Fabrication of HA/PEI-functionalized carbon dots for tumor targeting, intracellular imaging and gene delivery

M. Zhang,X. Zhao,Z. Fang,Y. Niu,J. Lou,Y. Wu,S. Zou,S. Xia,M. Sun,F. Du

Carbon quantum dots (CDs) as emerging carbon nano-materials have attracted tremendous attention in biomedical fields due to unique properties. In this study, hyaluronate (HA) and polyethylenimine (PEI) functionalized carbon dots (HP-CDs) were synthesized by a facile bottom-up method for tumor targeting and gene delivery. After HA modification, the HP-CDs exhibited superior dispersibility in water and good biocompatibility and were internalized readily into the cytoplasm of cancer via HA-receptor mediated endocytosis. Meanwhile, the HP-CDs with PEI functionalization were shown to have excellent gene condensation capability via electrostatic attraction and protective capacity by preventing nuclease degradation. By virtue of good photoluminescence properties, the HP-CDs as a gene carrier were successfully applied to intracellular imaging and gene delivery. Taken together, the resultant HP-CDs displayed great potential in tumor targeting, intracellular imaging and gene delivery.

Chitosan/Al2O3-HA nanocomposite beads for efficient removal of estradiol and chrysoidin from aqueous solution

Long Li,Jibran Iqbal,Ying Zhu,Fang Wang,Feiyu Zhang,Wanchao Chen,Ting Wu,Yiping Du

Alumina, as a support material, was loaded together with chitosan and hydroxyapatite to form chitosan/Al 2 O 3 -HA composite beads and was used for estradiol and chrysoidin removal from aqueous solution in the present work. The physicochemical properties of the beads were studied with Scanning Electron Microscopy (SEM), Fourier Transform Infrared Spectrometry (FTIR), thermogravimetric analysis (TGA) and Brunauer-Emmett-Teller (BET) surface area analysis. FTIR spectra confirmed that the chitosan was loaded successfully on Al 2 O 3 -HA, and functional groups were immobilized onto the surface of the beads after the synthesis. The adsorption condition including pH, the amount of adsorbent, initial concentration and time were evaluated during the batch experiments. Isotherm data best matched the Langmuir model and the pseudo-second-order model best described the adsorption kinetics . The maximum adsorption capacity was found to be 39.78 mg/g and 23.26 mg/g for estradiol and chrysoidine, respectively. The adsorbed estradiol and chrysoidin were completely eluted from the composite beads with the eluent of 0.1 M H 2 SO 4 /MeOH and the regenerated material was used in several cycles without deterioration in its initial performances. This study suggests that the developed composite beads have high potential for the efficient removal estradiol and chrysoidin from aqueous solution.

Sequential gastrodin release PU/n-HA composite scaffolds reprogram macrophages for improved osteogenesis and angiogenesis

Limei Li,Qing Li,Li Gui,Yi Deng,Lu Wang,Jianlin Jiao,Yingrui Hu,Xiaoqian Lan,Jianhong Hou,Yao Li,Di Lu

Wound healing is a highly orchestrated process involving a variety of cells, including immune cells. Developing immunomodulatory biomaterials for regenerative engineering applications, such as bone regeneration, is an appealing strategy. Herein, inspired by the immunomodulatory effects of gastrodin (a bioactive component in traditional Chinese herbal medicine), a series of new immunomodulatory gastrodin-comprising biodegradable polyurethane (gastrodin-PU) and nano-hydroxyapatite (n-HA) (gastrodin-PU/n-HA) composites were developed. RAW 264.7 macrophages, rat bone marrow mesenchymal stem cells (rBMSCs), and human umbilical vein endothelial cells (HUVECs) were cultured with gastrodin-PU/n-HA containing different concentrations of gastrodin (0.5%, 1%, and 2%) to decipher their immunomodulatory effects on osteogenesis and angiogenesis in vitro . Results demonstrated that, compared with PU/n-HA, gastrodin-PU/n-HA induced macrophage polarization toward the M2 phenotype, as evidenced by the higher expression level of pro-regenerative cytokines (CD206, Arg-1) and the lower expression of pro-inflammatory cytokines (iNOS). The expression levels of osteogenesis-related factors (BMP-2 and ALP) in the rBMSCs and angiogenesis-related factors (VEGF and BFGF) in the HUVECs were significantly up-regulated in gastrodin-PU/n-HA/macrophage-conditioned medium. The immunomodulatory effects of gastrodin-PU/n-HA to reprogram macrophages from a pro-inflammatory (M1) phenotype to an anti-inflammatory and pro-healing (M2) phenotype were validated in a rat subcutaneous implantation model. And the 2% gastrodin-PU/n-HA significantly decreased fibrous capsule formation and enhanced angiogenesis. Additionally, 2% gastrodin-PU/n-HA scaffolds implanted in the rat femoral condyle defect model showed accelerated osteogenesis and angiogenesis. Thus, the novel gastrodin-PU/n-HA scaffold may represent a new and promising immunomodulatory biomaterial for bone repair and regeneration.

NIR-II-Responsive CeO2–x@HA Nanotheranostics for Photoacoustic Imaging-Guided Sonodynamic-Enhanced Synergistic Phototherapy

Jinling Li,Hui-Ling Peng,Changchun Wen,Peijing Xu,Xing-Can Shen,Cunji Gao

The therapeutic effect of photothermal therapy (PTT) and photodynamic therapy (PDT) is severely limited because of the shallow tissue penetration depth of the first near-infrared (NIR-I) light. Multifunctional nanotheranostics irradiated by the second near-infrared (NIR-II) light have received wide interest with respect to deeper tissue penetration, and sonodynamic therapy (SDT) synergistic phototherapy can achieve the complete elimination of tumors. Herein, we successfully constructed a single NIR-II light-induced nanotheranostic using cerium oxide (CeO2–x) with abundant oxygen vacancies for photoacoustic imaging-guided SDT-enhanced phototherapy for the first time. CeO2–x with surface crystalline disorder showed extensive NIR-II region absorption and an outstanding photothermal conversion ability. In addition, the CeO2–x layer with numerous oxygen defects can promote the separation of holes and electrons by ultrasound irradiation, which can remarkably enhance the efficacy of phototherapy to achieve high-efficiency tumor ablation. CeO2–x was surface modified with hyaluronic acid (HA) to prepare CeO2–x@HA to allow active tumor targeting efficiency. Both cell and animal experiments confirmed that all-in-one CeO2–x@HA exhibited a high therapeutic efficacy of SDT-enhanced PDT/PTT under 1064 nm laser irradiation, which achieved complete tumor eradication without systemic toxicity. This study significantly broadened the application of NIR-II-responsive CeO2–x for photoacoustic imaging-mediated SDT-enhanced phototherapy to the highly efficient and precise elimination of tumors.

3D Printing and Performance Study of Porous Artificial Bone Based on HA-ZrO2-PVA Composites

Hongling Bie,Honghao Chen,Lijun Shan,C. Y. Tan,M. S. H. Al-Furjan,S. Ramesh,Youping Gong,Y. F. Liu,R. G. Zhou,Weibo Yang,Honghua Wang

An ideal artificial bone implant should have similar mechanical properties and biocompatibility to natural bone, as well as an internal structure that facilitates stomatal penetration. In this work, 3D printing was used to fabricate and investigate artificial bone composites based on HA-ZrO2-PVA. The composites were proportionally configured using zirconia (ZrO2), hydroxyapatite (HA) and polyvinyl alcohol (PVA), where the ZrO2played a toughening role and PVA solution served as a binder. In order to obtain the optimal 3D printing process parameters for the composites, a theoretical model of the extrusion process of the composites was first established, followed by the optimization of various parameters including the spray head internal diameter, extrusion pressure, extrusion speed, and extrusion line width. The results showed that, at the optimum parameters of a spray head diameter of 0.2 mm, extrusion pressure values ranging from 1–3 bar, a line spacing of 0.8–1.5 mm, and a spray head displacement range of 8–10 mm/s, a better structure of biological bone scaffolds could be obtained. The mechanical tests performed on the scaffolds showed that the elastic modulus of the artificial bone scaffolds reached about 174 MPa, which fulfilled the biomechanical requirements of human bone. According to scanning electron microscope observation of the scaffold sample, the porosity of the scaffold sample was close to 65%, which can well promote the growth of chondrocytes and angiogenesis. In addition, c5.18 chondrocytes were used to verify the biocompatibility of the composite materials, and the cell proliferation was increased by 100% when compared with that of the control group. The results showed that the composite has good biocompatibility. Keywords:artificial bone;biomanufacturing;finite element analysis;biological scaffold

Electroactive HA/PDA/PPy stents applied for bone tissue engineering by layer-by-layer strategy

Ching-Wen Lou,Mengdan Wei,Jiaxin Li,Ting-Ting Li,Bing-Chiuan Shiu,Jia-Horng Lin

Bioelectric signals are an important factor that maintain cell functions and affect cell behavior. Conductive polymers (CPs) can facilitate cell activity and reinforce the signal transmission among cells, so CPs have triggered attention to the tissue regeneration. In this study, layer-by-layer strategy is employed and polyvinyl alcohol (PVA)/polylactic acid (PLA) braids are polymerized in polypyrrole (PPy) by ice template method. The stents that are deposited with PPy are then immersed in dopamine solution, which during polydopamine (PDA) is coated over the stents naturally. Finally, PDA is used as a template to form hydroxyapatite (HA) coating layer with the aim of electrochemical deposition . The test results indicate that PPy forms an even conductive network over the surface of braids while the electrochemically deposited HA made with PDA is adsorbed over the scaffolds in a needle pattern. According to the cyclic voltammetry measurement, the CV curves of HA/PDA/PPy scaffolds basically retain specified and thus the scaffolds possess stabilized electrochemical activity. At last, HA/PDA/PPy scaffolds are immersed in a simulated body fluid (SBF) for ten days and the surface is completely wrapped in HA, indicating an excellent biomineralization capability. MTT assay results suggest that the cell viability of composite scaffolds is improved by 188 %. This study provides a new perspective, a layer-by-layer structured bone scaffolds, broadening the application range of textile structure in the bone tissue engineering field.

Synthesis of a novel TiO2/HA/RGO composite material with photocatalytic activity for dye degradation

Qiuhua Yuan,Yuan Yang,Wenshan Wu,Xiaoyi Dai,Junxi Zhong,Youliang Jian,Ruilong Li,Tao Wang,Hong Yu,Xianyou Xia

Titanium dioxide (TiO 2 ) is widely used as photocatalysts in the field of organic wastewater treatment in recent years. In this study, TiO 2 is combined with hydroxyapatite (HA) and reduced graphene oxide (RGO) to prepare a novel ternary composite material. And methylene blue(MB) was used as the target pollutant to evaluate the photocatalytic activity of TiO 2 /HA/RGO. The results show that the introduction of HA and RGO can effectively enhance the catalytic performance of TiO 2 . Under 120min of simulated sunlight, the composite showed excellent dye removal effect, the removal rate of MB can reach 99.8%, and its degradation rate is 2 times that of TiO 2 /HA and 7 times that of TiO 2 . The improvement of the photocatalytic activity of the ternary composite is mainly due to the synergistic effect between TiO 2 , HA and RGO. This work provides a new idea for the synthesis of practical photocatalysts.

NG@B87@HA Hydrogel regulates glycolysis bidirectionally to promote wound healing in type 2 diabetes mellitus

Xudong Xie,Weixian Hu,Yue An,Yuheng Liao,Shengming Zhang,Yanzhi Zhao,Kangkang Zha,Wenqian Zhang,Zhenhe Zhang,Fan Xia,Bobin Mi,Benjie Zhou,Hankun Hu,Mengfei Liu,Guohui Liu

Diabetic wounds are characterized by an imbalance between impaired angiogenesis and persistent pro-inflammatory immune responses, leading to chronic non-healing. Under hyperglycemic and oxidative stress conditions, the glycolytic function of endothelial cells is significantly compromised. However, solely enhancing angiogenesis does not effectively promote diabetic wound healing due to concomitant dysregulation of the local immune microenvironment, which further suppresses vascularization. Notably, Th1 cells dominate the diabetic wound immune milieu and exacerbate inflammatory responses, thereby aggravating immune imbalance. Thus, an optimal therapeutic strategy should simultaneously target both inflammatory dysregulation and vascular dysfunction. To address the phenomenon of cell-specific metabolic reprogramming within diabetic wounds, where glycolytic activity is impaired in endothelial cells yet enhanced in Th1 cells, this study developed a glucose-responsive NG@B87@HA hydrogel system designed to reprogram the local metabolic network and modulate Th1 cell metabolism while restoring endothelial function. The hydrogel incorporates nanoparticles functionalized with CXCR3 antibody to specifically target Th1 cells and deliver the glycolysis inhibitor Bay87-2243 (B87), thereby suppressing their glycolytic flux and inflammatory activity via inhibition of the HIF-1α–GLUT1 signaling axis. Simultaneously, naringin (NG), which acts as a crosslinker, forms dynamic boronic ester bonds with HA-PBA and PVA, enabling glucose-dependent responsiveness. This mechanism not only mitigates local oxidative stress but also enhances the tube-forming capacity of endothelial cells, ultimately promoting angiogenesis. The results demonstrated that the NG@B87@HA hydrogel effectively restores the dysregulated metabolic network in diabetic wounds. By concurrently rectifying immune microenvironment imbalance and endothelial dysfunction, this system provides a novel therapeutic strategy for accelerating diabetic wound healing.

Hierarchical Structure and Mechanical Improvement of an n-HA/GCO–PU Composite Scaffold for Bone Regeneration

Limei Li,Yi Zuo,Qin Zou,Boyuan Yang,Lili Lin,Jidong Li,Yubao Li

To improve the mechanical properties of bone tissue and achieve the desired bone tissue regeneration for orthopedic surgery, newly designed hydroxyapatite/polyurethane (HA/PU) porous scaffolds were developed via in situ polymerization. The results showed that the molecular modification of PU soft segments by glyceride of castor oil (GCO) can increase the scaffold compressive strength by 48% and the elastic modulus by 96%. When nano-HA (n-HA) particles were incorporated into the GCO–PU matrix, the compressive strength and elastic modulus further increased by 49 and 74%, from 2.91 to 4.34 MPa and from 95 to 165.36 MPa, respectively. The n-HA particles with fine dispersity not only improved the interface bonding with the GCO–PU matrix but also provided effective bioactivity for bonding with bone tissue. The hierarchical structure and mechanical quality of the n-HA/GCO–PU composite scaffold were determined to be appropriate for the growth of cells and the regeneration of bony tissues, demonstrating promising prospects for bone repair and regeneration.

Dynamic HA/D-mannose dual-ligand nanomedicine reprograms macrophage-epithelial crosstalk for precision therapy of colitis

Jie Shi,Hongyan Liu,Jiafan Bai,Wenzhen Peng,Jie Weng,Wei Zhi,Jianxin Wang,Min Jia

Engineered HA/D-Mannose nanoparticles with temporal dual-targeting for colitis stages. • Achieved colon-specific drug release via a pH-responsive shell overcoming oral barriers. • Simultaneously targets inflammatory macrophages and damaged intestinal epithelium. • A single nanoparticle enables triple therapy: antioxidant, anti-inflammatory, and barrier repair. • Achieved >80% mucosal healing in ulcerative colitis within 7 days. Ulcerative colitis (UC) is characterized by complex spatiotemporal heterogeneity, which poses significant challenges for effective treatment. Herein, we constructed DS-HDLTQs, a core-shell nanodelivery platform with dynamic dual-targeting capabilities. This platform utilizes hyaluronic acid (HA) and D-mannose (D-Man) to target acute inflammation and promote repair. It also features a dopamine-alginate (DA-SA, or DS) shell to achieve pH-responsive release specifically at colonic lesions. In animal experiments, DS-HDLTQs achieved 83.3% mucosal healing in just 7 days, significantly outperforming single-targeting systems. The DA-SA protective layer ensures pH-responsive release in the colon with an efficiency of up to 92.7%. Such therapeutic enhancement is driven by three synergistic mechanisms: molecular functional synergy (restoring antioxidants), spatiotemporal targeting-pathological process synergy (accelerating barrier reconstruction), and immune-epithelial repair synergy (promoting macrophage polarization and epithelial regeneration). DS-HDLTQs demonstrated a 12.7-fold increase in drug concentration at the lesion site. This design resolves the core contradiction between dynamic pathology and oral delivery efficiency, providing a new paradigm for UC treatment and potential applications in other inflammatory diseases. DLP-fabricated TPMS bionic gradient PEGDA/MgO/TA/HA scaffolds: antibacterial, anti-inflammatory, and osteogenic properties

Shuaihang Wang,Jingbin Hao,Siyu Liu,Haiyu Liu,Ding Shen,Zhonghao Ji,Kai Chen

Due to their internal continuity and smooth surface geometry, three-period minimal surfaces (TPMS) have been widely applied in the structural design and fabrication of bone tissue engineering scaffolds. To achieve a biomimetic simulation of natural bone tissue structure and enhance the antimicrobial and anti-inflammatory properties of the implant site, this study innovatively proposes a biomimetic gradient TPMS scaffold with antimicrobial and anti-inflammatory functions. First, based on the hierarchical structural characteristics of compact bone and cancellous bone in natural bone, four TPMS gradient structures were parametrically designed. These four structures were then subjected to comprehensive testing to identify the one with the optimal performance. Furthermore, to address the limitation of single-function traditional bone repair scaffolds, this study used polyethylene glycol diacrylate (PEGDA) as the matrix and prepared a multifunctional composite slurry by incorporating nano-hydroxyapatite (HA), nano-magnesium oxide (MgO), and tannic acid (TA). A series of test results indicate that the I-WP scaffold exhibits the optimal combination of mechanical and permeability properties, while also demonstrating excellent in vitro osteogenic differentiation capacity and antibacterial and anti-inflammatory effects. In summary, the gradient-structured composite scaffold prepared in this study achieves an integrated structure that enhances mechanical properties, boosts osteogenic mineralization activity, and provides synergistic antibacterial and anti-inflammatory functions. It offers new insights for the structural design and functional optimization of bone tissue engineering scaffolds and holds promising application prospects.

Porous PLLA Microspheres Dispersed in HA/Collagen Hydrogel as Injectable Facial Fillers to Enhance Aesthetic Effects

Zhao Miaoran,Chang Shuhua,Wang Yunpeng,Cao Jun,Pu Yuji,He Bin,Pan Shengsheng

Injectable facial fillers such as Sculptra® stimulate collagen regeneration to fill wrinkles; however, the collagen regeneration is not satisfactory due to the slow emergence of filling effect. In this study, we designed a regenerative dermal filler to provide both immediate and long-lasting filling effects. A hydrogel matrix composed of crosslinked hyaluronic acid (HA) and collagen was engineered to encapsulate porous poly(L-lactide) (PLLA) microspheres and tranexamic acid (TXA). The hydrogel matrix was administered via intradermal injection to achieve wrinkle filling. TXA is released to exert skin-whitening effects, while the porous PLLA microspheres and their degradation product, lactic acid, continuously stimulate collagen regeneration over an extended period. Facial volume increased immediately following hydrogel injection. Large amounts of new Type I and Type III collagens are generated. The porous structure of PLLA microspheres facilitated the “penetrating growth” of collagen fibers, which effectively filled facial depressions and smoothed wrinkles. Overall, the HA/collagen composite hydrogel filler exhibited excellent aesthetic effects.
