Promoting osseointegration by in situ biosynthesis of metal ion-loaded bacterial cellulose coating on titanium surface

Xiaoxia Liu,Danyang Wang,Shuai Wang,Wuzhe Fan,Yulu Yang,Pengfei Gao,Maowen Chen,Weihu Yang,Kaiyong Cai

The biologically inert and excessive elastic modulus of Ti implant surface, as well as the excessive gap between implant and host, will lead to poor bone integration even implant failure. To solve the above problems, in this study, a method for functional Ti implant is reported, in which metal ions-containing bacterial cellulose (BC) coating is introduced in situ on the surface of Ti with complex shapes. Magnesium and strontium ions can be loaded into BC by in situ biosynthesis , which have great effects on the growth of bacteria and the structure of cellulose. In addition, both in vitro and in vivo experiments confirmed that the in situ preparation of functional BC coating on the Ti surface can integrate the operative crevices and promote osteogenesis. This simple and novel method for functional Ti implants has potential application value in clinical bone tissue repair and regeneration.

Layer-by-layer electrodeposition of chitosan quaternary ammonium salt antibacterial composite coating loaded with gentamicin

Xiaoyan Li,Fuxun Qi,Jixiang Zhu,Xinsheng Niu,Shengbing Yang,Deng-Guang Yu

Constructing bio-based antibacterial composite multilayer coatings with sustained drug release is important for reducing the infection risk of bone tissue repair. In this paper, a layer-by-layer aqueous electrodeposition method was applied to prepare a bio-based composite coating of chitosan, hyaluronic acid, and chitosan quaternary ammonium salt on the titanium surface. By pretreating with poly (acrylic acid), controlling the electrochemical deposition process, and post-treating with glutaraldehyde, a multi-layer composite coating with a smooth surface, good interface bonding with the titanium substrate, and a thickness more than ten microns was obtained. The composite coating was used as a drug-carrying system for gentamicin sulfate, which showed obvious antibacterial effects against Escherichia coli and Staphylococcus aureus . The bio-based composite coating with a sandwich structure loaded with antibiotics effectively inhibited the burst release of antibiotics, and the 14-day release rate is only 68.4 ± 2.1 %, achieving sustained release of the drug. The hydrophilic chitosan quaternary ammonium salt on the surface of the composite coating can effectively inhibit the early adhesion of bacteria, while the slow release of gentamicin sulfate in the interlayer can exert a long-lasting antibacterial effect. The preparation of bio-based antibacterial composite coatings using a layer-by-layer electrodeposition strategy has promising applications in titanium orthopedic implants.

Preparation and biological properties of BCZT/TiO2 electrokinetic conversion coating on titanium surface in vitro for dental implants

Cong Wu,Chun Zhang,Xianyi Yan,Junhua Cheng,Lei Chen,Yufei Tang,Kang Zhao

Titanium implants have the risk of loosening or even failure, especially the force generated by chewing is not conducive to the use of conventional titanium bioactive coatings in the oral environment. In this work, a BCZT/TiO 2 bio-piezoelectric coating was prepared on titanium surface. The force generated by chewing is cleverly converted into the piezoelectric charge on the coating surface , and the formation of the combination of implant and alveolar bone is accelerated by its promoting effect on bone repair. The formation mechanism of the coating was studied. The BCZT/TiO 2 bio-piezoelectric coating showed good piezoelectric properties . The calcium deposition results of BCZT/TiO 2 coating with static , physiological loading and low-frequency ultrasonic conditions were compared. Interestingly, the effect of polarized BCZT/TiO 2 coating applied with low-intensity pulsed ultrasound on promoting calcium deposition was similar to that applied with physiological loading, suggesting that low-intensity pulsed ultrasound has the potential to stimulate piezoelectric effects. In addition, the BCZT/TiO 2 coating was non-toxic and has good biocompatibility and bioactivity. The polarized BCZT/TiO 2 coating showed the best proliferation results, and the osteoblast adhesion morphology on the coating surface was extended. The results provide data support for the application of titanium bio-piezoelectric coating on dental implants .

Simultaneous deposition of tannic acid and poly(ethylene glycol) to construct the antifouling polymeric coating on Titanium surface

Ling Li Guo,Yan Fang Cheng,Xu Ren,Kasi Gopinath,Zhi Song Lu,Chang Ming Li,Li Qun Xu

Titanium (Ti) and its alloys are primarily explored to produce biomedical implants owing to their improved mechanical stability, corrosion resistance, low density, and good biocompatibility. Despite, Ti substrate surfaces are easily contaminated by plasma proteins and bacteria. Herein, a simple one-step process for the simultaneous deposition of a polyphenol tannic acid (TA) and four-armed poly(ethylene glycol) (PEG 10k -4−OH) on the Ti substrate (Ti-TA/PEG) surface was described. Additionally, a two-step process has been employed to fabricate the Ti-TA-PEG surface via successive deposition of TA and PEG 10k -4−OH for comparison. The resultant Ti-TA/PEG surface prepared by simultaneous deposition of TA and PEG 10k -4−OH exhibits higher coating thickness and better surface coverage than the Ti-TA-PEG surface. The Ti-TA/PEG and Ti-TA-PEG surfaces could actively inhibit the non-specific adsorption of proteins, suppress the bacterial and platelet adhesion, and prevents biofilm formation. Moreover, the Ti-TA/PEG surface displays a better antifouling performance than the Ti-TA-PEG surface. Thus, the present study demonstrates a simple and convenient approach for constructing polymeric coating with good anti-adhesive properties on the Ti substrate surface.

Structural optimization of Sr/Zn-phosphate conversion coatings triggered by ions preloading on micro/nanostructured titanium surfaces for bacterial infection control and enhanced osteogenesis

Zuo Kangqing,Li Aonan,Si Taoning,Lei Weiyi,Liu Yusheng,Zhang Linbo,Zhang Taixing,Xiao Guiyong,Lu Yupeng,Li Ningbo

Phosphate conversion coatings on metallic implants can synergistically integrate functional components and structural regulation, offering excellent biocompatibility and osteogenic activity. However, the passive oxide layer on the titanium (Ti) surface impedes the following chemical reactivity, adversely affecting the microstructure and properties of phosphate coatings. This study proposes a strategy for achieving structural optimization and properties enhancement of strontium-zinc phosphate (SrZnP) conversion coatings on Ti via regulating interface chemical reaction between coatings and Ti substrates. The results indicated that Sr 2+ and Zn 2+ ions-preloading (IPL) treatment enhanced the interfacial reactivity, which can further achieve crystal refinement and uniform crystal size in nucleation. In contrast, microstructural modifications on Ti substrates induced by acid etching, sandblasting, and alkali etching had minimal effects on the phase composition and crystal morphology (irregular cubic) of the SrZnP coatings. The coatings on IPL-Ti exhibited better mechanical properties and corrosion resistance. Besides, the coatings with optimized structures and surface characteristics elicited bacterial growth inhibition rates of 91.09% and 84.04% against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ), respectively. Meanwhile, the crystal-refined coatings further significantly enhanced the adhesion, proliferation, and differentiation of bone marrow mesenchymal stem cells (BMSCs), proving anticipated osteogenic activity. Overall, the ions preloading strategy on variable micro/nanostructured Ti substrates facilitates the potential application of Sr/Zn-phosphate conversion coatings for repairing infected bone defects.

Antibacterial activity of an NIR-induced Zn ion release film

Tingting Yang,Donghui Wang,Xuanyong Liu

Photothermal therapy originated from using gold nanorods (GNRs) and near-infrared (NIR) irradiation has been widely used in the antibacterial field. Zn element exhibits antimicrobial activity against various bacterial and fungal strains. In this study, a bilayer film, consisting of GNRs as the inner layer and a polydopamine layer containing Zn element (PDA@Zn) as the outer layer, was deposited on the Ti surface. The results testified that all the GNR-modified surfaces had the same photothermal conversion efficiency. The Ti surface modified with GNR and PDA@Zn layers had better antibacterial activity against E. coli and S. aureus due to the GNR-induced photothermal effect and the antibacterial Zn element. Moreover, the accelerated release of Zn ions from the PDA@Zn layer was attributed to the GNR-induced high temperature under the NIR irradiation, which caused the concentration of Zn ions to be high enough to kill the surrounding bacteria. This study illustrates that a composite surface possessing both the contact and heat-responsive antibacterial property was constructed on titanium for potential clinical applications.

Sandblasted/Acid-Etched Titanium Surface Modified with Calcium Phytate Enhances Bone Regeneration in a High-Glucose Microenvironment by Regulating Reactive Oxygen Species and Cell Senescence

Shuo Dong,Tong Zhao,Wei Wu,Zhewei Zhang,Jin Wu,Kunzhan Cai,Guoqing Li,Jiaxin Lv,Heyang Zhou,Chunbo Tang

Hyperglycemia in patients with diabetes affect osteoblast function, leading to abnormal bone metabolism and implant failure. Adequate bone volume surrounding an implant is essential for osseointegration, which can be improved by implant surface modifications. In this study, titanium surfaces were hydrothermally treated with a mixture of phytic acid (PA) and calcium hydroxide to produce a calcium-decorated surface. The control group comprised pure titanium with a sandblasted/acid-etched (SLA) surface. The elemental composition, hydrophilicity, surface roughness, and morphology of the titanium surfaces were examined. Evaluation of in vitro osteogenic differentiation ability in a high-glucose environment using alkaline phosphatase (ALP) staining, ALP activity assays, Alizarin Red S staining, quantitative reverse transcription-polymerase chain reaction (qRT-PCR), and immunofluorescence staining revealed that Ca-PA-modified SLA titanium surfaces can promote osteogenic differentiation of human bone marrow mesenchymal stem cells (hBMSCs). Evaluation of oxidative stress and aging using reactive oxygen species (ROS), malondialdehyde (MDA), superoxide dismutase (SOD), and β-galactosidase staining revealed that Ca-PA-modified SLA titanium surfaces can reduce ROS production and ameliorate oxidative stress damage in hBMSCs. In vivo assessment of osteogenesis in a diabetic rat model revealed that Ca-PA coating promotes peri-implant osseointegration. Ca-PA-modified SLA titanium surface is a candidate for improving implant osseointegration in patients with diabetes.

Fabrication of CO-releasing surface to enhance the blood compatibility and endothelialization of TiO2 nanotubes on titanium surface

Wenfu Ma,Xuhui Liu,Minhui Yang,Qingxiang Hong,Lingjie Meng,Qiuyang Zhang,Jie Chen,Changjiang Pan

Although the construction of nanotube arrays with the micro-nano structures on the titanium surfaces has demonstrated a great promise in the field of blood-contacting materials and devices, the limited surface hemocompatibility and delayed endothelial healing should be further improved. Carbon monoxide (CO) gas signaling molecule within the physiological concentrations has excellent anticoagulation and the ability to promote endothelial growth, exhibiting the great potential for the blood-contact biomaterials, especially the cardiovascular devices. In this study, the regular titanium dioxide nanotube arrays were firstly prepared in situ on the titanium surface by anodic oxidation, followed by the immobilization of the complex of sodium alginate/carboxymethyl chitosan (SA/CS) on the self-assembled modified nanotube surface, the CO-releasing molecule (CORM-401) was finally grafted onto the surface to create a CO-releasing bioactive surface to enhance the biocompatibility. The results of scanning electron microscopy (SEM), X-ray energy dispersion spectroscopy (EDS), and X-ray photoelectron spectroscopy (XPS) revealed that the CO-releasing molecules were successfully immobilized on the surface. The modified nanotube arrays not only exhibited excellent hydrophilicity but also could slowly release CO gas molecules, and the amount of CO release increased when cysteine was added. Furthermore, the nanotube array can promote albumin adsorption while inhibit fibrinogen adsorption to some extent, demonstrating its selective albumin adsorption; although this effect was somewhat reduced by the introduction of CORM-401, it can be significantly enhanced by the catalytic release of CO. The results of hemocompatibility and endothelial cell growth behaviors showed that, as compared with the CORM-401 modified sample, although the SA/CS-modified sample had better biocompatibility, in the case of cysteine-catalyzed CO release, the released CO could not only reduce the platelet adhesion and activation as well as hemolysis rate, but also promote endothelial cell adhesion and proliferation as well as vascular endothelial growth factor (VEGF) and nitric oxide (NO) expression. As a result, the research of the present study demonstrated that the releasing CO from TiO 2 nanotubes can simultaneously enhance the surface hemocompatibility and endothelialization, which could open a new route to enhance the biocompatibility of the blood-contacting materials and devices, such as the artificial heart valve and cardiovascular stents.

Preparation and antibacterial properties of BCZT-Ag piezoelectric coating on titanium surface for orthopedic implants

Cong Wu,Chun Zhang,Xianyi Yan,Qian Liang,Kang Zhao,Yufei Tang

The titanium dioxide drug-loaded nanotube coating on titanium alloy surface was helpful to reduce the risk of infection after implantation in vivo. In this work, Ba 0.85 Ca 0.15 Zr 0.1 Ti 0.9 O 3 (marked as BCZT) with piezoelectric effect and Ag nanoparticles with antibacterial effect were introduced into the TiO 2 nanotube coating, and a piezoelectric and antibacterial BCZT-Ag coating was prepared on titanium surface. The drug release rate in the nanotubes was reduced by the design of the coating structure. The results showed that the Ag nanoparticles were uniformly distributed in the BCZT-Ag coating. The piezoelectric coefficient of BCZT-Ag2.5 coating reached 13.0 pC/N. The release of silver ions in the BCZT-Ag coating showed a significant slow release trend compared with that in the TiO 2 nanotube silver-loaded coating. Interestingly, the antibacterial rate of polarized BCZT-Ag2.5 coating reached 99% even after being immersed in simulated body fluids for 7 days. The BCZT-Ag2.5 coating has no osteoblast toxicity, and osteoblasts can adhere to the BCZT-Ag2.5 coating on titanium surface. The BCZT-Ag2.5 also shows good biological activity in the simulated body fluid immersion experiment. The results will provide a new idea and data support for the application of biological piezoelectric silver loaded coating on titanium surface in orthopedic implants.

Construction of poly (vinyl alcohol)/poly (lactide-glycolide acid)/vancomycin nanoparticles on titanium for enhancing the surface self-antibacterial activity and cytocompatibility

Zehui Liu,Yizhou Zhu,Xiangmei Liu,K.W.K. Yeung,Shuilin Wu

Comparing with traditional drug dosage form, controlled release systems offer more effective and favorable route to deliver drugs in optimum dose to specific sites with long term release duration. In this work, an effective drug delivery system composed of poly (vinyl alcohol) (PVA)/poly (lactide-glycolide acid) (PLGA) nanoparticles (NPs) with encapsulated vancomycin (Van), is constructed on the surface of biomedical titanium. The PVA/PLGA/Van NPs synthesized via double emulsion route are grafted onto the surface of titanium plates modified by alkaline-heat treatment and subsequent aminopropyltriethoxysilane (APTES) deposition. In vitro tests disclose that NPs can release a small amount of drugs continuously due to the slow swelling or hydrolysis of polymer chain segments as the immersion time increases. As the pH value reduces, the ester bonds rupture with releasing more drugs, which is why this drug delivery system exhibits the highest antibacterial efficiency at the lowest pH value of 4.5 in this work. Cell culture results reveal that this smart surface system on titanium facilitates the cell attachment and proliferation on implants. Hence, this pH controlled drug delivery system can be successfully applied as a bio-platform for improving both the osteoblasts adhesion and antibacterial activity of metallic implants.

ZnO@ZnS nanorod-array coated titanium: Good to fibroblasts but bad to bacteria

Qiming Luo,Huiliang Cao,Lanyu Wang,Xiaohan Ma,Xuanyong Liu

Cell-selective toxic titanium is highly desired in clinical dental practice. Herein, based on the in situ conversion of ZnO to ZnO@ZnS, nanorod-array structured coatings with a controllable release features of zinc (Zn), has been successfully fabricated by a two-step hydrothermal method to endow titanium surface with cell-selectivity, i.e. boosting the functions (attachment and migration) of human gingival fibroblasts (HGnFs) while acting against the invasion of pathogenic bacteria. The improved functions of HGnFs over the ZnO@ZnS nanorod-array were attributed to the material’s optimized zinc release, which was decreased from an order of 3.5 mg L −1 to about 0.3 mg L −1 (within the first week). But more importantly, this concentration still had a high antibacterial efficacy up to 100% (against both the S. aureus and E. coli , 10 7 CFU mL −1 ). This study demonstrated that a ZnO@ZnS nanorod-array coating could be a promising strategy to endow titanium dental implants with improved soft tissue sealing and effectively reduce peri-implantitis.

Deposition of catechol-functionalized chitosan and silver nanoparticles on biomedical titanium surfaces for antibacterial application

Yan Fang Cheng,Jie Yu Zhang,Yun Bing Wang,Chang Ming Li,Zhi Song Lu,Xue Feng Hu,Li Qun Xu

The titanium (Ti) and its alloys have been widely used for dental and orthopedic implants. However, the Ti-based implants may suffer from bacterial infection, which would result in insufficient healing, implant failure and repeated surgical intervention. It is of great interest to inhibit the bacterial adhesion and colonization on the Ti-based implants by introducing proper surface coatings. In this work, a simple method was employed to synthesize the water-soluble catechol-containing chitosan (CACS). The CACS coatings can be deposited onto various substrate surfaces and exhibit substrate-independent behavior. The CACS-coated Ti surfaces were further deposited with silver nanoparticles (Ag NPs) via in-situ reduction of Ag + ions using catechol moieties as the reducing agents. The resulting AgNPs/CACS-coated Ti surfaces exhibit antibacterial properties and can prevent the surface adhesion of bacterial cells, as evidenced by the inhibition zone test, live/dead bacterial staining assay and spread plate method. In addition, they show negligible cytotoxicity to L929 mouse fibroblast cells.
