Copper Nanocluster-Decorated Magnesium Silicate-Based Microneedle Enhances Antimicrobial Effects and Tissue Remodeling for Diabetic Wounds

Shuo Tan,Hua Zeng,Wenshuya Li,Haibo Liu,Xuefeng Gu,Xiong Luo,Xinyu Zhao

Diabetic skin lesions, as one of the most common complications of diabetes, present chronic nonhealing wounds that face dual challenges of antibiotic-resistant bacteria threat and insufficient microenvironment regulation due to hyperglycemic conditions, bacterial infections, and multiple pathological factors (e.g., hypoxia and reactive oxygen species (ROS) accumulation and growth factor deficiency). This study develops a microneedle (MN) system integrated with copper nanocluster-decorated magnesium silicate nanoparticles (denoted as MS@Cu MNs), which enables efficient diabetic wound healing via a synergistic multimechanism strategy. Leveraging the unique enzyme-mimetic activity of copper nanoclusters (CuNCs) and the angiogenic properties of magnesium silicate nanoparticles (MS NPs), the engineered MS@Cu nanocomposites demonstrate: 1) broad-spectrum antibacterial efficacy (sterilization rate >99.9%), 2) microenvironment regulation via simultaneous hypoxia mitigation, ROS scavenging, and angiogenesis promotion, and 3) enhanced fibroblast proliferation and migration through PI3K-AKT signaling pathway activation. The MN system using γ-polyglutamic acid (γPGA) as a matrix exhibits both superior mechanical strength and excellent biodegradability. In vivo studies demonstrated accelerated closure of infected diabetic wounds in animal models, with histological analysis revealing robust mature collagen deposition and tissue regeneration. This study develops an integrated strategy for chronic diabetic wound management, combining potentiated antibacterial activity with targeted microenvironment remodeling.

Multifunctional trilayer films based on gelatin-dextran incorporated with defatted rice bran-derived carbon dots for scallop preservation

Hongkai Xie,Hailu Liao,Xinhao Wan,Yinlei Mao,Fengjiao Fan,Yuhong Fu,Li Wang,Liwei Mao,Yan Li,Zhuoli Li

Single-layer films lack sufficient mechanical, barrier, and controlled-release properties. Thus, a three-layer gelatin/gelatin-dextran/gelatin film incorporated with defatted rice bran-derived carbon dots (DRB-CDs) was prepared to overcome these issues and preserve scallops. DRB-CDs were non-toxic to HaCaT cells (<2 mg/mL). They enhanced the film's thermal stability and formed hydrogen bonds with the matrix polymer. Increasing the DRB-CDs concentration improved the film's UV-blocking, antioxidant, and antimicrobial properties. Adding 3 % DRB-CDs reduced the film's water vapor permeability by 35 %, increased the tensile strength 2.09-fold, and increased the elongation at break 1.15-fold. The three-layer structure enabled the sustained release of DRB-CDs in food simulants. Scallops wrapped with 3 % DRB-CDs-loaded films showed enhanced chemical and microbial stability, extending their shelf life by ≥6 days. In conclusion, this study proposes a sustainable and effective active packaging strategy using food-byproduct-derived nanomaterials to maintain seafood quality, showing significant potential to replace petroleum-based packaging.

Promoting walnut protein as a precursor of bioactive peptides: The assisting role of high hydrostatic pressure

Xiaoling Wang,Shuguang Wang,Mouming Zhao,Yao Tang,Liping Sun,Yongliang Zhuang,Bifen Chen

Innovative high hydrostatic pressure (HHP) technology offers a promising approach to enhance the enzymatic hydrolysis efficiency of plant proteins and boost the bioactivity of hydrolysates. This study aimed to evaluate the impact of HHP (100–400 MPa) on the enzymatic hydrolysis efficiency of walnut protein, with a focus on its potential as a precursor for bioactive peptides. Results demonstrated that HHP significantly promoted the enzymatic efficiency. The degree of hydrolysis (DH) increased by 127.6 % compared to enzymatic hydrolysis at atmospheric pressure, and the proportion of the low-molecular-weight (low-MW) fraction ( < 1000 Da) increased by 16.0 %. Furthermore, HHP followed by enzymatic hydrolysis (HHPFE) was found to be more effective than HHP combined with enzymatic hydrolysis (HHPCE) in promoting enzymatic hydrolysis. Relatively lower pressure was favorable for HHPFE, whereas higher pressure was beneficial for HHPCE. Peptidomic analysis and bioinformatic approaches indicated that HHP enhanced the production of potentially bioactive peptides with antioxidant, ACE inhibition and DPP-IV inhibition. These potent bioactivities were attributed to the positive effects of HHP treatment on walnut protein hydrolysis, such as promoting DH and enriching peptides containing aromatic amino acids for antioxidant, moderate hydrolysis and abundance of vicilin-derived peptides for ACE inhibition, and aiding release of specific peptide fragments for DPP-IV inhibition. Moreover, molecular docking demonstrated that HHP could facilitate the release of specific bioactive peptides encrypted in walnut proteins, such as antioxidant peptides (FYMSWR and SWPAP), DPP-IV inhibitory peptides (RVAPF and RAWYPH), and ACE inhibitory peptides (VQHWIY and FHHPYP).

Schiff base formed functional hydrogel dressing via ϵ-poly-L-lysine modified chitosan and oxidized dextran with the incorporation of epigallocatechin-3-gallate

Lei Nie,Xinran Li,Taiping Zhang,Yuanyuan Lu,Peng Ding

Hydrogel-based functional dressings for wound healing have garnered increasing attention due to their excellent hydrophilicity, adjustable mechanical properties, and superior biocompatibility. In this study, a composite hydrogel was facilely fabricated through the Schiff base reaction between -poly-L-lysine modified chitosan (CS-PL) and oxidized dextran (Odex). The formed hydrogel displayed the interconnected microstructure (100–200 μm), injectability, and adjustable mechanical properties. Macroscopic observation and alternating strain rheological analysis confirmed the good self-healing ability of the hydrogel. Furthermore, with the incorporation of epigallocatechin-3-gallate, the composite hydrogel exhibited an improved reactive oxygen species (ROS) scavenging capability and good antibacterial activity against E. coli and S. aureus. The designed composite hydrogel dressings exhibited hemolysis rates of 0.75 ± 0.60% to 0.81 ± 0.31%, indicating their excellent hemocompatibility. Moreover, CCK-8 analysis and fluorescent images confirmed the excellent cytocompatibility of the hydrogels after co-culturing with NIH 3T3 cells for various periods. The above results offer a promising strategy for preparing functional hydrogel dressings via -PL modification on CS for wound healing applications.

Screening and mechanistic investigation of food-derived multifunctional ACE-inhibitory tri-peptides

Dankui Liao,Lei Wang,Qian Zhou,Lixia Sun,Jianhua Sun,Zhangfa Tong,Guangzhi Zhou

Food-derived angiotensin-converting enzyme (ACE)-inhibitory peptides (ACEIPs) with antioxidant and antibacterial properties can function as food additives to alleviate hypertension-induced oxidative stress and bacterial infections. This study used bioinformatics to analyze Gly-Xaa-Pro ACEIPs, identifying Gly-Pro-Pro (GPP) from buckwheat. GPP exhibited potent free-radical-scavenging capacity and at 4 mg/mL eradicated both E. coli and S. aureus (0 % survival vs 14.1 %/4.6 % with GRP; p  < 0.001), demonstrating superior antioxidant and bactericidal potency. Multispectral techniques, thermodynamic analysis, and molecular docking simulations were used for a detailed molecular-level investigation to better understand the binding mechanism between GPP and ACE. The results indicated that GPP's binding to ACE is non-competitive and acts as an inhibitor. The inhibitory effect of GPP on ACE from its moderate binding ( K a  = 1.525 × 10 4  M −1 ), which alters the microenvironment and conformation of ACE. The binding process is primarily driven by entropic forces arising from hydrophobic interactions and proceeds spontaneously due to ΔG<0. Through multi-molecular synergy (Hill coefficient n  = 513), GPP forms hydrogen bonds with the Asp 121, Glu 123, Tyr 135, and Trp 220 residue sites outside ACE's active center, thereby inhibiting ACE activity. In conclusion, the research of GPP provides a theoretical basis for developing multifunctional blood-pressure-lowering dietary supplements.

Exogenous anthocyanin regulated Maillard pathway and aroma formation in blackened jujube wine

Jing Zhang,Yuxiao Wang,Na Li,Lin Gao,Dongxu Zhang,Hongxun Tao,Rentang Zhang

Previous studies demonstrated that the blackening process could enhance the bioactivity of jujubes while it could also lead to uncontrolled Maillard reactions, potentially compromising product quality. To address this, we investigated how exogenous anthocyanin regulated the Maillard pathway and promoted desirable aroma formation during the fermentation of blackened jujube wine. Results showed that exogenous anthocyanin reduced the 5-hydroxymethylfurfural content in blackened jujube wine from 1.18 mg/L to 0.82 mg/L, suggesting a potential interaction with Maillard intermediates or 5-hydroxymethylfurfural. Moreover, the contents of bioactive compounds increased during the fermentation. HS-SPME-GC-MS analysis identified that a crucial phase for flavor development occurred on day 6 of fermentation, with ester compounds peaking at 7658.19 μg/L. Among these, ethyl caprylate and ethyl caprate were the predominant flavor compounds. The addition of exogenous anthocyanin increased the concentration of total volatile compounds in blackened jujube wine, resulting in a rich fruity and floral aroma at the end of fermentation. These results elucidate the mechanistic role of exogenous anthocyanin in modulating the Maillard pathway and aroma formation, providing a theoretical foundation for optimizing blackened jujube wine quality.

Antioxidant, antibacterial and anticancer activities of norfloxacin-kaempferol drug cocrystal hydrate

Xin Jiang,Siyu Yuan,Jie Yang,Hui Liao,Hansong Wu,Baolin Jiang,Kun Qian

Drug cocrystal can not only improve the physicochemical properties of drugs but also exert a synergistic effect without altering the original structure of drugs. Herein, we successfully synthesized a norfloxacin-kaempferol cocrystal hydrate (cocrystal 1 ) from an antibacterial drug and a flavonoid active ingredient. Compared with norfloxacin or kaempferol alone, cocrystal 1 enhanced antibacterial activity against Gram-positive bacteria ( S. aureus ), Gram-negative bacteria ( K. pneumoniae and P. aeruginosa ), and fungus ( C. albicans ). Meanwhile, cocrystal 1 enhanced the antioxidant activity of kaempferol and also improved the solubility of norfloxacin and kaempferol. Moreover, cocrystal 1 enhanced the anticancer activity against human breast cancer cells MDA-MB-231, colon cancer cells SW480 and lung cancer cells H1299.

Green synthesis of citric acid-crosslinked ethanol lignin/carboxymethylcellulose films for fresh-cut apple preservation

Xu Dong,Liping Chen,Danjie Xu,Jian Zhang,Fang Hu,Qingwei Ping

Considering the non-renewable and non-biodegradable characteristics of conventional petroleum-based plastic films, their widespread use contributes significantly to environmental degradation. This study utilizes a green synthesis citrate-based chemical cross-linking strategy to improve the dispersion of ethanol lignin within the film, thereby promoting the development of a natural, safe, renewable, biodegradable, and environmentally friendly biomass-based functional food packaging material. A novel, multifunctional green food preservation film (ELA x C) was synthesized by employing CMC film as the substrate, ethanol lignin (EL) as the functional agent, and citric acid (CA) as the cross-linking agent. The influence of CA addition on the properties of the film was systematically examined. The results indicated that the ELA 0.2 C film exhibited an optimal elongation at a break of 8.0 %, a tensile strength of 28.49 MPa, and outstanding UV-blocking performance, effectively blocking 100 % of UVB and 99.2 % of UVA radiation. Furthermore, the ELA 0.2 C film exhibits antioxidant properties, gas barrier performance, and biodegradability. This film demonstrates significant potential as a multifunctional cling film for apple slices. Moreover, EL can be directly applied as a functional agent without the need for re-dissolution, thereby offering an innovative pathway for the efficient and environmentally sustainable utilization of lignin.
