Enhanced Intestinal Barriers by Puerarin in Combination with Tryptophan

You Wu,Jiaojiao Li,Wenjiao Ding,Zheng Ruan,Li Zhang

The intestinal barrier is essential for maintaining human intestinal health. The growing number of studies has shown that both puerarin and tryptophan and its metabolites have a beneficial effect on the intestinal barrier. This study aims at the combination of puerarin and tryptophan or its metabolites for improving the intestinal barrier. In our study, 40 female Sprague–Dawley rats were randomly divided into five groups (n = 8) for a 4-week experiment and dextran sodium sulfate was used to induce an intestinal barrier injury in rats. Our results showed that puerarin combined with tryptophan or its metabolites (indole-3-propionic acid, IPA) improved the intestinal barrier by enhancing the mucus layer barrier, which was mainly achieved by increasing the number of goblet cells and promoting the secretion of MUC2. Both TRPM5 and VAMP8 promoted MUC2 secretion in goblet cells through exocytosis, but their mechanisms of action are different. In our study, we found that puerarin and tryptophan showed different effects on TRPM5 and VAMP8, respectively. Puerarin enhances the expression of TRPM5, and tryptophan inhibits the expression of TRPM5; however, puerarin and tryptophan have no significant effect on the expression of VAMP8.

Simultaneous Determination of Melatonin, l-Tryptophan, and two l-Tryptophan-Derived Esters in Food by HPLC with Graphene Oxide/SiO2 Nanocomposite as the Adsorbent

Niu Jiahua,Zhang Xiaoting,Qin Peige,Yang Yixin,Tian Shufang,Yang Hui,Lu Minghua

In this research, a dispersive solid-phase extraction (dSPE) with graphene oxide@SiO 2 (GO@SiO 2 ) nanocomposites as the adsorbent followed by high-performance liquid chromatography analysis was developed for simultaneous determination of melatonin, l -tryptophan, and two l -tryptophan-derived esters in food (black sesame seed ( Sesamum indicum L. ) was selected in this case). The GO@SiO 2 nanocomposite was prepared by one-pot aggregation in aqueous phase with sol-gel technique. The structure and morphology of the GO@SiO 2 were characterized by scanning electron microscopy, transmission electron microscopy, Fourier transform-infrared spectroscopy, and X-ray diffraction. The extraction conditions of dSPE including the ratio of material to liquid, adsorption and desorption time, desorption temperature, and desorption solvents were investigated, respectively. The detection limits of the developed method for the analysis of melatonin, l -tryptophan, l -tryptophan methyl ester, and l -tryptophan ethyl ester were achieved below 0.1 μg mL −1 . The established method was successfully applied to the analysis of the target analytes in black sesame seed, which provided a simple, low-cost, and sensitive approach for the determination of trace compounds in complex samples.

Degradation of Tryptophan by UV Irradiation: Influencing Parameters and Mechanisms

Kejia Zhang,Weicheng Fei,Jingxuan Ji,Yulong Yang

The chlorination of dissolved amino acids can generate disinfection by-products (DBPs). To prevent the formation of DBPs, we examined the UV-induced degradation of tryptophan (Trp). In order to further understand the impact of UV disinfection on Trp, the effects of initial concentrations of Trp, pH, temperature, concentrations of NO3−, HCO3−and Cl−on Trp removal were investigated, and a degradation mechanism was also proposed. The results demonstrated that degradation fitted a pseudo first-order reaction kinetic model. The degradation of Trp was mainly caused by direct UV degradation. The apparent rate constant kobsdecreased with the increase in initial Trp concentration and increased with increases in pH and temperature. The thermal degradation activation energy was 19.65 kJ/mol. Anions in water also had a significant influence on the degradation of Trp. HCO3−and NO3−contributed to the kobsof Trp, but Cl−inhibited the degradation rate. By electron paramagnetic resonance (EPR) spectroscopy, ·OH was proven to be formed during the degradation of Trp by UV. Based on the intermediate products of C11H15NO3, C10H15N and C9H13N detected by LC-MS-MS, the degradation pathway of Trp was speculated. Keywords:tryptophan;UV photolysis;pseudo first-order kinetics;·OH;degradation

Targeting tryptophan metabolism reveals Clematichinenoside AR alleviates triptolide-induced hepatotoxicity

Xin-Nan Wang,Wen-Rui Xia,Jian-Qun Liu,Fang-Yuan Sun,Zhu-Jun Zhong,Li-Fang Liu,Gui-Zhong Xin

Liver toxicity induced by Triptolide (TP) has limited its clinical application on rheumatoid arthritis (RA). Saponins have been proved as an efficacious remedy to mitigate hepatotoxicity. However, the mechanism of reducing hepatotoxicity by saponins intervention remains incompletely characterized. Tryptophan (Trp) metabolites activate transcriptional regulators to mediate host detoxification responses. Our study aimed to investigate whether Clematichinenoside AR (C-AR) could attenuate TP-induced liver damage by regulating Trp metabolism. We used targeted metabolomics to quantify Trp metabolites in the serum and liver samples of collagen-induced arthritis rats treated by TP. Multiple comparison analyses helped the evaluation of promising biomarkers. The pronounced changed levels of Trp, indole acetic acid, and indole-3-carboxaldehyde in the serum and indole acetic acid , indole , and tryptamine in the liver are relevant to TP-induced liver injury. Intervention with C-AR could relieve TP-induced hepatotoxicity evidenced by ameliorative serum parameters and hepatic histology. In addition, C-AR regulated the levels of these indoles biomarker candidates to normal. Therapeutic modulation with natural compounds might be a useful clinical strategy to ameliorate toxicity induced by TP. Deciphering Trp metabolism will facilitate a better understanding of the pathogenesis of diseases and drug responding.

Enantiomeric separation of tryptophan via novel chiral polyamide composite membrane

Xinwei Peng,Yongming Wei,Yangfeng Peng,Hongliang Zhao,Tianzhong Tong,Quan He

The separation of chiral drugs continues to pose a significant challenge. However, in recent years, the emergence of membrane-based chiral separation has shown promising effectiveness due to its environmentally friendly, energy-efficient, and cost-effective characteristics. In this study, we prepared chiral composite membrane via interfacial polymerization (IP), utilizing β-cyclodextrin (β-CD) and piperazine (PIP) as mixed monomers in the aqueous phase. The chiral separation process was facilitated by β-CD, serving as a chiral selective agent. The resulting membrane were characterized using SEM, FT-IR, and XPS. Subsequently, the chiral separation performance of the membrane for DL-tryptophan (Trp) was investigated. Lastly, the water flux, dye rejection, and stability of the membrane were also examined. The results showed that the optimized chiral PIP 0.5 β-CD 0.5 membrane achieved an enantiomeric excess percentage (ee%) of 43.0% for D-Trp, with a solute flux of 66.18 nmol·cm −2 ·h −1 , and maintained a good chiral separation stability. Additionally, the membrane demonstrated positive performance in the selective separation of mixed dyes, allowing for steady operation over a long period of time. This study offers fresh insights into membrane-based chiral separations.

Tween 80 Exposure Induces Colitis by Disrupting Intestinal Tryptophan Metabolism

Jia Zhang,Shuang Zhang,Xinxin Xu,Hua Kuang,Chuanlai Xu,Xiaoling Wu

Inflammatory bowel disease (IBD) is a widespread public health problem worldwide, and environmental pollutants are regarded as the cause of IBD. Tween 80 (P80) may trigger IBD; however, its underlying mechanism of action has not been fully clarified, especially with respect to diet-induced susceptibility. This study systematically investigated the effect of P80 on the IBD development in male mice on a normal diet and high-fat diet (HFD) at an acceptable daily intake level. After 6 weeks of oral exposure to P80 (0.1, 1, 6%), the high-dose group of mice exhibited a significant increase in the disease activity index, a significant body weight loss, and a severe inflammatory response, with HFD markedly augmenting these effects. Multiomics results indicated that P80 exposure disrupted intestinal tryptophan metabolism and significantly reduced indole-3-acetic acid (IAA) levels. In vivo intestinal inflammation was transcriptionally regulated by genes related to the AhR/NF-κB/NLRP3 signaling pathway. Supplementing with IAA can improve inflammatory effects. In vitro studies confirmed that P80 exposure inhibited Lactobacillus from producing IAA, leading to an increase in the level of intestinal inflammation. This study revealed the specific toxic mechanism of P80-induced IBD and found that dietary factors have a synergistic toxic effect, providing new insights into the health risk assessment of environmental pollutants.

Spherulitic Crystallization of L-Tryptophan: Characterization, Growth Kinetics, and Mechanism

Jingxiang Yang,Yongli Wang,Hongxun Hao,Chuang Xie,Ying Bao,Qiuxiang Yin,Junbo Gong,Chen Jiang,Baohong Hou,Zhao Wang

L-Tryptophan was successfully crystallized in the form of spherulites by introducing trace amounts of gelatin into the crystallization solution. The physicochemical properties of L-tryptophan products were significantly improved by using this polymer-induced spherulitic growth strategy. This newly developed polymer-induced spherulitic growth strategy provides another choice for controlling the size distribution of particles. Furthermore, the formation and growth of these spherulites under different conditions was monitored in situ by using particle vision measurement and microscopy. It was found that the morphologies of spherulites could be influenced by the additives, the crystallization temperature, and the initial concentration of L-tryptophan. The influence of temperature on the growth rate of spherulites was investigated. It was found that the growth rate is constant at given temperatures. A population-based empirical model was developed and used to explain the spherulitic growth of L-tryptophan spherulites. It was verified that the growth of L-tryptophan spherulites is interface kinetics controlled.

Ginsenoside Rk3 Regulates Tryptophan Metabolism along the Brain–Gut Axis by Targeting Tryptophan Hydroxylase and Remodeling the Intestinal Microenvironment to Alleviate Depressive-Like Behavior in Mice

Jingjing Shao,Linlin Qu,Yao Liu,Jingjing Zhang,Yannan Liu,Jianjun Deng,Xiaoxuan Ma,Daidi Fan

Depression is a neuropsychiatric disease that significantly impacts the physical and mental health of >300 million people worldwide and places a major burden on society. Ginsenosides are the main active ingredient in ginseng and have been proven to have various pharmacological effects on the nervous system. Herein, we investigated the antidepressant effect of ginsenoside Rk3 and its underlying mechanism in a murine model of depression. Rk3 significantly improved depression-like behavior in mice, ameliorated the disturbance of the hypothalamus–pituitary–adrenal axis, and alleviated neuronal damage in the hippocampus and prefrontal cortex of mice. Additionally, Rk3 improved the abnormal metabolism of tryptophan in brain tissue by targeting tryptophan hydroxylase, thereby reducing neuronal apoptosis and synaptic structural damage in the mouse hippocampus and prefrontal cortex. Furthermore, Rk3 reshaped the composition of the gut microbiota of mice and regulated intestinal tryptophan metabolism, which alleviated intestinal barrier damage. Thus, this study provides valuable insights into the role of Rk3 in the tryptophan metabolic cycle along the brain–gut axis, suggesting that Rk3 may have the potential for treating depression.

Tryptophan-sorbitol based carbon quantum dots for theranostics against hepatocellular carcinoma

Wang Yang,Chen Jun,Tian Jiekang,Wang Guanchen,Luo Weikang,Huang Zebing,Huang Yan,Li Ning,Guo Mingming,Fan Xuegong

Background Despite novel advances in screening, targeting and immunotherapies, early diagnosis and satisfactory treatments against hepatocellular carcinoma (HCC) remain formidable challenges. Given the unique advantages, carbon quantum dots (CQDs) become a smart theranostic nanomaterial for cancer diagnosis and therapy. Results In this work, a type of bio-friendly CQDs, trichrome-tryptophan-sorbitol CQDs (TC-WS-CQDs), is synthesized from natural biocompatible tryptophan via the one-pot hydrothermal method. Compared with normal hepatocytes, a much stronger green fluorescence is detected in HCC cells, indicating the ability of TC-WS-CQDs to target HCC cells. Furthermore, green-emitting TC-WS-CQDs generate large amounts of reactive oxygen species (ROS), leading to autophagy of HCC cells. Additionally, the green-emitting TC-WS-CQDs perform significant tumor inhibition by inducing autophagy via p53-AMPK pathway in vitro and in vivo studies with almost no systemic toxicity. Conclusions The results may highlight a promising anticancer nanotheranostic strategy with integration of diagnosis, targeting, and therapy. Graphical Abstract

Microbial tryptophan metabolism activates host lysosomal activity to facilitate lipid breakdown

Kenan Zhang,Zihan Luo,Yan Chen,Yan Li,Lang Wang,Yanan Liu,Ruizhi Yang,Qian Li,Jiahao Zhao,Bin Qi,Zhao Shan

Lysosomes are central to lipid metabolism, yet how gut microbiota-derived metabolites regulate lysosomal function to influence host lipid homeostasis remains unknown. Here, we identify a mechanism in which bacterial tryptophan metabolism activates lysosomal activity to promote lipid breakdown in Caenorhabditis elegans , and show that the bacterial tryptophan metabolite indole recapitulates these effects in mammalian hepatocytes. By developing a lysosomal-responsive lipid reporter in C. elegans to screen for bacterial metabolic states that modulate host lipid storage, we discover that Escherichia coli tryptophan catabolism via tryptophanase TnaA induces lysosomal lipid chaperone LBP-8, driving lipid mobilization. Moreover, tryptophan metabolite indole enhanced lysosomal acidification and degradation capacity, while genetic disruption of lysosomal regulators reversed these effects. Strikingly, bacterial tryptophan metabolism further promoted mitochondrial β-oxidation through lysosomal lipase activity. This pathway was conserved in mammalian hepatocytes, where E. coli -derived tryptophan metabolite indole enhances lysosomal function and reduce lipid accumulation. Our work uncovers microbiota-regulated lysosomal activation as a critical axis in lipid homeostasis, highlighting its potential as a therapeutic target for metabolic disorders linked to lysosomal dysfunction.

Selective Inhibition Mechanism of Mycobacterium tuberculosis Tryptophan-tRNA Synthetase by Chuangxinmycin

Xingli Han,Zhiyong Liu,Biao Zhou,Yuanyuan Shi,H.M. Adnan Hameed,Yamin Gao,Cuiting Fang,Xiongfang Zhao,Linzhuan Wu,Xiaoli Xiong,Wei Yu,Bin Hong,Tianyu Zhang

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), represents a global health challenge, necessitating new treatments with distinct mechanisms of action (MOA) to combat drug resistance. Chuangxinmycin (CM), characterized by its indole-dihydrothiopyran heterocyclic skeleton, exhibits potent antibacterial activity both in vitro and in vivo, with a minimum inhibitory concentration (MIC) of 0.25 μg/mL against Mtb. However, the MOA of CM against Mtb has remained obscure. Through comprehensive genetic, chemical rescue, and protein-drug interaction studies, coupled with biochemical analyses, we reveal that CM selectively binds and inhibits tryptophanyl-tRNA synthetase (TrpRS) encoded by trpS, rather than anthranilate synthase (TrpE). Overexpression of trpS in Mtb results in a 128-fold increase in the MIC of CM, indicating a fundamental cause of resistance, whereas overexpression of trpE leads to modest resistance, suggesting a secondary effect. Conversely, knockdown of trpS or trpE enhances the susceptibility of Mtb to CM. Meanwhile, promoters of trpS in CM-resistant Mtb mutants exhibit increased activity compared to the wild type. Furthermore, drug–protein interaction and biochemical assays have confirmed that while CM effectively inhibits TrpRS, mutants of TrpE show decreased affinity for tryptophan. These results establish that CM exerts its anti-Mtb effects by interfering with the tryptophan-tRNA linkage essential for protein synthesis.
