Fatty acid vesicles (FAVs) are promising nanocarriers, but their application is limited by a narrow, alkaline pH formation window that mismatches the weak acidity of physiological environments, such as skin. To overcome this, we developed composite vesicles using oleic acid (OA) and the non-ionic surfactant Tween 40 (TW40). pH titration confirmed that the OA/TW40 system successfully broadened the vesicle formation window from 8.2–10.08 to 3.1–7.2, aligning it with the physiological pH range. The bioactive flavonoid luteolin (LUT) was efficiently encapsulated into these OA/TW40-FAVs, achieving a high encapsulation efficiency (EE) of 87.13% and a drug loading capacity (DLC) of 9.58. The formulation demonstrated superior topical delivery performance: the cumulative transdermal flux (933.08 µg·cm−2) and skin retention (68.18 µg·cm−2) were both approximately double that of the free LUT solution. Furthermore, the OA/TW40/LUT-FAVs provided sustained drug release and exhibited synergistically enhanced antioxidant and antimicrobial activities compared to free LUT or blank vesicles. Collectively, these findings establish OA/TW40 composite vesicles as a robust and efficient nanoplatform for the topical delivery of bioactive compounds.

pH-responsive intelligent films for food freshness monitoring have attracted a great deal of attention in recent years. In this study, dragon fruit peel was used to develop a new type of active and functional film. The film was prepared by using residues extracted from dragon fruit peel (RPP) as the matrix, Curcumin (Cur) as the active ingredient and ZnO-NPs as the antibacterial material. The effect of the incorporation of Cur and ZnO-NPs on the properties of the films was studied. The addition of Cur increased the oxidation resistance of the films (more than 90 %), and ZnO-NPs increased the bacteriostasis and hydrophobicity of the films (water contact angle (WCA):122.00°). The colorimetry response of the RPP/Cur/ZnO-NPs film was performed, and the color of the film changed from brown to tawny to yellow. These properties highlight the potential of the RPP/Cur/ZnO-NPs film for practical applications, indicating its potential applications as smart packaging materials for the food industry. Therefore, the RPP/Cur/ZnO-NPs composite film based on dragon fruit peel can be used to monitor the freshness of milk, and to a certain extent contribute to the sustainable value of dragon fruit by-products in the food industry.

Ischemic stroke, a critical neurological disorder resulting from abrupt cerebral blood flow interruption, remains a leading cause of global mortality and chronic disability. Despite advancements in reperfusion therapies, many survivors still suffer significant neurological deficits, primarily attributed to ischemia-reperfusion injury-induced oxidative stress and neuroinflammation. To address these challenges, we designed cerium-curcumin hybrid nanoparticles (Ce-Cur NPs) by leveraging the coordination interaction between redox-active cerium ions and curcumin’s β-diketone moiety. These Ce-Cur NPs were engineered to alleviate ROS-mediated secondary injuries and promote neural repair by leveraging their remarkable ROS scavenging capabilities, which mimic enzymatic activities to effectively neutralize superoxide and hydroxyl radicals. Furthermore, M1 polarized macrophage-derived membranes were employed to enhance the targeted delivery of Ce-Cur NPs to ischemic regions, while transferrin receptor (TfR)-activated peptides enabled efficient receptor-mediated transcytosis across the blood-brain barrier. In vivo studies using a middle cerebral artery occlusion (MCAO) model demonstrated that TfR-M 1 -Ce-Cur NPs successfully localized to the ischemic brain, significantly reduced infarct volume, and preserved neuronal integrity. Enhanced neurogenesis and improved functional recovery were observed, underscoring the therapeutic potential of Ce-Cur NPs as a versatile platform for ischemic stroke treatment. This approach provides a robust and evidence-based solution to enhance clinical outcomes for patients with ischemic stroke.

Zanthoxylum schinifolium essential oil (ZSEO) exhibits remarkable antioxidant and antibacterial biological activities and is widely used in the food industry. However, its low water solubility, high volatility, and susceptibility to decomposition by light, heat, and oxygen complicate its transport and storage. Microencapsulation of ZSEO is an effective method that can overcome these limitations and expand its application scope. To enhance stability and preserve functionality of ZSEO, ZSEO-loaded microcapsules were produced by complex coacervation of whey protein isolate (WPI) and gum Arabic (GA), followed with freeze-drying. So far, no comprehensive research has been conducted on the method of microencapsulating ZSEO using WPI/GA. Through research, the optimal pH for the formation of WPI/GA (1:1, w/w) complex coacervates was found to be 3.9. The ZSEO-in-water (O/W) emulsions stabilized by whey protein isolate (WPI) were prepared, which exhibited a uniform size of around 200 nm and showed enhanced stability with increasing WPI contents (3.0–6.0 wt%). After complex coacervation with GA, the resulting microcapsules showed improved physical properties and higher encapsulation efficiency as the ratio of WPI/GA complex wall material increased. The ZSEO-loaded microcapsules still maintained their ability to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) free radicals compared to free ZSEO, especially performing the optimal antioxidant activity at a core-to-wall ratio of 5:8 (w/w). Release kinetics studies confirmed controlled and delayed release of ZSEO from the microcapsules in various food simulants (aqueous, ethanolic and aerial environments). This work demonstrated that microencapsulation via WPI/GA complex coacervation was an effective strategy to encapsulate essential oils with high antioxidant activity and controlled release.

Strawberry fruit quality is linked to its phytochemical composition, yet the diversity of non-volatile terpenoids remains largely unexplored. Therefore, a comparative LC-MS/MS-based metabolomic analysis of terpenoid profiles was conducted using three commercial cultivars (Fragaria×ananassa) and a wild accession ofFragaria nilgerrensis(HM). Results from this study showed that the HM cultivar had a total terpenoid abundance 5–6 times higher than the commercial cultivars. The HM cultivar was uniquely enriched in specific triterpenoids, such as 3β,6β,19α,24-Tetrahydroxyurs-12-en-28-oic acid and 13,27-Cyclo-2,3-Dihydroxy-11,19(29)-Ursadien-28-Oic Acid, which was over 450 times higher than the ‘Danxue’ and ‘Fenyu’ commercial strawberry cultivars. Conversely, sesquiterpenoids like Alismol and Pterocarpol were 100 times lower in HM than in the commercial cultivars. This enhanced triterpenoid accumulation strongly correlated with a superior capacity to scavenge free radicals in vitro, with HM showing maximum capacity, as measured by the DPPH assay. These findings highlight the value of wild strawberry germplasm as a reservoir of biochemical diversity for breeding strawberries with enhanced functional quality.

Hazardous substances such as acrylamide (AA) are easily produced by the Maillard reaction during coffee roasting. By adding l -cysteine ( l -Cys), the production of AA was inhibited whereas the profiles of the newly formed compounds remain unclear. To elucidate the influence of l -Cys addition on the main characteristic compounds of Yunnan Coffea arabica , investigations using LC-MSⁿ-IT-TOF, FTIR and assays of in vitro antioxidant and xanthine oxidase (XO) inhibitory activities were carried out. The result revealed that the most characteristic compounds represented by the chlorogenic acid analogs (CGAs) were critical to hazards formation during the Maillard reaction. Adding l -Cys before roasting enhanced coffee quality by reducing harmful intermediates such as 5-hydroxymethylfurfural (5-HMF) while increasing bio-potentials. New compound N6 which was formed by the addition of l -Cys and CGAs, together with the flavonoids generated through polymerization and simple phenols derived from thermal cleavage, were mainly contributed to the above potential.

Chronic wounds, especially diabetic foot ulcers, pose a major clinical challenge due to persistent inflammation, impaired angiogenesis, and cellular senescence. Senolytic therapies, which selectively eliminate senescent cells, have shown promise in promoting healing, but systemic toxicity limits their application. To address this, we developed an asymmetric fabric-based composite platform for localized senolytic delivery. Using single-cell RNA sequencing, we characterized senescence-associated alterations in human diabetic foot ulcer tissues and screened several senolytic candidates in skin fibroblasts and endothelial cells. Among these, navitoclax (ABT-263) emerged as the most effective senolytic. Incorporating ABT-263 into the fabric-based platform to prepare ABT-263-CGH, we confirmed its ability to reduce senescent cell burden while maintaining biocompatibility. In a diabetic mouse model, the ABT-263-CGH significantly enhanced wound healing, reduced senescence markers, and exhibited no detectable systemic toxicity. These findings highlight the potential of localized senolytic therapy using an asymmetric fabric-based wound dressing as a novel strategy for enhancing chronic wound healing, offering a promising therapeutic avenue for diabetic wound management, and paving the way for future clinical applications.

Jackfruit (Artocarpus heterophyllusLam.) phenolic compounds are chemically diverse. Previous studies have predominantly focused on easily extractable free polyphenols, with limited attention on bound (conjugated) polyphenols. The transformation mechanisms of these polyphenols during maturation, digestion, and colonic fermentation remain unclear, limiting the full utilization of jackfruit resources. This study provides a comprehensive portrait of how jackfruit polyphenols shift in abundance, chemical composition, and bioactivity throughout maturation, in vitro digestion, and colonic fermentation. It also quantifies their selective reshaping of the human gut microbiota and their enhancement of short-chain fatty acid production. The results indicate a significant conversion of bound to free polyphenols during maturation, with distinct behaviors observed across different digestion stages. More importantly, jackfruit polyphenols selectively enrich beneficial gut bacteria, promote short-chain fatty acid production, and positively regulate gut health.

Cellulose-based hydrogels have attracted more and more attention in recent years. This study aimed to develop a biodegradable, dual-responsive cellulose-based hydrogel with sustained drug release properties. To achieve this goal, hydrogels with an interpenetrating-network structure based on cellulose derivatives with varying mass ratios were designed, and their controlled release properties for tetracycline hydrochloride (TH) were also investigated. First, carboxymethyl cellulose (CMC) was grafted onto hydroxypropyl cellulose (HPC) via a mediated reaction between N-hydroxysuccinimide and 1-(3-dimethylaminopropyl)-3-ethylcarbodimide hydrochloride. Subsequently, an interpenetrating strategy was employed by introducing N-isopropylacrylamide (NIPAM) monomer as the second network through free radical polymerization into the preformed cellulose-based gel (HPC-co-CMC), thereby constructing a smart dual-network hydrogel (HPC-co-CMC/PNIPAM). The rheology tests, fourier transform infrared (FTIR) spectroscopy, scanning electron microscopy (SEM), thermal analysis, mechanical strength assessment, and drug loading capacity evaluations confirmed the stable formation of chemical cross-linking. The ratio of HPC to CMC had a significant effect on swelling capacity, TH release behavior, and antibacterial properties. In vitro studies, HPC-co-CMC/PNIPAM gels performed outstanding biocompatibility and antioxidant properties. The cumulative release behavior of TH indicated that an optimal HPC/CMC ratio could effectively prolong drug release time, and the dual-network HPC-co-CMC/PNIPAM hydrogels successfully prevented burst release. Furthermore, the pH/temperature-responsive behaviors were also evaluated through swelling ratio measurements. At a temperature of 37 °C and pH 5.0, the TH release rate reached its maximum cumulative release over 65 h. Therefore, HPC-co-CMC/PNIPAM gels have the great potential for advanced drug delivery applications.

Tea polyphenols (TPs) are promising natural bioactive compounds; however, their practical application is hindered by poor stability and low bioavailability. To address this challenge, we synthesized TP–iron nanoparticles (TP-Fe NPs) through coordination-driven self-assembly. Comprehensive characterization (SEM, TEM, FTIR, and XRD) confirmed the successful formation of stable TP-Fe NPs, primarily mediated by phenolic hydroxyl and carbonyl groups. Among TP-Fe NPs, the TP3-Fe1 NPs exhibited superior performance, achieving DPPH and ABTS radical scavenging rates of 65.71% and 89.64%, respectively, and inhibition rates of 91.44% against E. coli and 88.67% against S. aureus . Furthermore, TP3-Fe1 NPs demonstrated excellent biocompatibility, showing no significant cytotoxicity to L929 cells at 0.01–0.1 mg/mL. These findings highlight the potential of TP3-Fe1 NPs as a safe and effective material with dual functionality for antioxidant and antibacterial applications.

BACKGROUND Phyllanthus emblica L. is a medicinal and edible plant renowned for its potent antioxidant and anti-inflammatory activity. However, the digestive stability, bioaccessibility, and intestinal protective effects of its polyphenolic components remain unclear. This study aimed to evaluate systematically the in vitro digestion behavior, antioxidant capacity, and anti-colitis potential of P. emblica extract (PE). RESULTS The bioaccessibility of total phenolics in PE was 92.03%, indicating high digestive stability, whereas the bioaccessibility of flavonoids was 42.69%, indicating partial degradation during digestion. Liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (LC-Q-TOF-MS) identified 3- O -galloylmucic acid and gallic acid as the dominant and stable phenolics throughout all digestive phases. Phyllanthus emblica L. retained strong antioxidant capacity across the oral, gastric, and intestinal stages. In a zebrafish model of trinitrobenzenesulfonic acid (TNBS)-induced inflammatory bowel disease, PE significantly ameliorated intestinal injury, reduced gut lumen expansion, inhibited neutrophil recruitment, and restored goblet cell numbers. Gene expression analysis showed a dose-dependent downregulation of pro-inflammatory cytokines ( tnf-α, il-1β, il-6) ( P < 0.05). CONCLUSION Phyllanthus emblica L. extract exhibited high digestive stability and polyphenol bioaccessibility while maintaining antioxidant activity. Its pronounced anti-inflammatory and mucosal-protective effects in vivo provided evidence supporting the potential application of P. emblica as a functional food ingredient for intestinal health and inflammatory bowel disease prevention. © 2025 Society of Chemical Industry.

This work combined multi-scale characterization with molecular dynamics (MD) simulations to investigate how hydroxyl group number of polyphenol molecules regulate self-assembly of polyphenol-diacylglycerol (DAG)-β-cyclodextrin (β-CD) ternary complexes and formation of resulting oil-in-water (O/W) Pickering emulsions using p-hydroxybenzoic acid (PHBA), protocatechuic acid (PCA), and gallic acid (GA) as models. XRD and ATR-FTIR confirmed hydrogen bonding dominated the complex process. Increasing hydroxyl groups reduced particle size and induced β-CD transition from cage- to channel-type structure in the complexes. Interfacial rheology and SAXS revealed that PCA-DAG-β-CD emulsions exhibited the fastest self-assembly rate with excellent resistance of interface layer to deformation. MD simulations confirmed weak hydrogen bonding and poor interfacial adsorption for PHBA-DAG-β-CD, while GA preferred to form GA-β-CD complexes in aqueous phase due to high hydrophilicity. PCA showed the most favorable self-assembly dynamics and strongest interfacial adsorption. PCA-50 % DAG-β-CD emulsion had the smallest oil droplets (3.9 ± 1.2 μm), the highest stability and excellent oxidative stability. This study highlights that stable Pickering emulsions require balancing hydrogen-bonding strength with polyphenol partitioning at the interface, providing mechanistic insight for designing DAG-based emulsions with polyphenols.
