A multicolor electrochemiluminescence (ECL) device was constructed based on closed bipolar electrode (BPE) for visualized sensing of Salmonella typhimurium ( S. typhimurium). Since the emission color of concomitant electrochemiluminophores ([Ir(ppy) 3 ] and [Ru(bpy) 3 ] 2+ ) is potentially distinguish, selective excitation of ECL could be obtained by regulating the interfacial potential at the poles of BPE. Via modulating the resistance of BPE, bidirectional color change from dark-orange to yellow to forest-green could be observed at the anode, which was ascribed to the dual-potential excitation property of the concomitant electrochemiluminophores. The presence of S. typhimurium on the cathode of BPE increases the resistance in the circuit, generating ECL reporting color at the other pole switched. In accordance with this multicolor BPE-ECL sensor, a low detection limit of 10 CFU mL −1 of S. typhimurium could be identified in 25 mL (g) sample with naked eyes. In this work, physical isolation of the cathode and anode of closed BPE not only avoids target and electrochemiluminophores mutual interference , but also the surface of the BPE need nothing modified, just added immunomagnetic beads (IMBs)- S. typhimurium compound to the cathode. Moreover, without complex pre-enrichment bacterial, the whole experiment time also can be greatly shortened with only 0.5 h. On the other hand, the electrochemical signal is converted into a more intuitive optical readout for visualized sensing of S. typhimurium in food. The multicolor ECL device is merging the sensitivity of electrochemistry with the intuitive of an optical readout, which may apply in the site of rapid detection of food safety.

A bipolar electrode electrochemiluminescence (BPE-ECL) with β-cyclodextrin (β-CD) host–guest recognition was constructed for the rapid detection of Salmonella typhimurium ( S. typhimurium). Firstly, the sulfhydryl β-CD (SH-β-CD, with inner diameter size: 6.0–6.5 Å) was assembled on the gold-plated cathode of BPE. Ferrocene (Fc, length: 3.5 Å; width: 4.4 Å) labelled signal probe and parts Fc-aptamer complement each other to form a double-stranded DNA (dsDNA). Due to its large size, the dsDNA with a double Fc (width > 8.8 Å) could not be embedded into the cavities of β-CD. In the presence of S. typhimurium, Fc-aptamer bond specifically to S. typhimurium, and the signal probe was shed from the dsDNA. Then, the signal probe (guest) was captured by the SH-β-CD (host) on the surface of the BPE. Due to the principle of spatial matching between Fc and SH-β-CD, a stable inclusion complex could be formed, which made Fc close to the cathode surface of the BPE. When a certain driving voltage was applied, Fc was oxidized to Fc + . The cathode surface of Fc + would promote electron transfer, causing the [Ru(bpy) 3 ] 2+ at the anode emit light, and effectively convert the electrochemical signal into an ECL signal. 10 1 -10 5 CFU mL −1 S. typhimurium was positively correlated with Fc + and ECL intensity, with a detection limit of 10 1 CFU mL −1 . Therefore, S. typhimurium can be quantitatively detected by the electron transfer rate of the BPE.

A bipolar electrode-electrochemiluminescence (BPE-ECL) sensor, utilizing a polydopamine (PDA) bacterial surface-imprinted polymer (SIP), has been developed for the detection of Salmonella typhimurium ( S. typhimurium). To fabricate this sensor, dopamine (DA) served as the monomer, while S. typhimurium played a crucial role as the template molecule. Through a precision-tuned electropolymerization process, a polymer film template was seamlessly crafted on the cathode of BPE, encasing the target bacteria within its matrix. Once the template was delicately removed, a molecular surface-imprinted film emerged on the cathode, forming a PDA SIP capture probe that was remarkably adept at recognizing and binding to the target bacteria. When the target bacteria was present on the cathode, the Fc-aptamer- S. typhimurium acted as a signal probe to specifically recognized S. typhimurium. Applying a constant potential to the BPE, the oxidation product Fc + , generated at the cathode, accelerating the luminescence of the [Ru(bpy) 3 ] 2+ /TPrA system on the anode. The sensor exhibited a remarkable sensitivity, capable of detecting S. typhimurium concentrations ranging from 10 1 to 10 6 CFU mL −1 , with a detection limit of 10 CFU mL −1 .

An ultrasensitive electrochemiluminescence (ECL) biosensor was proposed based on a closed bipolar electrode (BPE) for the detection of alkaline phosphatase (ALP). For most of the BPE–ECL biosensors, an effective signal amplification strategy was the key to enhance the sensitivity of the system. Herein, the signal amplification strategy of the enzyme catalysis was utilized in the BPE–ECL system. Au nanoparticles (NPs) were electrodeposited on the cathode surface of the ITO electrode to improve the stability and sensitivity of the signal. Compared with the previous BPE–ECL biosensors, the sensitivity was increased by at least 3 orders of magnitude. The biosensor showed high sensitivity and specificity of ALP detection with a detection limit of as low as 3.7 aM. Besides, it was further applied to the detection of ALP in different types of cells and successfully realized ALP detection in single Hep G2 cell, which had a huge application prospect in single biomolecule detection or single cell analysis.

In this work, we developed an innovative closed bipolar electrode (BPE)-electrochemiluminescence (ECL) sensing strategy with generality for target detection. Based on charge balance and 100% current efficiency between the closed BPE poles and the driving electrodes, one of the driving electrodes in one cell of the closed BPE system was employed as ECL sensing surface to reflect the target on the BPE pole in the opposite cell. Compared with traditional BPE-ECL sensing method, which in general adopted the anodic ECL reagents such as Ru(bpy)32+ and its coreactant on one pole (anode) to reflect the target (occurring reduction reaction) on the other pole (cathode), the difference was that the targets occurring oxidation reaction could be detected by the anodic ECL reagents based on this strategy. To verify the feasibility of this strategy, the detection principle was stated first, and Fe(CN)64– as model target at anodic BPE pole were detected by anodic ECL reagents (Ru(bpy)32+ and TprA) on the driving electrode first. The ECL signals showed good performance for target detection. By changing the size and the material of the BPE pole where the targets were located, the detection of l-ascorbic acid (AA), uric acid (UA), and dopamine (DA) as other model targets with higher detection limit were accomplished. Visual and high-throughput detection of AA, UA, and DA were also successfully realized by an array of the closed BPE system. This closed BPE (array) system is an effective supplement of traditional BPE-ECL sensing and could greatly expand the scope of the detection target.

Pseudomonas aeruginosa ( P. aeruginosa ) is an opportunistic infection-causing pathogen that threatens human health. Accordingly, a rapid and ultrasensitive analytical method is required urgently. Herein, an ultrasensitive multicolor electrochromic sensing platform was established on the basis of a closed bipolar electrode (BPE). Prussian blue (PB), the blue spot that can be easily electrodeposited, was selected as an electrochromic indicator at the closed BPE cathode. Integrating with the anodic emitter, Ru(bpy) 3 2+ , which emitted optical red light, visualized multicolor electrochromism was achieved on closed BPE. Particularly, physical separation between the positive and negative poles of the closed BPE greatly prevented mutual interference between Ru(bpy) 3 2+ and PB. Consequently, the sensitivity and accuracy of the proposed biosensor considerably improved. Notably, owing to magnetic-separation technology, the closed BPE surface required no modification. Without any complex pretreatment, the entire experiment time could be greatly shortened because the [email protected] completely captured P. aeruginosa in food matrix within only 20 min. By comparing the visual electrochromic colors, ultrasensitive screening of P. aeruginosa was accomplished within 1–10 8 CFU mL −1 . Combining the merits of closed BPE, electrochemiluminescence (ECL), and electrochromic, this strategy provided an accurate and intrinsic way for visual detection of P. aeruginosa , as well as great potential in measuring other pathogens.

In this study, an energy-efficient divided bipolar electrolysis system was developed for water softening, where two PTFE membranes were used as the separating materials and a bipolar electrode was employed to enhance the H 2 O-splitting reactions. As compared with other two operation modes, the optimum calcium harness removal efficiencies of 85% and 57% could be reached in the induction cathode effluent and terminal effluent, respectively, at 8 mA cm −2 in the mode A. Increasing the current density from 5 to 20 mA cm −2 evidently promoted the removal of calcium hardness from 33% to 65% in the terminal effluent and the CaCO 3 precipitation rate from 743 to 1462 gCaCO 3 h −1 m −2 with the increased energy consumption from 0.53 to 2.2 kWh kg −1 CaCO 3 . The optimized Ca 2+ /HCO 3 − molar ratio was 1:1.2 for the calcium hardness removal. In addition, increasing the flow rate into each cathode chamber from 10 to 40 mL min −1 gradually decreased from 67% to 35%. The calcium hardness was mainly removed in the forms of vaterite and calcite in the alkaline effluents and was marginally precipitated as aragonite and calcite on the cathodes surface. Generally, present energy-efficient electrochemical water softening system showed great potential for application in industrial processes.

Chiral molecules are abundant in nature. Phenylketonuria (PKU) is caused by the abnormal transformation of chiral molecules L-phenylalanine (L-Phe) in the human blood , which can cause irreversible harm to the human body. In this work, we documented an electrochemiluminescent (ECL) dual-mode sensor platform based on molecularly imprinted polymer (MIP) modified closed bipolar electrodes for high sensitivity detection of L-Phe and D-phenylalanine (D-Phe). In the anode chamber of a bipolar electrode modified with phenylalanine imprinting, Ru (bpy) 3 2+ underwent a redox reaction to produce a chemiluminescence response under the stimulation of a driving voltage . At the same time, the reduction of the cathode film of the bipolar electrode was promoted, and the color changed from dark blue to nearly white. Thus, the dual-mode detection of target molecules is realized. The detection range of the sensor for phenylalanine reached 0.01–10,000 nM, and the detection limits of L-Phe and D-Phe were 3.9 pM and 4.6 pM (S/N = 3), respectively. This dual-mode system achieved high stability and high specificity, and also successfully realized the detection of actual samples, which is expected to achieve future clinical applications.

Herein, a novel bipolar electrode-electrochemiluminescence (BPE-ECL) biosensing strategy was developed, engineered to achieve highly and pathogen-specific detection of Escherichia coli O157:H7 ( E. coli O157:H7) through enzymatic cascade signal transduction. Crucially, this strategy employs β-galactosidase (β-gal) activity as a biomarker signature, wherein the enzymatic hydrolysis of p -nitrophenyl-β-D-galactopyranoside (PNPG) generates electroactive intermediates, p-nitrophenol (pNP). The pNP undergoes a six-electron reduction reaction at the closed BPE cathode electrode and simultaneously oxidizes phosphorus Ru(bpy) 3 2+ at the anode electrode to trigger obvious ECL signal. The ECL signal amplification of enzymatic substrates with mono-, bi-, and tetra-electronic systems is inherently constrained by inefficient electron transfer, resulting in suboptimal sensing performance. In contrast, the PNPG/pNP-based detection platform demonstrates superior sensing capabilities. For indirect quantification of E. coli O157:H7, the ECL intensity was linearly correlated with bacterial concentration. The optimized biosensor, through rigorous tuning of enzymatic catalysis and ECL signal amplification strategies, delivers a wide dynamic range (10(Zhang et al., 2023a 1) –10 6 CFU) and a low detection limit (LOD = 2 CFU) in 200 μL of bacterial samples. The real sample effect was evaluated, and the assay successfully detected E. coli O157:H7 in water, juice, and pure milk. This label-free platform integrates operational simplicity and high specificity, offering a promising solution for on-site monitoring of foodborne pathogens and clinical diagnostics.

Escherichia coli O157:H7 ( E. coli O157:H7) is a foodborne pathogen with high pathogenicity and lethality, capable of producing substantial amounts of Vero toxin (VT), posing a significant threat to human health. Consequently, a platform based on the bipolar electrode-electrochemiluminescence (BPE-ECL) and loop-mediated isothermal amplification (LAMP) was constructed for the detection of E. coli O157:H7. Firstly, LAMP technology is utilized to efficiently obtain the amplified product (double-stranded DNA, dsDNA) of E. coli O157:H7. On the one hand, SYBR Green was added to the amplified product to detect E. coli O157:H7 by colorimetric determination. The concentration of 10 CFU mL −1 E. coli O157:H7 can only obtain qualitative analysis, to determine the presence of the target bacteria. On the other hand, methylene blue (MB) molecules were embedded in the dsDNA through electrostatic interactions. Then, by dropwise adding the mixture onto the cathode of the BPE, the ECL signal of Ru(bpy) 3 2+ onto the anode was generated. Specifically, the BPE-ECL sensor displayed superior sensitivity for E. coli O157:H7 with a detection limit of 10 CFU mL −1 and a wide linear concentration range of 10 1 –10 6 CFU mL −1 . In summary, this sensor can effectively enhance the sensitivity and reliability of detection when compared to traditional colorimetric methods, and achieve qualitative and quantitative detection of E. coli O157:H7 by measuring the anode ECL signal of Ru(bpy) 3 2+ .

A simple, efficient and versatile biosensing platform capable of the multiplexed detection for glucose, lactate and choline was developed by the integration of bipolar electrochemistry and electrochemiluminescence (ECL) imaging. The sensing bipolar electrodes (BPEs) were simply modified via a one-step method adaptable to different enzymes. The biorecognition event happening between the substrate and the corresponding enzyme could be directly reported by the ECL emitted on the same pole from luminol and in situ generated H 2 O 2 . Under optimized conditions, the BPEs array was successfully applied for the determination of glucose, lactate and choline in the ranges of 0.01–1 mM, 0.01–1 mM and 0.02–5 mM, with the LOD of 7.57 μM, 8.25 μM and 43.19 μM, respectively. Owing to the improved stability of in situ generated H 2 O 2 , a whole series of analytes testing could be completed in the same BPE biochip. Subsequently, an array chip consisting of nine BPEs enabled the concomitant detection of glucose, lactate and choline, demonstrating the capability for multifunctional detection of biomolecules. This versatile analytical system could be easily extended to sensitive screening in a miniaturized device and point of care testing.

Sensitive and accurate detection of tumor markers is crucial for disease diagnosis, yet numerous challenges still exist due to limitations of single-mode assays. This work constructed a universal dual-mode sensing platform combining closed bipolar electrode (cBPE) and split-type immunoassay, which realized simultaneous electrochemiluminescence (ECL) and electrochromic detection of proteins upon a single potential stimulus. Specifically, on the cBPE, L012 served as the ECL emitter at the anode, while Prussian blue (PB) film at the cathode acted as the electrochromic sensing unit. Applying a single potential stimulus could trigger simultaneous ECL emission and the color change from blue PB to white Prussian white. By virtue of MgO 2 @GOx-Ab 2 signal probes as bioprobe, a split-type immunoassay was developed for prostate-specific antigen (PSA) detection. The target-induced production of H 2 O 2 from cascade glucose oxidation and MgO 2 hydrolysis greatly enhanced the anodic ECL signal and facilitated the cathodic PB reduction. Consequently, the strategy achieved the highly sensitive and visual dual-mode assay for PSA, yielding the limits of detection of 2.7 × 10 −12 g/mL for ECL detection and 3.1 × 10 −9 g/mL for electrochromic sensing, respectively. The study opened up an avenue for developing simultaneous multi-mode sensing platforms toward more biomarkers.
