Chitosan (CS) is a type of natural polysaccharide, widely used as a biomaterial on account of its great biocompatibility and the multitudes of functional groups on the long chitosan chain, making it easy for property augmentations. Nevertheless, the lack of workability and mechanical capacity limits the applications. Herein, electrospinning was chosen to fabricate oriented chitosan/polyethylene oxide (PEO) nanofibres decorated by integrin-targeting polypeptides (RGD, Arg-Gly-Asp), which is essential for cell adhesion. At the same time, porous PLGA-PEG membranes were taken as a shell of the composite film, which can provide mechanical support for the scaffold. Adequate topological anisotropic information from oriented nanofibres helps the scaffold simulate the natural axon structure, which can induce accelerated regeneration of neurotmesis. Moreover, β-Tricalcium Phosphate (β-TCP) nanoparticles were brought into nanofibers to neutralize the acidic environment produced by the degeneration of PLGA-PEG. Cell viability tests at the in vitro level and dorsal root ganglion (DRG, 1-day sucking SD rat) co-culture detection proved that the scaffold possesses excellent biocompatibility while guiding oriented regeneration of damaged nerves. In this study, a CS-RGD/PEO/β-TCP/PLGA-PEG porous composite scaffold with low cytotoxicity was fabricated. The scaffold can not only lead to the oriented growth of nerves but also has selectivity, which can help nutrient exchange while inhibiting the invasion of fibrous tissue.

Chronic hyperglycemia in diabetic patients induces a sustained systemic inflammatory microenvironment that critically impairs bone regeneration and delays healing. In this study, we developed a β-tricalcium phosphate (β-TCP) scaffold functionalized with the anti-inflammatory peptide KAFAKAARLYRKALARQLGVAA (KAFAK), aiming to regulate the local hyperglycemic inflammatory milieu and enhance bone regeneration. Polydopamine (PDA) was applied as a bioadhesive coating to facilitate the efficient immobilization of KAFAK on the scaffold surface. The physicochemical properties and biocompatibility of the modified scaffolds were systematically characterized. A hyperglycemic inflammatory model was established using lipopolysaccharide (LPS) and glucose to simulate diabetic conditions. In this model, KAFAK-functionalized β-TCP scaffolds significantly downregulated the expression of key pro-inflammatory cytokines, including IL-6, TNF-α, and IL-1β, in RAW264.7 macrophages. Furthermore, the scaffold effectively modulated macrophage behavior, exhibiting notable immunomodulatory and anti-inflammatory activity. Overall, this study demonstrates that KAFAK-functionalized β-TCP scaffolds hold strong potential for bone regeneration under hyperglycemic inflammatory conditions and offer a novel strategy for immunomodulatory therapy in metabolic bone disorders.

The use of membrane-based guided bone regeneration techniques has great potential for single-stage reconstruction of critical-sized bone defects. Here, a multifunctional bone regeneration membrane combining flexible elasticity, electrical stimulation (ES) and osteoinductive activity is developed by in situ doping of MXene 2D nanomaterials with conductive functionality and β -TCP particles into a Poly(lactic acid-carbonate (PDT) composite nano-absorbable membrane (P/T/MXene) via electrostatic spinning technique. The composite membrane has good feasibility due to its temperature sensitivity, elastic memory capacity, coordinated degradation profile and easy preparation process. In vitro experiments showed the P/T/MXene membrane effectively promoted the recruitment and osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) under ES and enhanced the angiogenic capacity of endothelial cells, which synergistically promoted bone regeneration through neovascularization. In addition, an in vivo rat model of cranial bone defects further confirmed the bone regeneration efficacy of the P/T/MXene membrane. In conclusion, the developed P/T/MXene membrane can effectively promote bone regeneration through their synergistic multifunctional effects, suggesting the membranes have great potential for guiding tissue regeneration and providing guidance for the biomaterials design.

Poly(ε-caprolactone) (PCL) and β-tricalcium phosphate (β-TCP) are promising materials for bone tissue engineering due to their complementary biodegradability and osteoconductivity. However, their individual limitations hinder clinical application. In this study, PCL@β-TCP composite scaffolds containing 5%, 10%, and 20% β-TCP were fabricated via fused deposition modeling (FDM). The effects of β-TCP content on the physicochemical, mechanical, and antibacterial properties of the scaffolds were systematically investigated. Results showed that increasing β-TCP content enhanced hydrophilicity (contact angle reduced from 89.1° to 66.9°), accelerated biodegradation (higher weight loss after 103 days in PBS), and improved antibacterial activity, as evidenced by larger inhibition zones against E. coli and S. aureus. These findings demonstrate that β-TCP incorporation effectively enhances the bioactivity and antibacterial performance of PCL scaffolds, highlighting their potential for bone tissue engineering applications, particularly in non-load-bearing contexts.

Alveolar ridge deficiency remains a significant clinical challenge in dental implantology, often necessitating bone augmentation procedures. While autogenous bone grafting is considered the gold standard, its limitations, including donor site complications and limited availability, have prompted the development of alternative bone graft materials. In this study, we fabricated a novel polycaprolactone (PCL)/β-tricalcium phosphate (β-TCP)@zinc oxide (ZnO) composite scaffold via direct ink writing (DIW) three-dimensional (3D) printing. The scaffold integrates a biomimetic porous architecture with a controlled ion-release strategy to enhance osteogenic performance. The characterization of the material confirmed excellent chemical stability, hydrophilicity, and tunable degradation behavior. The incorporation of ZnO facilitated the sustained release of bioactive ions (Ca 2+ , PO 4 3− , and Zn 2+ ), significantly improving the scaffold's bioactivity. In vitro assessments revealed that the scaffold with 2% ZnO (PTZ-2) exhibited optimal cytocompatibility and osteogenic differentiation capacity, promoting alkaline phosphatase activity and mineralized nodule formation in MC3T3-E1 cells. In a rat cranial defect model, the PTZ-2 scaffold achieved approximately 95% new bone area after 12 weeks. Histological analysis further confirmed the presence of continuous, highly mineralized bone matrix with excellent osseointegration capability. These findings collectively indicate that the 3D-printed PCL/β-TCP@ZnO composite scaffold has great potential for clinical translation in bone defect regeneration.

Monodispersed and spherical β-TCP granules for 3D printing were prepared by spray drying with the surface modification of PVB . Modifier PVB attached to the particle surface to form the crosslinked network structure and keep them combine with each other into aggregates. The morphology, size, forming performance and crushing behavior of granules as well as the microstructure and density of as-fabricated β-TCP ceramics were used to optimize the PVB contents. Promising results could be expected that PVB contents were 1.0 wt.% and the average particle size of granules was 37.36 μm. The modified granules exhibited excellent forming performance with monodispersed and spherical aggregates for 3D printing . They could be completely crushed under 80 MPa without obvious inter-granules void. These granules with 1.0 wt.% PVB had defined strengths for ceramic forming. The good dispersity and flowability assured the high density and uniform porous structure of β-TCP ceramics, which were used as biological ceramic materials and completely met the standard for bone tissue engineering .

Bone tissue engineering scaffolds are used to repair dimensional alveolar crest defects resulting from etiologies such as trauma, infection, or tumor resection, thereby enabling a more accurate, efficient, and predictable dental diagnosis and treatment. In this study, cone beam computed tomography (CBCT) data from patients with alveolar crest defects were utilized to design a personalized scaffold. Subsequently, the 3D PCL/β-TCP scaffold was successfully fabricated based on this design using 3D printing technology. In parallel, a comprehensive series of assays were conducted to evaluate the properties of the 3D PCL/β-TCP scaffold, including its ability to promote MC3T3-E1 cell adhesion and proliferation in vitro. Furthermore, its capacity to induce bone formation was assessed in an in vivo animal model. The results demonstrated that the scaffold exhibited excellent biocompatibility, as evidenced by CCK-8 and live/dead (AM/PI) fluorescence staining assays, and significantly facilitated the adhesion and proliferation of MC3T3-E1 cells. Additionally, ALP and Alizarin Red staining experiments indicated that the 3D PCL/β-TCP scaffold significantly enhanced ALP expression and calcified nodule formation. Finally, the in vivo experiments in rats with skull defects confirmed that the 3D PCL/β-TCP scaffold contributed to accelerated bone formation. In summary, owing to numerous favorable properties such as personalized customization, biomimetic porous architecture, and excellent osteogenic ability, the 3D PCL/β-TCP scaffold was confirmed to be an advantageous candidate, offering ideas and effective methods for the precise repair of the alveolar crest.

The minimal demands for scaffolds include a guaranteed strength over time. In this study, a novel porous HA/β-TCP bioceramic scaffold with micro-ribs structure was fabricated by extrusion deposition technique and microwave sintering. Micro-ribs were placed at the center and corners of scaffold along the direction of load. Mechanical behaviors were studied to verify the strengthening effect of micro-ribs. Compared to the scaffold without micro-ribs, the average compressive strength of newly developed scaffold was remarkably improved from 28.3 MPa to 45.6 MPa under the porosity of 50%. Moreover, it also exhibited more stable and longer lasting mechanical strength during degradation in vitro. The effectiveness of micro-ribs on improving the mechanical performance of scaffolds provided a structural design reference for bone tissue engineering.

The recently developed poly(ethylene glycol)ylated poly(glycerol sebacate)/β-tricalcium phosphate (PEGS/β-TCP) membrane possesses favorable mechanical properties and excellent osteogenic activity. This study is designed to evaluate the feasibility of applying PEGS/β-TCP membrane in guided bone regeneration (GBR) through investigating the in vitro and in vivo activities of macrophages promoted by PEGS/β-TCP membranes during osteogenesis. The behavior of macrophages seeded on PEGS/β-TCP and PEGS membranes is assessed in terms of cell viability, polarization pattern, and release of inflammatory and osteogenic cytokines. The in vitro osteogenic effect of PEGS/β-TCP membrane-associated macrophage polarization is evaluated by incubating mesenchymal stem cells (MSCs) with conditioned medium derived from macrophages stimulated with PEGS/β-TCP or PEGS membranes. The in vivo correlation between macrophage polarization and PEGS/β-TCP membrane-driven osteogenesis is investigated using a rat calvarial defect model. The results show that PEGS/β-TCP and PEGS membranes are capable of downregulating M1 polarization and upregulating M2 polarization. Compared to the PEGS membrane, the PEGS/β-TCP membrane can further alleviate local inflammatory response and significantly promote the in vitro osteogenic differentiation of MSCs and in vivo restoration of bone defects. In conclusion, the novel PEGS/β-TCP membrane can promote the osteogenic commitment of MSCs and restore calvarial bone defects by improving the macrophage polarization pattern.

Multimodal optimization improved scaffold structure, mechanics, and bioactivity. • Low-dose BT generated a detectable but mild local piezoelectric response. • PTB4 achieved the best overall balance of printability and biological performance. • PTB4 enhanced osteogenic differentiation and VEGF-related endothelial responses. • PTB4 promoted osteogenesis–angiogenesis coupling and calvarial bone regeneration. Critical-size bone defects (CSDs) remain a major clinical challenge. Although three-dimensional (3D) printing enables precise structural control and defect-specific scaffold fabrication, conventional polymer-based scaffolds often exhibit insufficient mechanical strength and limited osteogenic activity. In this study, poly(ε-caprolactone)/β-tricalcium phosphate (PCL/β-TCP) scaffolds containing 0, 1, 2, 4, or 8 wt% barium titanate (BaTiO 3 , BT) were fabricated by direct ink writing. Their rheology, microstructure, mechanical properties, local electromechanical response, cytocompatibility, angiogenesis-related activity, osteogenic differentiation, and bone regeneration in vivo were evaluated. All formulations exhibited suitable printability and regular porous structures. Moderate BT incorporation increased surface roughness and mechanical strength, with compressive strength remaining within the range of human cancellous bone. Piezoresponse force microscopy confirmed a detectable but mild local piezoelectric response. However, its direct osteogenic contribution was not isolated from other BT-related material effects and requires further verification. All scaffolds exhibited good cytocompatibility. PTB4 showed the most favorable biological performance, including enhanced MC3T3-E1 cell proliferation, increased ALP-positive area, greater mineralized deposition, and more pronounced VEGF-related fluorescence in HUVECs. After 12 weeks in a rat calvarial critical-size defect model, PTB4 exhibited significantly higher bone volume fraction and bone mineral density than the blank and PT groups. Histological and immunohistochemical analyses showed more extensive new bone formation, collagen deposition, and stronger osteogenesis–angiogenesis coupling. Overall, PTB4 achieved the best balance among printability, mechanical compatibility, cytocompatibility, angiogenesis-related activity, and osteogenic performance, supporting low-dose BT modification as a promising strategy for safe, printable scaffolds for critical-size craniofacial and oral bone defect repair. Reconstructing bone defects remains a significant challenge in clinical practice, driving the urgent need for advanced artificial grafts that simultaneously promote vascularization and osteogenesis. Addressing the critical trade-off between achieving high porosity/strength and effective bioactivity at safe ion doses, we incorporated strontium (Sr) into β-tricalcium phosphate (β-TCP) scaffolds with a triply periodic minimal surface (TPMS) structure using digital light processing (DLP)-based three-dimensional (3D) printing. Systematically screening Sr concentrations (0-10 mol%), we identified 10 mol% as optimal, leveraging the synergy between the biomimetic TPMS architecture, providing exceptional mechanical strength (up to 1.44 MPa at 80% porosity) and facilitating cell recruitment, and precision Sr-dosing to enhance bioactivity. In vitro assays revealed that the Sr-TCP scaffold dose-dependently stimulated osteogenic differentiation and mineralization in mouse osteoblastic cell line (MC3T3-E1) cells, while also significantly enhancing the angiogenic capacity in human umbilical vein endothelial cells (HUVECs). In vivo studies indicated that the scaffold demonstrated synergistic osteogenic and angiogenic effects in rat femoral condylar defects, leading to marked improvements in bone healing. Collectively, this study establishes a novel design paradigm combining biomimetic topology with optimized ionic doping, resolving key limitations of conventional grafts and advancing the development of safe, highly effective biomaterials for vascularized bone regeneration.
