Schiff base formed functional hydrogel dressing via ϵ-poly-L-lysine modified chitosan and oxidized dextran with the incorporation of epigallocatechin-3-gallate

Lei Nie,Xinran Li,Taiping Zhang,Yuanyuan Lu,Peng Ding

Hydrogel-based functional dressings for wound healing have garnered increasing attention due to their excellent hydrophilicity, adjustable mechanical properties, and superior biocompatibility. In this study, a composite hydrogel was facilely fabricated through the Schiff base reaction between -poly-L-lysine modified chitosan (CS-PL) and oxidized dextran (Odex). The formed hydrogel displayed the interconnected microstructure (100–200 μm), injectability, and adjustable mechanical properties. Macroscopic observation and alternating strain rheological analysis confirmed the good self-healing ability of the hydrogel. Furthermore, with the incorporation of epigallocatechin-3-gallate, the composite hydrogel exhibited an improved reactive oxygen species (ROS) scavenging capability and good antibacterial activity against E. coli and S. aureus. The designed composite hydrogel dressings exhibited hemolysis rates of 0.75 ± 0.60% to 0.81 ± 0.31%, indicating their excellent hemocompatibility. Moreover, CCK-8 analysis and fluorescent images confirmed the excellent cytocompatibility of the hydrogels after co-culturing with NIH 3T3 cells for various periods. The above results offer a promising strategy for preparing functional hydrogel dressings via -PL modification on CS for wound healing applications.

Quality Consistency Evaluation of Multiorigin Chinese Herbal Medicine by Using a Combination of Chemical Similarity and In Vitro Bioequivalence: Taraxaci Herba as a Case Study

Yan Jiang,Hui Ni,Wen-Jing Zhao,Long Wang,Cai Zhang,Ping Li,Hui-Jun Li

Introduction Multiorigin phenomenon is quite common in Chinese herbal medicine, whereas quality consistency evaluation among the involved plant species is seldom considered. Objectives The present study, taking Taraxaci herba as a typical case, aimed to develop a strategy based on a combination of chemical similarity and in vitro bioequivalence to evaluate the quality consistency of three Taraxacum species. Materials and Methods The chromatographic fingerprints were established through two approaches, namely, high-performance liquid chromatography coupled with photodiode array detector and quadrupole time-of-flight mass spectrometry (HPLC-PDA/QTOF-MS) and gas chromatography coupled with quadrupole time-of-flight mass spectrometry (GC-QTOF-MS). The common characteristic peaks from chromatographic fingerprints were quantified. The bioactivities were tested by antioxidant and anti-inflammatory activity assays. Quality consistency of the three Taraxacum species was globally evaluated by using a combination of chemical similarity and in vitro bioequivalence through univariate and multivariate statistical analysis. Results The statistical results showed no classification of the three species based on contents of quantified analytes and in vitro bioactivities, respectively. Conclusion This study not only demonstrated the medicinal interchangeability of three species of Taraxaci herba but also proposed a comprehensive strategy to evaluate quality consistency of multiorigin Chinese herbal medicines by integrating chemical similarity and in vitro bioequivalence.

Apoptotic-body-inspired nanoplatform targeting the lung-macrophage-mitochondrial axis for acute lung injury therapy

Baojin Tan,Weilin Yang,Guanglin Zhang,Rong Zeng

Acute lung injury (ALI), a life-threatening inflammatory syndrome driven by macrophage polarization imbalance, triggers uncontrolled cytokine storms and alveolar-capillary barrier collapse. While macrophage-targeted immunomodulation holds therapeutic promise, current strategies suffer from imprecise cellular targeting and inadequate functional reprogramming. To address this, we developed an inhaled biomimetic apoptotic body-mimicking nanoplatform (CeCPT@PSL). Its phosphatidylserine-containing liposomal (PSL) shell replicates the “eat-me” signaling of apoptotic bodies to enable macrophage targeting and immunomodulation. Inspired by the organelle-regulating capacity of apoptotic cargo, the encapsulated synthetic mitochondrial-targeting nanozyme complexes (CeCPT) execute precision mitochondrial restitution through redox homeostasis restoration. This dual-pathway strategy achieves synergistic therapy by concurrently modulating macrophage function and repairing damaged mitochondria, thereby enhancing macrophage repolarization. In murine ALI models, inhaled CeCPT@PSL concurrently attenuated inflammatory cascades and resolved mitochondrial impairment via targeting lung-macrophage-mitochondrial axis, and restored pulmonary function. This bio-inspired multi-function platform establishes a heuristic strategy wherein artificial nanovesicles recapitulate cellular functions to resolve dysregulated pathological cascades, offering a promising therapeutic avenue for intractable inflammatory diseases.

Chitosan-sodium tripolyphosphate-zinc nanogel for synergistic hydrogen and ion release to eradicate Helicobacter pylori and promote gastric mucosal healing

Xiaoyi Zheng,Yijie Xie,Linlin Zhao,Xiaojuan Huang,Shige Wang,Yongkang Lai,Peng Fan,Yanbo Zeng,Jiulong Zhao,Zhaoshen Li

This study developed a chitosan (CS)‑sodium tripolyphosphate (TPP)‑zinc (Zn) nanogel for treating Helicobacter pylori ( H. pylori ) through synergistic antibacterial, anti-inflammatory, and mucosal repair mechanisms. The nanogel was synthesized via an emulsion method, leveraging electrostatic interactions between CS amino groups and TPP phosphate ions, followed by Zn incorporation. Characterization revealed uniform spherical nanoparticles (~200 nm) with high Zn content (20.18 %) and controlled hydrogen (H 2 ) and Zn 2+ release (15 h) in acidic environments. In vitro assays demonstrated potent antibacterial activity against H. pylori (95.83 % inhibition at pH = 2; 28.54 % inhibition at pH = 7) attributed to Zn 2+ -mediated enzyme disruption. The nanogel exhibited significant antioxidant capacity, scavenging 82.3 % of PTIO and 65.48 % of DPPH radicals. Biocompatibility testing confirmed minimal haemolysis (<5 %) and high cell viability (>95 %). In vivo studies in H. pylori -infected mice demonstrated a bacterial inhibition rate of 97.54 % through upregulation of β-catenin and tight junction proteins. Concurrently, mucosal repair was enhanced, achieving migration rates of 44.2 ± 5.6 % and 72.34 ± 2.1 % at 12 and 24 h respectively. The nanogel also preserved gut microbiota diversity post-treatment, unlike conventional antibiotics. This multifunctional nanogel platform combines biocompatibility, ease of manufacture and dual therapeutic effects. In summary, this study proposes a multifunctional therapeutic strategy capable of simultaneously addressing H. pylori infection, gastric inflammation and mucosal damage. It offers a paradigm shift from traditional antibiotic-centred therapies, providing a promising strategy for eradicating Helicobacter pylori and restoring gastric injury.

Concentration-dependent fibrillation of whey protein isolate:Structural and antioxidant properties

Caiyun Cheng,Qian Xu,Yang Li,Eric Haubruge,Ning Liao,Guangsu Zhu,Kunlun Liu

Whey protein isolate amyloid fibrils (WPIF) have attracted extensive research interest due to their stable structural characteristics and desirable functional attributes. However, limited research has been conducted on the relationship between the structure and antioxidant activity of WPIF. WPIF with various flexible and linear structures were successfully prepared by adjusting the fibrillation concentrations. The structure and antioxidant properties of WPIF were characterized, revealing multiscale structural transformations of WPIF in response to different fibrillation concentrations. WPIF with low fibrillation concentrations (1–3 %) displayed short flexible structures. WPIF with high fibrillation concentrations (3–5 %) displayed long rigid structures. Long rigid amyloid fibrils possessed enhanced antioxidant properties compared to short flexible amyloid fibrils. The active amino acids were more easily exposed to the surface of long rigid amyloid fibrils, enhancing free radical scavenging activity and ferric reducing power. The partial gelation of WPIF occurred at high fibrillation concentrations (4–5 %), which prevented the exposure of oxidizable amino acids and thus reduced the antioxidant activity of WPIF. This study revealed the relationship between fibrillation concentrations and the structural and antioxidant activity of WPIF, thereby furnishing a theoretical basic for the optimized utilization of WPIF in formulated foods.

Cu–Fe3O4 Nanozyme Reverses Age-Impaired Alveolar Healing via Reactive Oxygen–Nitrogen Species Scavenging and Mitophagy Reactivation

Zheng Cheng,Jikang Cao,Haobo Lin,Yuchen Wang,Ran Qin,Yuhao Ruan,Xinyu Hu,Jingxuan Cang,Chang Sun,Dong Qiu,Yifeng Bian,Yifei Du,Yongchu Pan

Age-related decline in bone healing capacity poses significant challenges for tooth extraction recovery in the elderly. Elevated reactive oxygen–nitrogen species (RONS) and impaired mitophagy drive this pathology. We engineered bimetallic Cu–Fe3O4 nanoclusters with oxygen vacancies (OVs) via hydrothermal synthesis. The abundant oxygen vacancies and bimetallic synergy endow Cu–Fe3O4 with exceptional enzyme-mimicking activity, enabling efficient scavenging of excess RONS and activation of mitophagy in senescent cells in vitro, thereby reversing functional decline. Owing to its excellent biocompatibility and negligible in vivo toxicity, Cu–Fe3O4 accelerates tooth extraction wound healing in aged animal models.

Dragon fruit peel pigment extraction residue based intelligent and biodegradable film for indicating milk freshness

Liangting Deng,Qun Liu,Wuyin Weng,Yucang Zhang

pH-responsive intelligent films for food freshness monitoring have attracted a great deal of attention in recent years. In this study, dragon fruit peel was used to develop a new type of active and functional film. The film was prepared by using residues extracted from dragon fruit peel (RPP) as the matrix, Curcumin (Cur) as the active ingredient and ZnO-NPs as the antibacterial material. The effect of the incorporation of Cur and ZnO-NPs on the properties of the films was studied. The addition of Cur increased the oxidation resistance of the films (more than 90 %), and ZnO-NPs increased the bacteriostasis and hydrophobicity of the films (water contact angle (WCA):122.00°). The colorimetry response of the RPP/Cur/ZnO-NPs film was performed, and the color of the film changed from brown to tawny to yellow. These properties highlight the potential of the RPP/Cur/ZnO-NPs film for practical applications, indicating its potential applications as smart packaging materials for the food industry. Therefore, the RPP/Cur/ZnO-NPs composite film based on dragon fruit peel can be used to monitor the freshness of milk, and to a certain extent contribute to the sustainable value of dragon fruit by-products in the food industry.

Tannic acid-assisted fractionation of lignin with multifunctional properties for sunscreen formulations

Xinyue Xu,Ying Xu,Jialong Wen,Tongqi Yuan

Lignin contains aromatic rings, phenolic hydroxyl, and methoxy groups that confer promising potential for sunscreen applications. However, traditional extraction methods often lead to structural condensation, thus limiting the functional utilization of lignin. Herein, we propose a strategy for simultaneous lignin extraction and in-situ modification using tannic acid (TA) as both a functionalizing agent and a phenolic modifier. This approach enables efficient lignin isolation under mild conditions, while preserving reactive sites and enhancing multifunctional performance. Under optimized conditions (100 °C, 30 min), the pretreatment achieved a delignification efficiency of 89.1 %. The resulting TA-modified lignin (TA-Lig) exhibited excellent UV-blocking performance, antioxidant activity, tyrosinase inhibition, and favorable biocompatibility. Notably, incorporating 5 wt% TA-Lig into a commercial sunscreen (SPF 15) markedly enhanced the SPF to 145.3. This work demonstrates a sustainable extraction–modification strategy that yields multifunctional bio-based ingredients with strong potential for cosmetic applications.

Bacterial cellulose-based multifunctional films modified with natural Sapindus saponin: a sustainable food packaging material

Shuyang Gan,Tianran Zheng,Jiayu Miao,Yunhua Bai,Xiong-Fei Zhang,Jianfeng Yao

Escalating environmental concerns over plastic packaging have prompted extensive researches into renewable bio-based alternatives. In this study, bacterial cellulose (BC) films modified with natural Sapindus saponin (SS) and gelatin were developed as multifunctional active packaging materials for fruit preservation. SS and gelatin gave the composite a denser and more stable structure via the formation of covalent crosslinking, thereby strengthening the mechanical properties of the bio-based films (BFs). The tensile stress of the designed film (BF3) was 28.21 MPa, which was 89.93% greater than that of the BC film. Compared with the control, the BFs had better thermal stability and greater barrier properties, coupled with good natural affinity. Additionally, owing to the presence of SS, the BF3 showed remarkable ultraviolet-blocking capacity (92.24% UVA and 99.71% UVB shielding rates), antioxidant activity (68.47% DPPH and 74.38% ABTS scavenging activities), and anti-bacterial property ( Escherichia coli , 6.4 mm; Staphylococcus aureus , 22.8 mm), which corresponded to the excellent freshness preservation ability. The fresh blueberries packaged with BF3 had less weight loss, reduced postharvest decay, and delayed softening, thus achieving a longer shelf life. In summary, BFs have a promising application prospect as a sustainable and effective packaging material for fruit preservation.

The impact of S. cerevisiae strains on the aroma profile and physicochemical properties of Phyllanthus emblica L. fruit wine

Haibin Yuan,Yanlin Li,Jianhang Tu,Tianyang Wang,Kaixian Zhu,Hongfeng Jia,Huachang Wu,Ping Dong

Since the key aroma compounds and bioactive profiles of Phyllanthus emblica fruit wine (PFW) driven by different yeast strains remain unclear, this study aimed to systematically evaluate the effects of five Saccharomyces cerevisiae strains on physicochemical properties, bioactive components, and volatile organic compound (VOC) profiles in PFW. Using an integrated flavoromics approach with chemometric analysis, we identified 110 VOCs. FR and D254 enhanced ester concentrations and antioxidant activity respectively, whereas strain L2323 significantly altered aroma complexity by promoting specific esters and alcohols. Multivariate analyses identified methyl nitrate and n-propyl acetate as discriminant markers among strains. Aroma-activity analyses further highlighted ethyl hexanoate, isoamyl acetate, and n-propyl acetate as major aroma contributors with distinctive strain-dependent distributions. These findings provide critical guidance for yeast strain selection and fermentation optimization to improve PFW quality, offering new insights for the development of functional fruit wines.

Konjac glucomannan-based emulsion films with lysozyme-amyloid fibrils-chitosan complexes as dual-function antimicrobial emulsifiers for salmon preservation

Yangyang Guo,Xingru Yan,Jie Pang,Bo Liu,Yihua Bai,Di Wu,Yukun Song,Mingqian Tan,Wentao Su

Salmon is highly susceptible to spoilage due to microbial contamination and lipid oxidation, whereas active packaging can effectively delay quality deterioration during storage. This study employed chitosan-modified lysozyme amyloid fibrils (LC) as both antimicrobial agents and emulsifiers to prepare stable turmeric essential oil (TEO) Pickering emulsions with different LC-to-TEO volume ratios (1:1, 2:1, 3:1, 4:1), designated as TEOP1, TEOP2, TEOP3, and TEOP4 respectively. These emulsions were then incorporated into a konjac glucomannan (KGM) matrix to develop composite films. This study systematically evaluated the structural properties and freshness-preserving effects of composite films, using pure KGM film as the primary control and commercial polyethylene terephthalate (PET) film as the reference control. The results showed that strong interactions among components enhanced film properties: TEOP4/KGM reduced water vapor and oxygen permeability by 44 % (6.79 ± 0.59 g·mm/ (m 2 ·day·kPa)) and 31 % (6.55 ± 0.36 g/ (m·s·kPa)). TEOP3/KGM exhibited optimal tensile strength (26.63 ± 1.45 MPa) and antioxidant activity (DPPH: 47.5 ± 2.6 %, ABTS: 96.3 ± 0.1 %), while TEOP4/KGM had highest elongation at break (68.35 ± 0.81 %). This suggests a trade-off, where a higher oil content (TEOP4) plasticizes the film, increasing flexibility but reducing strength. TEOP/KGM films inhibited E. coli , S. aureus , and L. monocytogenes , with the TEOP3/KGM and TEOP4/KGM formulations achieving 100 % inhibition rates. In salmon preservation, TEOP3/KGM best maintained appearance, pH stability, and reduced microbial growth/TVB-N, confirming its potential in aquatic product preservation.

Senolytic-Loaded Asymmetric Wound Dressing for Targeted Senescent Cell Clearance in Diabetic Wound Healing

Ming Zhang,Yamei Wang,Yao Dai,Yan Hu,Wanting Fu,Yuhao Zhao,Hanyu Ma,Di Zhang,Ying Chen,Yixuan Zhou,Lei Du,Jing Chang,Fang Liu,Shuyan Chen,Fei Wang,Dongdong Xiao,Zhen Li

Chronic wounds, especially diabetic foot ulcers, pose a major clinical challenge due to persistent inflammation, impaired angiogenesis, and cellular senescence. Senolytic therapies, which selectively eliminate senescent cells, have shown promise in promoting healing, but systemic toxicity limits their application. To address this, we developed an asymmetric fabric-based composite platform for localized senolytic delivery. Using single-cell RNA sequencing, we characterized senescence-associated alterations in human diabetic foot ulcer tissues and screened several senolytic candidates in skin fibroblasts and endothelial cells. Among these, navitoclax (ABT-263) emerged as the most effective senolytic. Incorporating ABT-263 into the fabric-based platform to prepare ABT-263-CGH, we confirmed its ability to reduce senescent cell burden while maintaining biocompatibility. In a diabetic mouse model, the ABT-263-CGH significantly enhanced wound healing, reduced senescence markers, and exhibited no detectable systemic toxicity. These findings highlight the potential of localized senolytic therapy using an asymmetric fabric-based wound dressing as a novel strategy for enhancing chronic wound healing, offering a promising therapeutic avenue for diabetic wound management, and paving the way for future clinical applications.
