Biochemical activities of N,O-carboxymethyl chitosan from squid cartilage

Dongke Zhao,Jun Huang,Sheng Hu,Jianwei Mao,Lehe Mei

Chitosan was prepared by alkaline N -deacetylation of squid cartilage β-chitin and carboxymethylated derivatives with different degrees of substitution (DS) were synthesized. The DS of the derivatives calculated by pH titration were 0.64, 0.81, 1.0, 1.33 and 1.59. The antioxidant properties and bile acid binding capacity of the derivatives were studied in vitro . The carboxymethylation of chitosan caused enhancement of bile acid binding capacity. At 1   mg/mL, carboxymethyl chitosan (CMCS) showed a stronger scavenging effect than chitosan towards DPPH radicals. The CMCS scavenging effect on superoxide radicals was stronger than that of chitosan, and EC 50 values were below 5.6   mg/mL. The effectiveness of reducing power correlated with the DS of CMCS. At 1.2   mg/mL, the ability of CMCS to chelate ferrous iron was 100%, whereas that of chitosan was only 22.34%. This suggested that carboxymethylation is a possible approach to obtain chitosan derivatives with desirable biological properties.

Removal of tetracycline from water using ethylenediamine-modified magnetic chitosan

Yuankun Liu,Xinxia Zhang,Liyuan Zhao

In this work, the magnetic chitosan nanoparticles were prepared by in-situ co-precipitation method and then modified by glutaraldehyde and ethylenediamine. The effect dose of crosslinking agents for adsorbent preparation on tetracycline (TC) adsorption was investigated. The experiment results showed that the modified chitosan (EMMCS-G) had the best adsorption performance, which was more than twice that of raw magnetic chitosan. EMMCS-G was characterized by SEM, TEM, VSM, FTIR, XRD and XPS. The effects of experimental parameters such as time, temperature, pH, adsorbent dosage and Cu 2+ on TC adsorption by EMMCS-G were discussed. The result indicated that the optimal pH was 6.5. Under the conditions of an initial TC concentration of 50 mg/L and an EMMCS-G dosage of 50 mg, the removal rate reached an impressive 94%. The isotherm and kinetic studies of adsorption process illustrated that adsorption of TC onto EMMCS corresponded to Freundlich isotherm and pseudo-second-order kinetic models. The maximum adsorption capacity was 84 mg/g. Thermodynamic analysis revealed that adsorption process was both endothermic and spontaneous. The regeneration experiment suggested that the adsorbent possessed excellent reusability for further use. Moreover, the adsorption mechanisms of TC onto EMMCS-G mainly involved pore filling, π-π interactions, hydrogen bonding and electrostatic attraction.

Manufacture of layered collagen/chitosan-polycaprolactone scaffolds with biomimetic microarchitecture

Youjia Zhu,Ying Wan,Jun Zhang,Dengke Yin,Wenze Cheng

Chitosan-polycaprolactone (CH-PCL) copolymers with various PCL percentages less than 45   wt% were synthesized. Different CH-PCLs were respectively blended with Type-II collagen at prescribed ratios to fabricate a type of layered porous scaffolds with some biomimetic features while using sodium tripolyphosphate as a crosslinker. The compositions of different layers inside scaffolds were designed in a way so that from the top layer to the bottom layer collagen content changed in a degressive trend contrary to that of chitosan. A combinatorial processing technique involving adjustable temperature gradients, collimated photothermal heating and freeze-drying was used to construct desired microstructures of scaffolds. The resultant scaffolds had highly interconnected porous layers with a layer thickness of around 1   mm and porous interface zones without visual clefts. Results obtained from SEM observations and measurements of pore parameters and swelling properties as well as mechanical examinations confirmed that graded average pore-size and porosity, gradient swelling index and oriented compressive modulus for certain scaffolds were synchronously achieved. In addition, certain evaluations of cell-scaffold constructs indicated that the achieved scaffolds were able to well support the growth of seeded chondrocytes. The optimized collagen/CH-PCL scaffolds are partially similar to articular cartilage extracellular matrix in composition, porous microarchitecture, water content and compressive mechanical properties, suggesting that they have promising potential for applications in articular cartilage repair.

Amidoxime functionalized magnetic MXene/chitosan composites for efficient uranium extraction

Hao Liang,Nana Wang,Wei Tian,Hongping Zhang,Cuicui Li,Ruibin Guo,Zunli Mo,Nijuan Liu

Uranium extraction from unconventional uranium resources is among the major constraints to the development of the nuclear industry at present. In this dissertation, we here propose to effectively graft amidoxime groups on magnetic MXene and chitosan (CS) composites to extract uranium from low-enriched uranium solutions (MCF-AO). The data of adsorption experiments displayed that MCF-AO possessed fast adsorption kinetics (within 120 min), outstanding selectivity and salient adsorption capacity (1305.7 mg/g at pH=4, T=298.15 K) for uranium and could be easily separated with a magnet. Adsorption studies revealed a behavior consistent with the pseudo-second-order kinetic model and the Langmuir adsorption isotherm model, which is suitable for the spontaneous heat-absorption chemical monolayer adsorption process . Its sorption remains at more than 55 % of the original after 5 adsorption-desorption tests, and has great tolerance to interfering ions. Mechanistic research pointed out that the selective adsorption of uranium by MCF-AO was significantly enhanced by the presence of hydroxyl, amino and amidoxime groups. Meanwhile, MCF-AO exhibited superb resistance to two common bacteria (E. coli and B. subtilis ) and achieved more than 95 % uranium extraction capacity in simulated seawater , even more 4.12 mg/g of uranium extraction in Salt Lake water. Consequently, MCF-AO has a broad application prospect in the recovery and treatment of complex uranium systems.

Epichlorohydrin crosslinked carboxymethyl chitosan for uptake of heavy metal ions

Rubab Irshad,Muhammad Tahir Saddique,Shahid Nazir,Zaighum Tanveer

The carboxymethyl chitosan (CMCh) was synthesized and studied to carry out the process of wastewater treatment. The solubility of chitosan was enhanced by modifying its structural material with carboxymethyl chitosan. The present study was used to synthesize the novel adsorbent structure (CMCh/GLA/ECH) and its study was carried out by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), Scanning electron microscopy (SEM) characterization. Previously the study was carried out on glutaraldehyde (CMCh/GLA), a single crosslinker. The latest research is on dual crosslinker and its activity in removing metal ions from metal contaminated water. The CMCh was cross linked with glutaraldehyde (GLA) and epichlorohydrin (ECH). In this study, the single (CMCh/GLA) and dual cross-linked (CMCh/GLA/ECH) polymer adsorbents were used to study the adsorption capacity of the adsorbent. The metal ions removal efficiency for Co (Ⅱ) and Cu (Ⅱ) was studied. The results obtained were fitted to the Langmuir and Freundlich adsorption model. The Langmuir model showed monolayer adsorption on the homogeneous surface of the adsorbent. The cross-linked polymer (CMCh/GLA/ECH) showed the maximum efficiency of the adsorbent prepared. UV Curable Antibacterial Chitosan/Chlorogenic Acid/Polyacrylamide Hydrogel Packaging Film

Yang Yang,Sizhi Wu,Kailin Li,Xiao Wang,Haoling Huo,Peng Zhang,Qiwei Wang,Zhidan Lin,Lin Cao

Food spoilage, particularly in perishable meats, leads to significant economic loss and food safety concerns, while synthetic preservatives face increasing consumer scrutiny. To address the need for effective and biocompatible alternatives, this study developed a novel chlorogenic acid-grafted chitosan/acrylamide (CCAM) antibacterial hydrogel film. The film was synthesized through carbodiimide-mediated grafting followed by UV-initiated polymerization. Incorporation of chlorogenic acid produced a denser polymeric network in CCAM-1, lowering its equilibrium swelling ratio approximately 44% relative to CA‑free CCAM‑0 and elevating tensile strength to 411 kPa. The resulting CCAM‑1 exhibited potent broad‑spectrum antibacterial efficacy, inhibiting 97.97% of E. coli and 98.24% of S. aureus . The antibacterial effect was further enhanced by the pro‑oxidant activity of chlorogenic acid, which elevated intracellular ROS levels and disrupted bacterial redox homeostasis. In practical application, chicken breast wrapped with CCAM film and stored at 4 °C maintained acceptable microbial counts (< 7.0   log CFU/g) and sensory quality for up to 10 days, extending the shelf life by approximately 6 days compared to unpackaged samples. These findings demonstrate that the CCAM hydrogel film is a promising active packaging material, offering a green and efficient strategy for prolonging the shelf life of high-protein foods.

Lignin nanoparticle-reinforced chitosan hydrogels for enhanced solar evaporation performance

Siquan Wu,Xuan He,Xianjiao Wang,Liheng Chen,Jianglin Liu,Shuai Zhao,Xuliang Lin

Solar-driven interfacial evaporation requires photothermal materials that combine efficient solar absorption, continuous water transport, and resistance to salt accumulation. Herein, a lignin nanoparticle-reinforced chitosan hydrogel evaporator (LPCH) was constructed by integrating alkali lignin nanoparticles into a chemically cross-linked chitosan network. The uniformly dispersed lignin nanoparticles functioned as photothermal and nanoreinforcing domains, enhancing light harvesting and increasing the surface temperature to 39.5 °C under 1 sun irradiation. Interactions between the hydroxyl-rich lignin nanoparticles and chitosan regulated the water state within the hydrogel network, increasing the intermediate-water-to-free-water ratio from 0.35 for the chitosan hydrogel (CS) to 1.24 for LPCH and lowering the equivalent evaporation enthalpy to 2085 J g −1 . The optimized LPCH evaporator achieved an evaporation rate of 2.77 kg m −2  h −1 under 1 sun irradiation, together with an apparent solar-to-vapor efficiency of 152.4%. It retained an evaporation rate of 2.29 kg m −2  h −1 in 10 wt% saline water and maintained continuous operation over 10 h in simulated seawater. Outdoor tests using real seawater produced a peak evaporation rate of 3.47 kg m −2  h −1 and a daytime average rate of 2.38 kg m −2  h −1 . These results demonstrate a sustainable strategy for combining lignin-derived photothermal conversion, water-state regulation, and structural reinforcement in biomass-based solar evaporators.

Modified high-viscosity chitosan hydrogel for seawater immersion wound healing

Yihao Song,Yaodong Ning,Xiaoqiong Shi,Zhanying Lu,Wenli Jiang,Yao Xu,Xinlu Zhang,Yingxin Qu,Xiaoling Lu

Seawater immersion wounds carry high health risks owing to seawater's high-salt, ‌hypertonicity and diverse bacteria, and current therapies are limited and nonspecific, posing clinical challenges. In this study, high-viscosity chitosan (HVCS, viscosity >1000 mPa·s) from Antarctic krill shells was used as the matrix, glutamate as a molecular bridge linked dopamine and HVCS; high-viscosity dopamine-glutamate-chitosan (DGCS) was synthesized via amide bond-based covalent networks. DGCS inherits the rapid gel-forming ability of HVCS while showing its excellent favorable adaptability to seawater immersion wound surfaces. The introduction of dopamine and glutamic acid endows DGCS with more prominent mechanical properties and biological activities, including cell proliferation-promoting, antibacterial, and anti-inflammatory effects, and particularly inhibits the growth of specific Gram-negative pathogenic bacteria such as Vibrio vulnificus and Vibrio parahaemolyticus in vitro. More importantly, in seawater immersion wound rat models, DGCS hydrogel outperforms commercial chitosan dressings and silvadene in regulating wound inflammation, promoting collagen deposition, angiogenesis and healing. In conclusion, DGCS integrates antibacterial, anti-inflammatory and pro-healing functions, offering an effective multifunctional strategy for seawater immersion wound treatment, realizing the high-value-added valorization of shrimp shell waste and expanding HVCS's application in wound dressings.

Glycosylation-Engineered Chitosan Bioelectronic Interfaces for P. aeruginosa Gradient Analysis

Kun Yan,Zongrun Zhang,Xianzhu Yang,Chenguang Yang,Zhentan Lu,Dong Wang

Point-of-care diagnostics for pathogen detection are crucial for informed clinical pretreatment strategies, rendering the emergence of electro-fluidic systems increasingly attractive. However, both optical and electrical sensing strategies still face challenges in achieving high sensitivity and specificity. Herein, we present a facile and general approach for effectively integrating an electro-fluidic device that enables optical-electrical dual-mode gradient analysis of bacterial populations, mediated by a 2-lactobionamidoethyl methacrylate (SLM)-functionalized chitosan film (CS). Notably, this rationally synthesized CS@SLM exhibits robust interfacial monolayer adsorption performance, serving as a versatile sensing platform for specific bacterial recognition, uniform capture, and rapid accumulation of Pseudomonas aeruginosa. Furthermore, the biointerface demonstrates distinct concentration-dependent adsorption behavior, particularly in diluted bacterial solutions, achieving remarkably high removal efficiencies (78.3–100%) while facilitating bacterial detection at ultralow concentrations (≤101 CFU/mL, 0–10 min). These characteristics enable the creation of a pronounced concentration gradient through subsequent diffusion processes in microfluidic channels, allowing for differential analysis of bacterial populations with thousands of fold enhancement in signal variation. Importantly, the surface-immobilized bacteria retain their bioactivities and can be effectively integrated with electrodes via the engineered biointerface, highlighting significant potential for electro-fluidic multimodal sensing systems. Collectively, this general strategy and versatile biointerface provide new insights into designing advanced electro-fluidic devices for precise bacterial density control and effective infection management.

Enhancing triboelectric nanogenerator performance using chitosan-modified multiwalled carbon nanotubes

Irfan Khan,Saad Ahmed,Zeeshan Haider,Kashan Memon,Azam Fareed

The rising demand for a sustainable and immaculate energy source on a global scale is fascinating regarding the usage and potential effects of green energy. Triboelectric nanogenerators have emerged as some of the major green energy sources, converting mechanical energy into electrical energy by using different electropositive and tribonegative materials. We used surface-modified multiwalled carbon nanotubes incorporated with a chitosan membrane as a tribopositive material in triboelectric nanogenerators for improved efficiency. We demonstrate that the chitosan-modified multiwalled carbon nanotube-based tribopositive material with PDMS as the tribonegative layer with a constant device size of 2 × 2 cm2 produced a short circuit current density (Jsc) of 42.004 mA m−2, an open circuit voltage (Voc) of 244.90 volts, and a peak power density of 4.22 W m−2, which can light up more than hundreds of LED lights. The high electrical performance of chitosan-modified multiwalled carbon nanotubes as a tribopositive material is attributed to the increased surface roughness and charge separation without causing significant charge leakage, which is produced by carbon nanotubes. Henceforth, the present work successfully introduces the use of chitosan-modified multiwalled carbon nanotubes as a tribopositive material for enhancing the TENG's output performance.

Multi-factor protective chitosan/polyvinyl alcohol aerogel antibacterial buffer packaging

Jing Jin,Yu Ren,Hua Zhou,Yujie Hou,Peng Jin,Yonghua Zheng,Zhengguo Wu

Berries, being juicy and thin-skinned, are prone to mechanical damage and microbial contamination during transportation. Therefore, developing functional cushioning packaging becomes crucial to minimize product losses. Herein, modified MXene was used to immobilize silver nanoparticles (AgNPs) and graft trans -2-hexenal (E2H), synthesizing MXene-NH 2 /AgNPs/E2H preservative. Subsequently, the CPMAE aerogel buffer packaging was obtained by MXene-NH 2 /AgNPs/E2H, chitosan and polyvinyl alcohol, and then using to the strawberry preservation. The physical entangling, electrostatic and hydrogen bonding between chitosan and polyvinyl alcohol, as well as the interaction with MXene-NH 2 /AgNPs/E2H endow CPMAE aerogel excellent buffering properties. Besides, only about 67.40 % of E2H and 5.80 % of AgNPs were released from the CPMAE aerogel after 7 days. Although the immobilization of MXene-NH 2 significantly reduced the release rate of AgNPs and E2H, the CPMAE aerogel still had excellent antioxidant (36.5 μg FeSO 4 /5 mg) and antimicrobial (reaches 99 %) capabilities. Moreover, these features enable the CPMAE aerogel buffer packaging to have an efficient fresh-keeping effect on strawberries and extend the storage time at 4 ℃ to 12 days. This study offers a novel approach for applying chitosan aerogel in buffer and antimicrobial packaging for berries.

Multifunctional CuTax nanozyme-based chitosan edible coatings for fruit preservation

Fengxian Luo,Xinchuang Wang,Wei Tian,Beiwei Zhu,Jiangning Hu

The preservation of fresh fruits is critically challenged by oxidative degradation and microbial contamination, which lead to quality deterioration and reduced shelf life. In this study, we took advantage of the multiple benefits of Taxfolin (Tax) and copper to develop CuTax nanozymes, and which were showed to have excellent free radical scavenging ability, as evidenced by 90.3 % ± 0.2 % DPPH and 85.61 % ± 0.08 % ABTS free radical scavenging rates. Additionally, the CuTax exhibited peroxidase-like (POD-like) activity and effective glutathione (GSH) depletion. Moreover, the CuTax were found to effectively suppress the colony formation of E. coli and S. aureus , reduce bacterial viability, and disrupt bacterial structures. Ultimately, a CuTax/CS composite coating/films for food preservation was successfully developed using chitosan (CS) as a carrier and the protective efficacy against food spoilage was evaluated using bananas and apples as representative fruits. These findings suggest that CuTax/CS composite coatings offer a multifunctional approach to active food packaging that effectively extend the shelf life and preserve the quality of fresh fruits.
