Light-driven sextuple theranostics: nanomedicine involving second near-infrared AIE-active Ir(III) complex for prominent cancer treatment

Liu Xue,Li Jiangao,Zhang Fei,Yan Dingyuan,Wang Lei,Wang Dong,Tang Ben Zhong

Light-driven cancer theranostics has shown inexhaustible and vigorous vitality by virtue of its high efficacy, prominent controllability and noninvasiveness. Exploration of an all-round theranostic material simultaneously affording both multimodal diagnosis imaging and synergistic phototherapy would be an appealing yet significantly challenging task. Herein, a novel nanomedicine Ir@PPEG-MeEPO was ingeniously constructed by integrating beforehand 1 O 2 -charged amphiphilic polymer and well-tailored Ir(III) complex IrDPTP, which was featured by second near-infrared (NIR-II) aggregation-induced emission (AIE) tendency, efficient reactive oxygen species (ROS) generation, good photothermal conversion efficiency and high-performance hydrogen gas production. To the best of our knowledge, IrDPTP held the longest emission wavelength among all reported AIE Ir (III) complexes. Moreover, Ir@PPEG-MeEPO was capable of controllably releasing ROS via triggered photothermal effect upon NIR irradiation, making it well-adapted to hypoxic environment of tumor. Those distinctive characteristics of Ir@PPEG-MeEPO endowed it with unprecedented performance on sextuple theranostics comprised of NIR-II fluorescence-photoacoustic-photothermal trimodal imaging and photodynamic-photothermal-hydrogen trimodal therapy, witnessed by the precise tumor diagnosis and complete tumor elimination. The study would open up new perspectives for the exploration of superior nanomedicine for practical cancer theranostics.

Reversal of the Heavy-Atom Effect Isostructural AIE Metal-Organic Frameworks for Enhanced Radioluminescence and Nuclear Battery Performance

Xu Zhiyi,Xu Zhiheng,Chen Yong,Xu Tiancheng,Zhang Xuebin,Dong Zongyan,Tang Xiaobin

Incorporation of high-Z elements into conventional organic fluorophores generally induces severe fluorescence quenching owing to the classic heavy-atom effect. Nevertheless, recent advances in aggregation-induced emission (AIE) systems with through-bond/through-space conjugation (TBSC) have uncovered an anomalous heavy-atom-promoted fluorescence enhancement, which remains confined to covalently substituted organic small molecules with nonmetallic heavy atoms. Herein, we extend this unique principle to periodic metal-organic frameworks (MOFs) with coordination-bonded heavy metal nodes. By adopting an isostructural design strategy, two MOFs (Zr-TCPE and Hf-TCPE) are fabricated using a TBSC-type AIE ligand (H 4 TCPE). Remarkably, both MOFs exhibit hot-exciton thermally activated delayed fluorescence (TADF) features. Under X-ray excitation, Hf-TCPE exhibits substantially enhanced radioluminescence relative to Zr-TCPE, achieving an ultra-low detection limit of 2.53 μGy s –1 . Mechanistic investigations confirm that the higher atomic number of Hf enhances X-ray absorption, while the inherent TBSC and hot-exciton TADF properties enable efficient high-energy reverse intersystem crossing from high-lying triplet states to singlet states, effectively suppressing non-radiative T 1 decay. This synergistic mechanism fundamentally reverses the conventional heavy-atom quenching effect into a highly radioluminescence enhancement strategy. As a proof-of-concept, the Hf-TCPE-based radioluminescent nuclear battery delivers a maximum output power of 1.15 μW at a dose rate of 5.6 μGy s –1 , which is 17.8% higher than that of the Zr-TCPE counterpart, verifying its promising potential in radiation energy conversion. This work extends the heavy-atom-enhanced luminescence principle to periodic MOF systems, providing a universal strategy for the rational design of high-performance scintillators and radiation energy conversion materials.

An interface-confined ultrabright AIE nanoparticle-enhanced lateral flow immunoassay platform for full-range and accurate CRP detection

Yuehan Shu,Jie Cheng,Yuanyuan Zou,Jiancheng Zhu,Yi Wu,Jinhong Guo,Diangeng Li,Bin Dai,Yunhe Yuan

C-reactive protein (CRP) is a key biomarker for both infectious diseases and cardiovascular and cerebrovascular disorders. Thus, accurate full-range quantification is critical for early screening and timely intervention. However, limited sensitivity in conventional lateral flow immunoassays (LFIAs) and aggregation-induced quenching in traditional fluorescent probes collectively restrict analytical performance. In this work, a fluorescent LFIA platform based on P(St-AA)@AIENPs is established via an interfacial confinement–assisted loading strategy, enabling effective incorporation of aggregation-induced emission (AIE) luminogens into a poly(styrene-acrylic acid) matrix and delivering a detection limit of 0.66 ng/mL, a broad working range of 0–2000 ng/mL, high specificity, and strong correlation with ELISA (R 2  = 0.995) within a 15 min assay. Overall, this platform offers a rapid, accurate, and portable solution for full-range CRP detection. It is particularly suitable for community-level screening and home-based testing, supporting improved disease management and a reduced public healthcare burden.

CRISPR/Cas12a-enhanced electrochemiluminescence sensing based on solid-state AIE-active ATT-AuNCs for ultrasensitive N-nitrosodimethylamine detection

Zhenjie Zheng,Siqi Cai,Mengqi Ma,Xiaoyi Chen,Zhanglin Weng,Da Huang,Bin Qiu,Zhenyu Lin,Fang Luo

ATT-AuNCs provide intrinsically efficient solid-state AIE emission without additional material engineering. • CRISPR/Cas12a enables efficient target-triggered molecular signal amplification. • The biosensor achieves a 0.37 pg·mL −1 detection limit for NDMA in processed meat samples. N -Nitrosodimethylamine (NDMA) is a potent carcinogenic small-molecule contaminant, and its trace-level occurrence necessitates sensitive and reliable detection methods. Herein, we developed an electrochemiluminescence (ECL) aptasensor integrating a solid-state aggregation-induced emission (AIE)-active gold nanocluster (Au NCs) luminophore with clustered regularly interspaced short palindromic repeats (CRISPR)/Cas12a-mediated molecular signal amplification for ultrasensitive NDMA detection. 6-Aza-2-thiothymine (ATT)-protected Au NCs (ATT-AuNCs) served as the ECL luminophore with triethylamine as the coreactant, while ferrocene (Fc)-labeled single-stranded DNA (Fc-ssDNA) acted as the ECL quencher. Upon NDMA recognition, target-induced strand displacement released a DNA activator, which activated the Cas12a/crRNA complex and triggered trans-cleavage of electrode-bound Fc-ssDNA. Cleavage of the ssDNA backbone disrupted the linkage between the Fc moiety and the electrode-anchored terminus, allowing Fc-containing cleavage products to detach from the electrode interface and thereby reducing Fc-mediated quenching. As a result, the ECL emission of ATT-AuNCs was restored, translating target recognition into an enhanced ECL response. The proposed sensor exhibited a linear range of 5 pg·mL −1 to 500 ng·mL −1 with a detection limit of 0.37 pg·mL −1 . By exploiting the intrinsically efficient solid-state AIE emission of ATT-AuNCs and CRISPR/Cas12a-mediated signal amplification, this strategy enables ultrasensitive NDMA detection. Chitosan nanogels-mediated AIE self-assembly of copper nanoclusters for highly sensitive detection of the anticancer drug methotrexate

Juan Du,Jingyi Duan,Li Yao,Huixiao Tong,Jinwen Chen,Shenghua Liao

In this study, a facile and fast aqueous-phase synthetic method for preparing chitosan-based fluorescent nanogels (CS) by exploiting the aggregation-induced emission (AIE) property of copper nanoclusters (Cu NCs) is proposed. Owing to the spatial confinement provided by the crosslinking network structure in chitosan-based nanogels and the electrostatic interaction between positively charged chitosan nanogels and negatively charged Cu NCs, the fluorescent intensity of the as-prepared CS-Cu NCs was increased by approximately 17-fold compared with that of Cu NCs. The fluorescence quantum yield is increased by more than four times; the as-prepared CS-Cu NCs exhibited a quantum yield of 64.12%. Additionally, CS-Cu NCs exhibited significantly improved stability in aqueous solution, including excellent oxidation resistance, high anti-salt stability, good thermal stability, and enhanced capacity of anti-photobleaching. These properties provide a fundamental guarantee for the application of Cu NCs with AIE property in the biosensor and bioimaging. Upon the addition of the anticancer drug methotrexate (MTX) to CS-Cu NCs, their fluorescent intensity was markedly quenched. Based on the observed fluorescence-quenching phenomenon of CS-Cu NCs induced by MTX, a novel fluorescence quenching nanoprobe was designed for detecting the labeling amount percentage of commercially available methotrexate tablets. The experimental results validated that our proposed nanoprobe exhibits a wider dynamic linear range and excellent accuracy with a low limit of detection (LOD) of 4.16 μM, thereby expanding the potential application of chitosan-based nanogels encapsulating metal nanoclusters presenting AIE property in pharmaceutical quality control.

Loading of AuNCs with AIE effect onto cerium-based MOFs to boost fluorescence for sensitive detection of Hg2+

Li Zhang,Xiaoya Bi,Hui Wang,Libo Li,Tianyan You

Ligand-protected gold nanoclusters (AuNCs) have become promising nanomaterials in fluorescence (FL) methods for mercury ions (Hg 2+ ) monitoring, but low FL efficiency hinders their widespread application. Herein, AuNCs/cerium-based metal-organic frameworks (AuNCs/Ce-MOFs) were prepared by loading 6-aza-2-thiothymine-protected AuNCs (ATT-AuNCs) with aggregation-induced emission (AIE) effect on the surface of Ce-MOFs by electrostatic attraction. This strategy improved the FL intensity of AuNCs through two aspects: (i) the AIE effect of ATT-AuNCs and (ii) the confinement effect of Ce-MOFs, which improved the restriction of intramolecular motion (RIM) of ATT-AuNCs. In addition, Ce-MOFs could adsorb and aggregate Hg 2+ during detection, which might increase the local concentration. Therefore, based on the high FL signal of AuNCs/Ce-MOFs and enriched Hg 2+ , sensitive detection of Hg 2+ could be achieved. More importantly, the strong specific recognition between AuNCs and Hg 2+ could guarantee selectivity. The developed FL sensor exhibited superior detection performances with a wide linear range of 0.2–500 ng mL −1 and a low detection limit of 0.067 ng mL −1 . Furthermore, the FL sensor used for sensitive and selective detection of Hg 2+ in real samples, and the results agreed well with the standard method. In summary, this work proposed an effective and generalized strategy for improving the FL efficiency of AuNCs, which would greatly facilitate their application in pollutant monitoring.

A label-free fluorescent aptasensor based on the AIE effect and CoOOH for ultrasensitive determination of carcinoembryonic antigen

Yulong Ju,Qiukai Tang,Yiwen Yang,Yanbo Zeng,Yunyun Zhai,Hailong Wang,Zuguang Li,Lei Li

Highly sensitive and specific detection of cancer markers (such as carcinoembryonic antigen) is very important for early diagnosis and treatment of cancer. Herein, we developed a label-free fluorescent aptamer biosensor based on the aggregation-induced emission (AIE) effect and hydroxycobalt oxide (CoOOH) platform, and used it to detect carcinoembryonic antigen (CEA) with high sensitivity and specificity. Fluorescent ionic liquid Compound B can combine with a CEA aptamer (CEA-Apt) through electrostatic attraction and hydrophobic interaction to form an ionic liquid/aptamer (CEA-Apt/B) complex and produce the AIE effect, thereby enhancing the fluorescence intensity of B. CEA-Apt/B was adsorbed on the surface of CoOOH when CoOOH was added to the buffer solution, and the fluorescence of B was quenched. After adding CEA to the solution, CEA-Apt/B bound to CEA and separated from the surface of CoOOH because CEA-Apt had stronger affinity for CEA, resulting in fluorescence recovery of B. In the level range of 0.67–10000 pg mL−1, the fluorescence recovery intensity of the sensor had an excellent linear relationship with the level of CEA, and its LOD was 0.2 pg mL−1 (S/N = 3). In addition, the sensor had good selectivity and can be directly used to detect CEA in human serum with high accuracy.

Triphenylamine indanedione as an AIE-based molecular sensor with one-step facile synthesis toward viscosity detection of liquids

Lingfeng Xu,Fangzhi Xiong,Minqing Kang,Yanrong Huang,Kui Wu

Viscosity is one of the most important physical parameters in a liquid, noninvasive, and effective viscosity inspection method toward liquid safety that needs to be developed urgently. In this study, two kinds of novel molecular sensors, namely, DPBID and DPTMID, were strategically constructed by the triphenylamine indanedione derivates; the rotatable conjugate structure was utilized as the recognition site and fluorescence quencher. This couple of molecular sensors was synthesized in a one-step facile manner. DPTMID displayed longer emission wavelength and larger Stokes shift (195 nm in water, 138 nm in glycerol) with a narrower energy band. Moreover, DPTMID exhibited high selectivity, sensitivity, and significant fluorescence signal enhancement toward a higher viscous microenvironment. The molecular sensor displayed good photostability, selectivity, and universality in various commercial liquids and featured with typical aggregation-induced emission (AIE). With the aid of DPTMID, the thickening effects of liquid thickeners can be captured. More importantly, DPTMID was explored to visualize the viscosity fluctuations during the metamorphic stages of liquids, and it was found that the microenvironment viscosity level is closely related to the spoilage degree of liquids. The method with rapid detection, high sensitivity, cheap equipment, and fast results output toward food quality and safety inspection can be achieved through this study.

Haohui Shi,Na Liang,Jiyang Liu,Siyi Li,Xianfeng Gong,Pengfei Yan,Shaoping Sun

In this study, a novel polymer based on aggregation-induced emission (AIE) fluorogen , biotin and disulfide bonds modified chitosan (TPE-bi(SS-CS-Bio)) was designed and synthesized. The polymer could self-assemble into micelles in aqueous media and encapsulate paclitaxel (PTX) into the core with high drug loading. Fluorescence study indicated that the micelles exhibited excellent AIE feature with intense blue fluorescence emitted. In vitro drug release study indicated that the micelles could disassemble rapidly in the presence of high level of glutathione . The modification by biotin could enhance the cellular uptake of the micelles. The drug-loaded micelles possessed remarkable cytotoxicity against MCF-7 cells, and their distribution in the cells could be traced due to the excellent AIE feature. In vivo antitumor efficacy study demonstrated the superior antitumor activity of the PTX-loaded TPE-bi(SS-CS-Bio) micelles. These results indicated that TPE-bi(SS-CS-Bio) has the ability of biological imaging and can be used as a potential carrier for PTX.

A novel AIE active NIR fluorophore based triphenylamine for sensing of Hg2+ and CN− and its multiple application

Xiaoye Wen,Li Yan,Zhefeng Fan

New strategies still need to be proposed that can be used to sense and remove toxic environmental pollutants in a sensing system. In this research, a novel NIR fluorescence sensor 1 was designed and prepared with aggregation induced emission (AIE) property. The fluorescence intensity of the sensor 1 in DMSO/H 2 O mixed solvent was changed along with the proportion of water. The sensor 1 can be successfully used for real-time detection and removal of Hg 2+ in 20% DMSO aqueous solution with high selectivity, quick response and so on. Furthermore it can be efficiently reused and recycled without any loss through Na 2 S. In addition, the sensor 1 displayed high sensitivity and selectivity to cyanide ions in 1% DMSO aqueous solution with the presence of other interference anions. The sensing mechanism for Hg 2+ and cyanide ion was evaluated by 1 H NMR spectra, Mass spectrometry. The sensor 1 exhibited low cytotoxicity for biological applications, which was used as an outstanding fluorescent transducer for detection of cyanide ion in living cells. Based on the visible fluorescence change for the sensor 1 to cyanide ion, the measurement was performed for food materials containing cyanide, such as potato, cassava powder and almond. This research provides perspective potential in solving the problem of other pollution and stimulating new thinking for designing of the novel fluorescence materials.

Two birds one stone: Facile preparation of AIE-active fluorescent polymeric nanoparticles via self-catalyzed photo-mediated polymerization

Jiande Dong,Ruming Jiang,Weimin Wan,Haijun Ma,Hongye Huang,Yulin Feng,Yanfeng Dai,Hui Ouyang,Xiaoyong Zhang,Yen Wei

In this work, we report the first preparation of AIE-active FPNs via metal-free photo-mediated atom transfer radical polymerization (ATRP), employing 2-methacryloyloxyethyl phosphorylcholine (MPC) and benzyl methacrylate (BzMA) as the monomers. Herein, the AIE fluorogen, 10-phenylphenothiazine (PTH)-containing α-bromoester (named as PTH-Br), was simultaneously used as the initiator and photocatalyst. We found that the AIE-active PTH-P(BzMA-MPC)-20(40) FPNs in water can emit intense fluorescence, with high fluorescence quantum yields of 34.3% and 41.2%, respectively, and show excellent water dispersibility and a low critical micelle concentration (CMC). To explore their potential for biomedical applications, biocompatibility evaluation and the cell uptake behavior of these FPNs were further studied. The results demonstrated that these polymeric nanoparticles are biocompatible with L02 cells and are promising for bioimaging applications. Compared with the traditional strategies for preparing AIE-active FPNs, the method described in this work can occur at room temperature, does not require metal catalysts, and is highly efficient. This work lays the foundation for the fabrication of AIE-active functional materials and broadens the potential of AIE-active molecules.

Rapid preparation of branched and degradable AIE-active fluorescent organic nanoparticles via formation of dynamic phenyl borate bond

Zi Long,Meiying Liu,Liucheng Mao,Guangjian Zeng,Qing Wan,Dazhuang Xu,Fengjie Deng,Hongye Huang,Xiaoyong Zhang,Yen Wei

The fluorescent organic nanoparticles (FNPs) with aggregation-induced emission (AIE) feature have received increasing attention for their advanced optical properties. Although many efforts have been devoted to the fabrication and biomedical applications of AIE-active FNPs, the preparation of branched AIE-active FNPs with degradability through formation of dynamic bonds have rarely been reported. In this work, branched AIE-active FNPs were fabricated via dynamic linkage of hydrophobic hyperbranched and degradable Boltorn H40 (H40) with phenylboronic acid terminated AIE dye (PhB(OH) 2 ) and mPEG (mPEG-B(OH) 2 ), which relied on a facile one-pot strategy between phenylboronic acid and diol group of H40. The branched H40-star-mPEG-PhB(OH) 2 FNPs were characterized using nuclear magnetic resonance spectroscopy, Fourier transform infrared spectroscopy, and fluorescence spectroscopy. Benefiting from their highly branched structure and amphiphilic properties, H40-star-mPEG-PhB(OH) 2 could self-assemble into micelles and emit strong orange-red fluorescence. More importantly, cell viability results demonstrated that H40-star-mPEG-PhB(OH) 2 FNPs showed good biocompatibility and promising candidates for bio-imaging. Taken together, we developed a one-pot strategy for preparation of branched AIE-active FNPs through the formation of dynamic phenyl borate. The resultant H40-star-mPEG-PhB(OH) 2 FNPs should be promising biomaterials for different applications for biodegradability of H40 and responsiveness of phenyl borate.
