Incomplete phase inversion W/O/W emulsion and formation mechanism from an interfacial perspective

Xinyue Wang,Xinyue Lu,Lili Wen,Zongning Yin

Water-in-oil-in-water (W/O/W) double emulsion can be prepared by incomplete phase inversion method using both medium chain triglycerides (MCT) and isopropyl myristate (IPM) as oil phase, Span 85-Tween 80 (HLB values of 2.5-3.0) as mixed emulsifiers. The preparation method was simple, and the final double emulsions were proved of good microstructure and particle size distribution. Owning to the addition of Tween 80 to Span 85, interfacial tension, interfacial viscosity and modulus decreased, which contributed to the phase inversion. Furthermore, formation of reverse micelles under high-speed dispersion may be a hypothesis to explain the incomplete phase inversion phenomenon.

Fabrication of polymersomes with controllable morphologies through dewetting w/o/w double emulsion droplets

Wang Wei-cai,Shi Kai,Pan Yan-xiong,Peng Chao,Zhao Zi-liang,Liu Wei,Liu Yong-gang,Ji Xiang-ling

In this work, monodisperse giant polymersomes are fabricated by dewetting of water-in-oil-in-water double emulsion droplets which are assembled by amphiphilic block copolymer molecules in a microfluidic device. The dewetting process can be tuned by solvation between solvent and amphiphilic block copolymer to get polymersomes with controllable morphology. Good solvent (chloroform and toluene) hinders dewetting process of double emulsion droplets and gets acornlike polymersomes or patched polymersomes. On the other hand, poor solvent (hexane) accelerates the dewetting process and achieves complete separation of inner water phase from oil phase to form complete bilayer polymersomes. In addition, twin polymersomes with bilayer membrane structure are formed by this facile method. The formation mechanism for different polymersomes is discussed in detail.

Monodisperse Alginate Microcapsules with Spatially Confined Bioactive Molecules via Microfluid-Generated W/W/O Emulsions

Hejia Sun,Hui Zheng,Qiming Tang,Yuman Dong,Fengmei Qu,Yaolei Wang,Guang Yang,Tao Meng

A simple process is developed for the one-step preparation of dual-compartment alginate microcapsules with controlled size and structure from microfluid-generated water-in-water-in-oil (W/W/O) emulsion droplet. Unlike other methods that rely on transient W/W/O emulsion droplet, we introduce an aqueous two-phase system (ATPS) to form a stable W/W/O emulsion droplet as a template for preparing dual-compartment alginate microcapsules. Two different bioactive molecules are able to be spatially confined encapsulated in the shell and core of alginate microcapsules due to the partitioning effect of ATPS and the high viscosity of alginate solution. Moreover, an enzyme cascade reaction with a spatial confined glucose oxidase and horseradish peroxidase in the shell and core of alginate microcapsules confirms its excellent biocompatibility and high activity. This method provides a green platform for enzyme-catalyzed tandem reactions and controlled sequential release of multiple drugs based on alginate microcapsules.

Mechanical properties and microstructures of W–TiC and W–Y2O3 alloys fabricated by hot-pressing sintering

X.F. Xie,K. Jing,Z.M. Xie,R. Liu,J.F. Yang,Q.F. Fang,C.S. Liu,Xuebang Wu

Bulk pure tungsten (W), W-0.5 wt%TiC (WTC05) and W-1 wt%Y 2 O 3 (WY10) materials were prepared by a developed two-step hot-pressing (HP) sintering method. Metallographic observation, SEM and TEM characterization were conducted to obtain the information of material microstructures . The results indicate that a small amount of TiC or Y 2 O 3 addition could effectively improve the sinterability and reduce the grain size of the HP sintered WTC05 and WY10 alloys. The relative density of WTC05 and WY10 reaches as high as 99%, and the average grain size of WTC05 and WY10 are 2.61 μm and 3.57 μm, respectively, which are several times smaller than that of pure W. Among the three W materials, pure W has the poor tensile performance with insufficient strength and ductility, WTC05 has the highest tensile strength with the ultimate tensile strength of 1082 MPa at 300 °C, and WY10 has the best ductility and moderate strength. The relationship between the microstructures and the tensile properties has been investigated, and suggestions on microstructure parameters have been proposed for preparing high performance W-based materials.

Protocol for programming DNA crystal morphology using concentration-modulated W/O/W double-emulsion microfluidic droplets

Jia Liu,Xugen Chen,Lebing Wang,Feng Zhou,Jiantao Zhang

Summary Here, we present a protocol for generating concentration-modulated water-in-oil-in-water (W/O/W) double-emulsion droplets (DEDs) to program DNA crystal morphology. We describe assembly of a dual-inner-phase glass capillary microfluidic device, flow-rate-controlled co-encapsulation, osmotic shrinkage, and image-based morphology analysis. This protocol validates encapsulation predictability using methylene blue and enables aspect-ratio control of 2T crystals and directional morphology regulation in 13-mer systems. Qing-Yu Lu,Zhao-Jie Wang,Li-Yu Bai,Wen-Biao Zu,Zhong-Shun Zhou,Yan-Yan Zhu,Yun-Li Zhao,Xiao-Dong Luo

Ethnopharmacological relevance Caryopteris trichosphaera W. W. Sm., a traditional ethnic medicine, was recorded in the Compendium of Materia Medica for treating wound infection by pathogenic infection. However, its antibacterial potential and bioactive compositions against drug-resistant bacteria need to be validated. Aim of the study To investigate the chemical constituents of C. trichosphaera and explore its anti-MRSA component in vitro and in vivo , together with the antibacterial mechanism. Materials and methods Bioactive constituents investigation was carried out by phytochemical method and antibacterial screening. The antibacterial mechanism was predicted by network pharmacology, which was further validated by time-kill analysis, membrane function tests, multigenerational resistance induction assay and biofilm test, and metabolomics analysis in vitro. In addition, MRSA-induced epidermal infection in mice was selected to evaluate its pharmacological effect in vivo . Results Six antibacterial diterpenoids against MRSA and VRE with MIC values 4–32 μg/mL from C. trichosphaera were reported for the first time, in which the major compound cativic acid ( 1 ) disrupted MRSA cell membranes by modulating permeability, depolarization, and fluidity while increasing reactive oxygen species (ROS) and malondialdehyde (MDA) levels. It also displayed remarkable anti-biofilm activity without inducing bacterial resistance or cytotoxicity. Moreover, cativic acid affected MRSA biosynthesis of cofactors, amino acid biosynthesis, nucleotide metabolism by metabolomics analysis. Furthermore, cativic acid accelerated wound healing in MRSA-infected mouse skin wounds, even better than vancomycin. Conclusions The results supported the traditional use of C. trichosphaera , and presented unreported anti-MRSA agent, cativic acid, as a plant-derived bactericide in vitro and in vivo for the first time.

Efficient separation of O/W and W/O micro-emulsion by thermally responsive superantiwetting PVDF membrane

Yanhui Xiang,Yunze Wang,Haibo Lin,Yi Wang,Zhu Xiong,Fu Liu

ABSTRACT Efficient separation of both oil-in-water (O/W) and water-in-oil (W/O) micro-emulsion was accomplished by a novel thermally responsive PVDF membrane. PNIPAAm–PVP–VTES micro-hydrogel was elaborately synthesized and incorporated into PVDF membrane, which was validated by transmission election microscopy (TEM), dynamic light scattering (DLS), X-ray photoelectron spectroscopy (XPS), attenuated total reflection Fourier transformed infrared spectroscopy (FTIR-ATR), laser confocal microscopy and thermogravimetric respectively. The thermally responsive mechanism of the engineered PVDF membrane was proposed from aspects of molecule structure, micro-hydrogel, membrane topology and membrane performance. The permeability and separation efficiency for pure water, both O/W and W/O micro-emulsions were modulated by temperature. The engineered PVDF membrane exhibited superantiwetting, outstanding permeability, high separation efficiency (>   99.90%), excellent antifouling property and reliability for long-term application. Moreover, multistage separation of oil-in-water micro-emulsion was achieved at nano-scale. Dispersed micro-emulsion droplets bigger than 20   nm and 40   nm were completely removed by manipulating the temperature below and above LCST , respectively. The engineered PVDF membrane highlights its great potential in micro-emulsion separation.

Food-grade W/O/W emulsion-filled sodium alginate hydrogel for co-delivery of riboflavin and curcumin

Pengrui Wu,Xindi Wei,Zhihong Song,Chunling Nie,Zhonghui Shen,Yijun Liu,YanBo Wang,JianGuo Wang,Hong Zeng

Developing environmentally friendly delivery systems capable of encapsulating, protecting, and promoting the absorption of bioactive components in the human body is crucial for those with low oral bioavailability and poor stability. This study developed W/O/W emulsion-filled sodium alginate (SA) hydrogel for the co-delivery of hydrophilic riboflavin and hydrophobic curcumin. Firstly, W/O/W emulsion was prepared using peanut protein isolate (PPI)-bacterial cellulose nanofibers (BCNFs) as the hydrophilic emulsifier and lecithin as the lipophilic emulsifier, and the effect of stabilizer concentrations on emulsion stability was investigated. Results showed that PPI and BCNFs self-assembled into stable complexes through electrostatic interaction and hydrogen bonding, thereby improving the emulsification properties of PPI. Subsequently, SA was added to the outer aqueous phase of emulsions, and its influence on emulsion properties and stabilization mechanism was investigated. SA significantly improved the stability and rheological properties of PPI-BCNFs-stabilized W/O/W emulsions. Finally, the loading capacity of this system for riboflavin and curcumin and its gastrointestinal fate were evaluated. We found this delivery system efficiently co-delivered both hydrophilic and hydrophobic bioactive components and improved their photostability and thermal stability. Moreover, the bioaccessibilities of riboflavin and curcumin were significantly increased to 61.97% and 45.66%, respectively. This was attributed to SA forming a composite interfacial layer on droplets and a dense gel network in the continuous phase, thereby acting as a physical barrier to better protect the encapsulated components. This delivery system holds promise for the development of functional foods incorporating multiple nutraceuticals.

W-induced formation of Ir-WOx(W) flattened nanoparticles anchored on TiN support for acidic oxygen evolution

Bosheng Zhang,Na Zhou,Panchao Zhao,Junqiu Guo,Hemu Pi,Yuanbin Qin

W induces flattened Ir-WO x (W) nanoparticles for high Ir utilization. • Alloy-heterojunctional composite interfaces tune Ir electronic structure for OER. • W optimizes OER intermediates and restrains Ir agglomeration to amplify Ir catalytic activity. • TiN-supported Ir-WO x flattened nanoparticles synthesized through a green approach. • Enhanced acidic OER activity vs commercial IrO 2 . Reducing the Ir loading in anode reaction catalysts is crucial for the scalable commercialization of proton exchange membrane (PEM) water electrolysis. Herein, W-induced formation of Ir-WO x (W) flattened nanoparticles with alloy-heterojunctional composite structure anchored on TiN support (Ir-WO x (W)/TiN) catalyst via a spray drying and heat treatment process is designed. For Ir-WO x (W)/TiN catalyst, TiN serves as the catalyst support, effectively achieving high dispersion of Ir as nanostructure on the support and enhancing the atomic utilization of Ir. W induces the formation of Ir-WO x (W) flattened nanoparticles to further improve atomic utilization efficiency of Ir and optimize its surface electronic structure during the oxygen evolution reaction (OER). Meanwhile, alloy-heterojunctional composite interfaces formed between Ir-WO x (W) and Ir-Ti can further optimize the adsorption/desorption capability of Ir toward oxygen-intermediates, lowering the energy barrier of the rate-determining step in the OER and enhancing the overall activity of catalyst, which is confirmed by a DFT calculation. The Ir-WO x (W)/TiN catalytic material demonstrates excellent OER performance, achieving overpotential of 221 mV and 296 mV at current densities of 10 mA·cm −2 and 50 mA·cm −2 in 0.5 M H 2 SO 4 solution, respectively, with a low Ir loading of 0.15 mg·cm −2 . Furthermore, this work demonstrates a green and scalable synthetic route without wastewater generation.

Dual fluorescence images, transport pathway, and blood–brain barrier penetration of B-Met-W/O/W SE

Liang Fang,Junying Li,Hongyan Cheng,Huanhuan Liu,Caiyun Zhang

Borneol is an aromatic traditional Chinese medicine that can improve the permeability of the blood–brain barrier (BBB), enter the brain, and promote the brain tissue distribution of many other drugs. In our previous study, borneol-metformin hydrochloride water/oil/water composite submicron emulsion (B-Met-W/O/W SE) was prepared using borneol and SE to promote BBB penetration, which significantly increased the brain distribution of Met. However, the dynamic images, transport pathway (uptake and efflux), promotion of BBB permeability, and mechanisms of B-Met-W/O/W SE before and after entering cells have not been clarified. In this study, rhodamine B and coumarin-6 were selected as water-soluble and oil-soluble fluorescent probes to prepare B-Met-W/O/W dual-fluorescent SE (B-Met-W/O/W DFSE) with concentric circle imaging. B-Met-W/O/W SE can be well taken up by brain microvascular endothelial cells (BMECs). The addition of three inhibitors (chlorpromazine hydrochloride, methyl-β-cyclodextrin, and amiloride hydrochloride) indicated that its main pathway may be clathrin-mediated and fossa protein-mediated endocytosis. Meanwhile, B-Met-W/O/W SE was obviously shown to inhibit the efflux of BMECs. Next, BMECs were cultured in the Transwell chamber to establish a BBB model, and Western blot was employed to detect the protein expressions of Occludin, Zona Occludens 1 (ZO-1), and p-glycoprotein (P-gp) after B-Met-W/O/W SE treatment. The results showed that B-Met-W/O/W SE significantly down-regulated the expression of Occludin, ZO-1, and P-gp, which increased the permeability of BBB, promoted drug entry into the brain through BBB, and inhibited BBB efflux. Furthermore, 11 differentially expressed genes (DEGs) and 7 related signaling pathways in BMECs treated with B-W/O/W SE were detected by transcriptome sequencing and verified by quantitative real-time polymerase chain reaction (qRT-PCR). These results provide a scientific experimental basis for the dynamic monitoring, transmembrane transport mode, and permeation-promoting mechanism of B-Met-W/O/W SE as a new brain-targeting drug delivery system.

Preparation and drying of water-in-oil-in-water (W/O/W) double emulsion to encapsulate soy peptides

Xin Ying,Jiaxing Gao,Jing Lu,Changlu Ma,Jiaping Lv,Benu Adhikari,Bo Wang

Soy peptide solution (40%, w/w) was successfully encapsulated in a W 1 /O/W 2 double emulsion produced by a two-step emulsification process. Polyglycerol polyricinoleate (PGPR) was found to be an effective inner emulsifier compared to Span 60 and lecithin to produce stable W 1 /O primary emulsion. The primary emulsion was subsequently emulsified into an outer aqueous phase (W 2 ) containing octenyl succinic anhydride (OSA) starch and maltodextrin. The droplet size and encapsulation efficiency of the peptide solution in W 1 /O/W 2 emulsion were found to depend on the W 1 :O ratio, peptide concentration in the inner W 1 phase and homogenization condition of the secondary emulsification step. The double emulsion with the highest encapsulation efficiency (>80%) was prepared by: (i) using 40% (w/w) soy peptide solution as W 1 phase; (ii) controlling W 1 :O ratio at 3:7 (w/w) and (iii) homogenizing the emulsion at 10,000 rpm for 3 min. The freeze-dried microcapsule powder of W 1 /O/W 2 emulsion showed higher encapsulation efficiency (>70%) compared to spray-dried one. The freeze-dried microcapsule of W 1 /O/W 2 double emulsion developed in this study is a promising delivery matrix to encapsulate hydrophilic ingredients including peptides. Fourier-transform infrared spectroscopy (FTIR) spectra of the microcapsule powder indicated good compatibility between peptide and encapsulants.

Sunflower wax-structured oil phase confers robustness to W/O/W emulsion gel beads for enhanced probiotic viability

Yejun Zhong,Ziyi Zhu,Ruiyun Chen,Liqiang Zou,Wei Liu,Shan Sun,Jianyong Wu,Er Sheng Gong

Natural waxes in oil phase enhance structural integrity of W/O/W emulsion gels. • Sunflower wax yields superior probiotic survival during pasteurization and digestion. • Wax-driven oil solidification protects L. plantarum and S. thermophilus effectively. • Novel oil-phase gelled emulsion system advances probiotic delivery in food matrices. This study explored impact of natural waxes (beeswax, BW; rice bran wax, RBW; sunflower wax, SFW)-mediated oil-phase gelling on the physicochemical properties and probiotic-protective efficacy of W/O/W emulsion gel beads co-encapsulating Lactobacillus plantarum and Streptococcus thermophilus . Wax incorporation preserved “two-membrane, three-phase” emulsion architecture while increasing droplet sizes and viscosity, with SFW and RBW yielding denser, more uniform bead microstructures and reduced moisture mobility compared to controls (without wax, NW) and BW. XRD and DSC revealed β’-polymorphic crystals in RBW and SFW, correlating with higher melting points and thermal stability. During pasteurization, simulated gastrointestinal digestion, and 28-day storage in acidic orange juice, SFW beads exhibited superior probiotic viability, with log CFU reductions minimized to 2.6 for Lactobacillus plantarum and 2.0 for Streptococcus thermophilus , versus 3.4 and 3.1 in NW. This approach holds significant promise for improving probiotic encapsulation in functional foods, enabling targeted delivery and extended shelf-life under adverse conditions.
