Fabrication and characterization of electrospun zein nanofibers incorporated with citronellol-rich Origanum vulgare essential oil

Mayire Abulitifu,Wei Qin,Wang Yingbo,Bai Xi

The citronellol-rich Origanum vulgare essential oil (OEO), which exhibits excellent antioxidant and antimicrobial activities, has limited use in food packaging owing to its high volatility and instability. This problem can be solved using electrospinning. In this study, OEO was incorporated into the zein matrix via emulsion electrospinning. OEO was obtained by hydro-distillation; gas chromatography–mass spectrometry analysis revealed that citronellol (44.34%) was a predominant compound in OEO. Scanning electron microscopy images demonstrated that the fibers had a bead-free and uniform structure with a diameter ranging from 215 ± 36 to 260 ± 72 nm. The X-ray diffractograms indicate the amorphous structure of the zein fibers. Fourier transform infrared absorption spectroscopy revealed an interaction between OEO and zein. The addition of OEO to the zein nanofibers affected its secondary structure. Thermogravimetric analysis showed that the OEO-loaded (OEO/zein) nanofibers exhibited high thermostability and that the water contact angle was enhanced. The encapsulation efficiency and loading capacity were in the range of 71.56–85.8% and 8.88–40.93%, respectively. The antioxidant activity of the OEO/zein nanofibers ranged from 45.40% to 71.50%. The OEO/zein fibers displayed antimicrobial activity against Staphylococcus aureus and Escherichia coli . In conclusion, OEO/zein nanofibers are promising alternatives for active food packaging.

Hyaluronic acid-coated zein nanoparticle-mediated resveratrol therapy for the reduction of cisplatin-associated nephrotoxicity

Yuan Ning,Ping Chen,Zhengnan Shen,Xing Liu,Huan Gu

Cisplatin (CDDP) is commonly used as an anticancer drug in clinical practice, but severe nephrotoxicity restricts it from exerting anticancer effects. Natural drugs, such as resveratrol, can alleviate the side effects of cisplatin, but their low solubility and gastrointestinal effects prevent them from working. Herein, we developed nanoparticles for kidney injury consisting of a biocompatible material, zein, as a carrier. HA-Zein/Res NPs were fabricated using low-molecular-weight hyaluronic acid coatings. This preparation is non-cytotoxic to renal tubular epithelial cells and can be used with confidence. Low-molecular-weight hyaluronic acid has inflammation-targeting properties and CDDP damage causes renal inflammation. Owing to this property of the low-molecular-weight hyaluronic acid coating, in vivo imaging experiments in mice demonstrated that the HA-Zein/Res NPs enabled more nanoparticles to accumulate in the renal sites affected by inflammation. Efficient resveratrol delivery alleviated kidney injury, and experiments demonstrated that HA-Zein/Res NPs could treat kidney injury while reducing the serum creatinine and urea nitrogen levels in mice. Collectively, these results indicated that this nanomaterial is a promising agent for reducing the clinical nephrotoxicity of cisplatin.

Oral delivery of Silybin via HA-DOCA-functionalized zein nanocolloids for amelioration of liver fibrosis

Lin Wen,Jijiao Wu,Yuxin Zhao,Zihao Sun,Chuipeng Liang,Xiaolian Liu,Jia Luo,Xiaofang Li

Silybin (SIL), a natural flavonoid with recognized anti-hepatic fibrosis activity, suffers from poor aqueous solubility and low oral bioavailability, which restrict its clinical utility. To address these limitations, we developed a novel oral delivery system, termed HDZN/SIL, based on hyaluronic acid–deoxycholic acid (HA–DOCA)-modified zein nanoparticles loaded with SIL. This system was fabricated via an anti-solvent precipitation method. Under optimized conditions, specifically at a zein-to-SIL mass ratio of 10:1, the resulting nanoparticles displayed a uniform spherical morphology with a smooth surface and an average diameter of 127.9 ± 8.2 nm. The formulation also demonstrated high drug loading (97.55 ± 1.02 %) and excellent colloidal stability. Following oral administration, HDZN/SIL achieved effective liver-targeted delivery. In a mouse model of CCl₄-induced hepatic fibrosis, HDZN/SIL treatment resulted in significantly enhanced therapeutic efficacy compared to free SIL, along with an improved safety profile. These findings support HDZN/SIL as a promising strategy to advance the clinical application of SIL.

Electrospun Poly(vinyl alcohol)-Stilbazole Quaternized/Zein-Tetracycline Hydrochloride Core-Sheath Nanofibers for Drug Release

Cui Jing,Wang Qing-Qing,Qiu Yu-Yu,Wei Qu-Fu

We developed a novel biocompatible material in the form of core-sheath nanofibers using coaxial electrospinning with tetracycline hydrochloride (TCH) as the model drug. To prepare nanofibrous membranes, poly(vinyl alcohol)-stilbazole quaternized (PVA-SbQ) and zein were first dissolved in acetic acid as the sheath form while drug TCH as the core form. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) images demonstrated that the nanofibers had a round and smooth surface morphology with an average diameter of 980±252 nm. It showed distinct core-sheath structure at the sheath-to-core flow rate ratios of 1 and 0.5, respectively. Differential scanning calorimetry (DSC) and X-ray diffraction (XRD) results revealed that the tetracycline hydrochloride was well distributed in the PVA-SbQ/zein matrix in an amorphous state. Fourier transform infrared spectroscopy (FTIR) analysis was used to verify the second-order interactions of the components, which revealed good compatibility of the components of the nanofibers. Thermal gravimetric (TG) analysis demonstrated that the core-sheath nanofibers improved the thermal stability of TCH. The nanofibers showed good cytocompatibility toward L929 cells, further confirming their potential use as drug delivery and wound dressing. In vitro drug release studies indicated that the core-sheath nanofibers exerted sustained release instead of initial burst release effect, and also followed the Fickian diffusion mechanism. The present study shows a simple but promising approach for the fabrication of novel biomaterials with structural characteristics for drug release profiles using coaxial electrospinning.

Metal–Phenolic Network Covering on Zein Nanoparticles as a Regulator on the Oil/Water Interface

Di Wu,Yalei Dai,Yunan Huang,Jin Gao,Hongshan Liang,Mohamed Eid,Qianchun Deng,Bin Zhou

Interfacial self-assembly has become a powerful force for regulating the amphipathy of Pickering emulsions on the oil/water interface. Herein, metal–phenolic supramolecular coatings, acting as a regulator on the oil/water interface, were fabricated on the surface of zein nanoparticles (NPs), as a consequence of which the prepared Pickering emulsions stabilized by the decorated zein NPs exhibited diverse properties, decided by different concentrations of zein, tannic acid (TA), and metal ions (Fe3+). Metal–phenolic network-decorated zein NPs named ZTFex NPs (ZTFe NPs represented zein/TA/Fe3+ NPs, and x represented different concentrations of compounds) exhibited increasing diameters of 100–110 nm. Three-phase contact angles also showed that the strong hydrophobicity of zein NPs could be decreased as a result of the formation of metal–phenolic networks. As for corresponding Pickering emulsions, the covering of TA–Fe3+ networks on zein NPs could enhance the stability of zein NP-based emulsion obviously, which might be due to the fact that ZTFex NPs revealed the ability to form strong films on the oil/water interfaces. ZTFe4 was selected as an optimal concentration because of its minimum size and excellent storage stability. Besides, it was also found that the diameter of ZTFe4-based emulsion enhanced with the increase in the oil phase. The rheological measurement results showed that both G′ and G″ increased with the increase of x and the oil phase. In general, our paper not only highlighted a straightforward method for the interfacial nanofabrication of solid particles but also provided a novel and potential strategy in Pickering emulsion applications.

Effects of Surfactants on Zein Cast Films for Simultaneous Delivery of Two Hydrophilic Active Components

Dongwei Wei,Fanhui Zhou,Hongdi Wang,Guijin Liu,Jun Fang,Yanbin Jiang

In order to prepare edible films with outstanding antimicrobials and antioxidants utilized in applications of food and pharmaceutics, in this study, effects of surfactants on zein cast films for simultaneous delivery of lysozyme (LY) and ascorbic acid (AA) were investigated, where sodium alginate (SA), soy lecithin (SL), and Pluronic f-68 (PF-68) were selected as surfactants. FT-IR tests indicated that SL or PF-68 dramatically changed secondary structure of zein composite films, which heightened the irregularity of the composite film and inhibited LY crystallization. Mechanical tests showed that highly flexible films exhibiting elongations between 129% and 157% were obtained when adding PF-68. Compared with the film without emulsifier, zein film containing SL and PF-68 showed approximately 7.51 and 0.55 times lower initial release rates for LY and AA respectively, which significantly improved the controlled release and heightened the anti-microbial and anti-oxidant activities of the film. Finally, emulsified mechanisms of the surfactants in zein films were proposed. Keywords:zein;surfactant;lysozyme;ascorbic acid;controlled release;emulsified mechanism

Zein-based films with γ-cyclodextrin metal organic framework/oregano essential oil for fruit preservation

Zhenbin Liu,Fengbing Yang,Liangbin Hu,Dan Xu,Guoqiang Zhang,Yumei Liao,Min Cui,Hongbo Li,Haizhen Mo

Fruits and vegetables are highly susceptible to quality degradation, resulting in substantial economic losses. In this study, an edible γ-CD-MOF was prepared using an ultrasound-assisted method and employed as a carrier for oregano essential oil (OEO) to fabricate OEO@CD-MOF nanoparticles. These nanoparticles were subsequently incorporated into zein protein to develop a Zein-OEO@CD-MOF composite film. The incorporation of 5% OEO@CD-MOF into the zein matrix significantly enhanced the composite film's mechanical properties, ultraviolet resistance, water vapor barrier, and antioxidant property. Furthermore, the Zein-OEO@CD-MOF composite film was applied for the preservation of strawberries, cherry tomatoes, and blueberries. Results demonstrated that the composite film effectively mitigated nutrient loss in the fruits, thereby significantly extending their shelf life. Therefore, the Zein-OEO@CD-MOF composite film represents a sustainable and efficient active packaging material with substantial potential for practical applications in food preservation.

Chitosan-coated zein microcapsules for rumen-protected docosahexaenoic acid delivery and enrichment in goat milk

Jianmin Zou,Genna Ba,Yan Zhang,Jian He,Chong Chen,Kai Zhou,Wenhua Jin,Hejing Tang,Senyang Hu,Zihao Wang,Wei Xiong,Liyi Chen,Huichang Zhong,Dian Wang,Wei Tan,Hongsheng Du,Pengjie Wang,Baocai Xu,Yinhua Zhu

A zein-chitosan composite microcapsule was developed for rumen-protected DHA delivery. • Microcapsules exhibited high encapsulation efficiency (>90%) and strong structural stability. • In vitro digestion showed 85.76% ruminal passage and > 75% intestinal release. • Compared with algal oil, the DHA conversion rate of milk reached 21.82% after adding microcapsules. Docosahexaenoic acid (DHA) is a long-chain omega-3 polyunsaturated fatty acid with significant nutritional value. Natural DHA-enriched milk is an excellent source, but DHA is lost in the rumen, limiting its enrichment in milk. Herein, a novel zein-chitosan (CS) composite microcapsule system was developed to bypass ruminal degradation and enable controlled post-ruminal DHA release. Zein-CS microcapsules and two zein-only formulations, namely freeze-dried zein microcapsules (Zein-FD) and spray-dried zein microcapsules (Zein-SD), were physically and chemically characterized. Addition of chitosan increased particle size, surface positive charge, and polydispersity index of nanoemulsions. For dried microcapsules, chitosan increased encapsulation efficiency and decreased surface oil. Fourier transform infrared spectroscopy and X-ray diffraction confirmed strong hydrogen bonding, electrostatic, and hydrophobic interactions among zein, chitosan, and DHA. In vitro simulated rumen digestion assays revealed that zein-CS microcapsules achieved a ruminal DHA passage rate of 85.75%, markedly outperforming Zein-FD (34.07–63.70%) and Zein-SD (45.60–58.05%), because chitosan suppressed burst release and premature lipid exposure in the rumen. Total DHA release during simulated stomach and intestinal digestion reached ≥75% for zein-CS capsules. Only Zein-CS preserved alpha-diversity and community structure of rumen microbiota comparable to untreated controls; conversely, oil, Zein-FD, and Zein-SD treatments reduced microbial richness and diversity, and stimulated Lachnospiraceae . Animals fed Zein-CS produced milk containing 10.40 mg/100 mL DHA, higher than goats fed algae oil (3.57 mg/100 mL), with a DHA transfer efficiency of 21.82%. Therefore, Zein-CS microencapsulation effectively shields DHA from ruminal biohydrogenation, preserves rumen microbial homeostasis, enables targeted gastrointestinal release, and enhances milk DHA enrichment.

Rapamycin loaded TPGS-Lecithins-Zein nanoparticles based on core-shell structure for oral drug administration

Zhonghui Xie,Zhenhai Zhang,Huixia Lv

Rapamycin as a novel macrolide immunosuppressive agent has been commonly used in organ transplantation owing to its stronger immunosuppressive effect, non-nephrotoxicity and lower side effect. However its drawbacks of low bioavailability and big individual difference remain to be improved in clinical application. Here rapamycin loaded TPGS-Lecithins-Zein nanoparticles (RTLZ-NPs) with core-shell structure were prepared by the phase separation method. The RTLZ-NPs were approximately 190.3  nm in size, with PDI and zeta potential about 0.256 and −19.71 mV respectively. Drug entrapment and loading achieved were about 86.64 and 25.73% respectively. Meanwhile RTLZ-NPs exhibited favorable enzymolysis resistance abilities in gastrointestinal environments and enhanced uptake in Caco-2 cells. The optimum absorption sites of rapamycin in the intestine were duodenum and jejunum as single-pass intestinal perfusion assay. Upon also considering the results of Caco-2 cell assay, it could be speculated that the transport of rapamycin in vivo involved active transport as well as P-glycoprotein (P-gp) based efflux. Finally, the relative oral bioavailability of RTLZ-NPS was 4.33 fold higher than free rapamycin in SD rat . Altogether the designed nanoparticles can be an efficient oral delivery strategy for rapamycin analogues to prevent the attacks from destructive enzymes, reduce cell efflux, increase cell uptake, and then enhance the oral bioavailability.

Preparation of zein-grafted mesoporous silica nanoparticles for improving Lambda-cyhalothrin insecticidal activity and biosafety

Shaoyang Sun,Shengfu Zhang,Zhenxin Lv,Ning Wei,Ze Lv,Xiaohan Meng,Jianguo Feng

Currently, stimuli-responsive pesticide carriers can effectively enhance the insecticidal effectiveness against target pests and safety to nontarget organisms , and further reduce environmental pollution. In this work, a pesticide delivery system (LCN@MSN-SS-Zein) based on pH and redox dual-stimuli-responsive was prepared by loading Lambda-cyhalothrin (LCN) onto biodegradable disulfide-functionalized mesoporous silica nanoparticles and combining with gatekeeper zein. The results showed that LCN@MSN-SS-Zein had a high pesticide loading (24.7%, w/w). The nanoparticles have excellent pH and redox dual response behavior and have significant controlled release characteristics. Under the stimulation of pH and glutathione , the disulfide bond and zein structure of the nanoparticles are decomposed to release LCN on demand. Compared with LCN technical, the stability of LCN@MSN-SS-Zein nanoparticles under ultraviolet light was significantly enhanced, and the adhesivity of these nanoparticles on the cabbage leaf surface was significantly improved. Biological activity tests showed that LCN@MSN-SS-Zein nanoparticles had higher insecticidal activity against Spodoptera exigua than Lambda-cyhalothrin emulsion oil in water (LCN@EW), and the control effect was more lasting, while the toxicity to zebrafish was significantly reduced. In addition, MSN-SS-Zein nanoparticles were safe for the growth of cabbage and had good biocompatibility. Therefore, LCN@MSN-SS-Zein nanoparticles provide a new strategy for effective pest control and have broad application prospects.

Encapsulation of quercetin into zein-ethyl cellulose coaxial nanofibers: Preparation, characterization and its anticancer activity

Shu-Fang Li,Jia-Hui Wu,Teng-Gen Hu,Hong Wu

In order to efficiently improve the colon-targeted delivery of quercetin , the hydrophobic core-shell nanofibers were fabricated to encapsulate quercetin using ethyl cellulose as the shell and zein as the core by coaxial electrospinning. The encapsulation efficiency of coaxial nanofibers reached >97 %. FTIR and XRD results revealed the interactions between quercetin and wall materials and quercetin was encapsulated in an amorphous state. The thermal stability and surface hydrophobicity of coaxial nanofibers were improved compared to the uniaxial zein fibers. After in vitro gastrointestinal digestion, the quercetin release from core-shell nanofibers was <12.38 %, while the corresponding value for zein fibers was 36.24 %. DPPH and FRAP assays showed that there was no significant difference in the antioxidant activity of quercetin before and after encapsulation. Furthermore, the encapsulated quercetin exhibited similar anti-proliferative activity against HCT-116 cells compared to the free form. The results suggest these coaxial nanofibers have potential applications in functional foods.

Effect of tannic acid modification on the interface and emulsification properties of zein colloidal particles

Bojia Li,Xinyi Wang,Jin Gao,Hongshan Liang,Di Wu,Shang Chu,Xiangwei Zhu,Bin Zhou

BACKGROUND Interface modification driven by supramolecular self-assembly has been accepted as a valuable strategy for emulsion stabilization enhancement. However, there has been a dearth of comparative research on the effect of simple complexation and assembly from the perspective of the responsible mechanism. RESULTS The present study selected zein and tannic acid (TA) as representative protein and polyphenol modules for self-assembly (coined as TA-modified zein particle and TA-zein complex particle) to explore the surface properties and interfacial behavior, as well as the stability of constructed Pickering emulsions to obtain the regulation law of different modification methods on the interfacial behavior of colloidal particles. The results demonstrated that TA-modified zein colloidal particles potentially improved the emulsifying properties. When the TA concentration was 3 mmol L −1 , the optimized TA-modified zein particle was nano-sized (109.83 nm) and had advantageous interfacial properties, including sharply reduced surface hydrophobicity, as well as a low diffusion rate at the oil/water interface. As a result, the shelf life of Pickering emulsion containing 50% oil phase was extended to 90 days. CONCLUSION Through multi-angled research on the properties of the interfacial membrane, improvement of emulsion stability was a result of the formation of viscoelastic interfacial film that resulted from the decrease of absorption rate between particles and interface. Using refined regulation to investigate the role of different sample preparation methods from a mechanistic perspective. Overall, the present study has provided a reference for TA to regulate the surface properties and interface behavior of zein colloidal particles, enriched the understanding of colloidal interface assembly, and provided a theoretical basis for the quality control of interface-oriented food systems. © 2023 Society of Chemical Industry.
