Site-specific nanomodulator capable of modulation apoptosis for enhanced colorectal cancer chemo-photothermal therapy

Wang Shuqi,Zhou Li,Tian Hailong,Li Bowen,Su Miao,Li Qiong,Nice Edouard C.,Huang Canhua,Shao Jichun,He Tao

Background Colorectal cancer (CRC) is a common malignancy with the second highest mortality and the third highest morbidity worldwide. However, the overall survival of patients is unsatisfactory, thus requiring more effective clinical strategies. Celastrol (CLT), a natural bioactive compound, has been reported to induce reactive oxygen species (ROS)-mediated apoptosis to exhibit significant antitumor effects against CRC. However, the poor water solubility, low targeting ability, and bioavailability of CLT have limited its application, and CLT-induced protective autophagy weakens its therapeutic efficiency. Results We designed a targeted chemo-phototherapy nanoplatform (HCR NPs) to improve the application of CLT. The codelivery of IR820 and CLT in HCR NPs solved the water-soluble problem of CLT and enhanced apoptosis via IR820-mediated hyperthermia. In addition, hydroxychloroquine (HCQ) conjugated to hyaluronic acid (HA) not only increased the active targeting of HCR NPs but also inhibited CLT-induced protective autophagy to exacerbate apoptosis, thus achieving an amplified antitumor effect. Importantly, the HCR NPs exhibited an excellent therapeutic effect on CRC both in vitro and in vivo. Conclusion The HCR NPs presented in this study may not merely provide a new reference for the clinical application of CLT but also result in an attractive strategy for CRC treatment.

Laser-activatable oxygen self-supplying nanoplatform for efficiently overcoming colorectal cancer resistance by enhanced ferroptosis and alleviated hypoxic microenvironment

Jiang Hao,Tian Hailong,Wang Zhihan,Li Bowen,Chen Rui,Luo Kangjia,Lu Shuaijun,Nice Edouard C.,Zhang Wei,Huang Canhua,Zhou Yuping,Zheng Shaojiang,Gao Feng

Background Colorectal cancer (CRC) is the second most deadly cancer worldwide, with chemo-resistance remaining a major obstacle in CRC treatment. Notably, the imbalance of redox homeostasis-mediated ferroptosis and the modulation of hypoxic tumor microenvironment are regarded as new entry points for overcoming the chemo-resistance of CRC. Methods Inspired by this, we rationally designed a light-activatable oxygen self-supplying chemo-photothermal nanoplatform by co-assembling cisplatin (CDDP) and linoleic acid (LA)-tailored IR820 via enhanced ferroptosis against colorectal cancer chemo-resistance. In this nanoplatform, CDDP can produce hydrogen peroxide in CRC cells through a series of enzymatic reactions and subsequently release oxygen under laser-triggered photothermal to alleviate hypoxia. Additionally, the introduced LA can add exogenous unsaturated fatty acids into CRC cells, triggering ferroptosis via oxidative stress-related peroxidized lipid accumulation. Meanwhile, photothermal can efficiently boost the rate of enzymatic response and local blood flow, hence increasing the oxygen supply and oxidizing LA for enhanced ferroptosis. Results This nanoplatform exhibited excellent anti-tumor efficacy in chemo-resistant cell lines and showed potent inhibitory capability in nude mice xenograft models. Conclusion sTaken together, this nanoplatform provides a promising paradigm via enhanced ferroptosis and alleviated hypoxia tumor microenvironment against CRC chemo-resistance.Graphical Abstract

A Versatile Prodrug Strategy to In Situ Encapsulate Drugs in MOF Nanocarriers: A Case of Cytarabine-IR820 Prodrug Encapsulated ZIF-8 toward Chemo-Photothermal Therapy

Huiyuan Zhang,Qian Li,Ruiling Liu,Xinke Zhang,Zhonghao Li,Yuxia Luan

Zeolitic imidazolate framework-8 (ZIF-8) is an attractive metal organic framework (MOF) in drug delivery. Strong interaction between drugs and ZIF-8 is essential for high drug loadings through in situ construction of MOFs. However, only limited drugs with unique functional groups (  COOH,  SO 3 H, et al.) can interact with ZIF-8 and be encapsulated satisfactorily so far. Drugs without these functional groups are difficult to be loaded due to the lack of strong interaction. Herein a versatile prodrug strategy is proposed to solve the problems encountered by MOFs. Cytarabine (Ara) is chosen as a model drug since it cannot be loaded in ZIF-8 satisfactorily by itself. New indocyanine green (IR820) is utilized to bond with Ara for the formation of prodrug (Ara-IR820) and endows the prodrug with fluorescence imaging-guided chemo-photothermal therapy, in which sulfonic groups strengthen the interaction between prodrug and ZIF-8. This prodrug loaded ZIF-8 is further functionalized with hyaluronic acid (HA) to result in active-targeting HA/Ara-IR820@ZIF-8 nanoparticles. The in vitro and in vivo results demonstrate its excellent visual cancer therapy with tumor-targeted and pH-responsive release behavior. This design offers a new concept to solve the drug loading problem of MOFs, exhibiting a flexible strategy to expand the biomedical applications of MOFs.

Catalase-positive Staphylococcus epidermidis based cryo-millineedle platform facilitates the photo-immunotherapy against colorectal cancer via hypoxia improvement

Haijiao Wang,Ruiling Gou,Jiayu Chen,Qian Wang,Xiaoyu Li,Jiaxin Chang,Hongli Chen,Xianwen Wang,Guoyun Wan

The synergistic anti-tumor impact of phototherapy and a cascading immune response are profoundly limited by hypoxia and a weakened immune response. Intravenous and intratumoral injection of therapeutic drugs also cause pain, rapid drug clearance and low utilization rates. Here, a novel cryo-millineedle platform for intratumoral delivery of a phototherapy system, S. epi @IR820, is developed in this work, combining the properties of Staphylococcus epidermidis ( S. epidermidis ) and IR820 for photo-immunotherapy of colorectal cancer. In this cryo-millineedle platform, S. epidermidis enhances the near-infrared absorption and light stability of IR820 and catalyzes the decomposition of H 2 O 2 into O 2 via an endogenous catalase to relieve tumor hypoxia, improve phototherapy and enhance immunogenic cell death (ICD). More interestingly, the native immunogenicity of S. epidermidis and ICD elicited by phototherapy achieved a potent anti-tumor immune response. To the best of our knowledge, this is the first study to utilize native S. epidermidis to relieve hypoxia and facilitate phototherapy. Both in vitro and in vivo experiments showed that the millineedle based phototherapy system can efficiently catalyse the decomposition of H 2 O 2 into O 2 , facilitate phototherapeutic killing of CT26 tumor cells by S. epi @IR820 and enhance ICD, thus successfully activated the immune response and achieved the photo-immunotherapy against colorectal cancer. In conclusion, this study provides a novel strategy for enhanced anti-tumor efficiency of photo-immunotherapy, and develops an effective method for orthotopic administration of tumors.

Interface-Engineered Paclitaxel-Based Hollow Mesoporous Organosilica Nanoplatforms for Photothermal-Enhanced Chemotherapy of Tumor

Chunxi Qian,Bandar Al-Hamyari,Xiaofei Tang,Bo Hou,Shuaibo Yang,Guifang Zhang,Huijuan Lv,Zhigang Yang,Zhaohui Wang,Yanbin Shi

Having benefited from the combination of different therapeutic modalities, functionalized nanoplatforms with synergistic strategies have aroused great interest in anticancer treatment. Herein, an engineered, a biodegradable hollow mesoporous organosilica nanoparticle (HMON)-based nanoplatform was fabricated for photothermal-enhanced chemotherapy of tumor. For the first time, we demonstrated that HMONs could serve as nanocarriers for co-delivering of both the paclitaxel and photothermal agent new indocyanine green (IR820), denoted as Paclitaxel/IR820@ HMONs-PEG. The as-prepared nanosystem exhibited a high paclitaxel-loading capacity of 28.4%, much higher than most paclitaxel-loaded nanoformulations. Furthermore, incorporating thioether bonds (S-S) into the HMONs’ framework endowed them with GSH-responsive biodegradation behavior, leading to the controllable release of drugs under a tumor reducing microenvironment, and hindered the premature release of paclitaxel. Upon being irradiated with an NIR laser, the obtained co-delivery nanosystem exhibited great photothermal properties generated from IR820. The fabricated nanocomposites could significantly suppress tumor growth under NIR laser irradiation, as validated by in vitro and in vivo assessments. Combined with outstanding biocompatibility, the constructed nanosystem holds great potential in combinational antitumor therapy.

Cold to Hot: Rational Design of a Minimalist Multifunctional Photo-immunotherapy Nanoplatform toward Boosting Immunotherapy Capability

Di Zhang,Jing Zhang,Qian Li,Aixin Song,Zhonghao Li,Yuxia Luan

The concept of integrating immunogenic cell death (ICD) with tailoring the immunosuppressive tumor microenvironment (TME) is promising for immunotherapy. Photothermal therapy (PTT) could efficiently induce ICD, while an indoleamine 2,3-dioxygenase (IDO) inhibitor could convert the “cold” TME. Therefore, combination of PTT and the IDO inhibitor is an attractive approach for immunotherapy. Unfortunately, combination of PTT and the IDO inhibitor for tumor therapy is rarely reported. Herein, organic photothermal agent IR820 and IDO inhibitor 1-methyl-tryptophan (1MT) were, for the first time, designed to be an all-rolled-into-one molecule nanoplatform via a molecular engineering strategy. The designed IR820–1MT molecule could self-assemble into nanoparticles with remarkably high dual-therapeutic agent loading (88.8 wt %). Importantly, poor water solubility of 1MT and inadequate targeting and short lifetime of IR820 were all well solved within as-prepared IR820–1MT nanoparticles. The laser-triggered IR820–1MT nanoparticles remarkably enhanced accumulation of cytotoxic T cells, helper T cells, and memory T cells and simultaneously suppressed a proportion of regulatory T cells, resulting in excellent immunotherapy against tumor metastasis and recurrence. Our molecular engineering strategy provides a promising alternative option for design of a robust immunotherapy weapon against tumor metastasis and recurrence.

Chemo-photothermal nanoplatform with diselenide as the key for ferroptosis in colorectal cancer

Kaili Deng,Hailong Tian,Tingting Zhang,Yajie Gao,Edouard C. Nice,Canhua Huang,Na Xie,Guoliang Ye,Yuping Zhou

Colorectal cancer (CRC) is a prevalent clinical malignancy of the gastrointestinal system, and its clinical drug resistance is the leading cause of poor prognosis. Mechanistically, CRC cells possess a specific oxidative stress defense mechanism composed of a significant number of endogenous antioxidants, such as glutathione , to combat the damage produced by drug-induced excessive reactive oxygen species (ROS). We report on a new anti-CRC nanoplatform, a multifunctional chemo-photothermal nanoplatform based on Camptothecin (CPT) and IR820, an indocyanine dye. The implementation of a GSH-triggered ferroptosis-integrated tumor chemo-photothermal nanoplatform successfully addressed the poor targeting ability of CPT and IR820 while exhibiting significant growth inhibitory effects on CRC cells. Mechanistically, to offset the oxidative stress created by the broken Se Se bonds, endogenous GSH was continuously depleted, which inactivated GPX4 to accumulate lipid peroxides and induce ferroptosis . Concurrently, exogenously administered linoleic acid was oxidized under photothermal conditions, resulting in an increase in LPO accumulation. With the breakdown of the oxidative stress defense system, chemotherapeutic efficacy could be effectively enhanced. In combination with photoacoustic imaging, the nanoplatform could eradicate solid tumors by means of ferroptosis-sensitized chemotherapy. This study indicates that chemotherapy involving a ferroptosis mechanism is a viable method for the treatment of CRC.

Chemodynamic and Photothermal Combination Therapy Based on Dual-Modified Metal–Organic Framework for Inducing Tumor Ferroptosis/Pyroptosis

Huizi Deng,Jun Zhang,Yifan Yang,Jie Yang,Yawen Wei,Siyu Ma,Qi Shen

Single therapy for tumor therapy always exerts limited ability for the constraints on the reaction condition and the unavoidable multidrug resistance, which seriously influences the therapy effect in the clinic. Herein, a combination treatment nanosystem (MP@PI) based on chemodynamic therapy (CDT) and photothermal therapy (PTT) is constructed for triggering ferroptosis/pyroptosis, which is the metal–organic framework (MOF) modified with polydopamine (PDA) and IR820 to loaded with piperlongumine (PL). The MOF and PL respectively served as the iron source and H2O2 source, performing chemodynamic therapy (CDT) for eliciting ferroptosis. Meanwhile the iron source induces pyroptosis in tumor cells. PDA is not only pH responsive to release PL but also CDT-assisted which due to PDA consumes the glutathione to decrease the expression of glutathione peroxide 4. The photosensitizer IR820 exerts photothermal effects under near-infrared light and further facilitates the ferroptosis/pyroptosis. In addation, the MP@PI nanoplatform evokes the immune response in vivo and enhances the antitumor effects further. Overall, MP@PI is a kind of promising cancer therapy strategy through CDT and PTT combination, inducing ferroptosis and pyroptosis.

Functionalized Periodic Mesoporous Organosilica Nanoparticles for Combinational Chemo-Photothermal Antitumor Therapy

Chunxi Qian,Meng Wang,Lifang Wang,Haijiao Wang,Yun Li,Xiaofei Tang,Yanwen Zhou,Zhaohui Wang,Zhigang Yang,Yanbin Shi

Functionalized nanoplatforms show great promise in effectively delivering therapeutics to tumor sites with enhanced permeability and retention effect. Particularly, periodic mesoporous organosilica nanoparticles (PMONs) have become attractive nanocarriers in biomedical applications owing to their large specific surface area and tunable mesoporous channels. In the current work, innovative PMONs with double hybridization of both disulfide and tetrasulfide bonds were synthesized for improved biodegradability and biocompatibility. The constructed PMONs were applied to simultaneously encapsulate aloe-emodin and IR820 (denoted as Aloe-emodin/IR820@PMONs-PEG) for synergistic chemo-photothermal antitumor therapy. Incorporated thioether bonds endowed Aloe-emodin/IR820@PMONs-PEG with GSH-responsive biodegradation behavior, resulting in the controlled release of therapeutics in the tumor-reducing microenvironment. Furthermore, the as-prepared nanocomposites showed great biocompatibility, good photothermal conversion efficiency, and enhanced cellular uptake. The comprehensive in vitro and in vivo assessments exhibited that Aloe-emodin/IR820@PMONs-PEG had a satisfying synergistic chemo-photothermal antitumor effect. In short, this paper provides valuable insights into the rational design of periodic mesoporous organosilica nanoparticles for synergistic antitumor therapy.

Macrophage Membrane-Camouflaged Prussian Blue-Based Nanoparticles for Targeted Photothermal-Dynamic Therapy against ESKAPE Pathogens and Improved Skin Infection Outcome

Guangyun Hu,Xiaoqing Zhou,Baohua Zhao,Yuan Peng,Qiyun Shi,Zhiqiang Yuan,Qin Shu,Wei Qian,Yizhi Peng

Infections caused by Enterococcus fascism, Staphylococcus aureus, Klebsiella pneumonia, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter species (ESKAPE) bacteria and their biofilms considerably threaten human health. Developing antibacterial agents against ESKAPE bacteria and controlling related inflammation are urgently needed. Herein, we developed a nanosystem by coating M1 macrophage membranes onto the surface of Prussian Blue@polydopamine@IR820 nanoparticles (PDIM NPs) for determining the antimicrobial activity against skin and soft tissue infection. Upon administration, PDIM NPs actively aggregated in the infected regions of subcutaneous abscesses owing to their homologous targeting properties. Under irradiation with a near-infrared (NIR) 808 nm laser, PDIM NPs exhibited outstanding antibacterial effects via synergistic photothermal and photodynamic therapy. Thereafter, the naked Prussian Blue-based nanoparticles effectively scavenged excessive reactive oxygen species and alleviated inflammation. The mouse model demonstrated the regulation of angiogenesis, collagen deposition, and attenuated hyperinflammation. Furthermore, PDIM NPs exhibited excellent biosafety in vitro and in vivo. Therefore, PDIM NPs showed remarkable infectious site-targeting properties and antimicrobial performances and facilitated the healing of cutaneous abscesses, providing strategies for treating ESKAPE bacteria and biofilm-associated diseases.

Self-assembled nano-activator constructed ferroptosis-immunotherapy through hijacking endogenous iron to intracellular positive feedback loop

Hui Xiong,Cheng Wang,Zihan Wang,Haipeng Lu,Jing Yao

Ferroptosis had shown huge potential for antitumor treatment due to its capacity of improving the limited efficiency of traditional antitumor strategies. On the other hand, low confidence in clinical application prospects impeded their development as a result of use of toxic-dose iron. Herein, we prepared a nano-activator (DAR) which was assembled by doxorubicin (DOX), tannic-acid (TA) and IR820 as a photosensitizer to make full use of endogenous iron stored in endo -lysosome, realizing ferroptosis and its related oxidative stress through artificially intracellular positive feedback loop. Interestingly, this process could also promote immunogenic cell death (ICD)-associated immunotherapy through endoplasmic reticulum (ER) stress. After DAR +  laser treatment, the intracellular oxidative stress response was intensified. The produced ROS could be effectively distributed in intracellular lysosomes and ERs to facilitate ferroptosis and immunotherapy respectively. The pharmacodynamics study revealed that DAR + laser had excellent antitumor combination therapy efficiency even under the adverse combined drug ratio of DOX and IR820 due to the unique synergism activation effect of DAR mediated ferroptosis-immunotherapy. In summary, our study provided an innovative solution for the development of antitumor treatment based on ferroptosis-immunotherapy.

A Macrophage Membrane-Coated Cu–WO3–x-Hydro820 Nanoreactor for Treatment and Photoacoustic/Fluorescence Dual-Mode Imaging of Inflamed Liver Tissue

Yanni Luo,Caiying Li,Fanggui Ye,Jianniao Tian,Xuecai Tan,Shengqiang Hu,Shulin Zhao

A disease-targeting nanoplatform that integrates imaging with therapeutic activity would facilitate early diagnosis, treatment, and therapeutic monitoring. To this end, a macrophage membrane-coated Cu–WO3–x-Hydro820 (CWHM) nanoreactor was prepared. This reactor was shown to target inflammatory tissues. The reactive oxygen species (ROS) such as H2O2 and ·OH in inflammatory tissues can react with Hydro820 in the reactor to form the NIR fluorophore IR820. This process allowed photoacoustic/fluorescence dual-mode imaging of H2O2 and ·OH, and it is expected to permit visual diagnosis of inflammatory diseases. The Cu–WO3–x nanoparticles within the nanoreactor shown catalase and superoxide enzyme mimetic activity, allowing the nanoreactor to catalyze the decomposition of H2O2 and ·O2– in inflammatory cells of hepatic tissues in a mouse model of liver injury, thus alleviating the oxidative stress of damaged liver tissue. This nanoreactor illustrates a new strategy for the diagnosis and treatment of hepatitis and inflammatory liver injury.
