Synergistic antimicrobial effect of nisin−octanoic acid nanoemulsions against E. coli and S. aureus

Guo Hui,Yang Wei,Lei Bingshuang,Zhao Fengju,Guo Lili,Qian Junqing

Food safety is a major public health concern all over the world. Therefore, the prevention of food contamination is becoming extremely crucial. In this study, an antimicrobial nanoemulsion composed of water-soluble nisin and fat-soluble octanoic acid was successfully prepared. The results showed that the average particle size and the polymer dispersity index of the nisin−octanoic acid (NOA) nanoemulsion were around 52.21 nm and 0.253, respectively. The NOA nanoemulsion required less amounts of nisin and octanoic acid to achieve the effective antimicrobial effect against Escherichia coli and Staphylococcus aureus . In addition, the growth curves of E. coli and S. aureus were determined. The OD 600 of NOA nanoemulsion was significantly lower than free nisin after being incubated for 24 h ( p  < 0.001), indicating that the antimicrobial effect of NOA nanoemulsion was outstanding. Meanwhile, the synergistic antimicrobial property of NOA nanoemulsion against E. coli and S. aureus was significantly better than free nisin under nonacid conditions ( p  < 0.05). Overall, the results of this study may provide guidance for the further application of nisin in more forms.

Gelatinase-responsive biodegradable targeted microneedle patch for abscess wound treatment of S. aureus infection

Xiao-Ling Lei,Kai Cheng,Yong-Guo Hu,Yong Li,Xiao-Lin Hou,Fang Zhang,Lin-Fang Tan,Zi-Tao Zhong,Jian-Hao Wang,Jin-Xuan Fan,Yuan-Di Zhao

Abscess wound caused by bacterial infection is usually difficult to heal, thus greatly affect people's quality of life. In this study, a biodegradable drug-loaded microneedle patch (MN) is designed for targeted eradication of S. aureus infection and repair of abscess wound. Firstly, the bacterial responsive composite nanoparticle (Ce6@GNP-Van) with a size of about 182.6 nm is constructed by loading the photosensitizer Ce6 into gelatin nanoparticle (GNP) and coupling vancomycin (Van), which can specifically target S. aureus and effectively shield the phototoxicity of photosensitizer during delivery. When Ce6@GNP-Van is targeted and enriched in the infected regions, the gelatinase secreted by the bacteria can degrade GNP in situ and release Ce6 , which can kill the bacteria by generating ROS under laser irradiation. In vivo experiments show that the microneedle is basically degraded in 10 min after inserting into skin, and the abscess wound is completely healed within 13 d after applying Ce6@GNP-Van-loaded MN patch to the abscess wound of the bacterial infected mice with laser irradiation, which can simultaneously achieve the eradication of biofilm and subsequent wound healing cascade activation, showing excellent synergistic antibacterial effect. In conclusion, this work establishes a synergistic treatment strategy to facilitate the repair of chronic abscess wound.

Synthesis of sulfonated chitosan and its antibiofilm formation activity against E. coli and S. aureus

Jianying Huang,Yuhong Liu,Lijun Yang,Fengyan Zhou

A sulfonated chitosan (SCS) was prepared via nucleophilic substitution and characterized by Fourier transform infrared spectroscopy, 1 H and 13 C NMR spectra, gel permeation chromatography, elemental analysis, and thermo gravimetric analysis. The inhibition activities of bacterial adhesion and biofilm formation against E. coli and S. aureus of SCS were assessed in comparison with those of unmodified chitosan hydrochloride (WCS) which was commercially available. The metabolic activity and secretion of exopolysaccharide in biofilms of E. coli and S. aureus were significantly decreased after the treatment of SCS. Scanning electron microscopy and confocal laser scanning microscopy also demonstrated that SCS and WCS at 1MIC concentrations could obviously inhibit the formation of biofilm. Upon the experimental data obtained, it can be concluded that the alkylsulfonation of chitosan could significantly improve efficacy in killing biofilm-embedded bacteria, and the inhibition activities against biofilm formation of E. coli and S. aureus . The exploitation of SCS in this study is helpful to extend the understanding to an alternative to antibiotics and chemical preservatives in food and medicine fields.

Biomimetic dandelion-like magnetic nanoparticles for capture and detection of S. aureus and L. monocytogenes

Guanhua Chen,Zhengzheng Wang,Mei Deng,Fangbin Xiao,Xuekun Bai,Hengyi Xu

Foodborne pathogenic microorganism pose a serious threat to public health security; thus, constructing quick and sensitive methods for early screening foodborne pathogenic microorganism is necessary for human health. Teicoplanin (Teic) is a broad-spectrum biometric molecule, which can recognize Gram-positive (G + ) bacteria. In this study, using bovine serum albumin (BSA) and PEG 2k as “carrier” and “linker”, respectively. Teic was wired to magnetic beads (MBs) to form the biomimetic dandelion-like magnetic nanoparticles (MBs-PEG-BSA-Teic) to capture Staphylococcus aureus ( S. aureus ) and Listeria monocytogenes ( L. monocytogenes ). The MBs-PEG-BSA-Teic displayed excellent capture ability of S. aureus (> 93%) and L. monocytogenes (> 86%). After separation, the fluorescence probes were added, forming a “sandwich” structural compounds, namely, MBs-PEG-BSA-Teic~bacteria~fluorescence probes. The fluorescence intensity of the probes was measured by a multimode microplate reader. Consequently, this novel method showed high detection sensitivity to target model bacteria, and the detection limit of S. aureus and L. monocytogenes in PBS buffer solution and juice were achieved 10 1 CFU/mL, spinach and ground beef samples were achieved 10 2 CFU/g. The method has easy operation and high sensitivity, and it is less interfered by food matrix or negative bacteria; thus, this method is suitable for early screening G + pathogen in suspected food samples.

Antimicrobial/Anti-Inflammatory Peptidomimetic-Polyphenol Co-Assemblies with pH-Triggered Activation for S. aureus Abscess Therapy

Junfeng Song,Anming Yang,Yang Song,Xirui Huang,Xingjin Li,Tianzhao Xu,Tiancong Zhao

The treatment of Staphylococcus aureus subcutaneous abscesses is hindered by the protective abscess microenvironment and persistent bacterial colonization. Herein, we present a pH-responsive supramolecular nanoplatform fabricated via single-step coassembly of cationic arginine-derived antimicrobial peptidomimetics (AMPM-1) and natural polyphenols. This process, governed by hydrogen bonding, electrostatic interactions, and hydrophobic packing, effectively masks the cationic charges of AMPM-1, substantially reducing intrinsic hemolytic and cytotoxic properties. In vitro studies demonstrate that the assemblies maintain potent, membrane-disruptive activity against methicillin-resistant S. aureus (MRSA), including dormant and persister subpopulations. In a murine MRSA abscess model, local administration of the quercetin-based assemblies (AMPM-1-QA) simultaneously reduces bacterial burden and attenuates excessive inflammatory signaling. Histological examination confirms accelerated tissue regeneration, characterized by enhanced collagen deposition and anti-inflammatory macrophage polarization. By integrating acid-triggered bactericidal action with polyphenol-mediated immunomodulation, this dual-functional supramolecular strategy offers a promising approach for the stimuli-responsive treatment of recalcitrant localized infections.

A cell membrane inspired multifunctional hydrogel loaded with ginsenoside Rh1 for S. aureus infected wound healing

Chaorui Jiang,Yanan Yang,Zhuo Meng,Hao Wu,Kai Wang,Xufei Zheng,Minxiang Zhou,Lijing Zhao,Yanwei Du,Bo Liu,Zhihui Zhang

Graphene oxide enhanced hydrogel enables tunable mechanics. • Sustained Rh1 release promotes angiogenesis via VEGF/CD31 upregulation. • Hydrogel boosts anti-inflammatory cytokines (IL-4, IL-10) for faster healing. • The water gel dressing has an antibacterial rate of up to 98.69% against S. aureus . During the healing process of skin wounds, there are challenges such as slow tissue regeneration and bacterial infections. This paper has developed a drug-loaded composite hydrogel dressing that can promote angiogenesis, inhibit bacteria, and reduce inflammation. The hydrogel is composed of gelatin (Gel) and acrylamide (AM), and is formed into a biomimetic cell membrane structure through cross-linking. By adding different concentrations of graphene oxide (GO), the tensile strength of the hydrogel dressing can be significantly increased (from 57.54 KPa to 95 KPa), and the adhesion strength (shear strength) can be effectively enhanced (16.44 KPa). Additionally, Panax ginsenoside Rh1 has been successfully encapsulated in the hydrogel, achieving local controlled release. In vivo evaluation shows that the enhanced matrix by graphene oxide and the sustained release of Panax ginsenoside Rh1 have an antibacterial rate of up to 98.69% against S. aureus . It also stimulates the proliferation, migration, and tube formation of endothelial cells, thereby achieving strong vascularization at the wound site. Moreover, it can enhance the secretion capacity of anti-inflammatory factors (IL-4 from 19.54 to 105.61 pg/mL, IL-10 from 151.94 to 339.98 pg/mL), thereby accelerating the wound healing process. This hydrogel dressing provides a promising strategy for achieving high-quality wound repair. Download: Download high-res image (223KB) Download: Download full-size image

Synthesis of N-isonicotinic sulfonate chitosan and its antibiofilm activity against E. coli and S. aureus

Lin Wang,Yu Pang,Zhongwen Su,Meihua Xin,Mingchun Li,Yangfan Mao

N -(sodium 2-hydroxypropylsulfonate) chitosan (HSCS), N -sulfonate chitosan (SCS) and N -isonicotinic sulfonate chitosan (ISCS) were prepared. The structures of the prepared chitosan derivatives were characterized by nuclear magnetic resonance ( 1 H NMR) spectroscopy, Fourier transform infrared (FTIR) spectroscopy, ultraviolet–visible (UV–vis) spectroscopy and elemental analysis (EA). Antibacterial and antibiofilm activities of these chitosan derivatives were evaluated in vitro . The minimum inhibitory concentration (MIC) of HSCS and SCS against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) were 0.625 mg/mL and 0.156 mg/mL, respectively. ISCS exhibited MIC values of 0.313 mg/mL and 0.078 mg/mL against E. coli and S. aureus , respectively. ISCS demonstrated superior antibacterial and antibiofilm properties compared to SCS and HSCS. These findings suggest that the incorporation of a pyridine structure into sulfonate chitosan enhances its antibacterial and antibiofilm activities, and the prepared ISCS has a promising application prospect for controlling the reproduction of microorganisms in the field of food packaging.

A novel colorimetric sensing platform for the detection of S. aureus with high sensitivity and specificity

Yun Zhang,Shuyou Shi,Jiajia Xing,Wenqing Tan,Chenguang Zhang,Lin Zhang,Huan Yuan,Miaomiao Zhang,Jinjuan Qiao

In this study, a novel colorimetric sensing platform was developed for the detection of S. aureus using dog immunoglobulin G (IgG) as the capture antibody and chicken anti-protein A immunoglobulin Y labeled with horseradish peroxidase (HRP-IgY) as the detection antibody. Dog IgG labeled with magnetic beads was used to capture S. aureus through the interaction between the Fc region of dog IgG and Staphylococcal protein A (SPA). HRP-IgY was introduced to recognize the residual SPA on the surface of S. aureus and to create a sandwich format, after which a soluble 3,3′,5,5′-tetramethylbenzidine (TMB) substrate was added. A stop solution was utilized to cease the enzymatic chromogenic reaction, and then optical density was read at 450 nm. Under optimal conditions, the proposed method displayed a low detection limit of 1.0 × 103 CFU mL−1 and a wide linear range of 3.1 × 103 to 2.0 × 105 CFU mL−1. This detection method exhibited high specificity against other foodborne bacteria. The recovery rates ranged from 95.2% to 129.2%. To our knowledge, this is the first report to employ dog IgG and chicken IgY as an antibody pair to detect S. aureus. This technique exhibits high application potential for S. aureus monitoring in various kinds of samples.

Electrospun hierarchical structural films for effective wound healing

Haixia Xu,Feiyang Zhang,Menglong Wang,He Lv,Deng-Guang Yu,Xinkuan Liu,Hao Shen

Patients with acute and chronic wounds have been increasing around the world, and the demand for wound treatment and care is also increasing. Therefore, a new nanofiber wound dressing should be prepared to promote the wound healing process. In this study, we report the design and preparation of a hierarchical structural film wound dressing. The top layer is composed of profoundly hydrophobic polycaprolactone (PCL), which is used to resist the adhesion of external microorganisms. The bottom layer is made of hydrophilic gelatin, which provides a moist healing environment for the wound. The middle layer is composed of hydrophilic Janus nanofibers prepared with the latest side-by-side electrospinning technique. Gelatin and PCL are used as polymer matrices loaded with the ciprofloxacin (CIP) drug and zinc oxide nanoparticles (n-ZnO), respectively. Test results show that the dressing has outstanding surface wettability , excellent mechanical properties , and rapid drug release. The presence of biologically active ingredients provides antibacterial activity against Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ). Finally, the results of wound healing in mice show accelerated collagen deposition, promotion of angiogenesis, and complete wound healing within 14 days. Overall, this hierarchical structural dressing has a strong potential for accelerating wound healing.

Poly(Ionic Liquid) Nanofibers Suppress S. aureus Membrane Vesicle-Induced NETosis to Mitigate Wound and Lung Damage

Jiaying Lin,Jiali Duan,Jiangna Guo,Hui Xu,Rongwei Shi,Linhui Zhao,Yangyang Liu,Na Meng,Jiateng Zhou,Xiao Zhang,Shihui Lin,Feng Yan,Bin Wang,Hailei Mao

Staphylococcus aureus membrane vesicles (MVs) cause host injury and excessive inflammation, yet their pathological roles and clearance strategies remain undefined. Guided by molecular dynamics simulations of MV-polymer interactions, we engineered imidazolium-based poly(ionic liquid) (PIL) electrospun nanofibers for targeted MV interception. Among multiple formulations, PIL-C4 demonstrated optimal performance, combining potent antibacterial activity, robust adsorption of methicillin-resistant S. aureus (MRSA) MVs and their virulence factors, minimal cytotoxicity, and inhibition of resistance transmission. In vivo, PIL-C4 attenuated MV-induced neutrophil extracellular trap (NET) formation (NETosis) and vascular leakage, thereby reducing purulent-exudative wound injury and preventing systemic organ damage, including fatal lung injury. Notably, DNase-mediated NET degradation alone failed to rescue MV pathology, underscoring the necessity of direct MV clearance. This study uncovers previously unrecognized NETosis-driven phenotypes of S. aureus MVs—local purulent-exudative wound injury and systemic lethal lung damage—and establishes a polymer-based clearance strategy with translational potential for infection control.

Microwave-excited, antibacterial core-shell BaSO4/BaTi5O11@PPy heterostructures for rapid treatment of S. aureus-infected osteomyelitis

Yuelin Lv,Cuihong Chen,Liguo Jin,Yufeng Zheng,Shuilin Wu,Yu Zhang,Zhaoyang Li,Shengli Zhu,Hui Jiang,Zhenduo Cui,Xiangmei Liu

Owing to its deep penetration capability, microwave (MW) therapy has emerged as a promising method to eradicate deep-seated acute bone infection diseases such as osteomyelitis. However, the MW thermal effect still needs to be enhanced to achieve rapid and efficient treatment of deep focal infected areas. In this work, the multi-interfacial core-shell structure barium sulfate/barium [email protected] (BaSO 4 /BaTi 5 O 11 @PPy) was prepared, which exhibited enhanced MW thermal response via the well-designed multi-interfacial structure. To be specific, BaSO 4 /BaTi 5 O 11 @PPy achieved rapid temperature increases in a short period and efficient clearance of Staphylococcus aureus ( S. aureus ) infections under MW irradiation. After 15 min MW irradiation, the antibacterial efficacy of BaSO 4 /BaTi 5 O 11 @PPy can reach up to 99.61 ± 0.22%. Their desirable thermal production capabilities originated from enhanced dielectric loss including multiple interfacial polarization and conductivity loss. Additionally, in vitro analysis illuminated that the underlying antimicrobial mechanism was attributed to the noticeable MW thermal effect and changes in energy metabolic pathways on bacterial membrane instigated by BaSO 4 /BaTi 5 O 11 @PPy under MW irradiation. Considering remarkable antibacterial efficiency and acceptable biosafety, we envision that it has significant value in broadening the pool of desirable candidates to fight against S. aureus -infected osteomyelitis. Statement of significance The treatment of deep bacterial infection remains challenging due to the ineffectiveness of antibiotic treatment and the susceptibility to bacterial resistance . Microwave (MW) thermal therapy (MTT) is a promising approach with remarkable penetration to centrally heat up the infected area. This study proposes to utilize the core-shell structure BaSO 4 /BaTi 5 O 11 @PPy as an MW absorber to achieve localized heating under MW radiation for MTT. In vitro experiments demonstrated that the disrupted bacterial membrane is primarily due to the localized high temperature and interrupted electron transfer chain. As a consequence, its antibacterial rate is as high as 99.61% under MW irradiation. It is shown that the BaSO 4 /BaTi 5 O 11 @PPy is a promising candidate for eliminating bacterial infection in deep-seated tissues.

A portable 3D-printed pretreatment device combined with graded lateral flow assay for detection of S. aureus

Yang Chen,Min Zhang,Xin Wang,Xin Wang,Zainabu Majid,Kunlun Huang,Wentao Xu,Yunbo Luo,Cheng Nan

There is a need for facile yet reliable bacterial pretreatment and signal-graded strategies for current pathogens' point-of-care testing (POCT). Herein, we present a multifunction and portable 3D-printed magnetic separation device with graded lateral flow assay (LFA) for semi-automated pretreatment and rapid detection of Staphylococcus aureus ( S. aureus ) in food. The 3D-printed pretreatment device allows purification and enrichment of the [email protected] nanoparticles ( [email protected] NPs)-antibody- S. aureus complex. The recovery rate was 81.7–96.1% in actual food samples. [email protected] NPs-mediated signal amplification immunoassay achieved graded detection of S. aureus , reducing the detection limit from 10 3 CFU/mL to 10 2 CFU/mL, corresponding to the highest safety limit value (M) and acceptable level limit value (m) of S. aureus in many countries and organizations, respectively. This study paves the way for cost-effective and sensitive POCT with multiplexing potential in applications ranging from pretreatment of samples in the complex matrix as well as foodborne pathogens graded and rapid analysis.
