Degradation of chloramphenicol in soil with catechin-enhanced ferric iron/persulfate system

Tian Qiu,Linyi Li,Wenbo Dong,Gilles Mailhot,Yanlin Wu,Xu Zeng

Chloramphenicol (CAP) is a broad-spectrum antibiotic that has been commonly used in the world for the past few decades because of its high antibacterial activity. Although CAP has been banned, high concentration is still detected in the soil environment because of its low self-degradation. In the present research, the polluted soil was treated in situ in a sulfate radical (SO 4 •− ) based process. CAP was degraded by persulfate (PS) activated with Fe(III) and catechin (CAT). Fe(II) could be produced efficiently via complexation and reduction through Fe(III) and CAT reaction to enhance the PS activation efficiency. The optimal dosages of PS, CAT and Fe(III) were investigated. The degradation rate of CAP could reach 94.3% after 120 min of reaction. The contribution of free radicals on CAP degradation was also detected, and the results showed that SO 4 •− and HO • resulted in pollutant removal of 83.8% and 16.2%, respectively. The reaction between CAT and Fe(III) mainly occurred on the surface of soil particles because of the strong adsorption of CAT. However, the oxidation process between radicals and CAP mainly occurred in the liquid phase. Moreover, the biological toxicity of the reaction system and the total organic carbon of organic matter were also significantly reduced.

Regiospecific methylation of all the hydroxyls in (+)-catechin by a stepwise differentiation strategy

Shan Hong,Shanshan Wang,He Cai,Songbai Liu

BACKGROUND Methylated derivatives of catechins have received great attention for health beneficial effects, especially antiallergic activity. However, the scarce natural abundance of methylated catechins limits further bioactive studies. The objective of this work was to investigate regiospecific methylation of the hydroxyls of (+)-catechin through a stepwise differentiation strategy based on electronic difference between the hydroxyl groups. RESULTS Selective methylation of the hydroxyls on different rings was realized by employing Meerwein salt as the methylation reagent. Preferential acylation of the phenolic hydroxyls on A and B rings allowed selective exposure of the aliphatic hydroxyl on C ring to methylation. The vicinal phenolic hydroxyls on B ring were preferentially methylated under mild basic condition due to the acidic properties. Methylation of the phenolic hydroxyls on A ring was achieved by sequential protection and deprotection operations. Finally, antioxidant activities of all the individual methylated (+)-catechins were explored by 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) assay. CONCLUSION Regiospecific methylation of the phenolic and aliphatic hydroxyls was systematically achieved under mild conditions. Preparation of all the individual methylated (+)-catechins was accomplished with a greener methylation reagent: nonvolatile Meerwein salt. This work laid a solid foundation for preparation of diverse O -methylated catechins for bioactivity studies. © 2019 Society of Chemical Industry

Preparation, Characterization, and Performance Evaluation of Catechin-β-Cyclodextrin Oleate Nanoparticle Inclusion Complexes

Yongfu Li,Ju Qiu,Zhuxi Tian,Yaoqi Ran,Xi Chen,Mingxiu Long,Jing Wen

Catechins are polyphenolic antioxidants, widely distributed in tea and cocoa. They exhibit significant biological activities, including anti-inflammatory, antibacterial, and anticancer effects. However, they are characterized by poor water solubility, bitter taste, and sensitivity to light, thus limiting their application. In this study, the stability and bioavailability of CATs were enhanced by encapsulating them into β-cyclodextrin oleate nanoparticles through a co-precipitation method. The resulting CAT-CDO inclusion complexes were characterized using Fourier transform infrared spectroscopy, differential scanning calorimetry, thermogravimetric analysis, SEM and TEM. The encapsulation efficiency, drug loading capacity, adsorption thermodynamics, antioxidant activity, release behavior and cytotoxicity were systematically investigated. The results showed that CATs were successfully encapsulated within the hydrophobic cavity of CDO, forming spherical nanoparticles with an average size of approximately 175 nm. The encapsulation efficiency and drug loading rate were 81.43% and 27.14%, respectively. The water solubility was 13.39 times higher than that of free CAT. The inclusion complexes enhanced antioxidant activity against DPPH, OH·, and NO radicals, improved thermal and photostability, and provided a sustained release profile. Furthermore, cytotoxicity assays conducted on Caco-2 cells revealed reduced toxicity and potential proliferative effects, indicating that CDO is a promising carrier for improving the therapeutic and functional applications of CATs.

Isolation of (+)-Catechin from Food Waste Using Ionic Liquid-Modified ZIF67 Covered Silica

Mengshuai Liu,Xiaoman Li,Mengmeng Zhao,Xuyang Jiu,Chuang Yao,Minglei Tian

Background: Food waste contains abundant (+)-catechin, but its efficient recovery remains challenging. This study aimed to prepare ionic liquid (IL)-modified sorbents and establish an efficient method for (+)-catechin recovery from chocolate waste via solid-phase extraction (SPE). Methods: Three series of IL-modified sorbents (Sil-IL, ZIF67-IL, Sil@ZIF67-IL) were synthesized. Their adsorption performance was evaluated under different conditions; adsorption isotherm and kinetic data were fitted to Langmuir/Freundlich and pseudo-first/second-order models, respectively. Sorbent stability and (+)-catechin recovery from chocolate waste extracts were tested. Results: Sil@ZIF67-Hmim showed the highest adsorption capacity (154.4 mg/g) at 25 °C within 120 min. Adsorption followed the Langmuir model (R2= 0.99), indicating chemisorption. Sil@ZIF67-Hmim was subjected to repeated solid-phase extraction (SPE) for five consecutive days; the recovery rate ranged from 98.1% to 99.2%, and the relative standard deviation (RSD) was 3.2–4.4%. Conclusions: Sil@ZIF67-Hmim is a high-efficiency sorbent for (+)-catechin recovery from chocolate waste, providing a novel approach for food waste valorization and highlighting the application potential of IL-modified MOF-silica composites.

MXene/MWCNTs-COOH/MOF-808-based electrochemical sensor for the detection of Catechin

Jing Chen,Wenli Xue,Yaqin Shi,Wenjing Liu,Rongfang Ma

Catechin is a natural flavonoid with antioxidant, anticarcinogenic and anti-inflammatory properties. They can be used to treat or prevent human health problems. However, excessive intake of catechin can also be harmful to the human body. It is crucial to find a method for quantitative detection of catechin. The carboxyl-functionalized multi-walled carbon nanotubes were intercalated into the MXene layers to form a composite material with excellent electrical conductivity, and then MOF-808 with adsorption sites was further loaded on MXene/MWCNTs-COOH to synthesize MXene/MWCNTs-COOH/MOF-808 with excellent catalytic properties. Based on this, an electrochemical sensor was constructed for the detection of catechin. The CA sensor constructed in this work exhibited excellent analytical performance, good linear range (0.5–21.53 μM) and low detection limit (0.05 μM). It was also used for the detection of CA in green tea and beverages, which exhibited good recoveries (99.46–99.71 % and 96.46 % ~ 101.33 %) and the relative standard deviations (1.33–2.55 % and 0.07 % ~ 1.57 %).

Improving Fenton-like system with Catechin, an environmental-friendly polyphenol: Effects and mechanism

Zhongyi Fang,Jie Zhao,Yuan Li,Yu Wang,Tian Qiu,Yanlin Wu,Wenbo Dong,Gilles Mailhot

Fenton-like system had long been modified to promote its adaptability and efficiency by adding extra components like metal chelators or reducers. Nonetheless, chelator such as ethylene diamine tetraacetic acid (EDTA) was harmful in high dosage. While strong reducers could efficiently promote the generation speed of hydroxyl radicals (HO ), they could also get oxidized easily during storage time, for which further application was costly. In this study, (+)-catechin (CAT), an environmental- friendly component, was proved to be an efficient promoter of Fenton-like system on Bisphenol A (BPA) degradation. The modified system achieved 92% of BPA removal in one hour. The mechanism was studied, and the reaction procedure between CAT and Fe(III) together with the degradation pathways of CAT were identified. Fe(III) got chelated first by CAT and quickly reduced to Fe(II) via a single inner-compound electron transfer. The intermediates of CAT could further reduce one and a half more potion of Fe(III) on average. Consequently, massive amount of Fe(II) turned Fenton-like procedure to Fenton reaction and thus promoted the generation of HO . The main radicals contributed on BPA degradation was HO . BPA could be efficiently degraded at pH 6.5 or acidic pH range, but the degradation was completely inhibited with the pH higher than 8.0. The optimal mole ratio between Fe(III) and CAT was 6:1 and the degradation pathway of BPA was deduced.

Antidepressant potential of Uncaria rhynchophylla and its active flavanol, catechin, targeting melatonin receptors

Chang-An Geng,Tong-Hua Yang,Xiao-Yan Huang,Yun-Bao Ma,Xue-Mei Zhang,Ji-Jun Chen

Ethnopharmacological relevance Traditional Chinese medicines (TCMs) are fascinating sources for natural drug candidates. Uncaria rhynchophylla (Gouteng) is a famous TCM used for alleviating central nervous system (CNS) disorders, while its antidepressant constituents are still disputed. Aim of the study The present study was designed to assess the antidepressant property of U. rhynchophylla and characterize the active constituents targeting melatonin receptors which are closely related to CNS diseases. Materials and methods The total extract and each fraction of U. rhynchophylla were extensively assessed for their agonistic activity on melatonin receptors in vitro . The following bioassay-guided fractionation yielded the active constituents, whose activity was confirmed by dose-dependent bioassay and antagonistic experiment on HEK293 cells. Their antidepressant effects were evaluated on forced swimming test (FST), tail suspension test (TST) and open-field test (OFT) mice models in vivo . Their metabolic profiles in mice plasma were analyzed by LCMS-IT-TOF. Results The stems and hooks of U. rhynchophylla were revealed with agonistic activity on melatonin receptors (MT 1 and MT 2 ). Under the guidance of bioassay, two flavanols , catechin and epicatechin were obtained and showed obviously activity agitating MT 1 (EC 50 = 25.8 and 156.1 μM) and MT 2 (EC 50 = 47.3 and 208.8 μM) receptors. The agonistic activity of catechin on melatonin receptors can be antagonized by luzindole at the concentrations of 1.57–100 μM. Catechin could significantly reduce the immobility time in both FST and TST mice models at doses of 80 and 40 mg/kg, without obvious effect on locomotor activity in OFT mice model. Five phase II (M1-M5) and one phase I (M6) metabolites of catechin were detected in mice plasma after intragastric ( i.g .) administration. Conclusion Catechin is a potent antidepressant candidate from U. rhynchophylla by targeting melatonin receptors. The main metabolic pathways of catechin in mice plasma are glucuronidation (M3) and methylated glucuronidation (M4 and M5). This study provides valuable information for understanding the antidepressant potency of Gouteng and its active constituents.

Effect of catechin liposomes on the nitrosamines and quality of traditional Chinese bacon

Jieyu Wu,Rongfa Guan,Haizhi Huang,Zhenfeng Liu,Haitao Shen,Qile Xia

The instability of catechins (CT) is currently a challenge for their application in the food industry. This study explored whether or not the protective effects of liposome encapsulation on CT could enhance the role of CT as antioxidants in reducing nitrosamines (NAs) in traditional Chinese bacon (TCB). The samples were further characterized by the analysis of volatile compounds, color parameters, and texture profile during storage to assess the effects of catechin liposomes (CTL) on TCB. Results showed that the increase in NAs in fried TCB was strongly dependent on the storage time. CTL could reduce the production of NAs in fried TCB (40.45%) more strongly than CT (15.13%) after 49 days of storage. Higher phenol contents were found in CTL (1515.25 × 106 area unit (AU)) than those in control (1451.73 × 106 AU) and CT (1391.10 × 106 AU) TCB at the end of storage. The addition of antioxidants increased (p < 0.05) a* values in the following order: CTL > CT > control with mean values of 6.54, 5.09, and 3.92, respectively, after 49-day storage. Conversely, the addition of CTL decreased (p < 0.05) the hardness of TCB. Encapsulating CT in the liposomes can effectively reduce the NAs in TCB and positively affect its quality. This technique may guide the potential application of liposomes in the meat industry.

Self-assembled metal-phenolic nanocomplexes comprised of green tea catechin for tumor-specific ferroptosis

Min Wang,Aoling Yu,Wen Han,Jingyi Chen,Chunhua Lu,Xiankun Tu

Ferroptosis, a newly discovered form of regulated cell death, has garnered significant attention in the field of tumor therapy. However, the presence of overexpressed glutathione (GSH) and insufficient levels of H 2 O 2 in the tumor microenvironment (TME) hinders the occurrence of ferroptosis. In response to these challenges, here we have constructed the self-assembled nanocomplexes (FeE NPs) utilizing epigallocatechin-3-gallate (EGCG) from green tea polyphenols and metal ions (Fe 3+ ) as components. After grafting PEG, the nanocomplexes (FeE@PEG NPs) exhibit good biocompatibility and synergistically enhanced tumor-inhibitory properties. FeE@PEG NPs can be disassembled by H 2 O 2 in the TME, leading to the rapid release of Fe 3+ and EGCG. The released Fe 3+ produces large amounts of toxic •OH by the Fenton reactions while having minimal impact on normal cells. The generated •OH effectively induces lipid peroxidation, which leads to ferroptosis in tumor cells. Meanwhile, the released EGCG can autoxidize to produce H 2 O 2 , which further promotes the production of •OH radicals and increases lipid peroxide levels. Moreover, EGCG also depletes the high levels of intracellular GSH, leading to an intracellular redox imbalance and triggering ferroptosis. This study provides new insights into advancing anticancer ferroptosis through rational material design, offering promising avenues for future research.

Thermal activation in oxidative model drives catechin assembly into theaflavins with enhanced antioxidant capacity

Yuxuan Zhao,Jingyimei Liang,Rui Lu,Zebin Zou,Haoming Wu,Shengxiong Chen,Hui Cao,Jianbo Xiao

Theaflavins are important bioactive pigments in tea, but their non-enzymatic formation pathways under heating remain incompletely understood. This study investigated the temperature-dependent formation of four theaflavins using aqueous catechin model systems. Potassium ferricyanide was used as a controlled oxidant to initiate catechin oxidation and subsequent coupling reactions. Liquid chromatography-mass spectrometry (LC-MS), ultraviolet-visible spectroscopy (UV–Vis), DPPH and ABTS assays were applied to track product profiles and antioxidant capacity. Moderate temperatures promoted catechin oxidation to ortho-quinones and favored the accumulation of theaflavins, leading to higher antioxidant activity. Low temperatures limited conversion, whereas high temperatures accelerated precursor consumption and theaflavin degradation. Across all model systems, theaflavin levels peaked at 30 min and then declined with prolonged heating. Among the four products, TF3 showed the strongest thermoresponsive and the highest antioxidant capacity. These findings reveal a thermally activated catechin self-assembly mechanism in an oxidant-assisted model system.

Sustainable Production of Cyanidin-3-O-galactoside by Metabolic Engineered Escherichia coli from Catechin

Zhen Zong,Lianghua Xie,Jiaqi Fu,Zhongyang Liu,Wen-Wen Zhou,Wei Chen

Cyanidin-3-O-galactoside (C3Ga), a natural pigment, has various beneficial biological activities and is widely used as a food colorant. However, traditional plant extraction methods are time-consuming and unsustainable. Rapid and sustainable synthesis of C3Ga by engineered microorganisms offers a promising alternative to traditional plant-based methods and deserves to be explored. In this study, the bioproduction of C3Ga by Escherichia coli was achieved for the first time. The biosynthetic pathway of C3Ga from (+)-catechin was constructed by introducing anthocyanidin synthase (ANS) and UDP-galactose:cyanidin galactosyltransferase. Some strategies, including enhancement of the UDP-galactose biosynthesis pathway, identification of efficient ANS, overexpression of the C3Ga transporter, and modulation of multigene expression, were subsequently used to drive the metabolic flux toward C3Ga production. Next, the two-stage process for C3Ga production was optimized to mitigate limitations for further metabolic engineering. Combined with the knockout of β-phosphoglucomutase (ycjU), a newly identified competitive pathway for UDP-galactose, the production of C3Ga finally reached 217.9 mg/L. The strategies used in this study could be applied to the biosynthesis of other anthocyanins and galactosylated natural products.
