The grinding ofTremella fuciformisis a critical step for its value-added processing and the efficient utilization of its functional components, significantly impacting product quality and process adaptability. This study investigated ultrafine grinding (UFG) as a mechano-physical strategy to improve product quality, systematically analyzing its impact on physical properties (particle size, powder characteristics, color), extraction efficiency, chemical composition, and rheological behavior compared to conventional grinding (CG). The results revealed that UFG treatment induced an extensive disruption of the matrix, reducing particle size by 91.8% (D90 = 18.18 μm) and significantly increasing specific surface area. Notably, this physical modification directly translated into enhanced processing performance. UFG powder exhibited reduced powder flowability, superior solubility and improved color brightness. This structural degradation proved beneficial for extraction, unlocking a substantially higher yield (60.98–66.48%). Concurrently, the aqueous extracts of UFG powder exhibited more fluid-like rheological characteristics. This study confirms the potential of UFG as an effective pretreatment for the intensive processing ofT. fuciformisand indicates its promising application in functional food development and the extraction of bioactive components.

Tremella fuciformis polysaccharides (TFP) were evaluated for their gastrointestinal stability and microbiota-modulating effects using in vitro digestion and fecal fermentation. The results demonstrated that TFP possessed an average molecular weight of 2.21 × 103 kDa, primarily composed of glucose (63.67%) and mannose (19.85%), and exhibited resistance to simulated gastrointestinal digestion. During fermentation, TFP was selectively metabolized by the gut microbiota. Compared with the blank control, TFP increased the Shannon index by 7.74% and significantly enhanced SCFA production, with acetate, propionate, and butyrate reaching 21.52, 8.62, and 4.60 mM at 24 h, respectively. TFP fermentation selectively promoted beneficial taxa, including Megamonas (28.3-fold), Prevotella (6.1-fold), and Faecalibacterium (2.4-fold). Correlation analysis revealed positive associations between SCFA production and specific bacterial genera, indicating that TFP supports a healthy gut environment. These findings establish TFP as a novel prebiotic candidate and highlight its translational potential for functional food formulations targeting gut microbiome modulation.

Anti-tumor activity ofTremella fuciformispolysaccharides (TFPS) has been widely reported, but its mechanism remains poorly understood. In this study, we established an in vitro co-culture system (B16 melanoma cells and RAW 264.7 macrophage-like cells) to explore the potential anti-tumor mechanism of TFPS. Based on our results, TFPS exhibited no inhibition on the cell viability of B16 cells. However, significant apoptosis was observed when B16 cells were co-cultured with TFPS-treated RAW 264.7 cells. We further found that mRNA levels of M1 macrophage markers including iNOS and CD80 were significantly upregulated in TFPS-treated RAW 264.7 cells, while M2 macrophage markers such as Arg-1 and CD 206 remained unchanged. Besides, the migration, phagocytosis, production of inflammatory mediators (NO, IL-6 and TNF-α), and protein expression of iNOS and COX-2 were markedly enhanced in TFPS-treated RAW 264.7 cells. Network pharmacology analysis indicated that MAPK and NF-κB signaling pathways may be involved in M1 polarization of macrophages, and this hypothesis was verified by Western blot. In conclusion, our research demonstrated that TFPS induced apoptosis of melanoma cells by promoting M1 polarization of macrophages, and suggested TFPS may be applied as an immunomodulatory for cancer therapy. Keywords:Tremella fuciformispolysaccharide;immunoregulation;co-culture;macrophages;B16

The stem of Tremella fuciformis , an abundant byproduct of edible mushroom processing, is typically discarded despite containing higher levels of crude polysaccharide than the fruiting body. To valorize this agricultural residue, a series of polysaccharide-based hydrogels was fabricated via Schiff base crosslinking between carboxymethyl chitosan (CMCS) and oxidized T. fuciformis stem polysaccharide (OTP) with tunable aldehyde contents. The oxidation degree of OTP critically influenced the crosslinking density, thereby modulating the gelation kinetics, microstructure, rheological properties, and swelling behavior. Specifically, increasing the polysaccharide oxidation degree from 17.30 ± 1.66% to 32.85 ± 1.47% and subsequently to 40.75 ± 1.96% progressively shortened the gelation time from 12 min to 3 min and decreased the pore size from 219.2 ± 12.50 μm to 93.16 ± 16.60 μm. Concurrently, the storage modulus increased from 49 Pa to 388 Pa, and the CMOT3 hydrogel achieved the highest equilibrium swelling rate of 99.6%. All hydrogel formulations exhibited excellent cytocompatibility. In a full-thickness skin defect model, the optimally crosslinked CMOT3 hydrogel significantly accelerated wound closure, which was associated with reduced pro-inflammatory cytokine levels, elevated VEGF expression, and improved collagen organization. These findings suggest that the T. fuciformis stem polysaccharide-based hydrogel effectively promotes skin tissue repair, highlighting its potential as a sustainable functional biomaterial derived from agricultural byproducts.

Tremella fuciformis polysaccharides have diverse bioactivities and beneficial physicochemical properties, enabling broad use in pharmaceuticals and food. This study analyzed intracellular and extracellular polysaccharides (IPs and EPs) from three T. fuciformis spores, assessing their monosaccharide composition and in vitro antioxidant and hypoglycemic effects. Results showed that TWW01-AX and TWW02-TMS had significantly higher biomass and polysaccharide yields than TYH-SD1. IPs were predominantly composed of Glc, exceeding 60 mol%, whereas EPs were primarily composed of Man, accounting for over 64 mol%. Monosaccharide percentages served as key discriminators between IPs and EPs. EPs exhibited stronger DPPH, hydroxyl radical scavenging, CUPRAC, as well as α-glucosidase and α-amylase inhibition, while IPs showed superior ABTS radical scavenging activities and FRAP. Ridge regression showed key monosaccharide percentage variations notably influence bioactivities. This study provided a theoretical basis for the functional evaluation of high-quality T. fuciformis polysaccharides and the efficient screening of superior strains.

BACKGROUND Fortification is an effective approach to improve the nutritional value and functionality of noodles. However, the addition of non-gluten ingredients often impairs noodle quality by weakening the gluten network. This study investigated the potential of Tremella fuciformis pulp (TPG) as a quality improver in shiitake-fortified noodles. RESULTS The addition of TPG increased the viscous modulus and elastic modulus of the dough, reduced the gelatinization temperature of noodles by approximately 3 °C and enhanced the viscosity during gelatinization. It also decreased the water absorption and cooking loss of the noodles and improved the textural properties of noodles such as hardness, elasticity and chewiness. These effects were attributed to the high viscosity of TPG, reduced free sulfhydryl content and enhanced thermal stability. The incorporation of TPG promoted the formation of a more compact noodle microstructure, as confirmed by scanning electron microscopy, thereby improving noodle quality. A critical T. fuciformis -to-water ratio of 1:40 (w/v, equivalent to g mL −1 ) was identified. In addition, the pulp slightly enhanced 1,1-diphenyl-2-picrylhydrazyl (i.e. DPPH) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (i.e. ABTS) radical-scavenging activities and increased both the content and proportion of essential amino acids. CONCLUSION TPG is a feasible ingredient for improving the cooking and textural properties of shiitake noodles, at the same time as producing only slight improvements in selected antioxidant and amino acid indices, indicating its potential for use in fancy noodles and other wheat-based products. © 2026 Society of Chemical Industry.

Bioactive herbal carbon dots with inherited activities for multi-layer skin repair. • Achieving comparable anti-photoaging efficacy at half the concentration of vitamin E. • Breaking the ROS-organelle stress cycle while directly repairing three skin layers. • Concurrent barrier restoration, pigmentation inhibition, and collagen promotion. • Offering superior stability and natural mildness. To meet the substantial global demand for skin anti-aging, particularly for effectively against ultraviolet radiation (the primary exogenous driver of skin aging), this study successfully developed a novel tremella fuciformis polysaccharide derived carbon dots (TFP-CDs) via a hydrothermal method. The skin barrier function of the epidermis and the dense extracellular matrix of the dermis, poses a significant delivery challenge. It limits the penetration and bioavailability of most skincare agents, which would affect their intended multi-layer efficacy. To address this, the prepared TFP-CDs with ultrasmall size (∼2.4 nm) and amphiphilic functional groups, have been designed to sequentially overcome the barriers. As biologically responsive nanomaterials, they integrate the inherent bioactivity of herbal extract with excellent antioxidant properties, exhibiting superior efficacy over positive control drug. Through efficiently scavenging ROS, TFP-CDs may help break the vicious cycle of mitochondrial morphological and functional disorders, endoplasmic reticulum stress, and abnormal organelle contacts exacerbated. This study effectively achieves synergistic anti-aging repair in multi-layered skin repair, ranging from epidermal barrier repair and pigmentation inhibition to dermal collagen regeneration. This work provides a potential strategy for the intervention of skin aging, showing preliminary application promise. Download: Download high-res image (326KB) Download: Download full-size image

Here, the extraction of total flavonoids from Tremella fuciformis (TF) by aqueous two-phase extraction combined with ultrasound is presented. The extraction was optimized via a single-variable approach using the variables of mass fraction of ethanol and NaH2PO4, ultrasonication time, and TF sample amount. Response surface methodology (RSM) was then applied to determine the best conditions for extraction using the following three key parameters: ultrasonication time, ethanol mass fraction, and NaH2PO4 mass fraction. The optimum conditions were as follows: an aqueous two-phase system of ethanol/NaH2PO4, an ethanol mass fraction of 23%, a NaH2PO4 mass fraction of 27.18%, and an extraction time of 8.25 min. Under these conditions, the maximum extraction rate of total flavonoids was 0.158 mg/g. The scavenging rates for superoxide anion and hydroxyl radical, as well as the antioxidant activity of total flavonoids in TF, were studied. Total flavonoids from TF showed some scavenging ability, but the scavenging effects were lower than those of vitamin C (Vc) at the same concentrations. Nevertheless, ultrasound-assisted two-phase extraction was considered an efficient and rapid method that can be used to extract total flavonoids from TF.

Steam explosion (SE) technology is an effective modification method for improving resource utilization of edible fungi processing by-products. In this study, the effect of SE-modified Tremella fuciformis ( T. fuciformis ) stem soluble dietary fiber (SDF) on the quality and digestibility of biscuits was investigated. The results showed that the addition of SE-modified T. fuciformis stem SDF (M-SDF) changed the gluten network structure and moisture distribution in the biscuits, which improved the spread ratio of the biscuits and resulted in attractive colors. Meanwhile, as starch was embedded, the starch hydrolysis rate (from 60.9 ± 0.90 % to 43.01 ± 0.78 %) and estimated glycemic index (from 84.10 ± 4.39 to 68.45 ± 3.15) of 12 % M-SDF biscuits were reduced. Furthermore, 8 % M-SDF received the highest sensory scores. These results demonstrate the potential applicability of SE-modified edible fungi processing by-product SDF as an additive in functional foods.

High molecular weight and inert functional groups limit the biomedical utility of natural polysaccharides, such as Tremella fuciformis polysaccharides (TFPs). In this study, we describe a H 2 O 2 -driven, one-step method for the concurrent oxidation and degradation of TFPs, which reduces the molecular weight from 1079.6 to 4.6 kDa while introducing aldehyde (0 %–0.99 %) and carboxyl (1.22 %–9.80 %) groups. By altering the H 2 O 2 concentration, we obtained oxidized TFPs (OTFPs) with defined properties, namely OTFP-1 (13.5 kDa, 0.36 % aldehyde) exhibiting low cytotoxicity (73 % viability at 15 mg/mL); OTFP-3 (5.0 kDa, 0.80 % aldehyde) exhibiting enhanced antioxidant (DPPH/ABTS: 83 %/89 %), anti-inflammatory (IL-6/TNF-α reduction: 46 %/53 %), and ultraviolet B repair (36 % recovery) activities; and OTFP-4 (4.6 kDa, 0.99 % aldehyde) displaying broad-spectrum antibacterial activity (minimum inhibitory concentration: 7–9 mg/mL). The aldehyde groups also yielded injectable, self-healing hydrogels through dynamic Schiff base bonds. This green strategy decouples molecular weight and functionalization and provides a versatile platform for tailored bioactive materials.

This study aimed to establish a bidirectional fermentation system using Tremella fuciformis and Acanthopanax trifoliatus to promote the transformation and utilization of the synthesized antioxidant metabolites from fermentation supernatant. The effect of fermentation conditions on the total phenolic content was investigated using response surface methodology in terms of three factors, including temperature (22-28°C), pH (6-8), and inoculum size (2-8%, v/v). The optimized fermentation parameters were: 28°C, pH 8, and an inoculum size of 2%, which led to a maximum total phenolic content of 314.79 ± 6.89 μg/mL in the fermentation supernatant after 24 h culture. The content of total flavonoids and polysaccharides reached 78.65 ± 0.82 μg/mL and 9358.08 ± 122.96 μg/mL, respectively. In addition, ABTS+, DPPH⋅, and ⋅OH clearance rates reached 95.09, 88.85, and 85.36% at 24 h under optimized conditions, respectively. The content of total phenolics, flavonoids and polysaccharides in the optimized fermentation supernatant of T. fuciformis - Acanthopanax trifoliatus increased by 0.88 ± 0.04, 0.09 ± 0.02, and 33.84 ± 1.85 times that of aqueous extracts from A. trifoliatus , respectively. Simultaneously, 0.30 ± 0.00, 0.26 ± 0.01, and 1.19 ± 0.12 times increase of antioxidant activity against ABTS+, DPPH⋅, and ⋅OH clearance rates were observed, respectively. Additionally, the metabolite composition changes caused by fermentation were analyzed using ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) based on untargeted metabolomics and the phytochemical profile of fermentation supernatant differentiated significantly based on unsupervised principal component analysis (PCA) during fermentation from 24 to 96 h. Furthermore, a significant increase in antioxidant phenolic and flavonoid compounds, such as ellagic acid, vanillin, luteolin, kaempferol, myricetin, isorhamnetin, and (+)-gallocatechin, was observed after fermentation. Thus, these results indicated that the fermentation broth of T. fuciformis and A. trifoliatus had significant antioxidant activity, and may have potential application for health products such as functional beverages, cosmetics, and pharmaceutical raw materials.

Edible fungi by-products are rich in dietary fiber (DF). In this study, we used steam explosion (SE) to modify Tremella fuciformis ( T. fuciformis ) stem DF. The SE conditions were optimized using response surface methodology (RSM), and the soluble dietary fiber (SDF) extraction rate increased 1.42-fold (from 23.33 ± 0.42 % to 33.21 ± 0.28 %) under optimized conditions. SE destroyed the dense structure of SDF, which improved the specific surface area and thermal stability. Furthermore, the structural changes induced by SE resulted in improved functional properties, and SDF had better hydration properties (water holding capacity, oil holding capacity, and swelling capacity increased by 1.23, 1.59, and 1.24 times, respectively) and hypoglycemic capacity (glucose adsorption capacity increased 1.84-fold at 100 mmol/L glucose). Therefore, SE is an excellent modification method for improving quality of edible fungi processing by-products SDF.
