Colorimetric/fluorescent dual-mode assay for antioxidant capacity of gallnuts based on CuCo nanozyme and AIE luminogen

Yiming Yang,Junlei Liu,Wenying Li,Yueqing Zheng,Wei Xu

In this work, we present a dual-mode assay system consisting of a nanozyme and a luminogen with the aggregation-induced emission (AIE) feature. In the assay system, the chosen nanozyme named CuCo-0 catalyzes the substrate to produce colorimetric signals, while the aggregates of H 4 ETTC (4,4′,4″,4‴-(ethene-1,1,2,2-tetrayl) tetrakis ([1,1′-biphenyl]-4-carboxylic acid), a typical AIE luminogen, generate fluorescent signals. The peroxidase-like activity of the CuCo-0 nanozyme can be remarkably suppressed with sequential additions of antioxidants , leading to a dual-signal response characterized by enhanced fluorescence emission and reduced UV-vis absorbance. On this basis, a dual-mode assay capable of producing both colorimetric and fluorescent signals for the assessment of antioxidant capacity using gallic acid as a representative antioxidant was exploited. Good linearity can be obtained in the 0–60 μM range for both colorimetric analysis and fluorescent analysis, with detection limits of 1.3 μM and 0.35 μM, respectively. Furthermore, this dual-mode assay was successfully applied to real gallnut samples, yielding satisfactory results.

Application of Aggregation-Induced Emission (AIE) Technology in Monitoring of the Preparation of Spinning Solution for Electrospinning

Li Zhaopeng,Qiao Xiaoguang

Aggregation-induced emission (AIE) technology had already been applied in polymer science and provides a deeper understanding on polymer structure and formation processes. Here we prepared a copolymer of (2-(4-vinylphenyl)ethene-1,1,2-triyl)tribenzene (TPEE) and acrylonitrile (AN) (PTPEE- co -AN), which was utilized as the fluoresce probe for the monitoring of the electrostatic spinning process. Owing to the physical entanglement which restrincted the movement of TPE units, the PL intensity of the spinning solution increased with the increase of PAN in the solution. The viscosity and the concentration of PAN spinning solution was, therefore, could be monitored by PL intensity or by nake eyes. As the main component of fluoresce probes was PAN, the copolymer can be well integrated with spinning components, and has no impact on the electrospinning process. Moreover, the PTPEE-co-AN endowed photoluminescence properties to the formed fibers, as well as the fiber films. This method provides a new observation platform for the electrospinning process, and a variety of probes can be prepared through copolymerization to suit the spinning of different polymers.

Fabrication of water dispersible and biocompatible AIE-active fluorescent polymeric nanoparticles through a “one-pot” Mannich reaction

Guangjian Zeng,Meiying Liu,Ruming Jiang,Qiang Huang,Long Huang,Qing Wan,Yanfeng Dai,Yuanqing Wen,Xiaoyong Zhang,Yen Wei

Fluorescent nanoparticles have been attracting more and more interest from scientists in chemistry, materials, medical and biology fields. Among them, fluorescent nanoparticles with aggregation-induced emission (AIE) character stand out as the most promising candidates for different applications especially in the biomedical field. In this work, we reported for the first time that amphiphilic polymers containing a hydrophobic AIE dye and a hydrophilic poly(ethylene glycol) (PEG) segment can be facilely fabricated based on a Mannich reaction, which could conjugate the phenol hydroxyl-containing AIE dye (PhE-OH), paraformaldehyde and 4-arm poly(ethylene glycol)amine (4-arm PEG-NH2) in one pot. The obtained PEG-PhE fluorescent polymeric nanoparticles (FPNs) were ascertained by a series of characterization techniques. Moreover, the cell viability and cell uptake behavior of PEG-PhE FPNs were also investigated to evaluate their potential for biomedical applications. The results suggest that the PEG-PhE FPNs show great potential application in the field of biomedical science. More importantly, the strategy used in this work can also be extended for the preparation of many other various AIE-active FPNs with different functional polymers.

One bioprobe: a fluorescent and AIE-active macromolecule; two targets: nucleolus and mitochondria with long term tracking

Hengchang Ma,Zengming Yang,Haiying Cao,Lei Lei,Lu Chang,Yucheng Ma,Manyi Yang,Xiaoqiang Yao,Shaobo Sun,Ziqiang Lei

Specific organelle imaging and long-term cellular tracking are of paramount importance in monitoring biological processes, pathological pathways, and therapeutic effects, etc. Herein, we report a novel macromolecule fluorescent probe (TPPA–DBO), which is synthesized from tris(4-(pyridin-4-yl)phenyl)amine (TPPA) and 1,8-dibromononane (DBO) with a gram scale by a simple method. TPPA–DBO demonstrates a highly specific nucleolus-targeting ability, which is very challenging in the bioimaging research field. We have shown that the green nucleolus-specificity probe TPPA–DBO has advantages over the commercially available nucleolus-staining probes such as DAPI, Hoechst dyes and SYTOs in terms of its AIE-performance, large Stokes shift (175 nm), excellent photostability, and promising usefulness in live cell imaging experiments. Surprisingly, after internalizing TPPA–DBO into the nucleus region for a period of time, some TPPA–DBO are reversely diffused from nucleolus into the cytoplasm, thus resulting in the staining of mitochondria with a redder emission color. This research result may provide a new concept of cellular tracker design and provide insight into biological questions, understanding disease mechanismss, and designing new therapeutic strategies.

Microwave-assisted multicomponent reactions for rapid synthesis of AIE-active fluorescent polymeric nanoparticles by post-polymerization method

Qian-yong Cao,Ruming Jiang,Meiying Liu,Qing Wan,Dazhuang Xu,Jianwen Tian,Hongye Huang,Yuanqing Wen,Xiaoyong Zhang,Yen Wei

The development of simple and effective methods for synthesis of fluorescent polymeric nanoparticles (FPNs) with aggregation-induced emission (AIE) plays an important role for the biomedical applications of AIE-active FPNs. In present work, we developed a facile strategy for the fabrication of AIE-active FPNs by a post-polymerization method based on the microwave-assisted Kabachnik–Fields (KF) reaction, which can conjugate with poly(PEGMA-NH 2 ), AIE-active dye (TPE-CHO) and diethyl phosphate (DP) under microwave irradiation within 5   min. The characterization results confirm that PEGMA-TPE FPNs are successfully prepared through the microwave-assisted KF reaction. The resultant AIE-active FPNs show high water dispersity, intensive fluorescence and low cytotoxicity. These features make these AIE-active FPNs great potential for biomedical applications. Moreover, the microwave-assisted KF reaction is simple, fast, atom economy that should be a general strategy for the fabrication of various multifunctional AIE-active FPNs. We believe this work will open up a new avenue for the preparation of AIE-active functional materials with great potential for different applications.

Modulating the optical properties of the AIE fluophor confined within UiO-66's nanochannels for chemical sensing

Zhixiang Lu,Mingjiang Wu,Shuang Wu,Shaoxiong Yang,Yuan Li,Xiaolan Liu,Liyan Zheng,Qiue Cao,Zhongtao Ding

Transformation of aggregation induced emission (AIE) molecules into functional materials can greatly tune their fluorescence properties and expand their related potential applications. Here, we demonstrate a facile strategy to modulate the fluorescence properties of AIE molecules by confining them within the nanochannels of metal–organic frameworks (MOFs). AIE molecules (4,4′-(hydrazine-1,2-diylidene bis(methanylylidene)) bis(3-hydroxybenzoic acid), HDBB) with pH-dependent emission were successfully integrated into UiO-66 resulting in a UiO-66⊃HDBB complex while maintaining the crystal structure by the host–guest process. The fluorescence properties of HDBB can be modulated by virtue of the unique nanospace confining effect. Furthermore, UiO-66⊃HDBB was used as a sensitive and selective fluorescent probe for Cu2+ detection in aqueous solution. This study provides a facile strategy for the construction of high performance AIE based luminescent materials with adjustable properties for potential applications.

Regulation on the aggregation-induced emission (AIE) of DNA-templated silver nanoclusters by BSA and its hydrolysates

Wei-Xian Wang,Yuqing Wu,Hong-Wei Li

The aggregation-induced emission (AIE) phenomenon of the metal nanoclusters (NCs) has been discovered quite recently, which is considered to be creative to improve the optical properties of NCs upon ligand aggregation. In the present study, we report an AIE system for double-stranded DNA-templated silver nanoclusters (AgNCs-dsDNA) by using bovine serum albumin (BSA) in solution, which induce a moderate emission enhancement of AgNCs (5-fold) and a blue-shift through a loosen interaction. In addition, significant luminescence enhancement (30-fold) is further stimulated by the addition of digestive enzyme to the system by altering the surface structure of the aggregated particles. The processes are observed directly through transmission electron microscopy (TEM), where the dispersed AgNCs spheres are aggregated in a large incompact sheet-like film by BSA; while further trypsin addition lead them into larger particles (>50   nm) with higher density. And the intrinsic mechanism is explained well by UV–vis absorption and time-resolved luminescence spectra. The observed luminescence enhancement in step-II is attributed to the AIE through stronger interaction between the hydrolysates of BSA and the ligand dsDNA. Therefore, the optical properties of AgNCs-dsDNA are regulated well by the addition of BSA and trypsin in different amounts, which relates directly to the degree of aggregation and can be extended to other metal nanoclusters by employing the pairs of protein and related enzyme.

Jiahui Lin,Xing Feng,Jintao Huang,Yiwei Liu,Yongshuang Xiao,Yanfeng Li,Yonggang Min,Ben Zhong Tang

Alcohol sensing plays a critical role in medical detection and personal health management. AIE materials with high sensitivity, selectivity and fast response have been widely used in biosensing, but their application in the field of alcohol sensing still needs further research and development. Furthermore, developing flexible phase change materials (PCMs) is significant for the research of human-body thermal management. In this study, a kind of flexible polyacrylonitrile (PAN)/polyvinylpyrrolidone (PVP)/polyethylene glycol (PEG)/Py-CH (pyrene-based AIE molecule)/SiO 2 @h-BN composite fiber textile (PAB) with alcohol sensing performance, writable fluorescence property, and human body thermal management function has been prepared via electrospinning technique. The PAN/PVP fiber matrix successfully integrated AIE fluorescent sensing material and PCM into a multi-functional composite with great shape stability. Owing to the introduction of novel pyrene-based Py-CH with AIE characteristic, this innovative textile exhibited wonderful fluorescent properties, including sensitive alcohol fluorescence sensing, writable fluorescence performance and variable temperature fluorescence. Furthermore, proposed PAB textile delivered a high energy storage density of 87∼90 J/g, excellent thermal reliability, great comprehensive mechanical flexibility and enhanced thermal conductivity for flexible human body thermal management. Hence, this flexible multifunctional AIE/PCM composite sensing textiles can be widely used in alcohol sensing, fluorescence anti-counterfeiting and flexible body thermal management.

Shuai Zhang,Jincheng Xiong,Boyan Sun,Chunchao Yin,Yongjun Zheng,Xingxing Liu,Yanchen Gao,Xiaomei Wang,Sihan Wang,Haiyang Jiang

Emerging contaminants seriously jeopardize food safety and even human health. This study presents the formation of all-in-one aggregation-induced emission (AlE)-type immunomagnetic ZlF-8 (AlMZ) through an ultra-simple self-assembly method in an aqueous phase, successfully harnessing the functions of immuno-recognition, luminescence, and magnetism, Here, we have achieved a dual enhancement in detection efficiency rapidly enriching targets and performing a triple lateral flow immunoassay (LFlA) for ractopamine, diethylstilbestrol, and chloramphenicol within 40 minutes, The dual readout biosensor was then configured to automatically extract the color intensity values of the three test lines using a smartphone. A portable sensing platform ensures stable temperature and light intensity in non-laboratory environments, providing favorable conditions for immunological detection. This study demonstrated low detection limits of 0.11, 0.14, and 0.01ug/L for ractopamine, diethylstilbestrol, and chloramphenicol, respectively, Applications in pork and tap water showed excellent practicality and stability, with recovery rates ranging from 87.2 % to 112.7 %, emphasizing the potential for applications in food and environmental safety.

AIE fluorescent nanozyme-based dual-mode biosensor for analysis of the bioactive component hypoxanthine in meat products

Guojian Wu,Jiaqi Luo,Chenxing Du,Zhi Zheng,Yang Zhang,Pengjie Luo,Yongning Wu,Yizhong Shen

The development of facile, low-cost reliable, and precise onsite assays for the bioactive component hypoxanthine (Hx) in meat products is significant for safeguarding food safety and public health. Herein, we proposed a smartphone-assissted aggregation-induced emission (AIE) fluorogen tetraphenylethene (TPE)-incorporated amorphous Fe-doped phosphotungstates (Fe-Phos@TPE) nanozyme-based ratiometric fluorescence-colorimetric dual-mode biosensor for achieving the onsite visual detection of Hx. When the Hx existed, xanthine oxidase (XOD) catalyzed Hx into H 2 O 2 to be further catalyzed into •OH by the prominent peroxidase activity of Fe-Phos@TPE at pH = 6.5, resulting in the oxidization of nonfluorescent o -phenylenediamine (OPD, naked-eye colorless) to be yellow fluorescent emissive 2,3-diaminophenazine (DAP, naked-eye dark yellow) at 550 nm as well as the intrinsic blue fluorescence of Fe-Phos@TPE at 440 nm to be decreased via inner-filter effect (IFE) action, thereby realizing a multi-enzyme cascade catalytic reaction at near-neutral pH to overcome the traditional acidity dependence-induced time-consuming and low sensitivity troublesome.

Alkyl chain length-regulated in situ intelligent nano-assemblies with AIE-active photosensitizers for photodynamic cancer therapy

Lingyi Shen,Qi-Long Zhang,Yongchao Yao,Ya-Li Huang,Zhichang Zheng,Ming Li,Hong Xu,Lin Tan,Xukun Liao,Binyi Xia,Lin Li,Carl Redshaw,Yang Bai,Chengli Yang

Photodynamic therapy (PDT) brings new hope for the treatment of breast cancer due to few side effects and highly effective cell killing; however, the low bioavailability of traditional photosensitizers and their dependence on oxygen severely limits their application. Aggregation-induced emission (AIE) photosensitizers (PSs) can dramatically facilitate the photosensitization effect, which can have positive impacts on tumor PDT. To-date, most AIE photosensitizers lack tumor targeting capability and possess poor cell delivery, resulting in their use in large quantities that are harmful to healthy tissues. In this study, a series of AIE photosensitizers based on pyridinium-substituted triphenylamine salts (abbreviated to TTPAs 1 - 6 ) with different alkyl chain lengths are synthesized. Results reveal that TTPAs 1 - 6 promote the generation of type I and type II reactive oxygen species (ROS), including ·OH and 1 O 2 . In particular, the membrane permeability and targeting of TTPAs 4 - 6 bearing C8-C10 side-chains are higher than TTPAs 1 - 3 bearing shorter alkyl chains. Additionally, they can assemble with albumin, thereby forming nanoparticles (TTPA 4 - 6 NPs) in situ in blood, which significantly facilitates mitochondrial-targeting and strong ROS generation ability. Moreover, the TTPA 4-6 nanoparticles are pH-responsive, allowing for increased accumulation or endocytosis of the tumor and enhancing the imaging or therapeutic effect. Therefore, the in vivo distributions of TTPA 4 - 6 nanoparticles are visually enriched in tumor sites and exhibited excellent photodynamic cancer therapy efficacy. This work demonstrates a novel strategy for AIE PDT and has the potential to play an essential role in clinical applications using nano-delivery systems.

Rapid and simple fluorescent detection of chlorogenic acid in Aidi injection using aggregation-induced emission (AIE) nanoclusters

Pengwei Zhao,Zhixuan Song,Yunhan Li,Xiaorui Liu,Zhengjin Jiang,Qing Zhu,Jia-Huan Qu

Chlorogenic acid (CGA) is a key component in Aidi injection, known for its anti-cancer properties and ability to reduce toxicity. Therefore, accurate detection of CGA levels in Aidi injection is essential for monitoring therapeutic efficacy and minimizing adverse effects. This study presents a rapid and simple fluorescent method for detecting CGA in Aidi injection using aggregation-induced emission (AIE) nanoclusters, i.e. D(-)-penicillamine (DPA)-capped bimetallic gold/copper nanoclusters (DPA-Au/CuNCs). Upon the addition of CGA, the aggregation state of DPA-Au/CuNCs was disrupted through hydrogen bond formation and ligand exchange, leading to fluorescence quenching. The prepared DPA-Au/CuNCs exhibited a rapid response time of 0.5 min, and demonstrated good sensitivity for CGA, with a limit of detection of 3.75 μg/mL, and a linear detection range of 12.5–200 μg/mL. This method was successfully applied for the analysis of CGA in Aidi injection and plasma with good recovery rates and minimal matrix effect, highlighting its potential for the applications in pharmaceutical products and clinical samples.
