Dramatic fluorescence enhancement of PCN-224 and its application in “turn off” immunoassay for sensitive detection of E. coli O157:H7 in milk

Bolong Fang,Xin Liu,Juan Peng,Yuzhi Li,Zhiyong Gong,Weihua Lai

In this study, a type of luminescent porous coordination network-224 (PCN-224) in alkaline conditions was synthesized with the dramatic fluorescence enhancement by 20.4 times, which was explained by the fact that the decrease of Zr 4+ content in alkaline conditions resulted in the partial recovery of the electron cloud density of 4,4′,4′′,4′′′-(Porphine-5,10,15,20-tetrayl) tetrakis(benzoic acid) (TCPP). Given the large overlap between the excitation spectrum of PCN-224 and the absorption band of Ag nanoparticles (Ag NPs), the coating of the Ag layer on PCN-224 triggered the fluorescence quenching effect, which was applied to “turn off” fluorescence immunoassay for sensitive detection of Escherichia coli O157:H7 ( E. coli O157:H7) in milk. The proposed immunoassay reached a low limit of detection (LOD) of 3.3 × 10 2 CFU mL −1 , 29.7 times more sensitive than the conventional ELISA. It will provide a novel alternative strategy for sensitively detecting pathogenic bacteria in the field of food safety.

Porphyrinic Metal–Organic Framework PCN-224 Nanoparticles for Colocalization of Nanoparticle-Cargo in the Skin and Enhancement of Sonodynamic Efficacy

Xinxing Sun,Qiuyue Wang,Hongyang Bao,Yitong Liu,Sicheng Yao,Mingming Li,Naiying Chen,Yang Chen

Sonodynamic therapy (SDT) is a novel therapeutic modality that is effective for the noninvasive treatment of dermatological diseases. As a ubiquitous large class of sonosensitizers, however, porphyrins suffer from poor skin permeability and the inefficient generation of reactive oxygen species. Herein, different-sized porphyrinic metal–organic frameworks (MOFs), PCN-224 (90, 125, and 200 nm), were developed via the coordination of tetra-kis(4-carboxyphenyl) porphyrin (TCPP) and the metal zirconium to enhance the local delivery of porphyrins, and fluorescent sonosensitizers (methylene blue or rhodamine 6G) were further incorporated to improve the sonodynamic effect, as well as to visualize the delivery of PCN-224 and its cargo. The skin penetration of PCN-224 was found to be increased with the reduction of particle size, and all three-sized samples could overcome the barrier of stratum corneum, even achieving the dermis of the porcine skin. Both intercellular and follicular pathways were noticed for three-sized PCN-224, while the follicular one played a more significant role in the penetration of the smallest PCN-224. The intact nanoparticles were found in the skin. At the initial stage, the nanoparticle and its cargo penetrated the skin as an intact entity, while the encapsulated rhodamine 6G was slowly released to the microenvironment of the skin from the nanoparticle with the prolongation of application. The combination of PCN-224 and methylene blue could increase the acoustically triggered generation of 1O2, distinctly boosting the lethal effect against the cancerous cell. Collectively, the study revealed the feasibility of PCN-224 for topical use, suggested an innovative method to enhance the sonodynamic effect, and proposed a possible mechanism for skin drug delivery of MOFs.

Enhanced singlet oxygen generation in PCN-224-Zn without the need of electron-hole separation for efficient photocatalytic water disinfection

Xin Jiang,Sanmei Liu,Wen Wang,Shunli Shi,Zhenxing Zeng,Chao Chen

Photocatlytic water disinfection for pathengenic miroorgasnisms removal is of great importance to public health safety, however the photocataltyic performance is unsatisfactory due to the low reactive oxygen species (ROS) generation. In this work, an innovative energy-transfer-mediated oxygen activation system on PCN-224-Zn is proposed for selective singlet oxygen generation without the need of electron-hole separation. The as-prepared PCN-224-Zn exhibits remarkable singlet triggered photocatalytic E-coli inactivation and organic pollutant degradation performance. The enhanced photocatalytic activity is attributed to the introduction of Zn into the metal-organic frameworks (MOFs) that facilitates the transformation of electron-hole couple from singlet to triplet state for selective singlet oxygen generation with high efficiency. This work provides a new oxygen activation pathway for efficient and selective ROS generation without the need of electron-hole separation.

“Switch-On” Fluorescence Detection of Glucose with High Specificity and Sensitivity Based on Silver Nanoparticles Supported on Porphyrin Metal–Organic Frameworks

Peiyao Du,Qixia Niu,Jing Chen,Yang Chen,Jie Zhao,Xiaoquan Lu

As the energy source of living cells and the intermediate product of metabolism, glucose plays an important role in biological systems. Therefore, it is of great significance to establish a reliable and sensitive method for the detection of glucose, especially in blood. Herein, a “switch-on” fluorescence sensor for rapid, sensitive, and specific detection of glucose was successfully developed. In this strategy, PCN-224 served as the recognition unit, while AgNPs played the roles as both a “quencher”, to decrease the fluorescence intensity of PCN-224, and an H2O2 recognizer. After exposure to H2O2 that was produced in situ during GOx-catalyzed oxidation of glucose, AgNPs can be effectively etched into silver ions and released from PCN-224, thereby recovering the fluorescence of PCN-224. The present sensing strategy shows many merits including high sensitivity with a low limit of detection (0.078 μM) and excellent selectivity toward glucose over other saccharides. More importantly, the sensing platform we proposed was further extended to monitoring glucose in human serum samples with satisfactory recoveries, indicating its promising potential for diagnostic purposes.

Precise Temperature-Gradient Regulated Structure–Activity Relationship of PCN-224 as Bifunctional Nanocarriers

Lejie Tian,Huidan Xue,Michael Gradzielski,Li Shang,Shuanhong Ma,Peiwei Gong,Long Wang,Jianxi Liu,Weimin Liu

Metal–organic frameworks (MOFs) demonstrate considerable potential for biomedical applications. Due to their high porosity and photoactive porphyrin ligands, PCN-224 is considered an ideal drug delivery nanocarrier. However, the synthesis strategy for tailoring the application of PCN-224 in complex pathological diseases remains unclear. In this study, we investigated the structure–activity relationship of PCN-224 nanoparticles as bifunctional nanocarriers for osteoarthritis (OA) treatment under a precisely controlled temperature gradient. By tuning the temperatures, a series of PCN-224 nanoparticles with adjustable sizes were synthesized and their morphologies transitioned from a spindle-shaped to a spherical-shaped structure. The photoluminescence intensity of PCN-224 nanoparticles decreased as the synthesis temperatures increased. Using PCN-224 nanoparticles as aqueous lubricating additives, both the friction coefficient and wear volume were significantly reduced. Among the MOFs, PCN-224 (synthesized at 105 °C) exhibited excellent antifriction performances and acid-responsive drug release properties, demonstrating their promise as bifunctional OA treatment nanocarriers. Our work enhanced the understanding of the relationships between PCN-224 synthesis conditions and its physicochemical properties relationships, expanding their potential for multifunctional biomedical applications.

Zr-porphyrinic MOF PCN-224 enables sequential antibacterial and inflammation-modulating PDT for periodontitis

Qi Liu,Chenxi Tang,Jiafeng Liu,Yuan Gao,Lin Peng

Periodontitis is a chronic inflammatory disease driven by microbial dysbiosis and a dysregulated host response, leading to soft-tissue destruction and alveolar bone loss. Antibacterial photodynamic therapy (aPDT) can rapidly suppress pathogens via light-triggered reactive oxygen species (ROS). Here, we report zirconium-based porphyrinic metal–organic framework (MOF) nanoparticles, PCN-224, as a bifunctional platform that couples early antibacterial aPDT with subsequent immunomodulation within the periodontal microenvironment. PCN-224 demonstrated excellent biocompatibility in vitro and in vivo . Under 660 nm irradiation, PCN-224 effectively generated singlet oxygen ( 1 O 2 ) and reduced Porphyromonas gingivalis burden. After uptake by RAW 264.7 macrophages, PCN-224 attenuated lipopolysaccharide-induced inflammation by lowering intracellular ROS levels, restoring superoxide dismutase activity, decreasing nitric oxide release, suppressing NF-κB signaling, accompanied by reduced IL-6, IL-1β, and iNOS and partial recovery of anti-inflammatory mediators. In a ligature-induced mouse model, local PCN-224 injection combined with 660 nm illumination decreased inflammatory infiltration and alleviated alveolar bone resorption. Collectively, PCN-224 provides a time-sequenced antibacterial-to-anti-inflammatory therapeutic effect and represents a potential MOF-enabled strategy for periodontitis management.

Light-propelled actived Janus PCN-224@Au nanomotors overcome transport barriers for broad-spectrum sterilization

Xiao Wang,Yanan Ren,Yuhao Yang,Hui Tu,Dandan Fan,Yuchuan Zhuang,Jing Li,Yanbo Dong,Andrey Tulupov,Ying Hu,Yong Zhang,Jianfeng Bao

The accelerating global crisis of multidrug-resistant infections and the physical intractability of dense bacterial biofilms demand therapeutic platforms that are both functionally active and externally controllable. Here we report the rational design and synthesis of a fuel-free, near-infrared (NIR) propelled Janus nanomotor, PCN-224@Au, that unites photothermal therapy (PTT) with sonodynamic therapy (SDT) in a single construct. Asymmetric deposition of gold onto a porphyrinic PCN-224 metal-organic framework (MOF) establishes the geometric anisotropy required for propulsion and a metal–MOF interface that promotes charge separation. Under low-power 808-nm irradiation, the Au cap converts light to heat and drives self-thermophoretic motion, yielding directed transport with ensemble speeds of ∼10 μm s −1 and super-diffusive MSD of ∼60 μm 2 at 4 s while producing a local temperature rise of 35 + °C within 600 s at 100 μg mL −1 . Ultrasound (US) activates ROS formation in PCN-224, and the Janus architecture simultaneously delivers NIR photothermal input to amplify the effect. Dual activation with NIR and US therefore delivers localized thermal stress (PTT) together with high ROS flux (SDT), producing broad-spectrum antibacterial effects against methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli (E. coli) . Light-driven motility enhances deep biofilm penetration and nanomotor-bacteria contact, while US-activated ROS disrupt membranes and trigger cytoplasmic leakage, translating into rapid, depth-resolved sterilization with favorable hemocompatibility and cytocompatibility. By coupling controllable propulsion with sonocatalysis, this Janus nanomotor approach overcomes transport and resistance barriers and provides an effective, fuel-free platform for broad-spectrum antibacterial therapy.

Smartphone-assisted ratiometric fluorescence biosensor for mercury ion detection based on fluorescence-enhanced PCN-224

Meng Tian,Jiaxiong Li,Weiru Li,Zheng Liu,Rui Jin,Chunyan Sun,Junyang Wang

Fluorescence-enhanced PCN-224 (FE-PCN-224) was prepared by modifying porphyrin MOF PCN-224 by 1 M Tris-HCl. We found that FE-PCN-224 was able to significantly enhance the fluorescence intensity of 6-FAM-labelled aptamer (FAM-Apt) in pure water, while its own fluorescence intensity was not disturbed. Based on the above, a ratiometric fluorescence biosensing platform was constructed relying on the specific recognition of Hg 2+ by T (thymine)-rich aptamers and the PET effect between T-Hg 2+ -T and FAM. The emission of FE-PCN-224 was used as the reference signal, and the emission of FAM-Apt was used as the sensitive response signal for detecting Hg 2+ . Hence, the concentration gradient of Hg 2+ could be accurately identified by the difference of the ratiometric signals F 518 / F 650 . The sensing system has good reliability and high stability. With the support of ImageJ software and mobile phone colour analysis software, convenient quantitative detection was realized through RGB channel analysis of real colour images. Meanwhile the prepared fluorescent colourimetric card can be applied to achieve semi-quantitative detection by the naked eye. The developed convenient biosensor provides a potential route for rapid detection of mercury ion contamination.

Hydrothermal Deposition of PCN-224 on 3D-Printed Porous β-Ca2SiO4 Scaffolds for Bone Regeneration

Jianhang Du,Weiye Liu,Zhanrong Kang,Bin Yu,Dejian Li,Huifeng Ding,Jianming Huang

3D-printed bioceramics derived from silicone resin are of great interest in bone tissue engineering owing to their simple processes and low cost. Herein, PCN-224, as a subclass of metal−organic frameworks (MOFs), is successfully fabricated and deposited on the surface of 3D-printed porous β-Ca 2 SiO 4 (C 2 S) scaffolds. The composite scaffolds possess uniform interconnected macropores (≈400 μm), with increased porosity and slower degradation. The results indicate that deposition of PCN-224 significantly promotes cell proliferation and the expression of osteogenesis-related genes. Moreover, the composite scaffolds significantly improve calvarial defect healing in rats, compared with C 2 S scaffolds. Therefore, the PCN-224-modified 3D-printed porous C 2 S composite scaffolds are promising candidates for bone tissue engineering.

Novel CdS/MOF Cathodic Photoelectrochemical (PEC) Platform for the Detection of Doxorubicin Hydrochloride and Gentamicin Sulfate

Wenxia Dong,Zhongping Li,Wen Wen,Bin Liu,Guangming Wen

Nanomaterial selection is critical for photoelectrochemical (PEC) sensing. In this report, a novel cathodic photoelectrochemical (PEC) strategy was proposed for the detection of doxorubicin hydrochloride (Dox) and gentamicin sulfate (CN). The photocathode was synthesized by noncovalently coupling cadmium sulfide (CdS) to the porphyrin-derived metal–organic framework (CdS@PCN-224). This type of assembly created a pleasant interface for the combination of doxorubicin hydrochloride and gentamicin sulfate, resulting in a good CdS@PCN-224 donor–acceptor system. When compared to a single optoelectronic material, its photocurrent is enhanced by unprecedented nine times. This research could pave the way for the realization of PCN-224’s enormous potential in PEC sensing.

Injectable, pH-Responsive Chitosan/Tannic Acid/PCN-224 Nanocomposite Hydrogels with Synergistic Phototherapeutic and Antioxidant Properties

Wang Jiayu,Yan Herui,Li Fan,Chen Zhuoming,Xin Binjie,Wu Yu

Diabetic wounds remain difficult to treat due to bacterial resistance, a localized weakly acidic microenvironment, imbalanced oxidative stress, and poor dressing conformity. To address these challenges, this study developed an injectable pH-responsive nanocomposite hydrogel using phenylboronic acid-modified chitosan (PBCS), tannic acid (TA), and porphyrinic metal–organic framework PCN-224 nanoparticles, featuring with superior tissue adhesiveness, self-healing, and pH-responsiveness. The hydrogel was fabricated using PBCS as the matrix and TA as the cross-linker, establishing a dual-cross-linked network sustained by dynamic boronate ester bonds and multiple hydrogen bonds, into which the porphyrinic metal–organic framework (MOF) PCN-224 was subsequently loaded. This hydrogel precisely senses and exploits the endogenous weakly acidic microenvironment of the infected wound, triggering the dissociation of boronate ester bonds to achieve the on-demand release of TA, as well as the smart regulation of the network’s swelling behavior. The synchronously released polyphenol component, TA, imparts excellent antioxidant activity to scavenge excess free radicals and re-establish oxidative stress balance. Meanwhile, under 660 nm laser irradiation, PCN-224 generated reactive oxygen species and a mild photothermal effect, enabling combined photodynamic and photothermal antibacterial activity. Collectively, the PBCS-TA/PCN-224 nanocomposite hydrogel integrates pH-responsive behavior, antioxidant activity, and light-triggered antibacterial functionality, supporting its potential application in infected diabetic wounds.

Light responsive chitosan hydrogel incorporated with PCN-224@berberine for antibiotic resistant bacterial infected wound management

Zhiyue Feng,Shiqi Chen,Ke Han,Qiang Ma,Boyan Sun,Chujun Liu,Qiqi Wang,Hao Zhao,Zimo Liu,Junying He,Zhimeng Zhang,Haiyang Jiang

Infections caused by multidrug-resistant bacteria significantly hinder wound healing, highlighting the need for non-antibiotic alternatives. Here, we developed a multifunctional hydrogel platform, LAMC@PCN-224@BBR, by incorporating photodynamic PCN-224 and the natural product berberine (BBR) into lipoic acid-modified chitosan (LAMC). This design combines photo-crosslinking for enhanced tissue adhesion with photodynamic therapy (PDT) for antimicrobial activity. Upon light irradiation, the hydrogel generates reactive oxygen species (ROS), effectively eradicating methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli ( E. coli ) with an efficiency exceeding 99 %. Its excellent biocompatibility and adaptive adhesion allow conformal coverage of irregular wounds. In a full-thickness skin defect model, the hydrogel accelerated healing by suppressing inflammation, promoting collagen deposition, enhancing angiogenesis, and polarizing macrophages toward the pro-regenerative M2 phenotype. These findings demonstrate that the LAMC@PCN-224@BBR hydrogel is a promising clinical strategy for controlling resistant infections and promoting tissue regeneration.
