Zearalenone (ZEN) poses a potential threat on human and animal health partly through the nuclear factor (NF)-κB signaling pathway. In silico study suggested that rutin effective against TLR4 and NF-κB. A wetting test was designed to evaluate the effect and underlying mechanism of rutin in alleviating ZEN-induced inflammation in animals. Twenty-four female mice were randomly divided into 4 groups: control (basal diet), ZEN group (basal diet + ZEN), rutin group (basic diet + rutin), Z + R group (basal diet + rutin + ZEN). Results showed that rutin effectively alleviated ZEN-induced inflammation and damage of liver and jejunum in mice. Rutin addition reduced the content of lipopolysaccharide (LPS) in serum and liver mainly by improving the intestinal barrier function resulted from the production increase of short-chain fatty acids (SCFA). In sum, this study showed that rutin alleviated ZEN-induced liver inflammation and injury by modulating the gut microbiota, increasing the production of SCFA and improving intestinal barrier function, leading to the decrease of LPS in liver and the inhibition of MyD88 independent NF-κB signaling pathway in mice. Specifically, these findings may provide useful insights into the screening of functional natural compounds and its action mechanism to alleviate ZEN induced liver inflammation.

Rutin has attracted widespread attention due to its multiple pharmacological activities, but inherent defects such as poor water solubility and low bioavailability have greatly limited its clinical applications. Therefore, this study employed cyclodextrin glucanotransferase (CGTase) to catalyze the glycosylation modification of rutin, preparing rutin derivatives with improved water solubility. After optimizing the conditions, we isolated and purified 98.33 % of high-purity α-monoglucosyl rutin (α-GR), and its structure was also characterized. Subsequently, the cyclophosphamide (CTX)-induced murine liver injury model was used for activity evaluation. Our results showed that α-GR could significantly reduce liver function indicators (AST, ALT, and TBA), regulate antioxidant enzyme activities and inflammatory factor levels, and effectively attenuate CTX-induced liver injury. Furthermore, integrated metabolomics, transcriptomics, and RT-PCR analyses indicated that α-GR primarily exerts its hepatoprotective effects by intervening in key metabolites and gene expressions in CTX metabolic processes, related inflammatory responses, and oxidative stress pathways. Our results show that α-GR can effectively repair liver damage caused by CTX, thus laying the foundation for its use as a dietary supplement or therapeutic adjuvant.

The emergence of multidrug resistance (MDR) and extensive drug resistance (XDR) in Klebsiella pneumoniae strains has posed great threats to conventional antibiotics. Previous studies have shown that plant-derived flavonoids have inhibitory functions against pathogens . However, in K . pneumoniae , the antibacterial activity of different flavonoids against growth and biofilm formation remains a mystery. The aim of the present study was to evaluate the antioxidant abilities of different flavonoids, to screen active ingredients and to identify their inhibitory effects on K . pneumoniae growth and biofilm formation. In total, 10 flavonoids representing 4 major categories were screened and used in this study. The antioxidant capacity of each flavonoid was evaluated through a DPPH (2,2-diphenyl-1-picrylhydrazyl) assay. Rutin showed the highest level of free radical scavenging capacity, followed by kaempferol , luteolin , quercetin , apigenin , hesperidin , sinensetin, naringenin , naringin and 3,5,6,7,8,3′,4′-heptamethoxyflavone. The inhibitory effects of rutin and naringin on bacterial growth were also compared. The lowest MICs of rutin were found against K. pneumoniae ATCC700603 (1024 μg/mL) and E. coli ATCC25922 (512 μg/mL). However, the MBICs were not found. Rutin showed strong inhibitory ability against both the growth curve and biofilm production. The expression profiles of 15 biofilm-related genes were analyzed in biofilm cells both with and without rutin treatment . The luxS gene and wabG gene were downregulated significantly by rutin treatment. Correlation analysis showed that mrkA gene expression was positively correlated with biofilm biomass accumulation. Our study indicated that biofilm production is correlated with the expression of several genes rather than one. MrkA gene expression was positively correlated with biofilm biomass accumulation. Our study screened rutin as a potential agent to inhibit K . pneumoniae biofilm formation.

The development of promising environmentally friendly extraction tools has been one of the hottest topics in green chemistry. Among these, natural deep eutectic solvents (NADESs) as green extraction media have received widespread recognition. In this paper, with the aim of exploring the potential applications of NADESs in the extraction field, water-soluble n -propanol was used to form liquid–liquid biphasic systems with choline chloride/ maltose NADESs. The phase behavior of those systems was designed by adjusting the molar proportion of the hydrogen bond acceptors (HBA): hydrogen bond donors (HBD) and temperature. This allows for the systems tailored to the specific requirements of the target molecule, as shown by their application in the extraction of rutin. Investigation of the distribution behavior of rutin suggested that the partition coefficient and the extraction efficiency of rutin in the bottom phase increase significantly with decreasing temperature and increasing HBA : HBD molar ratio. And with the temperature of 288.15 K, the highest TLL of NADES (ChCl: Mal = 1:2) + NPA + water system achieved higher E% of 94.27 %. The interaction force between NADES and rutin was studied using the Hansen solubility parameter, it turned out that Δ δ i , j of the 2:1 and 1:2 M ratio of NADES was only 8.91, 5.12, which meaning that there may be strong interactions between NADES and rutin. In addition, the hydrogen bond ( Δ δ H ) of the NADES (HBA: HBD = 1:2) was 25.47, which is greater than other interaction force, revealing that the hydrogen bond is the maximum force between rutin and the NADES (HBA: HBD = 1:2). The energy ( Δ G T ), enthalpy ( Δ H T ) and entropy ( Δ S T ) calculated from the partition data of rutin was consistent with the free energy ( Δ G C ) , enthalpy ( Δ H C ) and entropy ( Δ S C ) changes of system, which confirmed that the driving force of the phase separation of the system drives the transfer of rutin to the lower phase. In addition, we used eco-friendly water to regenerate NADES. The recovery efficiency of rutin was explored while NADES was reused in three cycles. This result showed there was no change observed in the extraction capability for rutin in those system. Finally, a larger aggregate of size 1315 nm was observed by dynamic light scattering, which furthermore demonstrated that the hydrogen bonding interaction between NADES (HBA: HBD = 1:2) and rutin is the main driving force tool for enhancing rutin partitioning. This method presents a newly system constructed by NADES and NPA for the green extraction, revealing the feasibility of the system for rutin extraction and facilitating industrial applications.

BACKGROUND This study was conducted to investigate the effect of dietary rutin on the meat quality, antioxidant status and mitochondrial structure and function in the breast muscle of heat-stressed broilers. A total of 192 male broilers were randomly assigned into three groups and treated with normal control (CON), heat stress (34 °C, HS), and HS with 500 mg kg −1 rutin supplementation (HS + Rutin), respectively. RESULTS Dietary rutin significantly reversed HS-induced decrease in body weight, average daily feed intake, average daily gain, and feed efficiency. Rutin supplementation attenuated HS-induced impaired meat quality by decreasing the lightness, drip loss at 24 and 48 h, the peak time of free water ( T 22 ) and the peak area ratio of free water ( P 22 ), and increasing the pH 24h and peak area ratio of immobilized water ( P 21 ). Rutin supplementation promoted superoxide dismutase, glutathione peroxidase activities and total antioxidant capacity, and decreased malondialdehyde levels compared with the HS group. Moreover, rutin attenuated HS-induced mitochondrial damage by increasing the mitochondrial DNA copy number and improving mitochondrial morphology. Dietary rutin significantly increased mitochondrial biogenesis-related mRNA (proliferator-activated γ receptor coactivator-1α [PGC-1α], nuclear respiratory factor 1 [NRF1], and mitochondrial transcription factor A [TFAM]) expression via the AMP-activated protein kinase (AMPK) signaling pathway. HS significantly increased mitophagy-related genes and proteins (Parkin, PTEN-induced putative kinase 1 [PINK1], microtubule associated protein light chain 3-II [LC3-II]) expression, and dietary rutin significantly reversed these alterations. CONCLUSION Dietary rutin attenuated the HS-induced decline in meat quality and antioxidant capacity of broilers, which may be related to inhibition of the AMPK/PINK1–Parkin signaling pathway to attenuate mitochondrial damage. © 2023 Society of Chemical Industry.

Enzymatically modified isoquercitrin (EMIQ) is a glyco-chemically modified flavonoid exhibiting notably high biological activity, such as antioxidant, anti-inflammatory and anti-allergic properties. However, the utilization of expensive substrates such as isoquercitrin and cyclodextrin in the conventional approach has hindered the industrial-scale production of EMIQ due to high cost and low yields. Hence, the development of a cost-effective and efficient method is crucial for the biological synthesis of EMIQ. In this study, a natural cascade catalytic reaction system was constructed with α -L-rhamnosidase and amylosucrase using the inexpensive substrates rutin and sucrose. Additionally, a novel approach integrating gradient temperature regulation into biological cascade reactions was implemented. Under the optimal conditions, the rutin conversion reached a remarkable 95.39% at 24 h. Meanwhile, the productivity of quercetin-3- O -tetraglucoside with the best bioavailability reached an impressive 41.69%. This study presents promising prospects for future mass production of EMIQ directly prepared from rutin and sucrose.

The mycotoxin citrinin is often produced during fermentation of Monascus products. We studied the effects of flavonoids on citrinin production by Monascus aurantiacus Li AS3.4384 (MALA) by adding rutin, α-glucosylrutin, or troxerutin to the fermentation medium, in a first-of-its-kind study. Appropriate amounts of rutin, α-glucosylrutin, or troxerutin did not affect normal mycelial growth. Addition of 5.0 g/l of rutin only weakly reduced (29.2%) citrinin production, relative to inhibition by 5 g/l α-glucosylrutin or troxerutin (by 54.7% and 40.6%, respectively). In starch inorganic liquid culture media, addition of 20.0 g/l of troxerutin, followed by fermentation for 12 days, reduced citrinin yield by 75.26%. Addition of 15.0 g/l of troxerutin to low-starch peptone liquid fermentation media reduced citrinin yield by 87.9% after 14 days of fermentation, and addition of 30.0 g/l troxerutin to yeast extract sucrose liquid media for 12 days reduced citrinin yield by 53.7%.

Literature examples of non-aqueous Pickering emulsions stabilized by fat crystals are very rare. Moreover, the applications of rutin are limited due to its low solubility in both water and oils (less than 0.10 mg g−1 and 0.25 mg g−1, respectively). Thus, herein, we developed an optimum formulation of a non-aqueous self-double-emulsifying delivery system (SDEDS) containing rutin and evaluated its oral bioavailability. The new formulation stabilized by fat crystals (glycerol monostearate, GMS) and nonionic surfactants was prepared via a two-step emulsification process. The presence of a mixture of GMS crystals and nonionic surfactants effectively improves the stability of the emulsions. The non-aqueous SDEDS spontaneously forms oil-in-oil-in-water (O/O/W) double emulsions in the gastrointestinal environment with the inner oil phase mainly containing the active ingredients. It is stable at both 4 °C and 25 °C for 30 days and could enhance the dissolution properties of the active ingredients. Furthermore, the protection of rutin against digestion-mediated precipitation was observed when the formulation contained a high concentration of GMS crystals. The oral absolute bioavailability of rutin obtained from SDEDS (8.62%) is 1.76-fold higher than that of the actives suspension (4.90%). Thus, the non-aqueous SDEDS is an attractive candidate for the encapsulation of water-insoluble and simultaneously oil-insoluble nutrients (such as rutin) and for use in oral delivery applications.

This study investigated the effects of tanshinone IIA combined with rutin and squalene (RTS) on atherosclerosis (AS) associated with hyperlipidemia and the underlying mechanisms. Female New Zealand rabbits were subjected to a hyperlipidemia and AS model. Treatment with RTS for 4 weeks reduced the weight, and some serum lipid levels include serum TC, serum TG, serum LDL-C. RTS also improved lesions of the liver and aorta. In vitro analyses showed that RTS significantly activated LDLR-mediated hepatic lipid endocytosis, but blocking LDLR activation with si-SREBP2 attenuated the therapeutic effect, which demonstrated that RTS can activate the SREBP2/LDLR pathway mediating liver clearance of blood lipids. This study highlights the therapeutic potential of RTS in hyperlipidemia and AS. As a convenient form of a combination drug, RTS has shown considerable potential in the improvement of hyperlipidemia combined with AS.

Enzymatically modified isoquercitrin (EMIQ), a natural compound known for its antioxidant, anti-proliferative, and anti-inflammatory properties, exhibits remarkable bioavailability and low toxicity, making it a promising therapeutic agent for diabetic kidney disease. However, conventional chemical synthesis of EMIQ is hampered by inefficiency, high reagent consumption, and substantial by-product formation. In this study, EMIQ was produced from rutin and sucrose using a novel Heteropeptide Self-Assembly-Cascaded Dual-Enzyme (HSC-DE) system, which co-immobilized α -L-rhamnosidase and amylosucrase via SpyTag/SpyCatcher pairs. This system significantly enhances reaction efficiency by adding the substrate and gradient temperature, achieving a sucrose conversion rate of 98.3% within 24 h, which was increased by 5.7% compared to previous (92.6%). Notably, the yield of quercetin-3- O -tetraglucoside, the form with optimal bioavailability, reached 62.1%, an increase of 12% over previous methods. This study presents a novel, sustainable, and highly effective bioengineering strategy for the synthesis of EMIQ, highlighting its potential for scalable and environmentally friendly production.

Rutin, a natural flavonoid with anti-inflammatory and antioxidant properties, is widely found in plants and foods. To meet the demand for in situ detection of Ru in soft plant tissues, in this paper, a thin laser-induced graphene (LIG) electrode was prepared on polyimide (PI) and transferred to Ecoflex substrate, obtaining LIG/Ecoflex electrodes with good flexibility and tensile resistance. To further improve the electrochemical catalytic performance, nanocomposites of hafnium diselenide (HfSe 2 ), carboxylated single-walled carbon nanotubes (COOH-SWCNT), carboxylated graphene (COOH-GR) and nafion were also modified on the surface of LIG by a one-step dropping method. The HfSe 2 -COOH-SWCNT-COOH-GR-nafion/LIG/Ecoflex sensor could detect Ru in the range of 1 μM–700 μM under different pH values (4.5–7.4). It was also successfully used to in situ detection of Ru content in tomato leaves under different salt stress. The as-prepared sensor has important practical application prospects in the monitoring of plant physiological information in situ.

One-dimensional (1D) ordered mesoporous carbon hybrid nanotubes loaded with bimetallic oxide hollow nanospheres are expected to be very attractive electrocatalysts . In this study, through high-temperature pyrolysis , we generated Mo 2 C@C PNTs with outstanding electrical conductivity , a significant surface area , and effective catalytic activity by wrapping ordered mesoporous polypyrroles on the surface of 1D MoO 3 nanotubes using block polymers that self-assembled. FeMnO 3 hollow porous nanospheres have a large specific surface area, many electroactive sites, and excellent molecular/charge diffusion properties, which can be compounded with 1D ordered mesoporous nitride-rich carbon to solve the problem of poor electrical conductivity of metal oxides and to prevent the accumulation of 1D nanotubes. The composite showed excellent electrocatalytic activity towards rutin due to the synergistic interaction between Mo 2 C@C PNTs and FeMnO 3 . In the best case, rutin has a limit of detection (LOD) as low as 0.35 × 10 −9 M and a wide linear range (3 × 10 −9 to 3.1 × 10 −6 M). The sensor displays a remarkable level of selective response to rutin in the presence of other co-interfering species. Furthermore, it has superb repeatability and reproducibility. The sensor has shown outstanding recovery (95.7–104.1 %) in the practical viability of detecting rutin in beverages. This work not only provides a new candidate for the accurate detection of rutin but also provides insight into the exploration of advanced sensing materials.
