Multidirectional interstitial flow promotes microvascular network formation: insights from a square chip-based platform

Yang Qihang,He Yuening,Wang Shuo,Li Zengting,Wang Jiaxuan,Sun Zehao,Yang Wenbo,Zhong Xiang,Peng Bo,Chen Zaozao,Gu Zhongze,Zhu Dan,Yu Tingting

Microvascular network formation is governed by a variety of factors, with interstitial flow (IF) playing a pivotal role. However, the impact of multidirectional IF (MDIF) on microvascular network development remains insufficiently explored. In this study, we developed a platform consisting of a Square chip capable of generating MDIF and a deep learning-based Vasculature-on-a-Chip Analysis Tool (VoCAT) for high-efficient analysis of vascular morphology on the chip. Using this platform, we demonstrated that microvascular networks formed on the Square chip exhibited intricate structural features with enhanced functionality. We also demonstrated its utility in modeling a tumor microenvironment with complex microvascular networks and observed enhanced tumor cell migration. This study provides the first evidence that MDIF promotes microvascular network formation, offering new perspectives for advanced in vitro vascular and disease research.

Portable dual-mode microfluidic sensor for rapid and sensitive detection of DPA on chip

Lin Xiaodong,Cao Zhengyang,Zeng Shiyu,Zhu Haoyu,Zhai Kairui,Yin Binfeng,Zhang Cheng,Peng Tao,Cheng Teng,Zhang Biao

In this work, we developed a dual-mode portable device that integrated a 3D-printed microfluidic chip for detection of dipicolinic acid (DPA) on chip. The system uses a ratiometric fluorescence nanoprobe formed by embedding carbon dots (CDs) into an Eu 3 ⁺ metal–organic framework (Eu-MOF). Upon reaction with DPA in the microchannel, red fluorescence was enhanced and blue fluorescence suppressed, enabling sensitive ratiometric detection of DPA on chip with a detection limit (LOD) of 0.04 µM. Interestingly, the composite Eu MOF /CDs/DPA also exhibits peroxidase-like activity, catalyzing the oxidation of TMB into a blue-colored product (oxTMB), which allows for colorimetric detection with an LOD of 10.14 µM. To improve usability and reduce environmental or instrumental variability, incorporating a microfluidic chip into a semi-portable device and utilizing a smartphone, making the system portable and miniaturized for easy operation. In the smartphone-assisted mode, the LODs were 0.33 µM (ratiometric fluorescence) and 12.27 µM (colorimetry), determined by RGB signal analysis, respectively. Moreover, satisfactory recoveries (85–104.6%) were achieved in the spiked real samples. Overall, this platform offers a straightforward, cost-effective, and versatile approach for DPA detection, with promising applications in food safety, environmental monitoring, and clinical diagnostics.

An Integrated Luminescent Information Encryption–Decryption and Anticounterfeiting Chip Based on Laser Induced Graphene

Congcong Zhu,Lu-Qi Tao,Zhirong Peng,Guanya Wang,Yexiong Huang,Simin Zou,Hao Sun,Yunfei Zhao,Xianping Chen,Tian-Ling Ren

Visual optical information encryption–decryption and anti-counterfeiting (IEDAC) technology play a vital role in the field of information security. Recent luminescent information encryption technologies face the disadvantages of depending on external large-scale stimulus decryption equipment, inability to read out repeatedly, and information leakage, impeding the practical applications of luminescence encryption. Here, an integrated luminescent IEDAC chip is proposed, which provides a convenient approach to store and decipher pre-patterned luminescence information based on laser engraved template and film heater. The luminescent encryption chip contains a double-layer structure made up of long persistent phosphors based on SrCaGa 4 O 8 host and a laser induced graphene heater, which makes it possible to decrypt information on a single chip. This design enables dual-mode (photoluminescence/long persistent luminescence), dual-color (blue/yellow-green), and multi-level IEDAC function, providing a novel insight and integrated strategy for implementing advanced IEDAC technologies.

The Rapid Fabrication of Hydrogel Microfluidic Chip for Cell Capture Culture and Metabolites Detection

Ke Jing Fang,Chang Jun Hou,Cheng Hong Huang,Xiao Gang Luo,Su Yi Zhang,Cai Hong Shen,Dan Qun Huo

The microfluidic chip with well-defined structure is an important platform for cell research. The existing techniques for chip fabrication especially in cell biology and tissue engineering have many defects, for example, poor processing precision, high processing cost, as well as sophisticated manufacturing procedure. Thus, fabrication of simple and practicable microfluidic chip with highly efficient cell control ability and low-cost is turned to be the main target for bioengineering application. Poly(ethylene glycol) (PEG) is a hydrophilic polymer. Substituting terminal hydroxyl groups with acrylates, forming poly(ethylene glycol) diacrylate (PEGDA), allows the polymer to be cross-linked to form a three-dimensional polymer network. Meanwhile the use of photopolymerization can realize precise and temporal control of polymerization for formation of complex shapes. Herein, we utilize PEGDA hydrogel’s highly tunable characteristic, using photopolymerization method to obtain desirable micro-structure. Each chip has four of uniform micro-structures, which can carry multiple parallel experiments at the same time. We also add 2-Hydroxyethyl Methacrylate (HEMA) to the PEGDA prepolymer in order to increase the cell adhesion capacity of the microchip surface for cell culture. The experimental results showed that this method can achieve double-layer cell culture with short time treatment. Cells can be well captured and cultured in the hydrogel microfluidic chip with excellent activity. The hydrogel microfluidic chip has the potential of practicable application once large-scale preparation is accomplished.

Michael Coln,Qingdong Meng,Viorel Bucur,Ramji S. Lakshmanan,Aditya Yadav,David Lloyd,Nicolò Ferri,Mark Bignell,Daniele Di Nuzzo,Phillip Nadeau,Matthew Hayes,Roman Trogan

A chip platform for thermally controlled biochemistry is presented comprising postprocessed thermally isolated heated reaction sites atop a application-specific integrated circuit (ASIC) using proportional-integral-differential (PID) feedback temperature control. The 384 sites can be individually programmed over $25~^{\circ }$ C– $125~^{\circ }$ C, with thermal crosstalk below the $\pm 0.12~^{\circ }$ C precision. Double-strand deoxyribonucleic-acid (DNA) purification via selective melting is demonstrated, with 70% retention of perfectly matched sequences, while 55% of those with two mismatched base-pairs are removed.

Dynamic Single-Binding Event Profiling With on-Chip Microlenses for Wash-Free Digital Biosensing

Tingting Zhan,Lianyu Lu,Guoqiang Gu,Pengcheng Zhang,Jienan Shen,Xiaotian Tan,Wei Ye,Shi Hu,Yi Zhang,Hao Yu,Shaoqin Liu,Hairong Zheng,Hui Yang

Single-molecule detection enables precise biomarker quantification, yet conventional endpoint assays suffer from long incubation and nonspecific binding. Kinetic assays resolve these via real-time monitoring of molecular interactions, but their reliance on complex optics has restricted their practical adoption, especially in point-of-care settings where speed, portability, and ease of use are paramount. We present a microlens-assisted platform that enables real-time tracking of single-binding events under conventional bright-field microscopy. An on-chip microlens array amplifies scattering from gold nanoparticle labels, enabling kinetic monitoring with low-magnification optics. Dynamic fingerprints of individual events distinguish specific from nonspecific interactions, achieving wash-free digital biosensing with high sensitivity and specificity. This approach not only preserves the analytical advantages of kinetic profiling but also enables an exceptionally compact and portable optical configuration. Using cardiac troponin I, a time-critical biomarker for acute myocardial infarction, we demonstrate an ultralow detection limit of 0.051 pg mL −1 and validate clinical applicability in undiluted serum samples, paving the way for miniaturized, high-performance biosensors that can deliver laboratory-grade diagnostics at the point of need.

In situ monitoring PUVA therapy by using a cell-array chip-based SERS platform

Xiao-Peng Liu,Wen-Shu Zhang,Ya-Ning Wang,Wen-Qi Ye,Zhang-Run Xu

Psoralen ultraviolet A (PUVA) therapy has thrived as a promising treatment for psoriasis. However, overdose of PUVA treatment will cause side-effects, such as melanoma formation. And these side-effects are often ignored during PUVA therapy. Hence, in situ monitoring therapeutic response of PUVA therapy is important to minimize side-effects. Aberrant expression of tyrosinase (TYR) has been proved to be associated with melanoma, indicating that TYR is a potential target for evaluation of PUVA therapy. Herein, we reported a strategy for in situ monitoring TYR activity during PUVA therapy by using a cell-array chip-based SERS platform. The cell-array chip was used to simulate cell survival environment for cell culture . Capture of single cells and living cell analysis were realized in the isolated microchambers. An enzyme-induced core-shell self-assembly substrate was used to evaluate TYR activity in living cells during PUVA therapy. The gold nanoparticle modified with a SERS reporter, 4-mercaptobenzonitrile (4-MBN), was used as the core. In the presence of oxygen and TYR, hydroxylation of l -tyrosine occurred, leading to the reduction of silver ion on the surface of gold cores. The growth of silver shells was accompanied by the increased SERS intensity of the reporter, which is related directly to TYR activity. The detection limit for TYR activity is 0.45 U/mL. Upregulation of TYR activity was successfully monitored after PUVA therapy. Notably, real-time and in situ information of therapeutic response can be obtained through monitoring PUVA therapy by using a cell-array chip-based SERS platform, which has great potential to guide the clinical application of PUVA therapy.

Novel double perovskite Cs2Na0.7Ag0.3InCl6: Mn2+ for deep UV chip-based high color-rendering WLEDs

Yifei Xu,Pengjun Zhang,Shuwei Hao,Chongqiang Zhu,Chunhui Yang

Novel double perovskite Cs 2 Na 0.7 Ag 0.3 InCl 6 : Mn 2+ material is developed. • Tunable dual emission covering full visible light range is fulfilled. • Novel perovskite material is excited by deep-ultraviolet light. • White LED with high CRI of 87.9 is fabricated by coating double perovskites and commercial BaMgAl 10 O 17 :Eu 2+ blue phosphor on deep-UV chip. The commercial potential of traditional double perovskites in white light-emitting diodes (WLEDs) has been constrained by low photoluminescence quantum yield (PLQY) and narrow emission spectrum, mainly due to parity-forbidden transitions and indirect bandgap characteristics. To address these issues, Ag  +  doped Cs 2 NaInCl 6 (CNIC) are synthesized via solvothermal method. Compared to the undoped sample exhibiting a low PLQY of 2.76%, Ag + doped samples show a high PLQY of 77.56%, attributing to STEs emission of 540 nm. Additionally, a series of Cs 2 Na 0.7 Ag 0.3 InCl 6 : x Mn 2+ novel materials are designed. The results demonstrate that an energy transfer pathway from STEs to Mn 2+ in co-doped system, enabling tunable dual emission across the entire visible spectrum. Notably, Cs 2 Na 0.7 Ag 0.3 InCl 6 : x Mn 2+ materials can be effectively excited by deep-ultraviolet (deep-UV) light. A WLEDs device is fabricated by coating this novel material and commercial BaMgAl 10 O 17 :Eu 2+ blue phosphor on deep-UV chip, producing warm white light with a CIE coordinate of (0.3609, 0.3576), a correlated color temperature (CCT) of 4460 K and a high color rendering index (CRI) of 87.9. These results show that Cs 2 Na 0.7 Ag 0.3 InCl 6 : x Mn 2+ novel materials have potential for application in solid-state lighting.

A multimetallic nanozyme enhanced colorimetric biosensor for Salmonella detection on finger-actuated microfluidic chip

Fan Jiang,Nana Jin,Lei Wang,Siyuan Wang,Yanbin Li,Jianhan Lin

Salmonella screening is essential to avoid food poisoning . A simple, fast and sensitive colorimetric biosensor was elaborately developed for Salmonella detection on a microfluidic chip through limiting air chambers for precise air control, switching rotary valves for accurate fluid selection, a convergence-and-divergence passive micromixer and an extrusion-and-suction active micromixer for efficient fluid mixing, and immune gold@platinum palladium nanocatalysts for effective signal amplification. The mixture of bacteria, immune magnetic nanobeads and nanocatalysts was first rapidly mixed to form nanobead-bacteria-nanocatalyst conjugates and magnetically separated for enrichment. After washing with water, the conjugates were used to catalyze colorless substrate and blue product was finally analyzed using ImageJ for quantifying bacterial concentration. The finger-actuated microfluidic chip enabled designated control of designated fluids in designated places towards designated directions by simple press-release operations on designated air chambers without any external power. Under optimal conditions, this sensor could detect Salmonella at 45 CFU/mL in 25 min.

Valveless gated injection for microfluidic chip-based liquid chromatography system with polymer monolithic column

Xiu-Li Wang,Ying Zhu,Qun Fang

In this work, we developed a microfluidic chip-based liquid chromatography (LC) system with valveless gated sample injection method and monolithic columns. The valveless LC system consisted of a microchip with a separation column and a balance column, two syringe pumps and a laser induced fluorescence detector. Nanoliter-scale sample injection was achieved by simply switching the ON and OFF states of the syringe pumps under gated injection mode, without the need of any mechanical valve. The balance column facilitated the gated injection process by balancing the back pressure in the sample channel with that in the separation channel. Under the gated injection mode, the sample injection volumes could be varied in the range of 0.4–2.4   nL. We investigated the effects of the mobile-phase composition, flow rate , and the sample injection volume on the performance of the LC system. Under the optimized conditions, two fluorescently labeled amines were successfully separated in less than 220   s with a column length of 1   cm. Theoretical plates ranging from 35,700 to 29,400 per meter, corresponding to plate heights ranging from 28 to 34   μm were obtained. The advantages of the present system include simple system structure, ease of operation, convenience for varying injection volume, and high sample loading capacity.

Versatile titanium dioxide inverse opal composite photonic hydrogel films towards multi-solvents chip sensors

Min Guo,Xiao-Qing Yu,Jin Zhao,Jia-Wei Wang,Ren-Kun Qing,Ji-Dong Liu,Xingjiang Wu,Liangliang Zhu,Su Chen

Photonic crystals with orderly periodic structure and manipulated structural colors have garnered widespread attention in sensors field. However, the unsatisfactory detection range, low sensitivity and inconvenient integration highly limit their further practical applications. In this paper, by combining TiO 2 inverse opal photonic crystal with polyacrylamide hydrogel, a novel organic-inorganic composite solvents responsive photonic hydrogel (SRPH) was developed. By encapsulating TiO 2 inverse opal photonic crystal with high refractive index in this hydrogel, the photonic hydrogels, therefore, possess bright structural colors. Due to the discrepancy in swelling rate of SRPH film in various solvents (ethanol, dimethyl formamide , dimethyl sulfoxide , etc.), it can identify these organic solvents through the rapid change of structural colors and detect the concentrations of the solvent in a wide range. Even for solvents with similar refractive index, it can be clearly distinguished, along with the excellent repeatability. Benefiting from the wide detection range and high sensitivity, the chip sensors were then developed by integrating multiple hydrogel films into the microfluidic system, which have high efficiency in solvent naked-eye multi-identification and concentration multi-detection. The new composite photonic hydrogels provide a reference for the design of functional photonic crystals and next-generation application of sensors .

Modeling nonalcoholic fatty liver disease on a liver lobule chip with dual blood supply

Kun Du,Shibo Li,Chengpan Li,Ping Li,Chunguang Miao,Tianzhi Luo,Bensheng Qiu,Weiping Ding

Nonalcoholic fatty liver disease (NAFLD) has emerged as a public health concern. To date, the mechanism of NAFLD progression remains unclear, and pharmacological treatment options are scarce. Traditional animal NAFLD models are limited in helping address these problems due to interspecies differences. Liver chips are promising for modeling NAFLD. However, pre-existing liver chips cannot reproduce complex physicochemical microenvironments of the liver effectively; thus, NAFLD modeling based on these chips is incomplete. Herein, we develop a biomimetic liver lobule chip (LC) and then establish a more accurate on-chip NAFLD model. The self-developed LC achieves dual blood supply through the designed hepatic portal vein and hepatic artery and the microtissue cultured on the LC forms multiple structures similar to in vivo liver. Based on the LC, NAFLD is modeled. Steatosis is successfully induced and more importantly, changing lipid zonation in a liver lobule with the progression of NAFLD is demonstrated for the first time on a microfluidic chip. In addition, the application of the induced NAFLD model has been preliminarily demonstrated in the prevention and reversibility of promising drugs. This study provides a promising platform to understand NAFLD progression and identify drugs for treating NAFLD. Statement of significance Liver chips are promising for modeling nonalcoholic fatty liver disease. However, on-chip replicating liver physicochemical microenvironments is still a challenge. Herein, we developed a liver lobule chip with dual blood supply, achieving self-organized liver microtissue that is similar to in vivo tissue. Based on the chip, we successfully modeled NAFLD under physiologically differentiated nutrient supplies. For the first time, the changing lipid zonation in a single liver lobule with the early-stage progression of NAFLD was demonstrated on a liver chip. This study provides a promising platform for modeling liver-related diseases.
