The use of biodegradable materials combined with natural metabolites in wound dressings has received much attention. Flavonoids (FLs) from green cocoons, as metabolites, have antibacterial, antioxidant, anti-inflammatory, and other pharmacological effects. In this study, composite membranes of FL-loaded polylactic glycolic acid (PLGA)/silk fibroin (SF) were prepared by an electrospinning method. The prepared membranes, including SF, exhibited a good slow-release effect and cytocompatibility. An in vitro evaluation of the FL-loaded PLGA/SF membranes demonstrated good antioxidant, antibacterial, and anti-inflammatory properties. Animal experiments showed that the wound healing rate of PLGA/SF-2.5FL membranes within 15 days was 97.3%, and that of the control group was 72.5%. The PLGA/SF-2.5FL membranes shortened the inflammatory period of a full-layer wound model and promoted skin regeneration and wound healing by downregulating expression of the pro-inflammatory cytokines IL-1β and TNF-α and promoting expression of the growth factors VEGF, TGF-β, and EGF. In summary, the PLGA/SF-2.5FL composite nanofibre membrane with anti-inflammatory properties is an ideal wound dressing to promote acute wound healing.

This study investigates the use of flavonoid-rich extracts from Zanthoxylum bungeanum pericarps (ZBPFs) for sustainable dyeing and functionalization of cationized cotton fabrics. The extracts were obtained through ethanol/water extraction and purified using macroporous resin, with flavonoids such as rutin, astragalin, coumarin, baicalin, and quercetin making up over 72 % of the total composition. Stability tests revealed that ZBPFs were stable under neutral or mildly acidic conditions but were highly sensitive to reducing agents, prolonged light exposure, and metal ions. Cotton fabrics modified with 3-chloro-2-hydroxypropyl trimethylammonium chloride and dyed with ZBPFs reached a maximum color depth (K/S value) of 12.5 at 5 % owf. Fabrics dyed with 2 % owf ZBPFs demonstrated excellent UV protection (UPF of 115.54), antioxidant activity (98.1 % radical scavenging), and antibacterial efficacy (over 98 % against E . coli and S . aureus ). The adsorption of ZBPFs onto cationized cotton fitted both the pseudo-second-order and Sips models. This indicates that chemical adsorption is the predominant mechanism and suggests multiple interactions between the ZBPFs and the fabric. The dyed fabrics exhibited high stability in rubbing and washing fastness, although their light fastness was comparatively lower. Further, washing with potassium aluminum sulfate significantly diminished the antioxidant and antibacterial properties, while washing with other solutions, such as tap water and deionized water, showed minimal effects on performance. This research highlights the potential of ZBPFs as a stable, bioactive, and environmentally friendly dye, offering considerable functional and ecological benefits in textile applications.

Flavonoids are key bioactive compounds in plants that play important defense roles against abiotic stress and are involved in plant growth and development. Methyl jasmonate (MeJA) is a significant growth regulator that promotes the accumulation of flavonoids in a variety of plants, but the effect of MeJA inHedera helixremains poorly understood. In the present study, the flavonoid content was significantly increased after MeJA treatment and peaked at 6 h post-treatment. A total of 31,931 genes were identified using transcriptome, and 6484 DEGs were identified at 6 h post-treatment. Through GO and KEGG enrichment analysis, it was shown that DEGs were primarily enriched in phenylpropanoid biosynthesis pathways. Based on the putative transcription factors derived from DEGs, growth-regulating factor (GRF), a transcription factor potentially linking MeJA signaling to flavonoid accumulation and participating in plant growth and stress responses, was further identified. A total of 20 Hh-GRFd genes were identified on the whole genome level and clustered into five phylogenetic groups with conserved subfamily characteristics. Abundant MeJA-responsive cis-elements were presented in the promoter regions ofHhGRF1-HhGRF20. They exhibited a tissue-specific expression variation, andHhGRF10was dominantly expressed in leaves ofH. helix. Notably,HhGRF10exhibited MeJA-induced expression that correlated temporally with flavonoid accumulation, suggesting thatHhGRF10might play a potential role in promoting flavonoid biosynthesis, and overexpression and knockout assay substantiated this conclusion. The finding provides the first transcriptome-wide resource for flavonoid biosynthesis inH. helixand identifies the candidate GRF-mediated regulator for flavonoid accumulation.

Treating acute lung injury (ALI) presents significant challenges due to adverse drug reactions. This study systematically explored the protective effects and mechanisms of a flavonoid-rich extract fromBombyx batryticatus(FBB), a traditional Chinese medicine, in combating ALI. Through UPLC-MS/MS analysis, we identified 163 flavonoid components in FBB for the first time, including flavonoids, flavonols, and chalcones. Unlike single-component flavonoid therapies, FBB provides synergistic regulation across multiple targets and pathways. Network pharmacology predictions, supported by experimental validation, revealed that FBB primarily suppresses the expression of inflammatory factors (IL-1β, IL-6, TNF-α) and oxidative stress markers (iNOS, COX-2) by modulating the PI3K/Akt, MAPK, and NF-κB signaling pathways. FBB inhibits pro-inflammatory responses and upregulates chemokine receptors like Ccr1 and Ccr2, along with IL-2Rb, at the transcriptional level. This suggests its potential to promote inflammation resolution and tissue repair through immune microenvironment remodeling, rather than mere immunosuppression. Additionally, FBB demonstrated significant anti-apoptotic effects both in vitro and in vivo, effectively reducing pulmonary edema and vascular permeability. Its complex composition and multi-pathway synergistic mechanisms offer broader regulatory potential and unique therapeutic advantages in treating ALI compared to single flavonoid compounds or conventional hormone drugs like dexamethasone (DEX). This study reveals a novel mechanism by which FBB, a multi-component natural drug, exerts therapeutic effects in ALI, providing a theoretical and experimental foundation for developing flavonoid-based compound preparations from traditional Chinese medicine.

Safflower is featured with time-honored medical and economic values and developing into diverse phenotypic and genetic variations. In this study, to explore the critical genes associated with color phenotypes and flavonoid derivatives biosynthesis of safflower, BSA-seq, conflated with transcriptomic and metabolic methods were performed in two extreme colors (yellow and white) in the population of “ZHH0119” and “XHH007”. After crossing two parent plants reciprocally, the F4 generation of two accessions were used to construct near-isogenic gene pools for the two extreme traits of yellow and white safflower. BSA-seq results located five QTLs regions on chromosomes 2, 8, 9, 10 and 12 including six CtPALs, three CtC4Hs, two Ct4CLs, one CtCHS, thirty-two CtUGTs and seventy CtCYPs which tied to the yellow color phenotype of safflower. Through transcriptome analysis of two accessions and at different flowering stages, 1 CtPAL, 5 CtC4Hs, 4 CtCHSs, 3 CtCHIs, 3 CtFLSs, 48 CtUGTs, 51 CtCYPs and 75 transcription factors were revealed as significantly upregulated in the yellow accession compared to the white. Integrated analysis identified eight CtUGTs (CtUGT50-57) which exhibited significant positive correlations with chalcone glycosides of yellow safflower. Based on functional characterization, CtUGT52 was found to boost Hydroxysafflor yellow A (HSYA) content in yellow safflower which possessing substrate promiscuity (chalones, flavonols and flavonoids) and catalytic promiscuity (flavonols and flavonoids), revealing its vital role in the HSYA biosynthesis through transgenic overexpression. Combining catalytic mechanism verification of CtUGT52 towards phloretin, kaempferol and luteolin, our study to some extent, elucidated the modification function of CtUGTs for flavonoid aglycones in the flavonoid biosynthesis pathway of safflower.

Senescence is widely recognized as a key driving factor in the onset and progression of many chronic diseases. Although numerous animal studies have demonstrated that various drugs can delay senescence, their clinical application is severely limited due to significant toxic side effects. As a result, developing safer, natural anti-senescence agents has become an urgent priority. In this study, we identified mitochondrial dysfunction as a critical contributor to cellular senescence. In addition, we identified Shenrong Guben Huanshao Pill (SRW) as an effective agent that alleviates mitochondrial dysfunction and consequently delays cellular senescence by regulating mitochondrial metabolism. Mechanistically, SRW enhances mitochondrial function by upregulating the expression of NADH:ubiquinone oxidoreductase subunit V2 (NDUFV2), a core component of mitochondrial respiratory chain complex I. Simultaneously, SRW reduces lactate-induced mitochondrial dysfunction by regulating the glycolytic enzyme lactate dehydrogenase B (LDHB), thereby decreasing lactate accumulation. Collectively, our findings indicate that SRW alleviates cellular senescence by coordinately regulating NDUFV2 expression and LDHB-mediated lactate metabolism to improve mitochondrial function. In addition, several flavonoids in SRW were identified to possess potential anti-senescence activity. Molecular docking and cellular experiments confirmed that myricetin, luteolin, and gamma-mangostin effectively alleviate cellular senescence.

Icariside I, a prenylated flavonol glycoside derived from Herba Epimedii , has attracted significant interest due to its enhanced bioavailability and pharmacological activities. Traditional methods for producing icariside I, including direct extraction and chemical hydrolysis of icariin, are limited by low yields, harsh conditions, and environmental issues. In this study, we developed a biocatalytic approach for the efficient conversion of icariin to icariside I using naringinase immobilized on magnetic metal-organic framework (MOF) nanoparticles (Nar@MOF) via EDC/NHS cross-linking. The Nar@MOF exhibited improved pH and thermal stability, as well as significant recyclability. Nar@MOF also demonstrated a lower K m value compared to the free enzyme, indicating enhanced substrate affinity. The biocatalyst enabled continuous icariside I production with 99.8% conversion and 98.3% yield within 4 h. Additionally, Nar@MOF maintained over 75% yield after ten consecutive cycles at 60 °C, highlighting its excellent stability and recyclability. Notably, Nar@MOF also exhibited broad substrate applicability. These results indicate that MOF-based immobilization offers a cost-effective and sustainable strategy for large-scale enzymatic conversion of icariin, with the magnetic MOF nanoparticles providing a versatile platform for high-value flavonoid production.

In this study, ultrasonic-assisted deep eutectic solvent (DES) method was used to extract flavonoids from Dendrobium officinale . The mechanism of DESs extraction differences was investigated by physicochemical characterization and molecular dynamics simulation experiments. Moreover, flavonoids from Dendrobium officinale were rapidly identified and quantitatively analyzed by UPLC-Triple-TOF/MS without removing DES. The results showed choline chloride-lactic acid (Chol-Lac) had the best extraction effect among forty groups of DESs, the extraction yield was up to 35.23 ± 2.12 mg/g. It was speculated that the tight hydrogen bond structure was the key to the better extraction ability of Chol-Lac. Meanwhile, the viscosity was the main physical parameter reflecting the hydrogen bond structure of DES. Fourteen flavonoid glycosides were identified, among which the content of rutin (1.94 ± 0.24 mg/g) was the highest. And the results of cluster analysis showed that different DESs had great differences in the extraction of flavonoid glycosides.

Background Type 2 diabetes mellitus (T2DM) is a chronic metabolic disorder characterized by elevated blood glucose levels, posing a significant global health concern due to its increasing prevalence. Insulin resistance (IR) plays a major role in the development of T2DM and is often linked to factors such as obesity, physical inactivity, and a sedentary lifestyle. Recently, there has been growing interest in exploring the potential of natural products for improving insulin sensitivity and glucose metabolism. Among these, Cynomorium songaricum Rupr., an edible parasitic plant, has shown promising antidiabetic effects. However, research on its beneficial effects on IR is still nascent. Therefore, this study aims to investigate the application of a Cynomorium songaricum flavonoid-enriched fraction (CSF) in the treatment of IR in T2DM, along with elucidating the chemical and biochemical mechanisms involved. Method First, UHPLC/ESI-LTQ-Orbitrap-MS was utilized to perform a chemical profiling of Cynomorium songaricum extract (CSF). Subsequently, glycogen synthesis, gluconeogenesis and glucose consumption assays were conducted on HepG2 cells with a high glucose high insulin -induced IR model to illustrate the favorable impacts of CSF on IR. Then, an innovative network pharmacology analysis was executed to predict the potential chemical components and hub genes contributing to CSF's protective effect against IR. To further elucidate molecular interactions, molecular docking studies were performed, focusing on the binding interactions between active constituents of CSF and crucial targets. Additionally, an RNA-sequencing assay was employed to uncover the underlying biochemical signaling pathway responsible for CSF's beneficial effects. To validate these findings, western blot and qPCR assays were employed to verify the pathways related to IR and the potential signaling cascades leading to the amelioration of IR. Results The UHPLC/ESI-LTQ-Orbitrap-MS analysis successfully identified a total of thirty-six flavonoids derived from CSF. Moreover, CSF was shown to significantly improve glycogen synthesis and glucose consumption as well as inhibit gluconeogenesis in HepG2 cells of IR. An innovative network pharmacology analysis unveiled key hub genes—AKT1 and PI3K—integral to metabolic syndrome-related signaling pathways, which contribute to the favorable impact of CSF against IR. Noteworthy active ingredients including quercetin, ellagic acid and naringenin were identified as potential contributors to these effects. The results of western blot and qPCR assays provided compelling evidence that CSF improved insulin sensitivity by modulating the PI3K-Akt signaling pathway. Subsequent RNA-sequencing analysis, in tandem with western blot assays, delved deeper into the potential mechanisms underlying CSF's advantageous effects against IR, potentially associated with the enhancement of endoplasmic reticulum (ER) proteostasis. Conclusion CSF exhibited a remarkable ability to enhance insulin sensitivity in the IR model of HepG2 cells. This was evident through enhancements in glycogen synthesis and glucose consumption, along with its inhibitory impact on gluconeogenesis. Furthermore, CSF demonstrated an improvement in the insulin-mediated PI3K-Akt signaling pathway. The potential active constituents were identified as quercetin, ellagic acid and naringenin. The underlying biochemical mechanisms responsible for CSF's beneficial effects against IR were closely linked to its capacity to mitigate ER stress, thereby offering a comprehensive understanding of its protective action.

Flavonoid glycosides are formed by dehydration condensation of aglycones and sugar molecules. Therefore, discrimination of flavonoid glycosides from their corresponding aglycones is a challenging task because they contain the same aglycone part in their molecular structures. Herein, boric acid-functional Eu(III)-organic framework (BA-Eu-MOF) was applied to discriminate flavonoid glycosides including baicalin (Bai), wogonoside (Wog), rutin (Rut), puerarin (Pue), quercitrin (Que) and astragalin (Ast) from their corresponding aglycones for the first time. Besides as organic ligand to sensitize the luminescence of Eu 3+ through “antenna” effect, 5-boronobenzene-1,3 dicarboxylic acid provided recognition site for flavonoid glycosides. Infrared, fluorescence, UV–vis, and mass spectra were used to investigate the recognition reaction between BA-Eu-MOF and flavonoid glycosides. The data indicated that the cis -diols of flavonoid glycosides from sugars covalently bonded to boric acid group to form cyclic boronic esters, which quenched the fluorescence of BA-Eu-MOF at 620 nm through decreasing the intersystem efficiency, inner filter effect and photoelectron transfer. In contrast, aglycones could not alter the fluorescence of BA-Eu-MOF because of no covalent bond between them. This probe exhibited high sensitivity towards flavonoid glycosides with the low detection limits of 3.3 nM, 3.5 nM, 33 nM, 56 nM, 5.1 nM and 5.5 nM for Bai, Que, Wog, Ast, Pue and Rut, respectively. The unique recognition ability of boric acid group enables selective and sensitive detection of flavonoid glycosides without the interference of their corresponding aglycones.

Total flavonoid content is one of the most important quality indicators of Ginkgo biloba leaves and its products, but the complex pretreatment procedures and the excessive use of organic solvents complicate the detection process. In response, an innovative and efficient two-phase deep eutectic solvent (DES) system—composed of an acidic hydrophilic DES and a hydrophobic DES—was developed to quantify flavonoids for quality control of G. biloba products. High-performance liquid chromatography (HPLC) detection conditions were optimized, followed by systematic screening and design of the two-phase DES system through adjustments to polarity, hydrophilicity, and acidity. Optimal pretreatment conditions were established through operating condition optimization. This approach enabled simultaneous extraction, deglycosylation, and enrichment of all flavonoids from G. biloba samples into the hydrophobic phase in a single step. Method validation showed strong linearity with a correlation coefficient (R 2 > 0.9995). The detection (LOD) and quantification (LOQ) limits were between 0.35 and 0.37 mg/kg, and 0.92 and 0.96 mg/kg, respectively. The method also exhibited high sensitivity and accuracy, with a recovery rate of 96.91–100.39%. A greenness assessment with the ComplexGAPI tool confirmed this method’s eco-friendly advantages over conventional techniques, aligning well with green chemistry and economic principles. Ultimately, this technique effectively measured flavonoid levels in different G. biloba products. The method developed in this work not only enhances the efficiency of simultaneous extraction and deglycosylation by modifying the polarity, hydrophilicity, and acidity of the two-phase DES system, but also offers valuable insights for the advancement of environmentally friendly analysis technology for flavonoids.
