Salecan is a novel linear extracellular polysaccharide with a linear backbone of 1–3-linked glucopyranosyl units. Salecan is suitable for preparing hydrogels for biomedical applications due to its prominent physicochemical and biological profiles. In this contribution, a variety of innovative semi-interpenetrating polymer network (semi-IPN) hydrogels consisting of Salecan and poly(methacrylic acid) (PMAA) were developed via free radical polymerization for controlled drug delivery. The successful fabrication of the semi-IPNs was verified by Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and thermogravimetric (TGA) measurements. Scanning electron microscopy (SEM) and rheology analyses demonstrated that the morphological and mechanical behaviors of the resultant hydrogels were strongly affected by the contents of Salecan and cross-linker N,N′-methylenebis(acrylamide) (BIS). Moreover, the swelling properties of these hydrogels were systematically investigated, and the results indicated that they exhibited pH sensitivity. The drug delivery applications of such fabricated hydrogels were further evaluated from which doxorubicin (Dox) was chosen as a model drug for in vitro release and cell viability studies. It was found that the Dox release from the Dox-loaded hydrogels was significantly accelerated when the pH of the release media decreased from 7.4 to 5.0. Toxicity assays confirmed that the blank hydrogels had negligible toxicity to normal cells, whereas the Dox-loaded hydrogels remained high in cytotoxicity for A549 and HepG2 cancer cells. All of these attributes implied that the new proposed semi-IPNs serve as potential drug delivery platforms for cancer therapy.

Poly(N,N-diethylacrylamide) is not only a thermosensitive polymer, but also a good hydrogen bond acceptor. Therefore, drugs with carboxyl groups can serve as hydrogen bond donors and form interactions with the tertiary amide groups in N,N-diethylacrylamide. Herein, we report a novel drug delivery system for anionic drugs composed of poly(N,N-diethylacrylamide) and salecan. Salecan was used to improve the hydrophilicity and accelerate the responsive rate of this system. As expected, salecan-enriched hydrogels exhibited higher swelling ratios and were more sensitive to temperature. Moreover, scanning electron microscopy images showed that the hydrogels are superporous structures, with pore-sizes that increase with salecan concentration. The swelling ratios decreased continuously with the increase of temperature in the range 25–37 °C. MTT assay for cell viability and cell adhesion studies confirm the cell compatibility of the system. Delivery tests using diclofenac sodium, an anti-inflammatory drug, indicate that the thermosensitive property of this system is favorable for anionic drug delivery. Interestingly, the release rates of diclofenac sodium from the hydrogels were temperature dependent, with higher temperatures contributing toward faster release rate.

This study investigated the preparation, characterization, and capacity of polydopamine-modified salecan hydrogel (PMSH) to remove antibiotics from aqueous solutions. The incorporated polydopamine enabled the physical properties of PMSH to be regulated and enhanced the antibiotic adsorption capacity. The antibiotics adsorption by PMSH3 was relatively fast, reaching equilibrium within 10 h with the maximum adsorption capacity of 134.85, 134.65, and 82.24 mg/g for chlorhexidine acetate, minocycline, and erythrocin, respectively. The pseudo-second-order model exhibited the best kinetic fitting data, suggesting that all the PMSH3 adsorption sites had an equal affinity for the antibiotics and that the adsorption process was chemical. The Freundlich model best described the isotherm adsorption of antibiotics, and adsorption on PMSH3 was multilayer adsorption. The antibiotics were adsorbed on the PMSH3 through hydrogen bonding, electrostatic and π-π interactions based on the changes observed in the FTIR spectra and zeta potential of PMSH3 after adsorption. Moreover, it is found that PMSH3 possesses superior recyclability and biocompatibility. Also, antibiotics regenerated by PMSH3 showed high antimicrobial properties toward Staphylococcus aureus and Escherichia coli . Altogether, our data demonstrate that the proposed strategy has the potential as an antibiotic adsorbent for sustained wastewater treatment.

Salecan is a water-soluble microbial polysaccharide produced by Agrobacterium sp. ZX09, a salt-tolerant strain isolated from a soil sample in our laboratory. Previous work inspired us salecan is a good candidate to fabricate hydrogels. Poly( N , N -diethylacrylamide) is one type of thermo sensitive polymer which is not investigated extremely as poly( N -isopropylacrylamide). Here, we report a novel pH-thermo dual responsive hydrogel based on salecan and poly( N , N -diethylacrylamide-co-methacrylic acid) semi-interpenetrating polymer networks (semi-IPNs). The physicochemical property of this hydrogel was investigated by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR) and thermogravimetric analyses (TGA), rheological test and Scanning Electron Microscopy (SEM). It was interesting that the storage modulus (G′) and pore size of the hydrogel could be tuned by adjusting the content of salecan and crosslinker. The pH-thermo dual responsive property was demonstrated by swelling behavior test: the swelling ratio of the hydrogel decreased continuously as the temperature increased from 25 °C to 37 °C, while it was pH-dependent as well. Especially, when exposed to a higher temperature (37 °C) and acidic environment (pH 4.0), drug-loaded hydrogel would have a quick release. Finally, the cytotoxicity of drug-free hydrogels was investigated on A549 and HepG2 cells, results showed that it was non-toxic while the DOX released from hydrogels had comparable cytotoxicity with respect to free DOX. In conclusion, the novel salecan/poly( N , N -diethylacrylamide-co-methacrylic acid) semi-interpenetrating polymer network hydrogels were pH-thermo dual responsive and may be a promising candidate for drug delivery system.

Salecan is a novel microbial polysaccharide produced by Agrobacterium sp. ZX09. The salt-tolerant strain was isolated from a soil sample in our laboratory and the 16S rDNA sequence was deposited in the GenBank database under the accession number GU810841 . Salecan is suitable to fabricate hydrogel for biomedical applications due to the excellent hydrophilicity and biocompatibility. Here, salecan has been introduced into poly(N-isopropylacrylamide) (PNIPAm) network to form novel thermo-sensitive semi-interpenetrating polymer networks (semi-IPNs). The structure of salecan/PNIPAm semi-IPNs was confirmed by Fourier transformation infrared spectroscopy (FTIR) and X-ray diffraction (XRD). Thermogravimetric analysis (TGA) proved the stability of the semi-IPNs. Rheological and compressive tests revealed an elastic solid-like behavior and good mechanical properties of the hydrogels. Swelling behavior test showed the hydrogels possessed high water content at room temperature. An excellent thermo-sensitive property of fast response rates to temperature had been demonstrated as well. In vitro degradation measurements ensured the semi-IPNs were degradable. Cytotoxicity and cell adhesion study suggested the synthesized salecan/PNIPAm hydrogels were non-toxic and biocompatibility. The results indicated the novel thermo-responsive hydrogels could be a suitable candidate for biomedical applications.

Achieving the controllable detachment of polysaccharide-based wound dressings is challenging. In this study, a novel, photodetachable salecan-based hydrogel dressing with injectable, self-healing, antibacterial, and wound healing properties was developed using a green and facile approach. A salecan hydrogel with a uniform porous structure and water content of 90.4 % was prepared by simply mixing salecan and an Fe 3+ –citric acid complexing solution in an acidic D-(+)-glucono-1,5-lactone environment. Metal coordinate interactions were formed between the released Fe 3+ ions and carboxyl groups on the salecan polysaccharide, inducing homogeneous gelation. Benefiting from this dynamic and reversible crosslinking, the salecan hydrogel exhibited self-healing and injectable behavior, facilitating the formation of the desired shapes in situ. The exposure of Fe 3+ –citric acid to UV light (365 nm) resulted in the reduction of Fe 3+ to Fe 2+ through photochemical reactions, enabling phototriggered detachment. Moreover, the hydrogel exhibited excellent biocompatibility and satisfactory antibacterial efficacy against Escherichia coli and Staphylococcus aureus of 72.5 % and 85.3 %, respectively. The adhesive strength of the salecan hydrogel to porcine skin was 1.06 ± 0.12 kPa. In vivo wound healing experiments further highlighted the advantages of the prepared hydrogel in alleviating the degree of wound inflammation and promoting tissue regeneration within 12 days.

Expanding the application of polysaccharide material has attracted tremendous attention in the fields of wastewater treatment, agriculture and biomedical engineering, on account of its tunable and unique properties. Herein, we employ a water-soluble, sustainable and low cost bacterial polysaccharide, salecan as a matrix, poly(acrylamide-co-itaconic acid) (PAI) as a synthetic component to synthesize salecan-g-PAI hydrogels through a simple chemical crosslinking method. Their physicochemical properties were fully characterized by various methods including Fourier transformed infrared spectroscopy, X-ray diffraction, thermogravimetry, scanning electron microscope and rheometry. We found that salecan not only acted as the interaction sites to regulate the water content of the developing hydrogels, but also endowed them with tailorable morphology. The designed salecan-g-PAI hydrogels exhibited excellent adsorption properties toward methylene blue (MB) dye, and the adsorption process could be well described by the pseudo-second-order kinetic and Freundlich isotherm models. Altogether, this study broadens the application of salecan polysaccharides and provides a new device for dye decontamination.

Polysaccharides are an important class of materials that are often exploited in the fields of food, agriculture, biomedical engineering and wastewater treatment owing to their unique and tunable properties. In this work, we utilize an inexpensive and sustainable extracellular polysaccharide salecan (EPS), which is produced by bacterium Agrobacterium sp. ZX09 , as a hydrogel matrix, poly(3-sulfopropyl methacrylate potassium salt) (PSM) as side chains to fabricate EPS-grafted-PSM adsorbents through a simple one-pot approach. Scanning electron microscope, X-ray diffraction, Fourier transformed infrared spectroscopy, rheometry and thermogravimetry were conducted to characterize the physicochemical properties of resultant adsorbents. We noticed that EPS not only served as the host chains of network to adjust the water uptake ability of adsorbents, but also endued them with tunable polarity. Further, the adsorption behaviors of developed adsorbents to copper ions (Cu 2+ ) were explored: these gels present high absorption ability for Cu 2+ through a chemical adsorption process which well described by Freundlich isotherm and pseudo-second-order kinetic models. In summary, the approach exhibited in this work opens a new avenue to design polysaccharide-based materials for Cu 2+ adsorption.

Salecan is a novel water-soluble extracellular β-glucan and has excellent physicochemical and biological properties. A series of hydrogels were fabricated from Salecan and poly(2-acrylamido-2-methylpropanesulfonic acid- co - N -hydroxymethyl acrylamide) (poly(AMPS- co -HMAA), PAH) by semi-IPN technique. The introduction of Salecan into the PAH network endows the system with enhanced cell adhesion properties. These semi-IPN hydrogels with open interconnected pores had pH and salt-sensitive swelling behavior. The increase in the content of hydrophilic Salecan could enhance the water uptake of the hydrogels. Rheological results indicated that the incorporation of PAH into hydrogels increased the storage modulus. The degradation property of the hydrogels can be tuned by modulating the content of Salecan and BAAm. Cytotoxicity results revealed that the hydrogels were non-toxic to COS-7 cells. All these results suggested that Salecan/PAH semi-IPN hydrogels exhibited great potential for applications in soft tissue engineering.

Hydrogel formulations are considered to be promising dosage forms for clinical delivery of antibiotics but suffer from many limitations such as polymers selection and crosslinking methods. Herein, dynamic Schiff base reaction crosslinked hydrogels (denoted as CO) with ‘switchable’ channels were designed based on Chitosan and Salecan. Chitosan maintains the pH sensitive property while providing functional groups for the reversible bond formation. Equilibrium water content, swelling and degradation behavior, dynamic rheology and cytotoxicity assay were investigated to characterize the hydrogels’ properties. By simulating and comparing four classic models, the drug release kinetics and mechanism were studied in detail. HeLa cells were encapsulated to further exploring the feasibility of 3D cell culture. All the results revealed CO hydrogels possessed excellent biological functions to maintain cell growth. Therefore, CO hydrogels can be served as a promising carrier for drug delivery and also expected to apply in many other medical fields.

Salecan is a novel water-soluble β-glucan produced by a salt-tolerant strain Agrobacterium sp. ZX09 which was isolated from a soil sample in our laboratory and the 16S rDNA sequence of this novel strain was deposited in the GenBank database under the accession number GU810841. Salecan has excellent physicochemical properties and can be used in industries such as food and medicine. In this paper, novel semi-interpenetrating polymer network (semi-IPN) hydrogels based on salecan and polyacrylamide (PAAm) were synthesized by radical polymerization/cryopolymerization and semi-IPN techniques. The resulting hydrogels with different salecan/PAAm composition ratios and preparation temperatures were characterized using FTIR, XRD, TGA and SEM measurements. The semi-IPNs exhibited a homogeneous porous architecture with a tunable pore size in a very broad range of 5–150 μm. Furthermore, swelling behaviors of the hydrogels were also studied to investigate the response properties of the hydrogels. The hydrogels obtained at subzero temperature can attain the equilibrium state in water within 260 seconds. Mechanical measurements showed that all semi-IPNs possessed good mechanical properties. In vitro degradation was also studied in PBS solution. Cytotoxicity results suggested that semi-IPN hydrogels were non-toxic to COS-7 cells. A cell culture experiment performed using COS-7 cells revealed their appropriateness for cell adhesion. Together, these results make salecan/PAAm semi-IPNs promising materials for biomedical applications.

Polysaccharide-based wound dressings face challenges in mechanical properties and effective wound repair for infected wound surfaces. This study presents a novel polyvinyl alcohol (PVA)/Salecan (Sal) composite hydrogel dressing with high toughness, biocompatibility, and wound healing capabilities, developed using an interpenetrating polymer network strategy. The primary network was formed through electrostatic interactions between polydopamine (PDA) and biocompatible polysaccharide Salecan, followed by incorporation of AgNO3, which was in situ reduced to silver nanoparticles within the hydrogel. PVA was introduced as a secondary matrix, further reinforcing the hydrogel network through cyclic freeze–thawing. The resulting hydrogel exhibited a tensile strength of 0.31 MPa, an elongation at break of 158.9%, and a toughness of 31.16 J·m−2, demonstrating enhanced mechanical performance compared to both Salecan/PDA and previously reported Salecan/Fe3+hydrogel. Co-culture experiments showed the hydrogel’s strong antibacterial effects, inhibiting 80.1% ofEscherichia coli(E. coli) and 99.5% ofStaphylococcus aureus(S. aureus). Fibroblast culture tests confirmed its excellent cytocompatibility. In vivo studies on infected wounds showed nearly complete healing in theS. aureus+ hydrogel group within 12 days. Quantitative immunohistochemical analysis of CD31 revealed that hydrogel treatment significantly upregulated CD31 expression, indicating enhanced neovascularization. Complementary Western blot analysis further demonstrated that hydrogel-treated groups exhibited a marked downregulation of pro-inflammatory factors alongside CD31 upregulation. In summary, the PVA/Sal-based hydrogel represents a valuable strategy for reducing inflammation and promoting regeneration in the management of infected wounds.
