Photothermal hydrogel adhesives have yielded promising results for wound closure and infected wound treatment in recent years. However, photothermal hydrogel bioadhesives with on-demand removability without additional nanomaterials-based photothermal agents have rarely been reported in the literature. In this work, an injectable intrinsic photothermal hydrogel bioadhesive with an on-demand removal trait is developed through dynamic cross-linking of gelatin (Gel), tannic acid (TA) quinone, and borax for closing skin incisions and accelerating methicillin-resistant Staphylococcus aureus (MRSA) infected wound healing. The TA quinone containing polyphenol and quinone groups with multifunctional adhesiveness and intrinsic photothermal performance confer the hydrogel adhesive with near-infrared (NIR) responsive antibacterial activity. The cross-linking of pH-sensitive boronic ester (polyphenol−B) and Schiff base bonds endow the hydrogel with great self-healing capacity and on-demand removability. Moreover, the hydrogel possesses good biocompatibility, injectability, and hemostasis. The in vivo experiment in a rat cutaneous incision model and full-thickness MRSA-infected wound model indicate that the smart hydrogel can close wounds efficiently and treat infected ones, demonstrating its superiority in noninvasive treatment of cutaneous incisions and enhancing infected full-thickness wound healing.

The acquired drug resistance coupled with complex physiology and pathogenesis of biofilm challenges multidrug-resistant bacterial biofilm infection treatment. Herein, antimicrobial phytochemical isoliquiritigenin (ISL) loaded rhamnolipid (RL) modified chitosan nanoparticles (ISL@RL-CS) were constructed. The obtained ISL@RL-CS NPs were able to simultaneously eradicate methicillin-resistant Staphylococcus aureus (MRSA) biofilms at all stages and relieve accompanied inflammation. In the acid environment of biofilms, RL could be detached from the ISL@RL-CS and destroyed the biofilm structure. Then, the combination of phytochemical ISL and cationic chitosan could rapidly kill the dispersed bacteria, and inhibit the re-adhesion of residual bacteria, thus preventing the re-formation of new biofilms. Additionally, the inflammatory responses at the infection site were simultaneously alleviated by the anti-inflammatory features of ISL. In the MRSA-biofilm implant infected mice model, ISL@RL-CS demonstrated prominent biofilm eradication and anti-inflammation efficacy. Taken together, ISL@RL-CS with simple formulation but multi-functions provides a comprehensive and sustainable strategy for managing the intractable MRSA biofilm infections.

Comprehensive Summary The intensive use of antibiotics intensifies the development of bacterial resistance, which has become a serious problem globally. Methicillin-resistant Staphylococcus aureus (MRSA) has resulted in significant morbidity and mortality. Therefore, it is an urgent need to develop new antimicrobial drugs and administration methods. Herein, we report a dual functional diblock copolymer PLL 20 - b -PBLG 20 , which was prepared by superfast and water-insensitive polymerization on N -carboxyanhydrides (NCA) initiated by tetraalkylammonium carboxylate. In addition to direct antimicrobial activity, PLL 20 - b -PBLG 20 also exerts a synergistic bactericidal effect against MRSA with curcumin, a plant extract with antibacterial property. Moreover, PLL 20 - b -PBLG 20 successfully encapsulates curcumin to form nanoparticles via self-assembly. The combination of dual functional PLL 20 - b -PBLG 20 and curcumin holds promise in combating MRSA infections.

Wound healing is a complicated process that can be susceptible toward infection caused by bacteria. The surging drug resistance has become one of the biggest challenges to the treatment of these infections in public health. Herein, a new asymmetrical aza-boron-dipyrromethene (ABDP) probe is prepared and encapsulated with cholesterol-modified polyethylene glycol polymer (Chol-PEG 2000 -NH 2 ) to construct a PTT/PDT dual-modality antibacterial agent (the ABDP NPs) and provide a new material for the treatment of MRSA-based infections. The ABDP NPs show remarkable wide UV–Vis absorption at 300–850 nm and superior bactericidal effects against Staphylococcus aureus ( S. aureus ), Escherichia coli ( E. coli ), and Methicillin-resistant S. aureus (MRSA), which can be controlled by a phototheranostic antibacterial switch (808 nm laser). Histological analysis indicates that wounds treated by the ABDP NPs plus NIR laser irradiation exhibit more complete re-epithelialization, abundant collagen fibers, and higher hair follicle numbers than the vancomycin-treated group. This study provides a promising perspective using aza-BODIPY dye for the treatment of multidrug-resistant bacteria-based infections and clinical applications in the future.

The clinical treatment efficacy for implant-associated infections (IAIs), particularly those caused by Methicillin-resistant Staphylococcus aureus (MRSA), remains unsatisfactory, primarily due to the formation of biofilm barriers and the resulting immunosuppressive microenvironment, leading to the chronicity and recurrence of IAIs. To address this challenge, we propose a light-induced immune enhancement strategy, synthesizing BSA@MnO2@Ce6@Van (BMCV). The BMCV exhibits precise targeting and adhesion to the S. aureus biofilm-infected region, coupled with its capacity to catalyze oxygen generation from H2O2 in the hypoxic and acidic biofilm microenvironment (BME), promoting oxygen-dependent photodynamic therapy efficacy while ensuring continuous release of manganese ions. Notably, targeted BMCV can penetrate biofilms, producing ROS that degrade extracellular DNA, disrupting the biofilm structure and impairing its barrier function, making it vulnerable to infiltration and elimination by the immune system. Furthermore, light-induced reactive oxygen species (ROS) around the biofilm can lyse S. aureus, triggering bacterium-like immunogenic cell death (ICD), releasing abundant immune costimulatory factors, facilitating the recognition and maturation of antigen-presenting cells (APCs), and activating adaptive immunity. Additionally, manganese ions in the BME act as immunoadjuvants, further amplifying macrophage-mediated innate and adaptive immune responses and reversing the immunologically cold BME to an immunologically hot BME. We prove that our synthesized BMCV elicits a robust adaptive immune response in vivo, effectively clearing primary IAIs and inducing long-term immune memory to prevent recurrence. Our study introduces a potent light-induced immunomodulatory nanoplatform capable of reversing the biofilm-induced immunosuppressive microenvironment and disrupting biofilm-mediated protective barriers, offering a promising immunotherapeutic strategy for addressing challenging S. aureus IAIs.

Thyroid cancer treatment and bacterial infection management have long been challenging due to the limited effectiveness and side effects of conventional therapies. Here, we introduce molybdenum nitride (Mo₂N) nanomaterials as an innovative platform for dual-functional photothermal therapy, combining anti-tumor and antibacterial treatments. Mo₂N nanoparticles with an average diameter of ~ 2.45 nm were synthesized and systematically characterized. Upon near-infrared (NIR) laser exposure, Mo₂N demonstrated efficient photothermal conversion (efficiency ≈ 31.8%), leading to significant temperature elevation. In vitro studies confirmed a selective cytotoxic effect, with Mo₂N-mediated PTT inducing significant death in thyroid cancer cells while having a minimal impact on normal endothelial cells. In vivo , this platform led to significant tumor growth inhibition and accelerated healing of MRSA-infected wounds. Additionally, Mo₂N exhibited substantial antibacterial activity against Escherichia coli and methicillin-resistant Staphylococcus aureus ( MRSA ) under NIR irradiation, showcasing its effectiveness in combating bacterial infections. The combined anti-tumor and antibacterial actions of Mo₂N suggest a promising strategy for dual-functional photothermal therapy, where the photothermal effect not only inhibits tumor growth but also effectively eliminates bacterial infections during treatment. These findings indicate that Mo₂N-based photothermal therapy can serve as a versatile, multi-functional therapeutic tool, presenting a significant advancement in both oncology and infectious disease treatment. Graphical Graphical Headlights. Schematic illustration of the synthesis and applications of Mo₂N nanomaterials for antibacterial and anti-cancer therapy. (i) Material Synthesis: Mo₂N nanomaterials were synthesized by reacting aniline with HCl and GO, followed by stirring, centrifugation, addition of ammonium molybdate tetrahydrate, freeze-drying, and finally thermal treatment in a tubular furnace. (ii) Antibacterial and Wound Healing: Under 808 nm NIR irradiation, the Mo₂N nanomaterials accelerate wound healing by eliminating MRSA, reducing inflammation, and promoting tissue regeneration. (iii) Antineoplastic Activity: When exposed to NIR light, the nanomaterials induce selective cell death in thyroid cancer cells while showing minimal toxicity to normal cells, as further validated in an in vivo mouse model.

We engineer a hybrid bacterial vesicle nanoreactor for refractory MRSA pneumonia. • Hybrid membrane enables dual-targeting cascade and vaccine-like immune activation. • Cinnamaldehyde/ferrocene trigger ferroptosis to kill MRSA and disrupt biofilms. • Nanoreactor induces M2 polarization, reducing inflammation and boosting immunity. Intracellular methicillin-resistant Staphylococcus aureus MRSA pneumonia remains difficult to treat because bacteria can survive within macrophages, where poor antibiotic penetration, intracellular persistence, and dysregulated inflammation collectively drive recurrent infection. Here, we developed a hybrid bacterial vesicle-camouflaged cascade nanoreactor PCFP/CA@HMV for dual-targeted intracellular MRSA eradication and pulmonary immune remodeling. The hybrid membrane vesicle HMV integrates S. aureus membrane vesicles (SCMV), which provide homologous targeting and vaccine-like immunogenicity, with Lactococcus lactis vesicles (LMV), which contribute to anti-inflammatory immune regulation. This biomimetic coating enables a sequential targeting cascade: HMV first directs the nanoreactor toward MRSA-infected macrophages, and ROS-triggered membrane disassembly subsequently exposes cationic PEI-Fc to promote electrostatic binding to intracellular MRSA. After intracellular activation, cinnamaldehyde CA increases bacterial membrane permeability and depletes glutathione, while ferrocene Fc catalyzes endogenous H₂O₂ into hydroxyl radicals, thereby overwhelming bacterial antioxidant defenses and inducing catastrophic lipid peroxidation. This cascade-amplified ferroptosis-like antibacterial process efficiently eliminates intracellular MRSA. In parallel, the combined effects of bacterial ferroptosis and L. lactis vesicles promote macrophage polarization from the pro-inflammatory M1 phenotype toward the reparative M2 phenotype, suppress pro-inflammatory cytokines, with vaccine-like immunogenicity of SCMV, enhance anti-inflammatory cytokine production, and accelerate resolution of lung inflammation. By integrating pathogen-homing biomimetic delivery, cascade-responsive controlled release, ferroptosis-amplified antibacterial activity, and immune microenvironment remodeling, PCFP/CA@HMV provides a promising therapeutic strategy for refractory intracellular MRSA pneumonia. Co-infection with influenza A virus (IAV) and methicillin-resistant Staphylococcus aureus (MRSA) often causes severe pneumonia clinically; however, the role of innate immunity in this setting remains poorly understood. In our study, we established a murine co-infection model using low-lethality IAV and MRSA. Compared with single IAV or MRSA infection, co-infection with relatively low-lethality IAV and MRSA resulted in more severe pneumonia. Transcriptomic analysis indicated marked upregulation of genes involved in the pyroptotic signaling pathway. Consistently, flow cytometry and immunofluorescence analyses revealed caspase-1 activation and colocalization of gasdermin D (GSDMD) with macrophages in the lung. The RAW264.7 macrophage cell line was used for in vitro validation. Co-infection significantly enhanced the cleavage of caspase-1 and GSDMD in RAW264.7 cells. Furthermore, disulfiram, a pyroptosis inhibitor, was incorporated into the antiviral and antibacterial combination treatment. Although combined oseltamivir and linezolid treatment failed to fully alleviate lung injury, the inclusion of disulfiram, a GSDMD pore formation inhibitor, significantly ameliorated pneumonia symptoms and reduced inflammatory responses. Collectively, our findings highlight that macrophage pyroptosis contributes to the exacerbation of pneumonia induced by IAV and secondary MRSA co-infection. Inhibition of GSDMD-mediated pyroptosis may represent a viable therapeutic approach to alleviate disease severity and improve outcomes in lethal co-infection.

Background:Intracellular bacteria frequently result in chronic and recurrent infections. MRSA is one of the most prevalent facultative intracellular bacteria in clinical infections. The drug resistance of MRSA and the difficulty of most antibiotics in entering cells result in a suboptimal clinical efficacy of antibiotics in the treatment of intracellular MRSA. Bacteriophages represent a promising alternative therapy in the context of the current antimicrobial resistance crisis. Nevertheless, the low efficiency of phage entry into cells and their rapid inactivation remain challenges in the treatment of intracellular MRSA using phages. The utilization of functionalized carriers for the delivery of phages into cells and their protection represents a feasible strategy. Methods:In this study, a new MRSA bacteriophage (vB_SauS_PHM) was isolated from hospital sewage, exhibiting the characteristics of short incubation period, large lytic amount, and good environmental tolerance. Subsequently, vB_SauS_PHM was encapsulated by TAT peptide-functionalized liposomes through microfluidic technology and size-exclusion chromatography (SEC), forming a phage delivery system, designated TAT-Lip@PHM.Results:The encapsulation rate of the phage by TAT-Lip@PHM was 20.3%, and the cell entry efficiency was ≥90% after 8 h. The 24 h eradication rate of 300 μg/mL TAT-Lip@PHM against intracellular MRSA was 94.05% (superior to the 21.24% and 44.90% of vB_SauS_PHM and Lip@PHM, respectively), while the mammalian cell activity was >85% after 24 h incubation. Conclusions:The TAT-Lip@PHM effectively delivered the phage into the cell and showed an excellent killing effect on intracellular MRSA with low cytotoxicity. This work provides a technical reference for the application of phages in the treatment of intracellular bacterial infection.

Hyperglycemia usually causes microvascular damage, which leads to chronic wound healing and is highly susceptible to bacterial infections. Overdependence on antibiotics accelerates the evolution of drug-resistant strains, making the treatment of diabetes related infected wounds more complex and difficult. Therefore, it is urgent to develop new antibacterial materials and therapeutic regimens, especially for diabetic wound infected with drug resistant bacteria. Herein, we propose a strategy based on black phosphorus nano-snowflakes (BPSFs) enhanced hydrogel (BPSFs@H), in which the BPSFs function as photothermal-responsive nanozymes, dedicated to the repair of diabetic infected wounds. The designed BPSFs are near-infrared (NIR) triggered photothermal responsive, and are effective in inhibiting methicillin-resistant Staphylococcus aureus (MRSA) by generating reactive oxygen species (ROS). BPSFs@H hydrogel exhibited remarkable efficacy in promoting cell migration and neovascularization. Moreover, it demonstrated the capacity to modulate the secretion of bioactive components by macrophages, driving them toward an anti-inflammatory phenotype. Animal studies revealed that BPSFs@H hydrogel substantially promoted the regeneration of wound epithelial tissue and enhanced the healing efficacy of infected wounds. Through mechanisms including augmented collagen deposition, improved local inflammatory response, and accelerated angiogenesis, this hydrogel significantly enhances the wound repair process by promoting re-epithelialization.

Chitosan-based pluronic thermo-sensitive hydrogels represent a class of biomaterials characterized by their favorable biocompatibility and exceptional thermo-sensitive properties. The purpose of this study was to investigate their potential as carriers for embedding α-Mangostin (α-M), a bioactive compound derived from mangosteen peel, with the aim of inhibiting methicillin-resistant Staphylococcus aureus (MRSA). During the synthesis process, chitosan (CS), tannic acid (TA), pluronic F-127 (F127), and pluronic F-68 (F68) were combined to form the thermo-sensitive hydrogel, into which α-M was physically incorporated through crosslinking (designated as CTFF@α-M). We are very interested in the results obtained during the research process. The results of fluorescence imaging in small animals showed that CTFF extended the residence time of α-M in the body from 3 to 9 days, improved the bioavailability of α-M, and achieved the goal of sustained drug release. In vitro antibacterial experiments showed that CTFF@α-M (30 mg/mL) had an inhibition rate of 79.69% against MRSA, which was higher than CTFF@amoxicillin (AM) (5 mg/mL) increased by 71.68%. This study is expected to provide new insights into the integration of biomaterials and natural therapy, and emphasize the therapeutic potential of thermosensitive hydrogels and α-M in antibacterial treatment, especially in the fight against MRSA infection. Graphic In this study, a CTPP hydrogel was prepared using a physical cross-linking method, and α-M was encapsulated to create the CTPP@α-M drug-loaded hydrogel. Various characterization techniques, including SEM, FT-IR, and XRD, were employed to confirm its physicochemical properties. The hydrogel exhibited excellent water solubility, swelling capacity, sustained release characteristics, and biocompatibility, with the ability to transform into a semi-solid gel at 37 °C. In vitro experiments demonstrated that the CTPP@α-M hydrogel showed a powerful inhibitory effect on MRSA, exhibiting strong resistance to antibiotic resistance. Infection experiments using a mouse model confirmed that CTPP@α-M not only accelerated wound healing but also significantly reduced bacterial counts at the wound site. Moreover, the CTPP@α-M group significantly decreased the expression of pro-inflammatory cytokines while increasing the levels of anti-inflammatory factors, indicating its promising anti-inflammatory properties. Collectively, these data suggest that CTPP@α-M is a highly effective antibacterial thermo-sensitive hydrogel with excellent performance.

The rampant use of commercial antibiotics not only increases drug resistance but also causes a significant threat to human health. This study assessed the wound healing efficacy of hydrogels crafted from carboxymethyl chitosan (Cmc), polyglutamic acid (γ-PGA), tannic acid (TA), and carbazole (Car), with the aim of expediting the wound healing process. Hydrogels were formulated using Cmc/γ-PGA, Cmc/γ-PGA/TA, and Cmc/γ-PGA/TA/Car, followed by a thorough evaluation of their physicochemical attributes. Additionally, assessments encompassed cytotoxicity, antibacterial efficacy, wound contraction rates, histopathological parameters, immunofluorescent staining of CD31, CD86, and COL1A, along with the determination of serum concentrations of IL-1β, IL-6, and IL-10. The physicochemical analyses validated the successful synthesis of the hydrogels, which exhibited both safety and potent antibacterial properties. Topical application of Cmc/γ-PGA/TA/Car hydrogels notably accelerated wound contraction, as evidenced by heightened expression of CD31 and COL1A, alongside reduced serum concentrations of IL-1β and IL-6. In essence, the Cmc/γ-PGA/TA/Car hydrogel demonstrated a dual effect of mitigating inflammation and modulating the proliferative phase, that shows their abilities for application in the wound healing process.
