Antioxidative and Cytoprotective Effects of Rosa Roxburghii and Metabolite Changes in Oxidative Stress-Induced HepG2 Cells Following Rosa Roxburghii Intervention

Yangchen Mao,Ruyi Sha,Yuhao Sun,Zhenzhen Wang,Jun Huang

Rosa Roxburghii(RR), a traditional Chinese medicinal fruit, is rich in bioactive substances that make it a potential natural antioxidant resource. This research aimed to study the antioxidant properties of RR by in vitro experiments and through intracellular assessment in H2O2-induced HepG2 cells. A non-targeted metabolic analysis was conducted to indicate changes in intracellular and extracellular metabolites. Differential metabolites and metabolic pathways were explored using PCA, PLS-DA, and KEGG pathway analysis. The results showed that RR rich in bioactive substances exhibited a significant antioxidative property in vitro and intracellularly. This property may be achieved by scavenging free radicals, increasing the activity of catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and the levels of bicinchoninic acid (BCA) while reducing the reactive oxygen species (ROS) generation. This study identified 13 differential metabolites intracellularly and 7 extracellularly, among which the key differential metabolites included D-glucopyranose, D-mannose, fructose, citric acid, malic acid, cholesterol, and cholestenone. These key metabolites primarily regulated glucose-related metabolism, the citrate cycle, and the primary bile acid biosynthesis pathway in H2O2-induced HepG2 cells. These findings provide potential application evidence of RR in the development of natural resources for functional foods.

Dual-Responsive Nanogels with Cascaded Gentamicin Release and Lysosomal Escape to Combat Intracellular Small Colony Variants for Peritonitis and Sepsis Therapies

Shuang Xie,Yu Li,Wenxiong Cao,Jiawen Peng,Kun Huang,Jie Meng,Xiaohong Li

Intracellular bacteria are the major cause of serious infections including sepsis and peritonitis, but face great challenges in fighting against the stubborn intracellular small colony variants (SCVs). Herein, the authors have developed nanogels (NGs) to destroy both planktonic bacteria and SCVs and eliminate excessive inflammations for peritonitis and sepsis therapies. Free gentamicin (GEN) and hydroxyapatite nanoparticles (NPs) with GEN loading and mannose grafts (mHA G ) are inoculated into ε-polylysine NGs to obtain NG@G1-mHA G2 through crosslinking with phenylboronic acid and tannic acid. The H 2 O 2 consumption after reaction with phenylboronic esters and the elimination of free radicals by tannic acid alleviates the escalated inflammatory status to promote sepsis therapy. After mannose-mediated uptake into macrophages, the acid-triggered degradation of mHA G NPs generates Ca 2+ to destabilize lysosomes and the efficient lysosomal escape leads to reversion of hypometabolic SCVs into normal phenotype and their sensitivity to GEN. In a peritonitis mouse model, NG@G1-mHA G2 treatment provides strong and persistent bactericidal effects against both extracellular bacteria and intracellular SCVs and extends survival of peritonitis mice without apparent hepatomegaly, splenomegaly, pulmonary edema, and inflammatory cell infiltration. Thus, this study demonstrates a concise and versatile strategy to eliminate SCVs and relieve inflammatory storms for peritonitis and sepsis therapies without infection recurrence.

A Novel Label-Free Electrochemical Immunosensor for the Detection of Thyroid Transcription Factor 1 Using Ribbon-like Tungsten Disulfide-Reduced Graphene Oxide Nanohybrids and Gold Nanoparticles

Wenjing Wang,Huabiao Tang,Leiji Zhou,Zhaohui Li

Thyroid transcription factor 1 (TTF1) is an important cancer-related biomarker for clinical diagnosis, especially for carcinomas of lung and thyroid origin. Herein, a novel label-free electrochemical immunosensor was prepared for TTF1 detection based on nanohybrids of ribbon-like tungsten disulfide-reduced graphene oxide (WS2-rGO) and gold nanoparticles (AuNPs). The proposed immunosensor employed H2O2as the electrochemical probe because of the excellent peroxidase-like activity of ribbon-like WS2-rGO. The introduction of AuNPs not only enhanced the electrocatalytic activity of the immunosensor, but also provided immobilization sites for binding TTF1 antibodies. The electrochemical signals can be greatly amplified due to their excellent electrochemical performance, which realized the sensitive determination of TTF1 with a wide linear range of 0.025–50 ng mL−1and a lower detection limit of 0.016 ng mL−1(S/N = 3). Moreover, the immunosensor exhibited high selectivity, good reproducibility, and robust stability, as well as the ability to detect TTF1 in human serum with satisfactory results. These observed properties of the immunosensor enhance its potential practicability in clinical applications. This method can also be used for the detection of other tumor biomarkers by using the corresponding antigen–antibody complex.

Tumor-targeted supramolecular catalytic nanoreactor for synergistic chemo/chemodynamic therapy via oxidative stress amplification and cascaded Fenton reaction

Xiaoyu Xu,Zishan Zeng,Jie Chen,Binyao Huang,Zilin Guan,Yanjuan Huang,Zeqian Huang,Chunshun Zhao

Utilizing intratumoral iron mediated Fenton reaction to induce cancer cells destruction, chemodynamic therapy (CDT) is an emerging tumor-specific therapeutic strategy. However, the CDT efficacy is severely restrained by insufficient endogenous H 2 O 2 and catalytic ferrous ions in tumor site. Herein, based on innovative small molecule prodrug design concept, an acid-labile ferrocene-modified cinnamaldehyde prodrug (Fc-CA) was first synthesized. Skillfully combining ROS-generator CA with Fenton catalyst Fc in its molecular structure, Fc-CA could synchronously elevate H 2 O 2 level and catalytic iron amount for enhanced CDT efficacy. Then, hydrophobic Fc-CA was efficiently encapsulated into hydrophilic cyclodextrin-modified hyaluronic acid conjugate (HA-CD) via host-guest interactions, to spontaneously form pH/redox dual-responsive supramolecular catalytic nanoreactor (HA-CD/Fc-CA NPs). The nanoreactor with high drug loading and stability, excellent catalytic activity and tumor specificity could preferentially accumulate in tumor tissues via EPR effect and CD44 receptor-mediated internalization. Through the cleavage of acid-labile hydrazone bond under acidic lysosome environments, the nanoreactor would be dissociated and release pre-protected CA for reinforced oxidation therapy and amplified H 2 O 2 level, which would in turn react with Fc through cascaded Fenton reaction, to generate highly cytotoxic •OH for enhanced cancer killing in a positive feedback process. Both in vitro and in vivo results verified excellent cascade-amplifying therapeutic performance and favorable biocompatibility of this nanoreactor. This study provides an innovative strategy for exploiting versatile organic therapeutic nanoplatforms, simultaneously activating tumor-specific oxidative stress amplification and cascaded enhanced Fenton reaction for superior cancer therapy with excellent biosafety.

Inhibition effect of thiol-type antioxidants on protein oxidative aggregation caused by free radicals

Man Lyu,Huijun Liu,Yalin Ye,Zongning Yin

This study was aimed to investigate the inhibition effect of thiol-type antioxidants on protein oxidative aggregation caused by free radicals and the underlying mechanisms using six different thiol-type antioxidants ( N -acetyl-L-cysteine, methionine , taurine, alpha-lipoic acid, glutathione and thioproline), Cu 2+ –H 2 O 2 as a free radical generator (mainly a hydroxyl radical generator) and bovine serum albumin as the model protein. The inhibition effect of these antioxidants on protein oxidative aggregation and protective effect against oxidative damage in mouse brain tissues were investigated using SDS-PAGE, intrinsic fluorescence, simultaneous fluorescence, thioflavin T fluorescence, Congo red absorbance and inverted microscope. The results showed that all six antioxidants could inhibit protein oxidative aggregation by scavenging free radicals. In addition, alpha-lipoic acid could also bind to proteins via hydrophobic interactions and thioproline could bind to proteins via hydrogen bonds and van der Waals forces , thereby showing much stronger inhibition effect than others. Moreover, alpha-lipoic acid and thioproline could effectively prevent oxidative damage of mouse brain tissues. These results suggest that alpha-lipoic acid and thioproline can effectively inhibit free radical-induced protein aggregation and brain damage, which are worth testing for further anti-Alzheimer properties.

Cerium oxide nanoparticles-mediated cascade catalytic chemo-photo tumor combination therapy

Hu Zitao,Ding Yin

ROS-based tumor therapy based on nanocatalytic medicine has recently been proposed for its tumor-specificity. However, a safe and highly efficient strategy towards getting high enough ROS to kill the hypoxic cancer cells is still a great challenge. Herein, we report a simple pH/H 2 O 2 -activatable, O 2 -evolving, and ROS regulating doxorubicin (DOX) and indocyanine green (ICG) co-loading PEGylated polyaniline (PANI) coated CeOx@polyacrylic acid (PAA) nanoclusters for highly selective and optimized cancer combination treatment. It can selectively and greatly enhance intracellular O 2 and ROS levels in tumor region, which depends on two-step catalytic properties of nanoceria (Ce 4+ /Ce 3+ = 3.46, neutral surface charge, mostly localize into the cytoplasm, pH 7.4–6.5, catalase-like catalytic agents convert to Ce 4+ /Ce 3+ = 0.58, negative surface charge, mostly localize into the lysosomes, pH 5–4, oxidase-like catalytic agents, triggered by near infrared (NIR) laser irradiation). Furthermore, the protective effect of polyethylene glycol (PEG), PANI, and PAA ensure that the nanoceria can only play the role of catalase under the irradiation of NIR light arrived at the tumor area. Moreover, loading of nanoceria and ICG onto PANI greatly enhanced photo thermal effect of nanoparticles (NPs), which is useful for killing cancer cells by relieving hypoxia and promoting cross-membrane drug delivery to further enhance photodynamic therapy and chemotherapy efficiency. The chemo-photo combination therapies fantastically inhibited tumor growth and prevented tumor recurrence in vivo , suggesting a smart nanotheranostic system to achieve more precise and effective therapies in O 2 -deprived tumor tissue.

The role of linalool in managing Alternaria alternata infection and delaying black mold rot in goji berry

Huaiyu Zhang,Yueli Zhou,Yue Yang,Lunaike Zhao,Hongbo Li,Lijie Long,Junjie Wang,Xuyan Wu,Zhao-Jun Wei

Alternaria alternata , a common fungal pathogen causes black mold in postharvest goji fruit. To explore the in vitro and in vivo impacts of linalool on the fungal infection, we treated A. alternata and infected goji fruit with varying linalool concentrations (0, 0.15, 0.45, and 1.35 mL L −1 ). Linalool repressed hypha growth, spore yield and germination, and germ tube length of A. alternata . Furthermore, we observed abnormal spore and mycelium morphology in the treated samples using optical, scanning electron, and transmission electron microscopy. Linalool reduced lesion diameter and disease incidence in goji fruit. Moreover, transcriptomic analysis of pathogen revealed that linalool inhibited nitrogen metabolism, altered chitin and β -1,3-glucan metabolisms, downregulated the key gene expression related to ergosterol synthesis and sulfur and glutathione metabolisms, regulated the enzymatic antioxidant system, and impacted the proportion of unsaturated fatty acids. Furthermore, linalool damaged the fungal cell wall integrity by inducing alkaline phosphatase and chitinase activities and β -1,3-glucan content and repressing β -1,3-glucanase and chitin synthase activities and chitin levels. Linalool elevated superoxide dismutase and catalase activities and declined peroxidase and glutathione reductase activities and glutathione content in A. alternata , leading to increased H 2 O 2 levels and ROS stress. Our data exhibited the promising antifungal effects of linalool with a future application in management of the postharvest rot of goji fruit.
