Inhibition of carbonic anhydrase II by sulfonamide derivatives

Xuan G. S.,Zhan J. H.,Zhang A. M.,Li W.,Zheng K.

A series of sulfonamide derivatives were synthesized, and the enzyme inhibitory activity of the synthesized compounds on carbonic anhydrase II was evaluated. Through molecular docking studies, it was found that compounds 1b, 1e, 2a, 2b, 3a have a strong binding affinity to carbonic anhydrase II. The IC50 values of the four compounds 1e, 2b, 3a, and 3b were lower than that of the positive control drug acetazolamide. What's more, the compounds had a high inhibitory activity for A549 lung cancer cell growth, among them, 1e and 3a could inhibit both carbonic anhydrase II and lung cancer cell proliferation.

Immobilization of carbonic anhydrase on epoxy-functionalized magnetic polymer microspheres for CO2 capture

Guohua Jing,Fujun Pan,Bihong Lv,Zuoming Zhou

Carbonic anhydrase (CA) is the most efficient catalyst for CO 2 hydration and dehydration. In the present work, magnetic polymer microspheres functionalized with epoxy group were prepared, and CA enzyme was immobilized on the carries by selective covalent binding. The parameters affecting CA immobilization, such as pH, temperature and enzyme dose were investigated. Based on the investigation and the characterization of the microspheres, the kinetic parameters of the immobilized and the free CA were also evaluated. The value of the Michaelis–Menten constant ( K m ) and the maximum velocity ( V max ) of the immobilized CA was 8.077   mmol/L (mM) and 0.027   μmol/(min   mL), while that of the free CA was 6.091   mM and 0.091   μmol/(min   mL), respectively. Moreover, the performance of the thermal stability, storage stability and reusability of the immobilized CA confirmed that CA immobilized on the epoxy-functionalized magnetic polymer microspheres owned a stable and efficient catalytic ability on CO 2 hydration, which seemed to be a suitable candidate for CO 2 capture.

Dopamine/Polyethylenimine-Modified Silica for Enzyme Immobilization and Strengthening of Enzymatic CO2 Conversion

Tingting Zhai,Caihong Wang,Fengjuan Gu,Zi-hui Meng,Wenfang Liu,Yanzi Wang

Enzymatic conversion of CO2 to high-value chemicals is a green route of resource reutilization, which is limited by low solubility of CO2 in mild conditions. Here, silica microspheres were first modified by the co-deposition of polyethyleneimine (PEI) and dopamine and the resultant polydopamine (PDA)/PEI-SiO2 was then used as the strengthening material of the enzymatic conversion of CO2 to formate. The initial reaction rate increased from 13.4 to 27.2 times with the loading amount of PDA/PEI-SiO2 increasing from 0.1 to 0.4 g. Following that, PDA/PEI-SiO2 was used as the carrier for the immobilization of carbonic anhydrase (CA). As a result of synergism, the formate synthesis was accelerated up to 48.6 times with a low loading amount of 0.2 g of PDA/PEI-SiO2-CA. Although the promoting effect of PDA/PEI-SiO2 retained only 56.1% after being used once, PDA/PEI-SiO2-CA retained 95.1% activity after 10 cycles of use. It was presumed that CA boosted the impact and reusability of PDA/PEI-SiO2 by improving CO2 desorption. This work is much significant in providing a simple and high-efficiency strategy for process intensification of enzymatic CO2 conversion.

The development of kinetics model for CO2 absorption into tertiary amines containing carbonic anhydrase

Bin Liu,Xiao Luo,Zhiwu Liang,Wilfred Olson,Helei Liu,Raphael Idem,Paitoon Tontiwachwuthikul

CO 2 absorption into aqueous solutions of two tertiary alkanolamines, namely, MDEA and DMEA with and without carbonic anhydrase (CA) was investigated with the use of the stopped-flow technique at temperatures in the range of 293–313 K, CA concentration varying from 0 to 100 g/m 3 in aqueous MDEA solution with the amine concentration ranging from 0.1 to 0.5 kmol/m 3 , and CA concentration varying from 0 to 40 g/m 3 in aqueous DMEA solution with the amine concentration ranging from 0.05 to 0.25 kmol/m 3 . The results show that the pseudofirst-order reaction rate (k 0, amine ; s −1 ) is significantly enhanced in the presence of CA as compared with that without CA. The enhanced values of the kinetic constant in the presence of CA has been calculated and a new kinetics model for reaction of CO 2 absorption into aqueous tertiary alkanolamine solutions catalyzed by CA has been established and used to make comparisons of experimental and calculated pseudo first-order reaction rate constant (k 0, with CA ) in CO 2 -MDEA-H 2 O and CO 2 -DMEA-H 2 O solutions. The AADs were 15.21 and 15.17%, respectively. The effect of pKa on the CA activities has also been studied by comparison of CA activities in different tertiary amine solutions, namely, TEA, MDEA, DMEA, and DEEA. The pKa trend for amines were: DEEA > DMEA > MDEA > TEA. In contrast, the catalyst enhancement in amines was in the order: TEA> MDEA> DMEA> DEEA. Therefore, it can be seen that the catalyst enhancement in the amines decreased with their increasing pKa values. © 2017 American Institute of Chemical Engineers AIChE J , 2017

Alumina-Supported Porphyrin Zinc as a Carbonic Anhydrase Mimic: Enhanced CO2 Hydration Catalysis

Yuchen Zhou,Zezhi Chen,Huijuan Gong,Donglin Jiang,Huiqiang Yu,Lu Chen

Zinc porphyrin is a promising carbonic anhydrase (CA) mimic for promoting CO2 absorption, but its application is hindered by poor dispersibility in absorption solutions. To address these challenges, we developed a strategy to impregnate zinc porphyrin on an γ-Al2O3 carrier. The hydrophilic surface groups and porous structure of γ-Al2O3 were expected to enhance both the hydrophilicity and stability of zinc porphyrin. To verify its feasibility, zinc tetraphenylporphyrin (ZnTPP) was chosen as the representative to synthesize ZnTPP/Al2O3. Characterizations showed that ZnTPP could be loaded into the pores of γ-Al2O3 in a highly dispersed state. Also, ZnTPP/Al2O3 could be uniformly dispersed in the absorption liquid, effectively exposing the Zn2+ active sites and reducing diffusion resistance. CO2 absorption experiments revealed that ZnTPP/Al2O3 significantly enhanced CO2 absorption in water, 20 wt % K2CO3, and 20 wt % monoethanolamine (MEA) solutions, far outperforming original ZnTPP. Density functional theory (DFT) calculations further elucidated the interaction mechanisms, showing that oxygen atoms on γ-Al2O3 stabilize ZnTPP by forming van der Waals and coordination bonds with Zn2+ active sites. Additionally, these oxygen atoms donate electrons to Zn2+, enhancing its catalytic activity. These findings highlight the effectiveness of this strategy and provide a promising pathway for optimizing other CA-mimics for CO2 capture applications.

Co-precipitation synthesis of carbonic anhydrase-embedded magnetic zeolitic imidazolate framework-8 nanocomposite enzyme for enzymatic CO2 conversion and utilization in facilitating uranium extraction

Zhao Cui,Ting Li,Jing Sun,Qian Li,Hui Zhang,Guang-Yue Li

Carbonic anhydrase (CA)-mediated carbon conversion and utilization (CCU) technology is recognized as an eco-friendly and cost-effective strategy for carbon reduction. However, the broader application of this technology is significantly constrained by the operational instability and challenging recyclability of free CA . Fortunately, nano-integrated biocatalysis has emerged as an effective solution to these challenges. In this study, we immobilized CA for the first time as a CA-embedded ferriferous oxide-zeolite imidazolium framework-8 (Fe 3 O 4 @ZIF-8@CA) nanocomposite enzyme through a one-pot co-precipitation strategy, integrating both the in-situ growth of ZIF-8 around the Fe 3 O 4 core and the immobilization of CA within a single step. Subsequently, this nanocomposite was employed as a nano-integrated biocatalyst to facilitate an innovative CCU pathway. This pathway leverages bicarbonate, produced from the enzymatic hydration of carbon dioxide (CO 2 ), as a substantive uranyl complexing agent to facilitate the neutral leaching of sandstone uranium ore. Systematic characterization techniques, including SEM, TEM, XRD, FTIR, TGA, CLSM, and MPMS, confirmed that Fe 3 O 4 @ZIF-8@CA, which possesses a core–shell structure and excellent magnetic responsiveness, has been successfully synthesized. Enzymatic assays revealed that Fe 3 O 4 @ZIF-8@CA retains approximately 79% of the activity compared to its free counterparts while demonstrating significantly enhanced thermal and storage stability. The outcome of CO 2 hydration conversion for facilitating uranium extraction indicated that, compared with the non-enzymatic reaction group, the bicarbonate yield increased up to 1.5 times, and the uranium extraction efficiency was nearly 10% higher in the Fe 3 O 4 @ZIF-8@CA-mediated reaction group. These findings suggest that Fe 3 O 4 @ZIF-8@CA possesses excellent catalytic properties for CO 2 hydration and can achieve multiplied decarbonization by mediating this innovative CCU pathway. Furthermore, Fe 3 O 4 @ZIF-8@CA demonstrated superb reusability and durability, experiencing no significant degradation in its catalytic performance after eight consecutive hydration regeneration cycles. Our research is anticipated to be a viable option for advancing the goals of carbon neutrality and sustainable development.

Study on the zinc porphyrins as potential carbonic anhydrase mimics for promoting CO2 absorption in K2CO3 solution

Yuchen Zhou,Zezhi Chen,Huijuan Gong,Xiaoshu Wang,Lu Chen,Huiqiang Yu

Zinc porphyrins, boasting a coordination environment akin to that of natural carbonic anhydrase (CA), coupled with excellent structural stability, built-in hydrophobic pocket, and suitable charge of center Zn 2+ , were considered as potential CA-mimics for CO 2 capture. To validate this assumption, three zinc porphyrin representatives, zinc porphin (ZnP), zinc protoporphyrin IX (ZnPP), and zinc meso -tetrakis(N-methylpyridinium-4-yl) porphyrin (ZnTMPyP), were prepared, and tested performance in enhancing CO 2 absorption in water and K 2 CO 3 solutions. The results from CO 2 absorption experiments indicated that ZnTMPyP with the optimal dosage increased CO 2 absorption rate by 44.25 % in water, and ZnPP with the optimal dosage increased the CO 2 absorption rate by 63.67 % in 20 wt% of K 2 CO 3 solution, which are respectively 1.53 and 2.07 times of CO 2 absorption performance of zinc cyclen (ZnC), the most effective CA-mimic reported. Besides, the zinc porphyrins demonstrated excellent stability and reusability throughout five cycles of CO 2 absorption and desorption, which are vital aspects for practical applications. Through the calorimetric method and density functional theory (DFT) calculations, it was confirmed that the prepared zinc porphyrins had similar enzymatic behavior and catalytic CO 2 hydration pathway to natural CA. According to the results of characterization and DFT calculations, high dispersion, the lowest charge of the Zn 2+ center, and the existence of additional CO 2 adsorption sites (propionate ions), endow ZnPP with the highest catalytic activity in the K 2 CO 3 solution.

Synthesis of ZIF-90 based nanozyme with excellent carbonic anhydrase activity

Zemin Ji,Zijin Zhang,Weifeng Lv,Yanyao Shi,Kai Wang,Shumin Ni,Hongchao Ma,Meiwen Cao

Capturing CO 2 using natural carbonic anhydrase (CA) has advantages of environmental friendliness , low energy consumption, renewable and no secondary pollution. However, the high cost, poor stability and low reusability of free CA under harsh conditions limit its industrial applications. Great efforts have been devoted to developing CA mimetic nanozymes to solve these problems. In this work, ZIF-90-based nanoparticles were prepared to mimic the active center of natural CA. The excellent CA-like catalytic activity of ZIF-90 nanoparticles was confirmed by using hydrolysis of p-nitrophenyl acetate (p-NPA) as the model reaction. Moreover, modifying ZIF-90 with monoethanolamine (MEA) could further improve its affinity for CO 2 , which in turn enhances the CO 2 uptake capacity. Moreover, the hydrolysis of CO 2 was also investigated through the pH decrease of the reaction solution and the results indicated that CO 2 hydration and exchange were remarkably enhanced with the aid of ZIF-90 or ZIF-90 @MEA. This approach is facile and costless, which sheds light on the development of novel catalysts for industrial CO 2 capture.

Carbonic Anhydrase-Embedded Hydrogen-Bonded Organic Frameworks Coating for Facilitated Offshore CO2 Fixation

Bo-Yu Zhang,Zhen-Hua Wu,Zi-Yi Chu,Ke Li,Jia-Fu Shi

Graphical A monolithic catalyst of Filler-CA@Lys-HOF-1 (FCLH) was prepared by coating chemical sorption of CA on the surface of the filler followed by the coating of Lys-HOF-1. The highest catalytic activity of FCLH was obtained by regulating the amount of HOF-1 monomer added. Due to the protection of Lys-HOF-1, the FCLH showed good tolerance against acidity and salinity, which could retain about 80.2 % of the original activity after 9 h incubation in simulated seawater. The catalytic activity of FCLH could retain 85.4 % of the initial activity after 10 cycles. In short, we may offer a promising biocatalyst for carbon mineralization, which may drive down carbon emissions when used for CO 2 capture and conversion on offshore platforms. Exhausted emission of carbon dioxide (CO 2 ) from ships or offshore platforms has become one of the major contributors to global carbon emissions. Enzymes such as carbonic anhydrase (CA) have been widely used for CO 2 mineralization because of their high catalytic rate. However, CA in seawater is easy to inactivate and difficult to reuse. Immobilization would be a feasible solution to address the stability issue, which, however, may cause an increase of internal diffusion resistance and reduced catalytic activity. In this regard, design of high-performance biocatalysts for acquiring high catalytic activity and stability of CA is highly desirable. Herein, a monolithic catalyst of Filler-CA@Lys-HOF-1 (FCLH) was prepared by chemical sorption of CA on the surface of the Filler followed by the coating of Lys-HOF-1. Hopefully, our study can provide a promising biocatalyst for CO 2 mineralization, which may drive down carbon emissions when used for CO 2 capture and conversion on offshore platforms.

Highly Hydrophilic ZIF-8 as a Carbonic Anhydrase Mimetic Catalyst for Promoting CO2 Absorption

Yuchen Zhou,Yu Wang,Zezhi Chen,Huijuan Gong,Lu Chen,Huiqiang Yu

Carbonic anhydrase (CA) is an enzyme with unparallele d catalytic activity in CO2 hydration. However, its high cost and low stability seriously restrict its industrial application in CO2 absorption. Constructing CA mimics is a promising strategy to overcome the above limitations. However, to date, a majority of mimics reported are suffering from laborious synthetic processes, harsh synthetic conditions, and costly ligand materials. Therefore, developing novel CA mimics with higher applicability remains a high priority. Due to the structural similarity between zeolite imidazolate framework (ZIF)-8 and natural carbonic anhydrase (CA), ZIF-8 may be considered as a potential CA-mimetic catalyst for CO2 absorption. To validate this assumption, ZIF-8 and hydrophilic ZIF-8-TA were synthesized and the results from subsequent CO2 absorption experiments indicated that the synthesized catalysts could significantly enhance CO2 absorption in water and common alkaline solutions. The addition of 1.0 wt % ZIF-8 in water and different alkaline solutions respectively increased the average CO2 absorption rate by 42.35 and 6.78–24.51%, while the addition of 1.0 wt % of ZIF-8-TA increased the CO2 absorption rate by 81.80 and 18.16–34.39%, respectively. Besides, both catalysts possessed excellent reusability that they retained high performance even after five cycles of absorption and regeneration. Through esterase experiments, it was confirmed that ZIF-8 and ZIF-8-TA exhibited higher CA-mimetic activity than many reported CA mimics. Through density functional theory (DFT) calculations, it was indicated that the catalytic CO2 hydration pathway of ZIF-8 primarily involves four steps, including nucleophilic attack, rearrangement, replacement, and deprotonation, which is similar to that of natural CA, while ZIF-8-TA does not undergo the rearrangement step. Moreover, the electron-donation effect and van der Waals interaction of tannic acid endow ZIF-8-TA with higher catalytic activity.

Construction of alginate beads for efficient conversion of CO2 into vaterite CaCO3 particles

Hao Zhang,Tuo Zhang,Jiachen Zang,Chenyan Lv,Guanghua Zhao

CO 2 is a greenhouse gas and a major contributor to global warming . Efficient conversion of CO 2 into high value-added food materials represents a promising strategy to decrease CO 2 concentration. In this study, we reported that Ca 2+ -crosslinked alginate beads loaded with carbonic anhydrase (CA) can efficiently catalyze conversion of CO 2 into porous vaterite CaCO 3 particles via a carboxymethyl cellulose (CMC) inducing process. CA encapsulated within the Ca 2+ -crosslinked alginate beads maintained around 100% activity of its original activity after 10 assay cycles. Functionally, the CA-loaded alginate beads facilitated conversion of CO 2 into porous vaterite CaCO 3 particles with high CO 2 removal efficiency in the presence of carboxymethyl cellulose. Notably, such vaterite CaCO 3 particles can be explored for adsorption of astaxanthin with higher loading capacity (2.0 wt %), consequently improving the photo- and thermal-stability of this pigment.

Encapsulated deep eutectic solvent and carbonic anhydrase jointly by microfluidics for high capture performance of carbon dioxide

Bingbing Zhao,Kunfeng Zhao,Yuan Cheng,Simin Huang,Jiahao Xu,Yun Xu,Kui He,Dechun Huang,Hongliang Qian,Wei Chen

Excessive carbon dioxide (CO 2 ) emissions from fossil fuel combustion have a profound impact on environment, leading to different associated climate disasters. Although deep eutectic solvents (DES) have the advantage of high absorption capacity of CO 2 , their high viscosities sharply decrease absorption performance due to the low absorption rates. Herein, one of DES choline chloride / urea (CU) and carbonic anhydrase (CA) was encapsulated jointly by microfluidics and crosslinked thermally (microcapsule@(CU + CA)), which was designed for high capture performance of CO 2 . The absorption abilities of microcapsules have been significantly improved by the increase of specific surface area, in which the absorption capacity and absorption rate of microcapsule@(CU + CA) were 74 times and 253 times higher than that of neat CU. In addition, the selectivity of CO 2 /CH 4 also increased 62 times via encapsulation. The high cyclic performance of microcapsule@(CU + CA) was achieved for its high absorption performance and high ability to desorb CO 2 from microcapsule. DES and CA encapsulated jointly by microfluidics can realize high absorption capacity, high absorption rate and high absorption selectivity simultaneously, and it also provides a potential pathway for the efficient capture of various gases.
