virulence-associated genes in Xoo was repressed. Based on gas chromatography/mass spectrometry (GC/MS) analysis, 12 volatile compounds specifically produced by strain D13 were identified. Among them, decyl alcohol and 3,5,5-trimethylhexanol inhibited the growth of Xoo at minimum inhibitory amounts of 0.48 and 2.4 mg, respectively. Furthermore, transcriptional expression of virulence-associated genes was also repressed by decyl alcohol and 3,5,5-trimethylhexanol. These results are useful for a better understanding of the biocontrol mechanisms of Bacillus .

An ultraviolet (UV)-based advanced oxidation process (AOP) for disinfecting water is introduced in this paper. This study aimed to evaluate the potential of UV light-emitting diodes (UV-LEDs)/chlorine AOP (UV/Cl 2 ) for Bacillus subtilis spore inactivation. Chlorine was combined with 265 and 280 nm LEDs (UV 265 /Cl 2 , UV 280 /Cl 2 ) and investigated. The addition of 4.0 mg/L of free chlorine at pH 7.0 in the presence of 125 mJ/cm 2 of UV irradiation resulted in an additional 1.8-log reduction in UV 265 /Cl 2 and 1.5-log reduction in UV 280 /Cl 2 . There was no observed enhancement in spore inactivation with the addition of a radical scavenger, t-BuOH, which indicated the role of • OH in the synergistic effect. To quantitatively evaluate the synergism, the primary treatment with UV/Cl 2 was followed by further UV or Cl 2 treatment. After UV/Cl 2 pretreatment at different pH levels, the 265 and 280 nm LEDs treatment enhanced an approximate 0.4–0.5-log reduction compared to UV only, and Cl 2 treatment enhanced an approximate 0.7–1.1-log reduction compared to Cl 2 only. In addition, at pH 7.0, in UV 265 /Cl 2 -Cl 2 and UV 280 /Cl 2 -Cl 2 , the inactivation rate constant k increased by approximately 2 and 1.5 times, respectively. The CT for the lag phases ( CT lag ) reduced to approximately 67 and 58%, respectively. Similar results were obtained at pH 7.5 and 8.0, and in the secondary effluent. The synergistic effect on spore inactivation suggested that the pathogen inactivation efficiency of sequential UV and chlorine disinfection processes, which are commonly applied, can be significantly enhanced by adding chlorine prior to UV treatment .

Pullulanase, a typical debranching enzyme, specifically hydrolyzes α-1,6 glycosidic linkages in pullulan, starch and so on. There is accumulated knowledge about the catalysis of pullulanase, but functional roles of acceptor subsites get less attention. According to the crystal structure of pullulanase from Bacillus subtilis str. 168, stacking interaction between tryptophan437 and glycosyl unit of α-CD was found. Trp437 located in conserved region III was highly conserved in pullulanases. But functional role of this residue remains unclear. Site-directed mutagenesis was used to determine the function of Trp437. Replacement of Trp437 with glycine, phenylalanine, proline, arginine resulted in mutants with undetectable hydrolysis activity and reverse hydrolysis activity. The secondary structure of mutated proteins showed no difference from the wild-type pullulanase, but they lost the capability to bind β-CD based on the ITC measurement. Molecular docking was performed to investigate the affinity of proteins to ligands including maltotriose, 6 2 -α-D-maltotriosyl-maltotriose and α-CD, showing the affinity of mutants to ligands became weaker compared to that of PulA. And the hydrogen bond between Trp437 and Glu525 in PulA only was found in mutant W437R. These results show that Trp437 at subsite +2 plays crucial role in catalysis of PulA, and it has little tolerance to change.

The alkaline protease fromBacillus licheniformisstrain 2709 (AprE 2709) is widely used in Chinese industries but faces stability challenges under high-temperature conditions. This study employed molecular modeling and mutagenesis to identify Asn residues at positions 61, 160, and 211 as key sites affecting the stability of AprE 2709. By leveraging the additive and cooperative effects of mutations, the mutant enzyme AprE 2709 (N61G/N160G/N211G) was engineered, exhibiting enhanced thermostability and catalytic activity. The mutant demonstrated a 2.89-fold increase in half-life at 60 °C and a 1.56-fold improvement in catalytic efficiency compared to the wild-type enzyme. Structural analysis revealed that the improved thermostability was due to altered electrostatic interactions and strengthened hydrophobic contacts. Targeting Asn residues prone to deamidation presents a promising strategy for improving protein heat tolerance. These findings not only enhance our understanding of enzyme stability but also lay a foundation for future research aimed at optimizing alkaline proteases for diverse industrial applications, particularly in high-temperature processes.

This article has been retracted: please see Elsevier Policy on Article Withdrawal ( https://www.elsevier.com/about/our-business/policies/article-withdrawal ). This article has been retracted at the request of the Executive Editor-in-Chief. Concern was raised about the EDX spectra shown by the insets of Figure 1D and the top two XPS spectra of Figure 9A (Fe 0 pH 3.5 and Fe 0 + B. Subtilis pH 3.5), as well as the FTIR spectra from Figure S6 and the XPS spectra from Figure S7B-E, which contained features and noise that indicated data fabrication. The corresponding authors were not able to provide reasonable explanations for these features and the Editor decided to retract the article. Moreover, the top EDX spectrum shown by the insets of Figure 1D is highly similar to the spectrum shown by the inset of Figure 1C of the article that the authors have previously published in the Journal of Hazardous Materials 280 (2014) 399–408 http://dx.doi.org/10.1016/j.jhazmat.2014.08.023 . One of the conditions of submission of a paper for publication is that authors declare explicitly that their work is original and has not appeared in a publication elsewhere. Re-use of any data should be appropriately cited. As such this article represents an abuse of the scientific publishing system. The scientific community takes a very strong view on this matter and apologies are offered to readers of the journal that this was not detected during the submission process.

For a broader understanding of uranium migration affected by microorganisms in natural anaerobic environment, the bioreduction of uranium(Ⅵ) (U(Ⅵ)) was revealed in Bacillus thuringiensis , a dominant bacterium strain with potential of uranium-tolerant isolated from uranium contaminated soil. The reduction behavior was systematically investigated by the quantitative analysis of U(Ⅳ) in bacteria, and mechanism was inferred from the pathway of electron transmission. Under anaerobic conditions, appropriate biomass and sodium lactate as electron donor, reduction behavior of U(Ⅵ) induced by B. thuringiensis was restricted by the activity of lactate dehydrogenase, which was directly affected by the initial pH, temperature and initial U(Ⅵ) concentration of bioreduction system. Bioreduction of U(Ⅵ) was driven by the generation of nicotinamide adenine dinucleotide (NADH) from enzymatic reaction of sodium lactate with various dehydrogenase. The transmission of the electrons from bacteria to U(Ⅵ) was mainly supported by the intracellular NADH dehydrogenase-ubiquinone system, this process could maintain the biological activity of cells.

Microorganisms can produce many antibiotics against bacteria and fungi, which have been used as a potential choice of new antibiotics. In this paper, we studied the characteristics of antibacterial substances by Bacillus cereus BC1. The results showed that the acid-precipitated substance played the main role in antibacterial activity, and further characterization indicated that the antibacterial substance might be a lipopeptide substance. Then the antibacterial spectrum suggested that the antibacterial substance had an inhibitory effect on Gram-positive bacteria and fungi, while selenium-riched antibacterial substance of Bacillus cereus BC1 could significantly enhance the inhibition. Then the morphological effects of the antibacterial substance to indicator bacteria were determined. The effects of different treatment methods on the stability of antibacterial substances were studied and the results showed that the antibacterial substance was stable to heat, ultrasonic, and ultraviolet treatment, and their antibacterial activity would not be greatly affected. However, they were sensitive to pepsin. The optimum pH range of antibacterial activity was 3–5. This study may contribute to reusing the fermentation supernatant often discarded in the previous fermentation process. At the same time, the lipopeptide antibacterial substance extracted from the fermentation broth of selenium-enriched Bacillus cereus BC1 can be used in the development of antibiotics and biopesticides, and open up a new way for the control of plant diseases.

Microbial degradation of chlortetracycline (CTC) is an effective bioremediation method. In the present study, an enrichment technique was used to isolate a Bacillus cereus LZ01 strain capable of effectively degrading CTC from cattle manure. Response surface methodology was used to identify optimized conditions under which strain LZ01 was able to achieve maximal CTC removal (83.58%): temperature of 35.77 °C, solution pH of 7.59, CTC concentration of 57.72 mg/L and microbial inoculum of 0.98%. The antibacterial effect of CTC degradation products on Escherichia coli was investigated by the disk diffusion test, revealing that the products by LZ01 degradation of CTC exhibited lower toxicity than parent compound. Shake flask batch experiments showed that the biodegradation of CTC was a synergistic effect of intracellular and extracellular enzymes, and intracellular enzyme had a better degradation effect on CTC (77.56%). Whole genome sequencing revealed that genes associated with ring-opening hydrolysis, demethylation, deamination and dehydrogenation in strain LZ01 may be involved in the biodegradation of CTC. Subsequent seven possible biodegradation products were identified by LC-MS analyses, and the biodegradation pathways were proposed. Overall, this study provides a theoretical foundation for the characterization and mechanism of CTC degradation in the environment by Bacillus cereus LZ01.

Doxycycline hydrochloride (DCH) could be continuously removed by Bacillus thuringiensis S622 with the in-situ biogenic manganese oxide (BioMnOx) via oxidizing/regenerating. The DCH removal rate was significantly increased by 3.01-fold/1.47-fold at high/low Mn loaded via the integration of biological (intracellular/extracellular electron transfer (IET/EET)) and abiotic process (BioMnOx, Mn(III) and •OH). BioMnOx accelerated IET via activating coenzyme Q to enhance electrons transfer (ET) from complex I to complex III, and as an alternative electron acceptor for respiration and provide another electron transfer transmission channel. Additionally, EET was also accelerated by stimulating to secrete flavins, cytochrome c (c-Cyt) and flavin bounded with c-Cyt (Flavins & Cyts). To our best knowledge, this is the first report about the role of BioMnOx on IET/EET during antibiotic biodegradation. These results suggested that Bacillus thuringiensis S622 incorporated with BioMnOx could adopt an alternative strategy to enhance DCH degradation, which may be of biogeochemical and technological significance.
