Organic NIR-II dyes with ultralong circulation persistence for image-guided delivery and therapy

Yang Li,Jianfeng Gao,Shuping Wang,Shijun Li,Xiaowen Hou,Yanna Pan,Jialu Gao,Xue Qiao,Zhiquan Tian,Deliang Chen,Hai Deng,Zixin Deng,Xuechuan Hong,Yuling Xiao

Nanocarriers hold great promise for the controlled release of therapeutic payloads to target organs/tissues and extended duration of anticancer agents in the bloodstream. However, limited data on their in vivo pharmacokinetics and delivery process hamper clinical applications. Here we report a series of micellar nanocarriers self-assembled from new-generation thiophenthiadiazole (TTD)-based NIR-II fluorophores HLAnP ( n  = 1–4) for simultaneous bioimaging and drug delivery. The NIR-II HLA4P nanocarrier displays exceptional non-fouling performance, minimal immunogenicity, ultralong blood half-life, and high tumor accumulation even with different administration routes. When used as a drug carrier, HLA4P with encapsulated doxorubicin (DOX) realized accurate tumor targeting and continuous real-time in vivo NIR-II tracking of drug delivery and therapy, showing a sustained release rate, improved therapeutic effect, and diminished cardiotoxicity as compared to free DOX. This study provides a new perspective on the design of dual-functional NIR-II fluorophores for diagnostic and therapeutic applications.

A High-efficiency blue-LED-excitable NIR-II-emitting MgO:Cr3+,Ni2+ phosphor for future broadband light source toward multifunctional NIR spectroscopy applications

Bo-Mei Liu,Xiao-Xuan Guo,Lu-Yu Cao,Lin Huang,Rui Zou,Zhi Zhou,Jing Wang

The development of phosphor-converted light-emitting diodes (pc-LEDs) in the second near-infrared window (NIR-II, 1000–1700 nm) represents an important, newly emerging, and dynamic field in NIR spectroscopy. Unfortunately, lacking efficient NIR-II phosphors that can be excited by commercial blue LED chips impedes NIR spectroscopy applications. Herein, a donor–acceptor strategy is developed by introducing Cr 3+ sensitizers to greatly enhance the blue-light excitation efficiency of NIR-II-emitting MgO:Cr 3+ ,Ni 2+ phosphor. Consequently, a high-efficiency blue-light-excitable MgO:Cr 3+ ,Ni 2+ phosphor that gives a NIR-II emission at 1335 nm with a full width at half-maximum (FWHM) of 235 nm, is demonstrated. The product has a high internal quantum efficiency of 92.7 % and excellent thermal stability, maintaining 83.0 % of the room temperature emission intensity at 150 °C. Excitingly, the fabricated NIR-II pc-LED device shows a high NIR-II optical power (27.4 mW@350 mA). The performances of the achieved NIR-II pc-LED are almost the best results until now. Additionally, multifunctional applications including nondestructive detection and anti-counterfeiting of the NIR-II light source are demonstrated. These results are pretty crucial for the further development of NIR-II spectroscopy and imaging technology.

NIR-II-responsive AuNRs@SiO2–RB@MnO2 nanotheranostic for multimodal imaging-guided CDT/PTT synergistic cancer therapy

Changchun Wen,Xiaolu Guo,Cunji Gao,Zhongkai Zhu,Nianqi Meng,Xing-Can Shen,Hong Liang

Specific tumor-responsive capabilities and efficient synergistic therapeutic performance are the keys to effective tumor treatment. Herein, AuNRs@SiO2–RB@MnO2 was developed as a new type of tumor-responsive nanotheranostic for multimodal imaging and synergistic chemodynamic/photothermal therapy. In AuNRs@SiO2–RB@MnO2, the SiO2 layer wraps the AuNRs, providing light absorption in the second near-infrared (NIR-II) region. The SiO2 layer also adsorbs the MnO2 nanosheets, which have Fenton-like activity, resulting in a fluorescent sensing platform based on the fluorescence quenching properties of MnO2 for rhodamine B dye. The fluorescence can be recovered by the consumption of MnO2 by glutathione, which simultaneously produces Mn2+ in the tumor region. The recovery of fluorescence reflects the consumption of glutathione and the increase in Mn2+, which produces hydroxyl radicals via Fenton-like reaction in the tumor microenvironment to realize chemodynamic therapy. Meanwhile, the AuNRs are a good photothermal reagent that can effectively absorb NIR-II light and convert it into heat energy to kill tumor cells via photothermal therapy. The NIR-II absorption performance of the AuNRs provides good photoacoustic imaging and deep photothermal performance, which is favorable for efficient NIR-II photoacoustic imaging-guided photothermal therapy. As a result, the AuNRs@SiO2–RB@MnO2 nanotheranostic exhibits outstanding imaging and synergistic chemodynamic/photothermal therapeutic performance for tumor imaging and treatment.

Miniature Hollow Gold Nanorods with Enhanced Effect for In Vivo Photoacoustic Imaging in the NIR-II Window

Kai Cai,Weiyun Zhang,Mohamed F. Foda,Xuyu Li,Jin Zhang,Yeteng Zhong,Huageng Liang,Huiqiao Li,Heyou Han,Tianyou Zhai

The miniaturization of gold nanorods exhibits a bright prospect for intravital photoacoustic imaging (PAI) and the hollow structure possesses a better plasmonic property. Herein, miniature hollow gold nanorods (M-AuHNRs) (≈46 nm in length) possessing strong plasmonic absorbance in the second near-infrared (NIR-II) window (1000–1350 nm) are developed, which are considered as the most suitable range for the intravital PAI. The as-prepared M-AuHNRs exhibit 3.5 times stronger photoacoustic signal intensity than the large hollow Au nanorods (≈105 nm in length) at 0.2 optical density under 1064 nm laser irradiation. The in vivo biodistribution measurement shows that the accumulation in tumor of miniature nanorods is twofold as high as that of the large counterpart. After modifying with a tumor-targeting molecule and fluorochrome, in living tumor-bearing mice, the M-AuHNRs group gives a high fluorescence intensity in tumors, which is 3.6-fold that of the large ones with the same functionalization. Moreover, in the intravital PAI of living tumor-bearing mice, the M-AuHNRs generate longer-lasting and stronger photoacoustic signal than the large counterpart in the NIR-II window. Overall, this study presents the fabrication of M-AuHNRs as a promising contrast agent for intravital PAI.

Ultrasmall AgBiSe2 nanodots for CT/thermal imaging-guided photothermal tumor therapy in the NIR-II biowindow

Jingge Zhang,Enna Ha,Danyang Li,Luyang Wang,Junqing Hu

Stimulus-responsive ternary chalcogenide nanomaterials are regarded as promising ‘all-in-one’ nanotheranostics agents on account of their tunable band structures and multi-metal intrinsic properties. Herein, ultrasmall AgBiSe2 nanodots are prepared by a simple thermal injection method. It shows a narrow band gap of 0.91 eV and high absorption coefficient in the NIR-II biowindow, resulting in excellent photothermal performance. Under the irradiation of a 1064 nm laser, AgBiSe2 can induce the overexpression of intracellular heat shock protein (Hsp70) and cell apoptosis to inhibit the growth of tumor cells. The strong signal from CT/thermal imaging also provides guidance for tumor diagnosis. Importantly, AgBiSe2 can be rapidly excreted from the body, thus avoiding long term toxicity. This study presents the first biomedical application of AgBiSe2 nanodots in cancer treatment and extends the development of ternary chalcogenide-based semiconductor nanomedicine.

Blue-Light-Excitable Broadband Ca3Ga2Ge3O12:Cr3+,Ni2+ Phosphor for the Applications in NIR-II Window

Fangyi Zhao,Yuhe Shao,Quanlin Liu,Jiasong Zhong

Near-infrared (NIR) phosphor-converted light-emitting diode (pc-LED) light sources in the second NIR window (NIR-II, 1000–1700 nm) have sparked great interest for their emerging applications in non-destructive detection and medical diagnostics fields. However, the development of efficient NIR-II phosphors that can be excited by commercial blue LED chips remains a significant challenge, and thus impeding the applications of NIR pc-LED. Herein, a blue-light-excitable broadband NIR-II luminescence of Ni 2+ is achieved upon incorporating Cr 3+ and Ni 2+ into the Ca 3 Ga 2 Ge 3 O 12 (CGGO) host simultaneously. Through the energy transfer from Cr 3+ to Ni 2+ , CGGO:Cr 3+ ,Ni 2+ phosphor presents the improved NIR-II emission peaking at 1470 nm under 460 nm blue light excitation. The internal/external quantum efficiency values are significantly improved from 13.8%/3.3% to 24.6%/13.3% along with the enhanced luminescence thermal stability. Finally, a NIR pc-LED is fabricated by combining CGGO:Cr 3+ ,Ni 2+ phosphor with 450 nm blue chip, and its potential applications in non-destructive examination and biomedical imaging fields have been demonstrated. The strategy of Cr 3+ and Ni 2+ co-doping solves the problem that Ni 2+ luminescence cannot be effectively excited by blue light and provides the design insights to exploit more ultra-broadband NIR pc-LEDs based on blue LED chips.

Targeting TNFAIP2 with NIR-II CRISPR-Cas9 nanosystem to overcome cisplatin resistance in laryngeal cancer

Li Xiaoli,Wang Jiashuo,Guo Jiaxing,Zhang Ming

Cisplatin resistance is a major challenge in laryngeal cancer treatment. Tumor necrosis factor-alpha-induced protein 2 (TNFAIP2) has been implicated in chemoresistance, though its role in laryngeal cancer remains unclear. We identified TNFAIP2 as a key gene associated with cisplatin resistance and developed a second near-infrared (NIR-II) light-responsive CRISPR-Cas9 nanosystem (APC@RBCs) to target it. In cisplatin-resistant Tu177/CDDP cells, light-activated knockout of TNFAIP2 significantly increased cisplatin sensitivity (IC50 reduced from 12.55 to 4.37 µg/mL). Mechanistic studies revealed that TNFAIP2 modulates cisplatin resistance via the NRF2 pathway, affecting oxidative stress response and epithelial-mesenchymal transition. TNFAIP2 deletion also suppressed cancer cell migration and invasion. Our results establish TNFAIP2 as a critical mediator of chemoresistance and demonstrate the potential of APC@RBCs as a precise, non-invasive gene therapy platform. This strategy offers a promising approach to overcoming cisplatin resistance and improving treatment outcomes in laryngeal cancer patients.

Cholesterol-Modified Black Phosphorus Nanospheres for the First NIR-II Fluorescence Bioimaging

Yifan Xu,Feng Ren,Hanghang Liu,Hao Zhang,Yaobao Han,Zheng Liu,Wenliang Wang,Qiao Sun,Chongjun Zhao,Zhen Li

Black phosphorus (BP) nanostructures with unique layer-dependent properties have been extensively applied in the fields of electronic devices, energy conversion and storage, and nanomedicine. As a narrow band gap semiconductor, they are expected to show strong second near-infrared (NIR-II) fluorescence. However, there is no report on the NIR-II fluorescence of free-standing BP nanostructures, which have great potential in the NIR-II fluorescence bioimaging because of their excellent biocompatibility and biodegradability. Here, for the first time, we report that the BP nanoparticles modified with cholesterol exhibit strong NIR-II fluorescence and can be encapsulated with the PEGylated lipid to form BP@lipid-PEG nanospheres for in vitro and in vivo NIR-II imaging. The resultant BP@lipid-PEG nanospheres exhibit broad emissions from 900 to 1650 nm under excitation by an 808 nm laser and have 8% quantum yield of that of standard dye IR-26. We also show that the NIR-II fluorescence image acquired with emission beyond 1400 nm has the sharpest contrast and can be used to in situ measure the diameter of blood vessels. In addition to NIR-II fluorescence imaging, we also show the potential of BP@lipid-PEG nanospheres in photoacoustic (PA) imaging. Both the long-wavelength NIR-II fluorescence imaging and PA imaging reveal that the as-fabricated BP@lipid-PEG nanospheres can be gradually metabolized by the liver in 48 h, thus making them promising for bioapplications.

Constructing a Self-Referenced NIR-II Thermometer with Energy Tuning of Coordinating Water Molecules by a Minimalist Method

Jianhao Zheng,Pengye Du,Ran An,Yuan Liang,Yi Wei,Shuyu Liu,Xinyu Wang,Pengpeng Lei,Shuyan Song,Hongjie Zhang

Fluorescence thermometry based on metal halide perovskites is increasingly becoming a hotspot due to its advantages of high detection sensitivity, noninvasiveness, and fast response time. However, it still presents certain technical challenges in practical applications, such as complex synthesis methods, the use of toxic solvents, and being currently mainly based on the visible/first near-infrared light with poor penetration and severe autofluorescence. In this study, we synthesize the second near-infrared (NIR-II) luminescent crystals based on Yb3+/Nd3+-doped zero-dimensional Cs2ScCl5·H2O by a simple “dissolve–dry” method. The whole synthesized process does not involve high temperatures or high pressures. Cs2ScCl5·H2O/Yb3+,Nd3+ has an optimum fluorescence performance when the Yb3+/Nd3+ doping amount is 15%/20%. The emission intensity ratio attributed to Yb3+ and Nd3+ varies with temperature, and this variation is exacerbated due to the fact that the 2F5/2 energy level of Yb3+ can be effectively aligned with the O–H bond stretching vibration energy of coordinating water molecules to facilitate energy transfer. Ultimately, the crystals can act as self-referenced ratiometric NIR-II luminescent thermometers with a maximum relative sensitivity of 1.66% K–1 at 323 K. This work highlights the advantages of NIR-II luminescent materials for temperature sensing, which is significant for advancement in the field of noncontact thermometers.

Noninvasive Detection and Monitoring of the Integrity of the Intestinal Barrier through NIR-II Fluorescence Imaging and Colorimetric Urinalysis

Li Zhang,Lei Feng,Lin Yang,Hao Wan

The variation of the integrity of the intestinal barrier is closely related to the occurrence and progression of various diseases, making its accurate detection and monitoring essential to provide reliable information for guiding the refinement of treatment scenarios. Herein, we developed a strategy utilizing glutathione-capped gold clusters (Au-GSH) to achieve simultaneous in vivo second near-infrared (NIR-II) fluorescence imaging and in vitro colorimetric urinalysis for noninvasive detection and monitoring of intestinal barrier integrity. After oral administration, Au-GSH can demonstrate different distribution behaviors in terms of the variation of intestinal barrier integrity. Specifically, Au-GSH could selectively permeate through compromised intestinal barrier to be distributed into the bladder in view of its ultrasmall size below the glomerular filtration cutoff, leading to the rapid excretion through urine. Such process can be visually profiled by collecting NIR-II fluorescence (>1100 nm) emitting from Au-GSH in a noninvasive real-time manner. By virtue of the peroxidase-like activity of Au-GSH, the simple colorimetric urinalysis was further established for evaluating the integrity of the intestinal barrier, advancing the detection and monitoring performance. As a result, our developed strategy successfully detected the damage of intestinal barrier integrity in ulcerative colitis (UC) mice, a common disease characterized by compromised intestinal barrier integrity. Excitingly, the monitoring of restoration of intestinal barrier integrity in both therapeutic and preventative modes for UC mice was also realized by our strategy, making it a promising diagnosis scenario for diseases related to the variation of intestinal barrier integrity.

Biomimetic gold nano-modulator for deep-tumor NIR-II photothermal immunotherapy via gaseous microenvironment remodeling strategy

Honglin Huang,Zhengxin Xie,Ningxi Li,Li Zeng,Qianyi Zeng,Ziman Yang,Jinyang Shen,Hong Yang,Yiyao Liu,Chunhui Wu

Introduction Effective immunotherapeutic treatment of solid tumors has been greatly challenged by the complex hostile tumor immunosuppressive microenvironment (TIME), which typically involves hypoxia and immunosuppression. Methods Herein, a multifunctional biomimetic gold nano-modulator (denoted as GNR-SNO@MMT ) was developed to realize the efficient second near-infrared (NIR-II) photothermal immunotherapy via tumor targeting and deep penetration, vascular normalization and immune reprogramming. NIR-II photothermal agent gold nanorods (GNR) were grafted with thermosensitive S-nitrosothiol (SNO) donors and camouflaged with the tumor-penetrating peptide tLyp-1-modified macrophage membrane (MM) to yield GNR-SNO@MMT . Results The engineered membrane coating increased the capacity for tumor inflammatory tropism and deep penetration, which aided GNR-SNO@MMT in ablating tumors together with NIR-II laser irradiation. Moreover, hyperthermia-stimulated nitric oxide (NO) release in situ acted as a gas immunomodulator to effectively enhance blood perfusion and reprogram the TIME via multiple functions (e.g., decreasing PD-L1, repolarizing tumor-associated macrophages, and revitalizing cytotoxic T cells). Ultimately, the inhibition rate against 4T1 mouse mammary tumor model mediated by GNR-SNO@MMT plus NIR-II laser was 94.7% together with 2.4-fold CD8 + T cells infiltrated into tumors than that of the untreated counterpart. Conclusions The engineered biomimetic nano-modulator of GNR-SNO@MMT provides an effective and novel photoimmunotherapy candidate against deep-sited solid tumors through immune reconfiguration via NO-involved nanomedicine and external NIR-II laser assistance. Triggered Cascade-Activation Nanoplatform to Alleviate Hypoxia for Effective Tumor Immunotherapy Guided by NIR-II Imaging

Yu Ji,Suchen Qu,Gaoyu Shi,Liansheng Fan,Jing Qian,Zhaorui Sun,Feng Lu,Xin Han

Hypoxia is one of the most typical features among various types of solid tumors, which creates an immunosuppressive tumor microenvironment (TME) and limits the efficacy of cancer treatment. Alleviating hypoxia becomes a key strategy to reshape hypoxic TME which improves cancer immunotherapy. However, it remains challenging to perform tumor precision therapy with controllable switches through hypoxia-activated gene editing and prodrugs to alleviate hypoxia. In this study, silica-coated second near-infrared window (NIR-II) emitting silver sulfide quantum dots are used as the carrier to load the Clustered Regularly Interspaced Short Palindromic Repeats/Cas9 (CRISPR/Cas9) system to target hypoxia-inducible factor-1 (HIF-1α) and guide tumor-targeted imaging. To reduce the off-target effects in nontumor cells and better control safety risks, a TME-triggered cascade-activation nanodiagnostic and therapeutic platform (AA@Cas-H@HTS) is designed, which achieves the hypoxia activation of prodrug tirapazamine (TPZ) and spatiotemporal release of CRISPR/Cas9 ribonucleoprotein. Tumor hypoxia is greatly alleviated by the synergistic function of HIF-1α depletion by gene editing and TPZ activation. Importantly, targeting HIF-1α disrupts the programmed cell death 1/programmed cell death ligand 1 (PD-1/PD-L1) signaling pathway, which effectively reshapes the immune-suppressive TME and activates T cell-mediated antitumor immunity. Taken together, we have provided a TME-triggered cascade-activation nanoplatform to alleviate hypoxia for improved cancer immunotherapy.
