An aggregation-induced emission (AIE)-featured amphiphilic polymer was synthesized by linking hyaluronic acid (HA) with a tetraphenylethene (TPE) derivative via hydrazone bonds (HA-hyd-TPE). The polymer exhibited a powerful AIE effect and could self-assemble to form polymeric micelles. The paclitaxel (PTX)-loaded micelles had a mean size as low as 82.2 nm, with a drug loading capacity of 10.7% and an entrapment efficiency of 79.8%. According to in vitro drug release testing, the micelles exhibited a rapid release of PTX under acidic conditions. In vitro cell viability experiments indicated the significant inhibition ability of the micelles on the growth and proliferation of MCF-7 cells. Cellular uptake assessment confirmed the excellent cellular imaging capability of HA-hyd-TPE micelles, and the micelles could be specifically internalized by MCF-7 cells. Furthermore, the superior antitumor activity and negligible systemic toxicity of the micelles were demonstrated by an in vivo antitumor test. All results suggested that HA-hyd-TPE micelles might be promising carriers for both bioimaging and efficient cancer treatment.

A synthetic polypeptide-based bioconjugate bearing tetraphenylethylene (TPE) at the polypeptide side chains, named mPEG-PPLGgTPE, was synthesized by ring-opening polymerization (ROP) of α -amino acid- N -carboxyanhydride (NCA) along with post-polymerization modification method. Interestingly, the bioconjugate enabled both the aggregation induced emission (AIE) effect at 469 nm and the aggregation caused quenching (ACQ) effect around 365 nm depending on water fraction in the tetrahydrofuran/water (THF/H 2 O) mixed solution, and this feature was very different from the commonly used TPE-based AIE polymers. The strong emission at the 365 nm was a rarely seen monomeric emission for the TPE units. Further studies on the fluorescence spectra indicated that the monomeric emission was not related with the secondary structure of polypeptides, but caused by the TPE unit itself. The mPEG-PPLGgTPE could self-assemble to AIE nanoparticles (NPs) in water in a mean size of 253 nm with polyethylene glycol (PEG) as the shells in surface. In addition, HeLa cell uptake level of the NPs was unsatisfactory according to the cell imaging results, which was independent of the polypeptides conformation, the PEG length and the size of NPs.

Organophosphorus pesticides (OPs) are frequently for pest control in the agriculture industry. Accumulation of OPs is harmful to the environment and human health. Thus, facile and portable detection of organophosphorus pesticides is of great importance. Among these methods, the fluorescence assay holds the advantages of high sensitivity, simplicity, nondestructive properties. Conventional fluorophores have the drawbacks of poor photostability and low signal-to-noise ratio due to their aggregation-caused quenching drawbacks at high concentration or in the aggregate state. Aggregation-induced emission fluorogens (AIEgens) are one key to develop next-generation fluorescence sensor due to their high emission efficiency in the aggregated state. 1,2-bis[4-(3-sulfonatopropoxyl) phenyl]-1,2-diphenylethene (BSPOTPE) is a typical AIE molecule containing two hydroxyl group . In this study, a fluorescence sensor based on BSPOTPE-SiO 2 –MnO 2 sandwich nanocomposites was fabricated. Thiocholine (TCh), which produced from acetylthiocholine(ATCh) by the hydrolysis of acetylcholinesterase (AChE), can “turn on” the fluorescence sensor. Based on the inhibition effect of OPs on AChE activity and the corresponding “turn off” effect on the fluorescence sensor, an AIE-based assay for OPs determination was developed. The fabricated sensor for paraoxon determination has a good linear relationship in the range of 1–100 μg/L and the LOD of 1 μg/L. Moreover, a simple, convenient fluorescence strip for visual semi-quantitative of OPs was fabricated, indicating this “on-off” fluorescent sensor is promising for on-site and infield detection.

This study aimed to add two functional components—antibacterial 45S5BGs particles and AIE nanoparticles (TPE-NIM+) with bioprobe characteristics—to the guided tissue regeneration (GTR) membrane, to optimize the performance. The PLGA/BG/TPE-NIM+membrane was synthesized. The static water contact angle, morphologies, and surface element analysis of the membrane were then characterized. In vitro biocompatibility was tested with MC3T3-E1 cells using CCK-8 assay, and antibacterial property was evaluated withStreptococcus mutansandPorphyromonas gingivalisby the LIVE/DEAD bacterial staining and dilution plating procedure. The fluorescence staining of bacteria was observed by Laser Scanning Confocal Microscope. The results showed that the average water contact angle was 46°. In the cytotoxicity test, except for the positive control group, there was no significant difference among the groups (p> 0.05). The antibacterial effect in the PLGA/BG/TPE-NIM+group was significantly (p< 0.01), while the sterilization rate was 99.99%, better than that in the PLGA/BG group (98.62%) (p< 0.01). Confocal images showed that the membrane efficiently distinguished G+bacteria from G−bacteria. This study demonstrated that the PLGA/BG/TPE-NIM+membrane showed good biocompatibility, efficient sterilization performance, and surface mineralization ability and could be used to detect pathogens in a simple, fast, and wash-free protocol.Keywords:aggregation-induced emission nanoparticles;antibacterial;antibacterial differentiation;bioactive glass;bioprobe;guided tissue regeneration (GTR)

Polysiloxane plays an irreplaceable role in aviation, automobile, flexible electronic devices, electrical engineering, etc. However, the poor mechanical strength makes it prone to damage during the working process. Therefore, it is of great significance to develop self-healing polysiloxane material. In this work, we prepared a silicon elastomer (CA-PDMS) by introducing citric acid (CA) and urea-group into the polydimethylsiloxane polymer chains, which featured a dynamic cross-linking network of multiple types of hydrogen bond synergies. The elastomer was mechanically tunable by adjustment of the ratio of the hydrogen bonds between the two parts. With an increase in the CA ratio, the elastomers showed a trend from “hard” to “soft”. The maximum elongation at break achieved was 1076.3%, revealing excellent stretchability. Due to the dynamic reversibility of hydrogen bonds, CA-PDMS showed conspicuous self-healing capability and CA had a positive impact on self-healing. Self-healing efficiency was >95% at 60 °C, while the self-healing efficiency reached 81.4% at room temperature. Compared with PDMS-0 without CA, the improvement was respectively about one and seven times. What's more, CA is commonly prepared as carbon quantum dots, compounding with polymer to obtain luminescent materials. Unlike previous research, this study introduced CA into the PDMS polymer chains through polymerization, generating untraditional light-emitting amide groups to achieve luminescence. As far as we know, this is the first time that CA has been reported to improve self-healing and ductility and realize AIE emission by co-polymerization with PDMS.

Mesenchymal stem cells (MSCs), due to their tumor tropism, are strongly recruited by various solid tumors and mobilized by inflammatory signals in the tumor microenvironment. However, effective cellular uptake is critical for MSC-based drug delivery. In this study, we synthesized a spherical copolymer, polyethylenimine–poly(ε-caprolactone), with aggregation-induced emission (AIE) material and the anticancer drug, paclitaxel, coloaded onto its inner core. This was followed by the addition of a transactivator of transcription (TAT) peptide, a type of cell-penetrating peptide, to modify the nanoparticles (NPs). Finally, the MSCs were employed to carry the TAT-modified AIE-NPs drug to the tumor sites and assist in simultaneous cancer diagnosis and targeted tumor therapy. In vitro, the TAT-modified AIE-NPs showed good biocompatibility, targeting, and stability in an aqueous solution besides high drug-loading and encapsulation efficiency. In vitro, the AIE-NPs exhibited a controllable release under a mildly acidic environment. The in vivo and in vitro studies showed high antitumor efficacy and low cytotoxicity of the AIE-NP drug, whereas biodistribution confirmed the tumor tropism of MSCs. To summarize, the MSC-based AIE-NP drugs loaded with TAT possessed good biocompatibility and high antitumor efficacy via the enhanced NP-drug uptake. In addition, the tumor tropism of MSCs provided selective drug uptake by the tumor cells and thus reduced the systemic side effects.

Using acyclic diene metathesis (ADMET) polymerization, we have synthesized a series of random conjugated copolymers from diene monomers based on tetraphenylethene (TPE), which is an aggregation-induced emission (AIE)-active molecule, and comonomers such as 1,9-decadiene, 2,7-divinyl-9,9-di-n-octylfluorene, and 1,4-dihexyl-2,5-divinylbenzene. These all-hydrocarbon copolymers exhibit good solubility and high molecular weights. As proved by 1H NMR spectra, chain segments composed of two monomers are randomly distributed in the polymers. Absorption and emission features from both TPE units and those conventional diene segments in conjugated copolymers are observed in their optical spectra. Because of the presence of AIE-active TPE units, emission intensities of the copolymers are greatly enhanced in tetrahydrofuran/water mixtures with increased water content. Moreover, the fluorescence of ADMET polymers can be quenched by palladium ions in quasi-aqueous solutions following a dynamic quenching mechanism. The fluorescence quench degrees are affected by both chemical structures of the copolymers and the solvent polarity. Our findings demonstrate that ADMET conjugated polymers have enormous potential as fluorescent “turn-off” probes specific for palladium ions even in the absence of a particular coordination interaction.

Herein, a series of conjugates combining bile acid and terpyridine units were developed. Based on the unique rigid skeleton and excellent amphiphilicity of bile acid fragment, the aggregation properties of three conjugates are finely regulated and the resulted aggregates can strongly confine and tune the rotation of terpyridine units to endow the three conjugates with excellent different AIE properties. Furthermore, with the excellent metal coordination properties, terpyridine units endow the conjugates with excellent metal response properties and lead to various colourful complexes. The supramolecular assembly morphology can also be well controlled by the metal ions . These biological conjugates with controllable AIE properties and metal tunable fluorescence and supramolecular assembly properties may provide a new avenue for the bioimaging and drug controllable delivery system.

A new nanoprobe, the luminescent diblock copolymer PNIPAM(MAh-4)-b-P4VP (PN4P), with pH- and thermo-responsive deprotonation-driven emission decay (DDED) and aggregation-induced emission (AIE) features was designed and synthesized. The nanoprobe PN4P can form micellar structures in water with reversible dual-responsive fluorescence (FL) behavior within a wide pH range of 2–11. The critical solution temperature was found at about 32, 30 and 27 °C as the pH switched from 2, 7 to 11. The critical pH value of the probe was about 4.0, and the micelles showed a core–shell inversion in response to pH and thermal stimuli, accompanied by a desirable emission tunability. P4VP as the micellar shell at pH = 2 was more easily dehydrated with the increase in temperature as compared to PNIPAM as the micellar shell at pH > 4. The strongest dehydration of the P4VP shell would make PN4P the most strongly aggregated and the most AIE-active, which supports the 2.10-fold most distinguished thermal-responsive emission enhancement at pH = 2. Moreover, a dramatic acidochromic redshift of the emission band from 450 (pH > 4) to 490 nm (pH = 2) was observed, and the maximum emission at pH = 2 was enhanced by about 2.07-fold as compared with that at pH = 7. Therefore, the probe displays the desired dual responses and good reversibility. AIE and DDED are the two major mechanisms responsible for the dual-responsive emission change, with AIE playing a more important role than DDED. This work offers a promising approach to interpreting temperature (range from 28 to 40 °C) and pH changes (range from 2 to 7) in water.

Photodynamic therapy (PDT) takes advantage of reactive oxygen species (ROS) to trigger the apoptosis for cancer therapy. Given that cell apoptosis is a form of programmed cell death involved with multiple suborganelles and cancer cells are more sensitive to ROS than normal cells, early confirmation of the apoptosis induced by ROS would effectively avoid overtreatment. Herein, we highlight an aggregation-induced emission (AIE)-based theranostic agent (TPA3) to in situ dynamically track mitophagy prior to late apoptosis. TPA3 showed high specificity to autophagy vacuoles (AVs), of which appearance is the signature event of mitophagy during early apoptosis and delivered photocytotoxicity to cancer cells and skin cancer tumors in nude mice under irradiation of white light. Furthermore, in situ monitoring of the dynamical mitophagy process involved with mitochondria, AVs, and lysosomes was performed for the first time under confocal microscopy, providing a real-time self-monitoring system for assessing the curative effect prior to late apoptosis. This fluorescence imaging guided PDT witness great advances for applying in the clinical application.

A novel technique of modulating the emission behavior of aggregation-induced emission (AIE) polymeric nanoparticles (AIE-PNPs) based on the polymer matrix was provided. White-light-emitting (WLE) AIE-PNPs were facilely synthesized through miniemulsion copolymerization of methyl methacrylate (MMA) and comonomers with various polarities by using a single luminogen with AIE and twisted intramolecular charge transfer (TICT) properties, (2-(3-cyano-5,5-dimethyl-4-(4-(1,2,2-triphenylvinyl)styryl)furan-2(5H)-ylidene)malononitrile (TPE-TCF)). The increased or reduced polarity of the polymer matrix could lead to red-shifted or blue-shifted emission of polymer/TPE-TCF AIE-PNPs, respectively. The polarity-dependent emission behavior of polymer/TPE-TCF AIE-PNPs was generally validated in the AIE-PNPs with various polymer matrices. The WLE polymer/TPE-TCF AIE-PNPs could be generally synthesized via miniemulsion copolymerization of MMA with isobornyl methacrylate, stearyl methacrylate, lauryl methacrylate, n-hexyl methacrylate, and cyclohexyl methacrylate at an appropriate monomer composition. By simply coating a layer of prepared WLE AIE-PNPs onto UV LED chips, the UV light could be successfully converted to white light, clearly demonstrating their great application potential in electronic and photonic devices.

Temperature-responsive nanomicelles with aggregation induced emission (AIE) property were prepared by the host–guest complexation of ferrocene functionalized tetraphenyl (TPE-Fc) and β-cyclodextrin-poly ( N -isopropylacrylamide) (β-CD-(PNIPAM) 7 ). The AIE chromophore TPE-Fc bound to the hydrophobic cavity of cyclodextrin serves as the core of micelles, and temperature sensitive PNIPAM serves as the shell to give the micelles good solubility. The size of the nanomicelles is about 100 nm. At the excitation wavelength of 340 nm, the strongest fluorescent emission peak was 421 nm. The introduction of cyclodextrin star polymer increased the fluorescence intensity of nanomicelles, thus improving the recognition of probe to Fe 3+ and Fe 2+ . The fluorescent probe can quickly detect Fe 3+ and Fe 2+ in water within 5 min even in the presence of various interfering ions. The detection limits of Fe 3+ and Fe 2+ were 1.04 μM and 0.78 μM, respectively in the range of 10–90 μM. The formation of complex between the probe and Fe 3+ /Fe 2+ was supported by Job’s plot. The probe was successfully applied to the detection of Fe 3+ and Fe 2+ in actual water sample with a good recovery. In addition, a possible sensing mechanism for the interaction of iron ions with amide bond groups of nanomicelles was proposed.
