Baicalin-Ternary LDH-Modified Magnesium Alloy with Anti-Corrosion and pH-Responsive Controlled Release, Near-Infrared-Enhanced Catalytic Property

Yuhan Liang,Lijie Chen,Mingyue Feng,Tong Zhang,Rongbang Sun,Yang Liu,Yifu Fu,Yunxiang Chen,Lan Chen

HighlightsWhat are the main findings? The Mg-Mn-Fe LDH grown in situ on the PEO coating-modified Mg alloy effectively blocks pores and introduces metal catalytic active sites.Baicalin achieves stable immobilization by forming coordination bonds with the metal ions in LDH through the oxygen-containing functional groups. The LDH/BA composite coating significantly improved corrosion resistance, along with pH-responsive drug release and near-infrared-enhanced enzyme-like catalytic performance. What are the implications of the main findings?A multi-functional modification strategy integrating “corrosion resistance, drug loading and control release” was established, which provides a novel technical solution for the long-term corrosion protection of magnesium alloys. The revealed interfacial coordination immobilization mechanism can provide theoretical references for the structural design of organic–inorganic composite protective coatings. The synergy of corrosion resistance and microenvironment stimulus-responsive functions is realized, which expands the application of LDH coatings in the modification of metal implants.Aiming at the rapid corrosion of degradable magnesium alloys in physiological environments and the insufficient long-term protection of single plasma electrolytic oxidation (PEO) coatings, Mg-Mn-Fe layered double hydroxide (LDH) and baicalin (BA)-loaded LDH/BA composite coatings were prepared in situ on PEO-pretreated ZE21C magnesium alloy substrates. BA was stably anchored on the LDH surface via coordination bonds between its oxygen-containing functional groups and laminate metal sites. Benefiting from the physical barrier of the LDH lamellar structure and the corrosion inhibition effect of baicalin, the LDH/BA coating significantly improved the corrosion resistance of the magnesium alloy matrix. The composite coating exhibited peroxidase-like catalytic activity for reactive oxygen species generation, which could be enhanced by near-infrared irradiation. It also possessed stable photothermal conversion performance and pH-responsive drug release behavior under acidic conditions. Biological characterization demonstrated that BA-loaded LDH composite coatings exert potent inhibitory effects on 143B cell proliferation. This work integrates long-term corrosion resistance, controlled drug release, and photoresponsive catalytic functions onto magnesium alloy surfaces, providing an effective strategy for developing high-performance biodegradable magnesium alloys.

The Effects of Baicalin in Combination with Cefotaxime on the Biofilm and Metabolic Reprogramming of Multidrug-Resistant Pseudomonas aeruginosa

Xin Meng,Chao Ning,Xinyu Lu,Mengna Kang,Yuxuan Yang,Zhiyun Yu,Yu Wang,Yantong Sun,Haiyong Guo

Baicalin, a natural plant-derived compound, holds promise in addressing clinical bacterial resistance when combined with antibiotics. This study evaluated the antibacterial activity of the combination of baicalin and cefotaxime and explored its mechanism of action on the cell wall and biofilm of multidrug-resistantPseudomonas aeruginosa(MRPA). The results showed that the combination of baicalin and cefotaxime exerted a synergistic inhibitory effect on the growth of MRPA, with a fractional inhibitory concentration index (FICI) of 0.28. Mechanistically, compared with cefotaxime alone, the combination of baicalin and cefotaxime enhanced the permeability of the cell membrane and cell wall of MRPA, thereby increasing cell damage. It also exhibited stronger antibiofilm activity by inhibiting numerous virulence factors (pyocyanin, elastase, lectin), reducing cellular metabolic activity, and downregulating the expression of biofilm genes (pslA,pelA,algD) and quorum-sensing genes (lasl, lasR,rhll, rhlR, pqsA, pqsR). The molecular docking results revealed that baicalin could stably bind to wbpE, LasR, and RhlR. Therefore, this interaction may indirectly influence the processes related to antibiotic resistance and biofilm formation in bacterial cells. Metabolomic analysis revealed that the combination of baicalin and cefotaxime upregulated 863 metabolites and downregulated 587 metabolites. These metabolites mainly included amino acids, lipids, nucleotides, carbohydrates, and secondary metabolites. The combination primarily enriched key pathways such as amino acid metabolism, lipid metabolism (sphingolipid metabolism) and secondary metabolite biosynthesis. Through these pathways, it triggers significant metabolic reprogramming, thereby interfering with the supply of cell wall synthesis precursors, membrane structural stability, and the generation of biomembrane matrix. Ultimately, it synergistically enhances the effects of cell wall damage and biomembrane inhibition. In conclusion, this study confirms that the combination of baicalin and cefotaxime exerts significant synergistic antibacterial activity against MRPA. It also reveals the mechanism of action of the combination on the cell wall and biofilm of MRPA at the metabolic level, providing theoretical support for the development of novel strategies to combat MRPA.

Effect of Hepatic Impairment on the Pharmacokinetics of Baicalin in Rats: Critical Roles of Gut Microbiota and Hepatic Transporters

Ping Li,Yihua Tian,Hong Wang,Yuting Ji,Huiying Zeng,Shengman Zhang,Xiuli Gao,Xiaoyan Chen

Background: Baicalin (BG) has been used in the treatment of many diseases. However, the effect of hepatic insufficiency on its pharmacokinetics has not been reported, and there is a lack of clinical guidance for the use of BG in patients with hepatic impairment. Methods: Carbon tetrachloride (CCl4)-induced rat models were used to simulate hepatic failure patients to assess the effect of hepatic impairment on the pharmacokinetics and distribution of BG. In vitro metabolism and transporter studies were employed to elucidate the potential mechanisms. Results: After intragastric administration of 10 mg/kg of BG, the peak plasma concentration and exposure (AUC0–t) of BG decreased by 64.6% and 52.6%, respectively, in CCl4-induced rats. After intravenous administration, the AUC0–tdecreased by 73.6%, and unlike in the control group, the second absorption peak of BG was not obvious in the concentration–time curve of CCl4-induced rats. The cumulative excretion of BG in the feces increased, but that in the bile decreased. In vivo data indicated that the absorption and enterohepatic circulation of BG were affected. In vitro studies found that the hydrolysis of BG to the aglycone baicalein decreased significantly in the intestinal tissues and contents of the CCl4-induced rats. And BG was identified as a substrate for multiple efflux and uptake transporters, such as breast cancer resistance protein (BCRP) and multidrug resistance-associated proteins (MRPs), organic anion transporting polypeptides (OATP1B1, 1B3, 2B1), and organic anion transporters (OATs). The bile acids accumulated by liver injury inhibited the uptake of BG by OATPs, especially that by OATP2B1.Conclusions: Hepatic impairment reduced BG hydrolysis by intestinal microflora and inhibited its transporter-mediated biliary excretion, which synergistically led to the attenuation of the enterohepatic circulation of BG, which altered its pharmacokinetics.

Zwitterionic polymers grafting of metal organic framework encapsulated boronic acid carbon dots as antibiofouling fluorescent probe for baicalin monitoring

Xiaofan Tang,Yinhai Li,Weilong Zhao,Chunmiao Bo

The sensitivity and accuracy of fluorescence probes for biological samples are affected by not only interfering molecule compounds but also the nonspecific adsorption of proteins and other macromolecules . Herein, fluorescence probe based on zwitterionic sulfobetaine methacrylate polymer (PSBMA) as an antibiofouling layer and amino boric acid carbon dots encapsulated in the metal-organic framework UiO-66-NH 2 (UiO-66-NH 2 /BN-CDs) as a target recognition site was designed for the detection of baicalin (BAI). Owing to the introduction of BN-CDs into UiO-66-NH 2 with high specific surface area , the prepared UiO-66-NH 2 /BN-CDs@PSBMA probe exhibited a high adsorption capacity of 78.9 mg g −1 , while presented fluorescence enhancing and superior fluorescence selectivity to BAI at excitation and emission wavelengths of 400 and 425 nm, respectively. Connecting PSBMA with good hydrophilicity to UiO-66-NH 2 , resulted in an anti-protein capacity of over 96.3 %, effectively inhibiting protein interference with the fluorescence signal. By virtue of its good antibiofouling and recognizing capacities, the fluorescence probe exhibited a satisfactory detection range of 10–80 nmol L −1 , with a fairly low detection limit of 0.0064 μmol L −1 . Using the method to detect BAI in Goji berry , Sophora and Yinhuang oral solution, demonstrating its potential for the accurate and quantitative detection of BAI in complex biological samples.

Unveiling Anti-Diabetic Potential of Baicalin and Baicalein from Baikal Skullcap: LC–MS, In Silico, and In Vitro Studies

Wencheng Zhao,Huizi Cui,Kaifeng Liu,Xiaotang Yang,Shu Xing,Wannan Li

Type 2 diabetes mellitus (T2DM) is marked by persistent hyperglycemia, insulin resistance, and pancreatic β-cell dysfunction, imposing substantial health burdens and elevating the risk of systemic complications and cardiovascular diseases. While the pathogenesis of diabetes remains elusive, a cyclical relationship between insulin resistance and inflammation is acknowledged, wherein inflammation exacerbates insulin resistance, perpetuating a deleterious cycle. Consequently, anti-inflammatory interventions offer a therapeutic avenue for T2DM management. In this study, a herb called Baikal skullcap, renowned for its repertoire of bioactive compounds with anti-inflammatory potential, is posited as a promising source for novel T2DM therapeutic strategies. Our study probed the anti-diabetic properties of compounds from Baikal skullcap via network pharmacology, molecular docking, and cellular assays, concentrating on their dual modulatory effects on diabetes through Protein Tyrosine Phosphatase 1B (PTP1B) enzyme inhibition and anti-inflammatory actions. We identified the major compounds in Baikal skullcap using liquid chromatography–mass spectrometry (LC–MS), highlighting six flavonoids, including the well-studied baicalein, as potent inhibitors of PTP1B. Furthermore, cellular experiments revealed that baicalin and baicalein exhibited enhanced anti-inflammatory responses compared to the active constituents of licorice, a known anti-inflammatory agent in TCM. Our findings confirmed that baicalin and baicalein mitigate diabetes via two distinct pathways: PTP1B inhibition and anti-inflammatory effects. Additionally, we have identified six flavonoid molecules with substantial potential for drug development, thereby augmenting the T2DM pharmacotherapeutic arsenal and promoting the integration of herb-derived treatments into modern pharmacology.

Caffeic acid-Fe3+ nanohybrids loaded with berberine/baicalin for NIR-activated antibacterial therapy via ROS generation and photothermal effects

Meixuan Zhang,Qin Zhang,Xuemin Mu,Xiaolin Wang,Shuhui Zhang,Wenyi Zhao,Xiaoming Song,Lulu Pan,Yanyan Li

The escalating threat of bacterial infections underscores the pressing need for innovative antibacterial solutions. In this study, BA-BBR@MPN nanoparticles (BA-BBR@MPN NPs) were synthesized through the self-assembly of berberine (BBR) and baicalin (BA), followed by modification with a metal-phenolic network (MPN) derived from caffeic acid and Fe 3+ . In the slightly acidic environment of the infected site, the MPN decomposes to release iron ions, BBR, and BA. The iron ions catalyze the generation of hydroxyl radicals (·OH) from hydrogen peroxide (H 2 O 2 ), enabling chemodynamic therapy (CDT) while simultaneously depleting glutathione levels. When combined with 808 nm laser-induced photothermal therapy (PTT), the system demonstrates synergistic antibacterial effects. Both in vitro and in vivo studies confirmed the platform's broad-spectrum antibacterial activity against Gram-positive and Gram-negative bacteria, along with excellent biocompatibility and significant wound-healing promotion. This multifunctional nanoplatform represents a promising strategy for combating bacterial infections while simultaneously promoting tissue repair.

Baicalin Promotes Recovery Following Intracerebral Hemorrhage by Inhibiting Ferroptosis in Brain Tissue Through Modulation of AKT/Nrf2/GPX4 Axis

Zhenfei Yu,Xiaoling Qian,Weihua Wu,Meiqi Zhang,Ying Li

Intracerebral hemorrhage (ICH) is a serious acute cerebrovascular disease with a high death and disability rate. Baicalin plays a neuroprotective role in various diseases, but its regulatory mechanism on ICH remains unclear. In this study, we investigated the protective effects and mechanisms of baicalin in ICH using an ICH mouse model. ICH mouse model was established by injection of collagenase type IV into intracranial in C57BL/6 mice. Neurological function was evaluated by neurological severity scores and the rotarod test. Hemorrhagic foci of brain was evaluated by TTC and hematoxylin−eosin staining. Iron ion deposition in brain was detected by Prussian blue staining. Ferroptosis was evaluated by measuring expression of FTH-1, SLC7A11, GPX4, and TFRC, as well as detecting iron content and levels of glutathione (GSH) and malondialdehyde (MDA). GPX4 expression and apoptosis of brain were detected by immunofluorescence staining and TUNEL assay. Results showed that baicalin improved neurological function and reduced the area of hemorrhagic foci of brain in ICH mouse model. Baicalin decreased iron ion deposition, inhibited ferroptosis and apoptosis, and upregulated GPX4 in brain of ICH mouse model. Moreover, baicalin increased AKT1 phosphorylation and the protein level of Nrf2 in brain of ICH mouse model. Notably, AKT1 inhibitor LY294002 and Nrf2 inhibitor reversed the effects of baicalin on the activation of AKT1/Nrf2/GPX4 axis and the inhibition of ferroptosis in brain of ICH mouse model. Collectively, we demonstrated that baicalin promotes ICH recovery by inhibiting ferroptosis in brain tissue through activation of AKT/Nrf2/GPX4 axis. These results may provide new insights for the study of baicalin in the treatment of ICH.

Solubility and bioavailability enhancement of baicalin by modified citrus pectin: preparation, in silico, in vitro and in vivo evaluation

Wenxin Ma,Fangling Du,Tongcheng Xu,Xiuxiu Wang,Baorui Li,Hao Yue,Min Jia,Aizhen Zong

Baicalin, a flavonoid compound with various health benefits, faces significant limitations in its application due to poor solubility. Identifying a nontoxic, biodegradable, and readily available solubilizer is therefore critical for improving the solubility of baicalin and related analogues. This study aimed to evaluate the potential of citrus pectin and its acid-hydrolyzed derivatives to improve baicalin solubility, elucidating the underlying mechanisms of solubilization. Three fractions, MCP-1, MCP-2, and MCP-3, were obtained via partial acid hydrolysis followed by anion exchange chromatography. The results demonstrated that MCP-3, composed of 91.2 % galacturonic acid and 8.8 % rhamnose, exhibited the highest efficacy in enhancing baicalin solubility. Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), thermodynamic, molecular dynamics (MD) simulation confirmed that MCP-3 adopted a helical structure in solution, encapsulating baicalin within its helical cavity through hydrogen bonding between carboxyl and hydroxyl groups, as well as hydrophobic interactions and van der Waals forces. The MCP-3/baicalin complex exhibited good pH and temperature stability. Pharmacokinetic studies demonstrated that MCP-3 significantly enhanced the in vivo bioavailability of baicalin by 1.95-fold, and the complex exhibited low cytotoxicity. This study offers a promising strategy for the development of more effective flavonoid-based products.

Synergistic passivation of black phosphorene with both silver nanoparticles and rice husk biochar for enhanced electrochemical detection of baicalin

Xiangrong Zeng,Zhongshu Duan,Yifu Zhu,Xiaoting Lei,Zhenzong Fan,Hongyu Xiao,Yangping Wen

Recent advancements in two-dimensional materials have highlighted black phosphorene (BP) as a promising candidate for optical and electrical applications. However, its practical utility is hindered by ambient instability and limited conductivity. This study aims to solve these problems with a surface passivation strategy. Silver nanoparticles (AgNPs) and rice husk biochar (RHB) are combined to prepare a BP-based nanocomposite (RHB-BP-AgNPs). AgNPs effectively occupy oxidation sites on BP’s surface, mitigating degradation while enhancing conductivity with RHB synergy. The morphology, microstructure and chemical composition of RHB-BP-AgNPs nanocomposite was comprehensively characterized by diversified techniques. The RHB-BP-AgNPs exhibited exceptional ambient stability (>16 days) and superior electrocatalytic activity, enabling its application as a sensing platform for baicalin detection. The sensor demonstrated a wide linear response (0.007–8.0 μM), a low detection limit (5.45 nM), and high selectivity against interferents. Validation in real pharmaceutical samples yielded satisfactory recovery rates of 95.4–104.6 % with RSD from 1.27 to 2.13, underscoring practical reliability. This work not only presents a robust strategy to stabilize and functionalize BP but also advances rapid, sensitive detection methodologies for bioactive compounds in traditional medicine, bridging material innovation with analytical applications.

Treating Bacterial Keratitis during Sleep through Placing Hydrogel Beads with Sustained Tobramycin/Baicalin Delivery Capability at the Palpebra Inferior

Shiyi Song,Zhiwei Ye,Dadong Jia,Mingshuang Li,Changying Yang,Mian Qin,Kaili Wu,Jiabing Ran,Liang Liang

Simultaneously restricting bacterial proliferation while mitigating inflammatory reactions and excessive reactive oxygen species (ROS)-mediated corneal damage during sleep represents a crucial therapeutic strategy for managing bacterial keratitis (BK). In this study, we developed micron-scale PCBT hydrogel beads for nocturnal BK treatment by integrating two classical ophthalmic drugs: tobramycin (TOB) as the antibacterial component and baicalin (BA) as the anti-inflammatory/antioxidant agent. The formation of hydrogel beads was facilitated by the synergistic interplay between: (i) TOB solution-induced crystallization of chitosan (CS) chains and (ii) benzene-1,4-diboronic acid (1,4-BDBA) mediated ester crosslinking of polyvinyl alcohol (PVA) chains. TOB and BA were encapsulated within the hydrogel beads, with BA molecules grafted onto PVA chains via reversible phenylboronic acid ester bonds. When applied to the inferior palpebra, mechanical friction easily transformed the beads into a smooth/soft paste that sustainedly released therapeutic concentrations of TOB and BA for over 24 hours in simulated BK microenvironments. Inhibition zone assays, macrophage (Mφ) polarization analysis, and ROS scavenging tests collectively demonstrated the simultaneous antibacterial, anti-inflammatory and anti-oxidative properties of the hydrogel beads. A Staphylococcus aureus (S. aureus) infected cornea model in rabbits was employed to assess the clinical therapeutic potential of the hydrogel beads against BK during the sleep phase. The results of visual scoring, H&E staining, Masson staining, etc. indicated that the PCBT hydrogel beads exhibited superior therapeutic efficacy compared to conventional commercial eye drops, highlighting their promising application for BK treatment during nocturnal periods.

Glycyrrhizin micellar nanocarriers for topical delivery of baicalin to the hair follicles: A targeted approach tailored for alopecia treatment

Quanfu Zeng,Zhuxian Wang,Zhaoming Zhu,Yi Hu,Yuan Wang,Yaqi Xue,Yufan Wu,Yinglin Guo,Peiyi Liang,Hongkai Chen,Zeying Zheng,Chunyan Shen,Cuiping Jiang,Hongxia Zhu,Qun Shen,Yankui Yi,Hui Li,Zhijun Yang,Li Liu,Qiang Liu

Alopecia affected approximately 16.6% of all people in China, however, treatment options remain limited due to the side effects. Plant bioactive compound baicalin (BC) possesses hair growth-promotion activity, but poor water solubility and unsuitable log P value restrict its topical application, and natural Glycyrrhizin (GL) can exactly overcome these drawbacks. Here, BC was encapsulated in GL to form GL-BC micelles for alopecia treatment. Simultaneously, tween 80 (TW) as carriers was incorporated in the GL-BC to form GL-TW-BC micelles. The topical penetration, penetration pathways, cellular uptake and the underlying mechanisms behind the hair loss reconstruction of the GL micelles were investigated. We found the optimal GL-BC and GL-TW-BC formulations significantly improved the penetration and accumulation of BC in the porcine skin predominantly through the hair follicles pathways without causing skin irritation , which resulted in a targeted treatment. The proliferation of human dermal papilla cells (hDPCs) and effective cellular uptake was also enhanced. Moreover, the activation of the Wnt/β-catenin pathway, up-expression of vascular endothelial growth factor (VEGF), α-melanocyte-stimulating hormone (α-MSH) and interleukin-10 (IL-10) were the mechanisms of micelles for the hair recovery. Interestingly, GL and BC exhibited a synergistic treatment of alopecia. Collectively, GL-BC and GL-TW-BC can be used as promising approaches for the treatment of alopecia.

ROS-responsive multifunctional hyaluronic acid/gelatin based adhesive hydrogel encapsulating baicalin and cerium dioxide nanozymes accelerates infected diabetic wounds healing

Wei Lu,Yanming Zhao,Hao Wen,Jianxin Liu,Yulong Niu,Zhenwei Yang,Yongwei Su,Shuai Li,Nan Xia,Quan Chen

A multifunctional adhesive hydrogel with outstanding ROS-responsive release of baicalin was prepared. • The HA/Gel@CeO₂@BA hydrogel achieved dual immunomodulatory and reparative functionality. • The HA/Gel@CeO₂@BA hydrogel exhibited broad-spectrum antimicrobial efficacy • The HA/Gel@CeO₂@BA hydrogel effectively suppresses inflammation, promotes the healing of infected diabetic wounds. The management of chronic wounds in diabetic patients continues to pose a substantial clinical challenge, especially when complicated by methicillin-resistant Staphylococcus aureus (MRSA) infection, which exacerbates inflammation and impedes the healing process. To address this, we developed a Reactive Oxygen Species(ROS)-responsive, self-healing multifunctional hydrogel (HA/Gel@CeO₂@BA) incorporating baicalin (BA) and cerium dioxide (CeO₂) nanozymes with superoxide dismutase and catalase activities to synergistically provide antibacterial, antioxidant, anti-inflammatory, and pro-regenerative effects. The hydrogel comprises a dual-network structure: (1) a dynamic Schiff base-crosslinked network formed between hydrazine-modified hyaluronic acid (HA-DTPH) (containing disulfide bonds) and aldehyde-modified HA (HA-CHO), enabling tissue adhesion through amino-aldehyde bonding; and (2) a secondary network of gelatin methacrylate (GelMA) and acryloylated cyclodextrin, which anchors hydrophobic BA for sustained release. Dynamic Schiff base bonds and disulfide bonds undergo diabetes-responsive degradation via pathways involving reactive oxygen species, enabling drug release in response to the local environment. In vitro, HA/Gel@CeO₂@BA was found to scavenge ROS in macrophages, promote M2 polarization and enhance angiogenesis. It also exhibited strong antibacterial effects against Escherichia coli and S. aureus . In a diabetic rat model of infected wounds, the hydrogel significantly reduced the bacterial load and accelerated the healing process without the need for external intervention. This strategy combines ROS-triggered drug release and multifunctional therapeutic effects with biodegradability, demonstrating its high potential for clinical translation in the treatment of infected chronic wounds.
