Effects of resveratrol on pectin with different esterification degrees: Structure, function properties, and fermentation characteristics

Pan Wang,Bangfeng Yin,Ping Zou,Zhenjia Zheng,Dan Wang,Peiyou Qin,Wenting Zhao,Yuanyuan Zhao,Ye Liu,Xiaoyan Zhao

To address the limitation of resveratrol (Res) related to its low bio-accessibility and poor stability, Res-pectin complexes with varying degrees of esterification were developed. The encapsulation efficiency of Res in these complexes was determined, revealing the following order: Res-L106 > Res-L13 > Res-H121 > Res-H115. Upon non-covalent interaction with pectins, Res exhibited enhanced stability, reduced crystallinity, and modified microstructure. The complexes exhibited stable particle sizes across a pH range of 4 to 8, with the Res-L106 complex showing minimal sensitivity to variations in ionic strength and temperature. All complexes exhibited improved antioxidant activities compared to pectins alone, with the Res-L106 complex displaying the highest activity. The complex enhancing the bio-accessibility of Res follows the order of Res-L106 > Res-L13 > Res-H121 > Res-H115. Additionally, all complexes promoted the production of short-chain fatty acids (SCFAs), with butyric acid being the predominant product. Notably, the Res-L106 complex exhibited the highest SCFAs-producing capacity. In conclusion, this study provides valuable insights into the design and optimization of strategies to enhance the bioavailability of Res, highlighting the potential of low-esterified pectin as an effective delivery carrier for bioactive compounds.

Extraction and Characterization of Clausena lansium (Lour.) Skeels Fruit Pectin Based on Deep Eutectic Solvent

Jiafu Lin,Jihui Lin,XiaoYu Tao,Dan Zhang,Siwei Chen,Xiujuan Fu,Yu Luo,Jiaqi Zheng,Linyu Liu,Hong Niu

Pectin derived from the fruit of Clausena lansium (Lour.) Skeels, commonly known as Wampee, possesses diverse bioactivities and holds significant potential for applications in food products. To broaden Wampee's application range, deep eutectic solvent (DES) was optimized for pectin extraction using response surface methodology. The fundamental and functional properties of DES-extracted pectin (D-CP) were compared to water-extracted pectin (W-CP). Extracted under the optimal extraction conditions (oxalic acid:choline chloride ratio was 1:1, reagent content was 10.2% (w/v), extraction time was 154 min, temperature was 82.5°C, and liquid: solid was 10.0 mL/g), D-CP was identified as Rhamnogalacturonan-dominated pectin. D-CP's molecular weight (129 kDa) and degree of esterification (61.05%) were lower than those of W-CP. However, D-CP exhibited a superior yield (18.09%) and enhanced functional properties, including higher purity, improved emulsifying stability and activity, better foaming capacity, and stronger foam stability. Additionally, D-CP demonstrated excellent solubility alongside notable antioxidant activities. Overall, these advanced properties highlight D-CP's promising potential for applications in the food and pharmaceutical industries, with DES proving to be an effective solvent for efficient extraction of bioactive pectin from Wampee. Modified sunflower pectin with low molecular weight and methylation targets gut microbiota to alleviate obesity

Wenfei Wang,Huiting Yang,Shengrong Xiang,Siliang Jiang,Haifeng Zhang,Mengqi Zhao,Zihan Jiang,Ya Gao,Jie Li

Obesity has become a global public health crisis, and the development of safe and effective interventions are urgently needed. In this study, sunflower head pectins with different molecular weights and degrees of methylation were synthesized via enzymatic modification. A high-fat diet was used to induce obesity in a mouse model. The effects of different modified pectins on the regulation of lipid metabolism disorders were systematically evaluated, and 16S rRNA sequencing was used to elucidate the mechanism by which different modified pectins regulate gut microbiota homeostasis and affect body metabolism disorders. The results showed that low-molecular-weight and low-methoxyl pectin could significantly reduce the weight growth rate ( P  < 0.05) and reverse hepatic steatosis. Its mechanism was closely related to the improvement of microbiota composition and recovery of short-chain fatty acid levels. This research has laid a theoretical foundation for precisely regulating the structure of pectin to develop pectin-enriched functional foods

Mussel-Inspired Biodegradable Ca2+ Complex Double Network Pectin-Based Hydrogel for Hemostasis and Tissue Regeneration

Yanai Chen,Limin Chang,Xiangyu Liang,Ziheng Zhang,Jianglei Qin,Shenzhou Lu

Inspired by mussel adhesion mechanisms and the structural advantages of double network (DN) hydrogels, this study developed a catechol- and polyphosphate-modified natural biomacromolecular-based DN hydrogel to tackle critical wound healing challenges, including persistent inflammation, oxidative stress, and impaired angiogenesis. The hydrogel exhibits tailored mechanical adaptability to wound microenvironments, ensuring conformal coverage under high-glucose conditions. Its inherent hemostatic capacity stems from rapid interfacial adhesion and coagulation activation, addressing the bleeding complications commonly observed in wounds. Furthermore, the hydrogel actively modulates pathological microenvironments via ROS scavenging and anti-inflammatory effects while facilitating sustained release of bioactive components to synergistically promote angiogenesis, collagen deposition, and epithelial regeneration. In summary, this mussel-inspired glycosyl cyclic hydrogel integrated multifunctional therapeutic advantages including microenvironment regulation, dynamic adaptability, and pro-regenerative signaling into a single platform, demonstrating great potential as a next-generation dressing for refractory diabetic wound management.

Eco-friendly extraction and physicochemical properties of pectin from jackfruit peel waste with subcritical water

Wen-Jia Li,Zhi-Guo Fan,Ying-Ying Wu,Zhi-Guo Jiang,Rui-Cheng Shi

BACKGROUND Water is generally considered to be a safe and green solvent suitable for use in natural product extraction. In this study, an eco-friendly subcritical water method was used to extract pectin from waste jackfruit peel (JFP-S), which was compared with pectin obtained by the traditional citric acid method (JFP-C). RESULTS The extraction process was optimized using response surface methodology (RSM), and the optimum process parameters were as follows: extraction temperature 138 °C, extraction time 9.15 min, liquid / solid (L/S) ratio 17.03 mL g −1 . Under these conditions, the pectin yield was 149.6 g kg −1 (dry basis). Pectin obtained from the two extraction methods displayed a high degree of esterification and the monosaccharide composition was consistent. The galacturonic acid content of JFP-S and JFP-C was 52.27% and 56.99%, respectively. JFP-S had more hairy regions and side chains than JFP-C. The molecular weight of JFP-S was 113.3 kDa, which was significantly lower than that of JFP-C (174.3 kDa). Fourier-transform infrared spectroscopy (FTIR) indicated that two samples had similar pectin typical absorption peaks. According to differential scanning calorimetry (DSC), both JFP-S and JFP-C had relatively good thermal stability. JFP-S demonstrated lower apparent viscosity and elasticity than JFP-C. Meanwhile, the G′ and G′′ moduli of JFP-S were lower, which found expression in the gel textural characterization of the samples. CONCLUSION This work showed that the subcritical water method is an efficient, time-saving, and eco-friendly technology for the extraction of pectin from jackfruit peel compared with the traditional citric acid method. The physicochemical properties of pectin could be changed during subcritical water extraction. © 2019 Society of Chemical Industry

Regulatory Roles of Pectin Oligosaccharides on Immunoglobulin Production in Healthy Mice Mediated by Gut Microbiota

Shanshan Zhang,Haijuan Hu,Wanying He,Zafarullah Muhammad,Lufeng Wang,Fengxia Liu,Siyi Pan

Scope The prebiotic regulation of the gut microbiota is a promising strategy to induce protective humoral and mucosal immune responses. The potential immune-improving effects of pectin oligosaccharides (POS) in healthy mice and the potential mechanism mediated by specific intestinal bacteria are investigated. Methods and results POS is prepared using a hydrogen-peroxide-assisted degradation. Mice that consumed diets containing POS are tested for microbial community shifts, short-chain fatty acids (SCFAs), and immunoglobulin (Ig) production using quantitative real-time polymerase chain reaction, gas chromatography, and ELISA kits. Pearson's correlation analyses are performed between Ig production and specific intestinal bacteria or SCFAs. POS treatment significantly improves the growth of healthy mice. Moreover, 4-week POS administration results in a profound change in intestinal microbial composition and a significantly higher fecal concentration of acetate, which leads to substantial increases of the levels of fecal secretory immunoglobulin A and serum IgG. Conclusions The results suggest that the inclusion of POS in a diet can increase Ig production and optimize the composition of the gut microbiota. A significant correlation is observed between changes in Ig production and specific intestinal bacteria or acetate, providing insight into the mechanism of POS as a potential immune-enhancing supplement.

Effect of pulsed electric field on assembly structure of α-amylase and pectin electrostatic complexes

Weiping Jin,Zhifeng Wang,Dengfeng Peng,Wangyang Shen,Zhenzhou Zhu,Shuiyuan Cheng,Bin Li,Qingrong Huang

Pulsed electric field (PEF) could change the charge distribution of proteins and polysaccharides and affect their interactions and complexes aggregation, but those influences are not enough evaluated. Here, the effects of PEF on the complexes of α-amylase and pectin driven by electrostatic binding were studied. Changes in molecular conformation of α-amylase and assembly structure of α-amylase/pectin complexes were orderly assessed by fluorescence, FTIR , DSC , enzyme activities, particle size, ζ-potential, CLSM, and SEM. After PEF treatment ( E ~20 kV/cm, t exp ~1 ms, and 5 cycles), the intrinsic fluorescence of α-amylase was quenched, the content of β-sheet increased, enzyme activities lose almost 80%, and the denatured temperature increased. Ζeta-potential of α-amylase/pectin complexes did not change significantly, but the particle size rose gradually. The particle revolution of α-amylase/pectin complexes was recorded by Turbiscan, and the size growth model fit the Allometric function well. Finally, the complexes of α-amylase and pectin after PEF treatment tended to the branched, ring, or circles-like shape.

Characterization of pectin from grapefruit peel: A comparison of ultrasound-assisted and conventional heating extractions

Wenjun Wang,Xiaobin Ma,Peng Jiang,Lyulin Hu,Zijian Zhi,Jianle Chen,Tian Ding,Xingqian Ye,Donghong Liu

In the present study, properties of pectin from grapefruit ( Citrus paradisi Macf. cv. Changshanhuyou) peel extracted by ultrasound-assisted extraction (UP), were compared with those of pectin extracted by conventional heating extraction (CP). UP exhibited a lower molecular weight ( M w ) but more concentrated M w distribution than CP. Although both UP and CP were high-methoxyl pectin, UP showed significantly lower degree of methoxylation but higher degree of acetylation. Monosaccharide composition analysis showed that the molecular structure of UP possessed more branch chains than CP, but little difference was observed in the chemical structures between UP and CP from FT-IR and NMR spectra analysis. X-ray diffraction (XRD) analysis further confirmed that UP had decreased level of crystallinity than CP. Moreover, thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) indicated UP had higher thermal stability. UP exhibited lower apparent viscosity and elasticity than CP, showing that ultrasound affected the rheological properties of pectin. Besides, antioxidant activity and lipase inhibitory capacity of UP significantly enhanced compared with CP.

Effect of interaction between ovotransferrin fibrils and pectin on properties of oleogel-based Pickering emulsions

Yue Dong,Zihao Wei,Changhu Xue

The effect of interaction between ovotransferrin fibrils (OVTFs) and citrus pectin (CP) on the properties of oleogel-based Pickering emulsions was innovatively explored. Firstly, the interaction between OVTFs and CP was investigated by constructing OVTF–CP complexes. OVTF–CP complexes with better stability were obtained when OVTFs to CP mass ratio of 3:1 at pH 5.0, which exhibited pearl chain-like structures by electrostatic attractions. Subsequently, to discuss the effect of interaction between OVTFs and CP on the properties of oleogel-based Pickering emulsions, oleogel-based OVTF-stabilized Pickering emulsion (OE) and oleogel-based OVTF–CP complex-stabilized Pickering emulsion (OCPE) were developed. The interaction between OVTFs and CP endowed OCPE with better stability, smaller droplet size, more prominent gel-like structure, higher viscosity and superior texture properties compared with OE. To construct an efficient delivery system for nutraceuticals, OE and OCPE were used to load curcumin . Compared with curcumin-loaded OE, curcumin-loaded OCPE had excellent protection for curcumin, higher rate of lipolysis and bioaccessibility of curcumin. Our work demonstrates that the interaction between OVTFs and CP can regulate the properties of oleogel-based Pickering emulsions, which will provide new insights into the formation and application of protein fibril–polysaccharide complexes. This may contribute to the precise development of Pickering emulsions with protein fibril–polysaccharide structures and ideal properties.

Tough, high conductivity pectin polysaccharide-based hydrogel for strain sensing and real-time information transmission

Yiying Liu,Ruonan Liu,He Liu,Deliang Li,Simian Fu,Kaiming Jin,Yugui Cheng,Zhiwei Fu,Fei Xing,Ye Tian

Hydrogels from natural polymers are eco-friendly, biocompatible and adjustable for manufacturing wearable sensors. However, it is still challenging to prepare natural polymer hydrogel sensors with excellent properties (e.g., high conductivity). Here, we developed a physically cross-linked, highly conductive and multifunctional hydrogel (named PPTP) to address this challenge. The natural renewable pectin-based PPTP hydrogel is synthesized by introducing tannic acid (TA), calcium chloride (CaCl 2 ), and sodium chloride (NaCl) into the pectin/polyvinyl alcohol (PVA) dual network structure. The hydrogel exhibits excellent characteristics, including unique tensile strength (2.6155 MPa), high electrical conductivity (7 S m −1 ), and high sensitivity (GF = 3.75). It is also recyclable, further enhancing its eco-friendly nature. The PPTP hydrogel can be used for monitoring human joint activities, as flexible electrodes for monitoring electrocardiogram (ECG) signals, and touchable screen pen for electronic skin. Moreover, when combined with Morse code and wireless Bluetooth technology, PPTP hydrogels can be used for underwater and land information encryption, and decryption. Our unique PPTP hydrogel offers promising opportunities for medical monitoring, information transfer, and human-computer interaction.

Novel Pickering emulsion gels stabilized solely by phenylalanine amidated pectin: Characterization, stability and curcumin bioaccessibility

Chenmin Zheng,Yufan Huang,Xiaojing Liang,Bihua Shen,Guoguang Zhang,Peng Fei

Pickering emulsion gels represent a novel class of non-toxic and biocompatible emulsions, offering extensive applications in the pharmaceutical and food additive sectors. This study delineates the synthesis of Pickering emulsion gels utilizing native and amidated pectin samples. Phenylalanine amidated pectin (AP) was procured via an ultra-low temperature enzyme method, while the control group (LP) adhered to an identical procedure without papain catalysis. Experimental outcomes revealed that the AP Pickering emulsion gel manifested superior stability compared to pectin emulsion samples (PE and LP). The Pickering emulsion gel from 5 % amidated pectin (5AP) retained stability throughout a 14-day emulsion stability assessment. Furthermore, all emulsion samples were evaluated for their capacity to deliver and sustain curcumin within an in vitro digestion simulation. Rheological properties and oil droplet size results indicated that the 5AP Pickering emulsion gel exhibited optimal cream index and emulsion stability, effectively inhibiting premature water-oil stratification within the emulsion and augmenting curcumin bioaccessibility. Within the in vitro digestion simulation, the 5AP Pickering emulsion gel demonstrated the highest curcumin bioaccessibility, measured at 17.96 %.

Mussel-inspired self-healing hydrogel form pectin and cellulose for hemostasis and diabetic wound repairing

Yanai Chen,Yu Zhang,Limin Chang,Weichen Sun,Wenhao Duan,Jianglei Qin

Diabetic wound is considered as a kind of chronic wound prone to infection and difficult to repair due to high glucose level in the blood of patients. In this research, a biodegradable self-healing hydrogel with mussel inspired bioadhesion and anti-oxidation properties is fabricated based on Schiff-base cross-linking. The hydrogel was designed from dopamine coupled pectin hydrazide (Pec-DH) and oxidized carboxymethyl cellulose (DCMC) for mEGF loading as a diabetic wound repair dressing. The Pectin and CMC as natural feedstock endowed the hydrogel with biodegradability to avoid possible side effects, while the coupled catechol structure could enhance the tissue adhesion of the hydrogel for hemostasis . The results showed the Pec-DH/DCMC hydrogel formed fast and can cover irregular wounds with good sealing effect. The catechol structure also improved the reactive oxygen species (ROS) scavenging ability of the hydrogel, which can eliminate the negative effect of ROS during wound healing. The in vivo diabetic wound healing experiment revealed the hydrogel as mEGF loading vehicle greatly enhanced the diabetic wound repairing rate in mice model. As a result, the Pec-DH/DCMC hydrogel could show advantages as EGF carrier in wound healing applications.
