TPGS assists the percutaneous administration of curcumin and glycyrrhetinic acid coloaded functionalized ethosomes for the synergistic treatment of psoriasis

Teng Guo,Jianying Lu,Yunlong Fan,Yongtai Zhang,Shuo Yin,Xianyi Sha,Nianping Feng

Combined therapy with anti-inflammatory drugs is preferred for the topical treatment of psoriasis , but the codelivery of drugs is restricted due to the lack of a suitable delivery system. Ethosomes with excellent   transdermal properties   are perfect as carriers for hyperplastic skin. Therefore, glycyrrhetinic acid-D-α-tocopherol acid polyethylene glycol succinate (GA-TPGS) was synthesized, which prevented the inflammation and lipid peroxidation damage, thus effectively stabilizing the psoriasis . Then GA-TPGS was surface-modified on the curcumin (Cur) loaded ethosomes to construct curcumin-loaded GA-TPGS-modified multifunctional ethosomes (Cur@GA-TPGS-ES), exerting synergistic treatment for psoriasis. Using an interleukin-6-induced cell model, we found that Cur@GA-TPGS-ES displayed desirable suppression of inflammation response and oxidative stress damage. Compared with the ethanol solution, the percutaneous penetration rates of Cur and GA in Cur@GA-TPGS-ES were superior. In vivo microdialysis revealed similar results, suggesting an increase of transcutaneous absorption in Cur@GA-TPGS-ES. Fluorescence staining revealed that the cellular uptake and skin distribution were distinctly enhanced with the delivery by Cur@GA-TPGS-ES. After topical administration to imiquimod-induced psoriatic mice, the Cur@GA-TPGS-ES group showed powerful treatment from inflammatory infiltration inhibition of Cur, glucocorticoid-like effects of GA and anti-lipid peroxidation of TPGS. Overall, GA-TPGS mediated ethosomes possess more advantageous transdermal properties and synergistic antipsoriatic efficacy.

Cur@SF NPs alleviate Friedreich’s ataxia in a mouse model through synergistic iron chelation and antioxidation

Xu Li,Sun Zichen,Xing Zhiyao,Liu Yutong,Zhao Hongting,Tang Zhongmin,Luo Yu,Hao Shuangying,Li Kuanyu

Abnormal iron metabolism, mitochondrial dysfunction and the derived oxidative damage are the main pathogeneses of Friedrich's ataxia (FRDA), a single-gene inherited recessive neurodegenerative disease characterized by progressive cerebellar and sensory ataxia. This disease is caused by frataxin (FXN) mutation, which reduces FXN expression and impairs iron sulfur cluster biogenesis. To date, there is no effective therapy to treat this condition. Curcumin is proposed harboring excellent ability to resist oxidative stress through Nrf2 activation and its newly found ability to chelate iron. However, its limitation is its poor water solubility and permeability. Here, we synthesized slow-release nanoparticles (NPs) by loading curcumin (Cur) into silk fibroin (SF) to form NPs with an average size of 150 nm (Cur@SF NPs), which exhibited satisfactory therapeutic effects on the improvement of FRDA manifestation in lymphoblasts (1 μM) derived from FRDA patients and in YG8R mice (150 mg/kg/5 days). Cur@SF NPs not only removed iron from the heart and diminished oxidative stress in general but also potentiate iron-sulfur cluster biogenesis, which compensates FXN deficiency to improve the morphology and function of mitochondria. Cur@SF NPs showed a significant advantage in neuron and myocardial function, thereby improving FRDA mouse behavior scores. These data encourage us to propose that Cur@SF NPs are a promising therapeutic compound in the application of FRDA disease.Graphical Abstract

Bimetallic Carbon Dots Cur/Cu, Fe@CDs Nanozymes Enhanced Luminol Chemiluminescence for the Determination of Magnesium Ions

Gu Jianlong,Gao Li,Cai Jieyun,Liu Yuchen,Meng Hongming,Long Jie,Ma Jing,Yang Bing,Wang Chunqiong,Yang Yaling

Carbon dots-based nanozymes show great potential for chemiluminescence (CL) detection due to their stability, excellent catalytic properties, good biocompatibility, and facile synthesis. In this study, bimetallic carbon dot nanoenzymes (Cur/Cu, Fe@CDs) that promote the chemiluminescence signal of luminol were synthesized. Cur/Cu, Fe@CDs possess high peroxidase (POD) activity and can catalyze the luminol/hydrogen peroxide reaction to generate a strong chemiluminescence intensity, which is approximately 8.98 times higher than the chemiluminescence spectral signal produced by the same concentration of K 3 [Fe(CN) 6 ]. Additionally, a sensitive and rapid method for detecting magnesium ions (Mg 2+ ) was developed by leveraging the enhancing effect of Mg 2+ on the CL signal in this system. The proposed CL technique for Mg 2+ demonstrated a linear response range from 3.33 to 166.67 µM and a detection limit of 0.99 µM. Moreover, the method was successfully applied to analyze Mg 2+ in cigarette paper samples through a spiking procedure, achieving a recovery rate of 91.1% to 109.0%. This research offers substantial evidence supporting the advancement of efficient and sensitive platforms for Mg 2+ CL detection.

The PTT, SDT, and CDT synergistic antitumor effect of Fe3+-coordinated PDA@PLGA-Cur nanoparticles in vitro

Fanfan Zhang,Xuemei Hu,Yue Zhang,Baiyuan Sha,Fengshi Shang,Wenjie Wang,Rui Liu,Hao Wang

Hepatocellular carcinoma (HCC) poses a severe threat to public health, with clinical therapy limited by narrow surgical indications, chemotherapeutic side effects, and targeted drug resistance. Herein, we developed Fe 3+ coordination Polydopamine@Poly (lactic-co-glycolic acid)-Curcumin (Fe 3+ -PDA@PLGA-Cur, FePPC) nanoparticles integrating photothermal therapy (PTT), chemodynamic therapy (CDT), sonodynamic therapy (SDT), and curcumin (Cur)'s intrinsic anticancer activity for synergistic HCC treatment. FePPC was fabricated via emulsion solvent evaporation, in-situ dopamine polymerization, and Fe 3+ coordination chelation, with its structure and physicochemical properties characterized by FT-IR, UV-vis, SEM, XPS, and particle size analysis. FePPC exhibited uniform spherical morphology (198.58 nm), favorable stability, 73.60 ± 0.51% encapsulation efficiency, and NIR-triggered temperature-dependent Cur release. Under 808 nm NIR light (2 W/cm 2 , 4 cm), FePPC achieved a 24.4 °C temperature elevation with robust photothermal stability. Catalytic assays confirmed FePPC generated ·OH via Fe 3+ -mediated Fenton-like reactions, 1 O 2 under NIR irradiation, and ·O₂⁻ upon ultrasound stimulation, realizing synergistic reactive oxygen species (ROS) production. In vitro experiments on HepG2 cells showed time-dependent FePPC internalization; the FePPC + PTT + CDT + SDT group reduced cell viability to 53.45%, induced 47.55% apoptosis, elevated intracellular ROS, disrupted mitochondrial membrane potential, downregulated LDH, modulated Bax/Bcl-2 expression, and inhibited cell migration. This study highlights FePPC's potential as a multifunctional nanoplatform for HCC therapy via coordinated chemical-biological interactions.

Advanced Nanopharmaceutical Intervention for the Reduction of Inflammatory Responses and the Enhancement of Behavioral Outcomes in APP/PS1 Transgenic Mouse Models

Jun Li,Dongqing Huang,Wanchen Liao,Yulin Wang,Yibiao Liu,Ping Luan

Background: The excessive accumulation of Aβ plays a critical role in the development of Alzheimer’s disease. However, the therapeutic potential of drugs like curcumin is often limited by low biocompatibility and BBB permeability. In this study, we developed a nanomaterial, BP-PEG-Tar@Cur, which was designed to enhance the biocompatibility of (curcumin) Cur, target Aβ, and augment BBB permeability through near-infrared (NIR) photothermal effects. Methods: Soluble Aβ, ThT fluorescence, and Aβ depolymerization fluorescence experiments were conducted to evaluate the ability of BP-PEG-Tar@Cur to inhibit Aβ aggregation and dissociate Aβ fibrils. Cell uptake assays were performed to confirm the targeting ability of BP-PEG-Tar@Cur towards Aβ. In vitro mitochondrial ROS clearance and in vivo detection of inflammatory factors were used to assess the anti-inflammatory and antioxidant properties of the nanodrug. Water maze behavioral experiments were conducted to evaluate the effect of BP-PEG-Tar@Cur on spatial memory, learning ability, and behavioral disorders in AD mice. Results: The nanodrug effectively inhibited Aβ aggregation and dissociated Aβ fibrils in vitro. BP-PEG-Tar@Cur demonstrated efficiency in curbing ROS overproduction in mitochondria and dampening the activation of microglia and astrocytes triggered by Aβ aggregation. Water maze behavioral experiments revealed that BP-PEG-Tar@Cur enhanced spatial memory, learning ability, and alleviated behavioral disorders in AD mice. Conclusions: Collectively, these findings demonstrate that BP-PEG-Tar@Cur has the potential to be an effective targeted drug for inhibiting Aβ aggregation and improving cognitive impairment in AD mice.

Cu,Fe,B/Cur-CDs with peroxidase- and ascorbic acid oxidase-like catalytic activity for dual-signal fluorescence sensing of metronidazole

Linqian Hou,Qiulan Li,Jitao Li,Dezhi Yang,Yaling Yang

A curcumin based iron, copper and boron doping carbon dots (Cu,Fe,B/Cur-CDs) with ascorbic acid oxidase (AAO)- and peroxidase (POD)-like activities were synthesized via microwave digestion. The Cu,Fe,B/Cur-CDs oxidize ascorbic acid (AA) to dehydroascorbic acid (DHAA), which reacts with o -phenylenediamine (OPD) to generate fluorescent 3-(1,2-dihydroxyethyl) furo [3,4-b]quinoxaline-1-one (DFQ). Simultaneously, it catalyzes OPD/H₂O₂ to fluorescent 2,3-diaminophenazine (DAP) via POD-like activity. In the presence of metronidazole (MNZ), it induces competitive inhibition by competing with OPD for the active sites of Cu, Fe, B/Cur-CDs. This reduces the generation of •OH and •O₂ − radicals, hindering the oxidation of OPD to form DAP and thus decreasing the system's fluorescence intensity. Additionally, MNZ quenches the fluorescence of DFQ at 420 nm via the inner filter effect. Therefore, a dual-signal fluorescence sensor for MNZ detection was constructed. The sensor achieved 91.6–109.9 % and 86.2–116.2 % recovery in food samples using standard addition, demonstrating high sensitivity for dual-signal detection systems.

Poly-l-lysine modified MOF nanoparticles with pH/ROS sensitive CIP release and CUR triggered photodynamic therapy against drug-resistant bacterial infection

Ya-li Xiang,De-yu Tang,Ling-ling Yan,Luo-lin Deng,Xing-han Wang,Xin-yang Liu,Qing-han Zhou

The challenge of drug resistance in bacteria caused by the over use of biotics is increasing during the therapy process, which has attracted great attentions of the clinicians and scientists around the world. Recently, photodynamic therapy (PDT) triggered by photosensitizer (PS) has become a promising treatment method because of its high efficacy, easy operation, and low side effect. Herein, the poly- l -lysine (PLL) modified metal-organic framework (MOF) nanoparticles, ZIF/PLL-CIP/CUR, were synthesized to allow both reactive oxygen species (ROS) responsive drug release and photodynamic effect for synergistic therapy against drug resistant bacterial infections. The PLL was modified on the shell of the zeolite imidazole framework (ZIF) by the ROS-responsive thioketal linker for controllable CIP release. CUR were encapsulated in ZIF as the photosensitizer for blue light mediated photodynamic effect to produce singlet oxygen ( 1 O 2 ) and superoxide anion radical ( O 2 − ) for efficient inhibition towards methicillin-resistant Staphylococcus aureus ( MRSA ). The charge conversion from negative charge (−4.6 mV) to positive charge (2.6 mV) was observed at pH 7.4 and pH 5.5, and 70.9 % CIP was found released at pH 5.5 in the presence of H 2 O 2 , which suggests the good biosafety at physiological pH and ROS-responsive drug release of the as-prepared nanoparticle in the bacterial microenvironment. The as-prepared nanoparticles could effectively kill MRSA and disrupt bacterial biofilm by combination of chemo- and photodynamic therapy. In mice model, the as-prepared nanoparticles exhibited excellent biosafety and synergistic effect with 98.81 % healing rate in treatment of MRSA infection, which is considered as a promising candidate in combating drug resistant bacterial infection.

Enriching and Smart Releasing Curcumin via Phenylboronic Acid-Anchored Bioinspired Hydrogel for Diabetic Wound Healing

Bangjiao Zhao,Shuangli Zhu,YuWei Liu,Jiayi Zhu,Huitong Luo,Maocai Li,Hao Wang,Qi Feng,Xiaodong Cao

Diabetic wounds suffer from bacterial infections, reactive oxygen species, and inflammatory reactions. Curcumin (Cur) is a natural diketone with antibacterial, anti-inflammatory, antioxidant activities, but poor water solubility and toxicity at high local concentration. Herein, inspired by the oxygen transport process of red blood cells via the dynamic interaction of hemoglobin and oxygen, phenylboronic acid (PBA)-anchored hydrogel (GOHA-PBA) is utilized to achieve active enrichment and smart release of Cur, which can reverse the inflammatory microenvironment and promote wound healing. First, PBA-modified gelatin (Gel-PBA) and oxidized hyaluronic acid (OHA) crosslink and form GOHA-PBA by borate ester reaction and Schiff base reaction. GOHA-PBA possesses excellent curcumin loading efficiency with 130 000 times than that in water and forms Cur-laden GOHA-PBA (GOHA-Cur). GOHA-Cur has injectability, self-healing, and self-adaptive properties, which are conducive to surgical procedure. GOHA-Cur owns reversible adhesion properties, facilitating it be peeled off and remove harmful substances, including excess glucose, acids, bacteria, and ROS at the wounds. GOHA-Cur can pH and glucose responsively release Cur; hence, it shows excellent antibacterial, antioxidant, anti-inflammatory effects in vitro. Animal experiments show that GOHA-Cur can inhibit inflammation and promote wound regeneration. This study can provide a valuable concept for Cur delivery and diabetic wound healing.

Biocompatible, adhesive and stable GelMAc/PVAMA/MPDA@Cur hydrogels regulate immune response to improve endoscopic submucosal dissection-induced gastric ulcer healing in vivo

Xu Zhang,Ye He,Xuan Li,Chuanchuan Lin,Zhang Yuan,Liangliang Dai,Feng Ma,Yi Lv,Tony Jun Huang,Mudan Ren,Kaiyong Cai,Shuixiang He

Endoscopic submucosal dissection (ESD) is one of the most important strategies for early gastrointestinal tumor treatment. However, the ideal healing of artificial ulcers produced after ESD procedure are still hard to achieve. In a gastric ulcer environment, the inherent peristole, acid condition and excessive oxidative stress seriously affect the therapeutic outcome of traditional medication. Curcumin (Cur) is a promising anti-inflammatory drug for gastric ulcer treatment, but its long-term use is still challenging. A polymer-assisted drug delivery system in gastrointestinal disease treatment is a new direction but is still at an initial stage. Herein, we report a hybrid hydrogel consisting of catechol motif-modified methacrylated gelatin (GelMAc) and methacrylated poly (vinyl alcohol) (PVAMA). GelMAc brought significant adhesive property and biocompatibility. PVAMA could effectively provide stability and mechanical strength. Moreover, mesoporous polydopamine nanoparticles were prepared as the drug carrier to load Cur via π-π stacking and hydrogen bonding interactions, and they were encapsulated in the hybrid GelMAc/PVAMA hydrogel. The prepared sample was proved to have a unique stability on gastric tissue even bearing acid (pH 2) and oxidative stress (H 2 O 2 ) conditions. The slowly released Cur from the hydrogel was capable of inducing M2 polarization of macrophages. In an in vivo gastric ulcer model, the prepared sample remarkably improved gastric ulcer healing by curbing the pro-inflammatory response and alleviating oxidative stress. An accelerated reepithelization and angiogenesis were also observed. Thus, this study provides a promising hydrogel for the treatment of ESD-induced ulcers, which may attract more attentions to explore the deeper combination between biomaterials and gastrointestinal research.

Transcutaneous amorphous preparation co-delivering curcumin and modified aptamer as a synergistic approach for the amelioration of psoriasis-like skin inflammation

Nan Jin,Ruotong Chen,Yuling Li,Ziyi Zhang,Baomei Zhang,Jianmin Chen

The aim is to achieve the targeting function of Aptamer 21-2 (Apt) by modifying glycyrrhizic acid (GA) to obtain Apt-GA and by delivering it through mesoporous silica in μm-size (μmS). Moreover, it is planned to induce synergistic effects of the novel amorphous preparation of Apt-GA with curcumin (Cur) in the treatment of psoriasis. UV/vis spectrophotometric analysis and Fourier transform infrared spectroscopy were performed to demonstrate the linkage of Apt and GA. The replacement of GA with Apt-GA in GA-Cur-μmS did not make a difference in the amorphous state, but showed superior fluorescence of Cur in the inflamed dermis similar to psoriatic induced by imiquimod. Consequently, compared to GA-Cur-μmS and Apt, Apt-GA-Cur-μmS exhibited less pronounced psoriatic characteristics in vivo , including Psoriasis Area and Severity Index, histological image, immunohistochemistry and immuno-fluorescence labeling neutrophils. The enzyme-linked immunosorbent assay showed a similar phenomenon that less IL-17A was observed after treatment with Apt-GA-Cur-μmS than with GA-Cur-μmS or Apt. Our finding suggests that this novel Apt delivery system achieved the Apt targeting function in the dermis by taking advantage of GA and μmS, and exhibited synergistic anti-psoriatic effect by co-delivering curcumin .

3D-Printed Biomimetic Hydroxyapatite Composite Scaffold Loaded with Curculigoside for Rat Cranial Defect Repair

Yiping Weng,Xiuchen Yuan,Shijie Fan,Weihao Duan,Yadong Tan,Ruikai Zhou,Jingbin Wu,Yifei Shen,Zhonghua Zhang,Hua Xu

The treatment of various large bone defects has remained a challenge for orthopedic surgeons for a long time. Recent research indicates that curculigoside (CUR) extracted from the curculigo plant exerts a positive influence on bone formation, contributing to fracture healing. In this study, we employed emulsification/solvent evaporation techniques to successfully fabricate poly(ε-caprolactone) nanoparticles loaded with curculigoside (CUR@PM). Subsequently, using three-dimensional (3D) printing technology, we successfully developed a bioinspired composite scaffold named HA/GEL/SA/CUR@PM (HGSC), chemically cross-linked with calcium chloride, to ensure scaffold stability. Further characterization of the scaffold’s physical and chemical properties revealed uniform pore size, good hydrophilicity, and appropriate mechanical properties while achieving sustained drug release for up to 12 days. In vitro experiments demonstrated the nontoxicity, good biocompatibility, and cell proliferative properties of HGSC. Through alkaline phosphatase (ALP) staining, Alizarin Red S (ARS) staining, cell migration assays, tube formation assays, and detection of angiogenic and osteogenic gene proteins, we confirmed the HGSC composite scaffold’s significant angiogenic and osteoinductive capabilities. Eight weeks postimplantation in rat cranial defects, Micro-computed tomography (CT) and histological observations revealed pronounced angiogenesis and new bone growth in areas treated with the HGSC composite scaffold. These findings underscore the scaffold’s exceptional angiogenic and osteogenic properties, providing a solid theoretical basis for clinical bone repair and demonstrating its potential in promoting vascularization and bone regeneration.

Hybrid Mesoporous–Microporous Nanocarriers for Overcoming Multidrug Resistance by Sequential Drug Delivery

Liucan Wang,Haidi Guan,Zhenqiang Wang,Yuxin Xing,Jixi Zhang,Kaiyong Cai

Combination chemotherapy with a modulator and a chemotherapeutic drug has become one of the most promising strategies for the treatment of multidrug resistance (MDR) in cancer therapy. However, the development of nanocarriers with a high payload and sequential release of therapeutic agents poses a significant challenge. In this work, we report a type of hybrid nanocarriers prepared by polydopamine (PDA) mediated integration of the mesoporous MSN core and the microporous zeolite imidazolate frameworks-8 (ZIF-8) shell. The nanocarriers exploit storage capacities for drugs based on the high porosity and molecular sieving capabilities of ZIF-8 for sequential drug release. Particularly, large amounts of an anticancer drug (DOX, 607 μg mg–1) and a MDR inhibitor curcumin (CUR, 778 μg mg–1) were sequentially loaded in the mesoporous core via π–π stacking interactions mediated by PDA and in the microporous shell via the encapsulation during ZIF-8 growth. The sustained release of DOX was observed to follow earlier and faster release of CUR by acid-sensitive dissolution of the ZIF-8 shell. Furthermore, the nanoparticles showed good biocompatibility and effective cellular uptake in in vitro evaluations using drug-resistant MCF-7/ADR cancer cells. More importantly, the preferentially released CUR inhibited the drug efflux function of the membrane P-glycoprotein (P-gp), which subsequently facilitated the nuclear transportation of DOX released from the PDA-MSN core, and, in turn, the synergistic effects on killing MDR cancer cells. The hybrid mesoporous–microporous nanocarrier holds great promise for combination chemotherapy applications on the basis of sequential drug release.
